Chinese Journal of Biologicals 2024;37(07):866-872
Objective To compare and analyze two methods for determination of vaccine particle content in freeze-dried human rabies vaccine(Vero cells)bulk and final bulk, and to provide experimental basis for establishing standard detection method for particle content and purity of rabies vaccine products. Methods The samples were subjected to vaccine particle cracking,BCA quantification, and size exclusion chromatograghy-high performance liquid chromatography(SEC-HPLC)analysis. BCA-SEC relative area method and BCA-SEC vaccine particle standard curve method were used to detect the total protein concentration of the bulk and final bulk, separately, and the content of vaccine particles was quantitatively analy-zed. Finally, the detection results of the two methods were compared and analyzed. Results The content of vaccine particles in freeze-dried human rabies vaccine bulk was determined to be within 256-305 μg/mL by the two methods, with an average value of 267-285 μg/mL,and the relative standard deviations(RSDs)ranged from 5. 8% to 10. 2%,with good consistency between two methods. The content of vaccine particles in freeze-dried human rabies vaccine final bulk was determined to be in the range of 149-169 μg/mL by the two methods, with an average value of 152-164 μg/mL,and the two methods showed the RSDs between 0. 9%-4. 7% with good consistency. Conclusion The measured value by BCA-SEC standard curve method deviates less from the expected value and is closer to the actual situation of samples,so it is recommended to use this method as a reference for enterprises.

5.Advances in pathogenesis, diagnosis and treatment of paroxysmal nocturnal hemoglobinuria
Journal of Leukemia & Lymphoma 2016;25(4):252-256
Paroxysmal nocturnal hemoglobinuria (PNH) is a rare disorder of hematopoietic stem cells due to acquisition of somatic mutations.Somatic mutations in phosphatidylinositol glycan class A (PIGA) account for intravascular hemolysis and other PNH manifestations, but the pathophysiology of clonal expansion of PNH cells cannot be elucidated clearly.PNH is closely related to aplastic anemia and myelodysplastic syndromes.Today, the gold standard for PNH is flow cytometry to detect the absence or severe deficiency of glycosylphosphatidylinositol (GPI)-anchored proteins on white and red blood cells. However,PNH diagnosed by phenotype is a group of heterogeneous disease in pathogenesis.Eculizumab,a first-in-class monoclonal antibody that inhibits terminal complement,is highly effective in stopping intravascular hemolysis and improving quality of life.Further research on the pathogenesis of PNH would be helpful to understand the underlying reasons for PNH phenotype cells in different patients, improve differential diagnosis and more targeted and specific therapy.Research progress in recent years will be reviewed in this article.
6.THE DISTRIBUTION OF INTRAMUSCULAR NERVESIN DIFFERENT SKELETAL MUSCLES OF THE RABBIT
Acta Anatomica Sinica 1957;0(04):-
Objective To investigate the distribution of intramuscular nerves in the different skeletal muscles of the rabbit. Methods Muscle architecture and modified sihler's neural staining methods were used. Results The nerve branches of innervating flat M.pectoralis major arose from the anterior pectoral and posterior pectoral nerve. The former innervated cross fibers mainly,it penetrated the middle of muscle belly and formed a “U-Shaped” nerve ansa. The latter innervated oblique fibers mainly. There were many anastomoses between them. The nerve of pennate M.plantaris derived from tibial nerve. After entering muscle, the nerve trunk gradually divided into many primary branches toward medial and lateral fibers, these branches then subdivided into numerous arboroid second and terminal branches toward all of muscle fibres; The nerve of innervating spindle M.extensor digitorum longus came from N.fibularis communis. Two extramuscular nerve trunk were seen.Superior trunk mainly innervated those muscle fibers of inserting in the second toe,Inferior one mainly distributed to the fibers of rest within this muscle. Conclusion The spatial arrangement of the muscle fibres were related to distribution of the intramuscular nerves;Walking of intramuscular nerve branches had two patterns which run perpendicular and/or parallel to muscle fasciculi.

9.Corneal high order aberrations in adults with myopia:correlations with myopic degree and keratometry
International Eye Science 2016;16(6):1043-1047
?AIM: To investigate the corneal high order aberrations in adults with myopia and to study its correlations with myopic degree and keratometry.?METHODS: A total of 63 patients ( 126 eyes ) from the ophthalmology department of our hospital were included in this study during May 2014 to June 2015. Visual acuity, intraocular pressure ( IOP ) , diopter, slit lamp examination, central corneal thickness ( CCT ) , funduscopic examination, corneal topography and corneal total higher order aberrations ( THOAs ) were recorded. Right eye was selected. According to the spherical equivalence ( SE) , 63 eyes were divided into two groups, the mild to moderate myopia group (M-M group, SE<-6. 00D) and the high myopia group (H group, SE≥-6. 00D). The correlations of corneal THOAs with diopter, keratometry, IOP and CCT were analyzed. Kolmogorov-Smirnov Test was used for normality test. Independent-samples t test was used for comparison between two groups. Pearson correlation analysis was for comparison between two continuous variables.?RESULTS: Z44 was found a statistic difference between the M-M group and H group (t=2. 51,P=0. 02). IOP was negatively correlated with Z5-3(r= -0. 52,P=0. 01), and CCT was positively correlated with Z4-4(r=0. 43,P=0. 03) in H group. There was no correlation between higher order aberrations and SE in M-M group ( all P>0. 05 ) Total coma aberration ( TCA ) was negatively correlated with SE (r=-0. 57,P=0. 002) and spherical degree (Sph, r=-0. 55,P=0. 003) in H group. Z40 was positively correlated with average K (AK) (M-M group:r=0. 40,P=0. 02. H group:r=0. 55, P=0. 03), K steep (Ks) (M-M group:r=0.39, P=0.02. H group:r=0.51, P=0.01) and K flat (Kf) ( M-M group:r=0. 40, P=0. 02. H group:r=0. 57, P=0. 01) in both groups. Z53was negatively correlated with Ks(r=-0.4, P=0. 04) in H group.?CONCLUSION: The THOAs parameters are influenced by diopter and keratometry. TCA is negatively correlated with SE and Sph in patients with high myopia, and spherical aberration is positively correlated with keratometry.
