2.Research on correlation factors of breast cancer stem cell and breast cancer metastasis

Objective To examine the expression of VEGF,MMP-1,and CXCR4 in breast cancer stem cells and its significance.Methods Flow cytometry was employed to separate breast cancer stem cells from MCF-7 cell.Then the expression of VEGF,MMP-1,and CXCR4 was detected by PCR in different subsets of cells.ResultsCompared with the non-stem cells of breast cancer,the expression of VEGF,MMP-1,and CXCR4 in breast cancer stem cells was significantly higher,but the expression of MMP-1 was not different.ConclusionsThe breast cancer stem cells could achieve increased metastasis ability by enhanced expression of VEGF and CXCR4,which are the key factors in metastasis of breast cancer.
3.Effects of electromagnetic field activated-ERK signaling pathway on proliferation and osteogenic differentiation of rat bone marrow mesenchymal stem cells

BACKGROUND: It has been demonstrated that electromagnetic field (EMF) can adjust proliferation and differentiation of bone marrow mesenchymal stem cells, but the specific mechanism is not clear. OBJECTIVE: To investigate the effects of EMF-activated ERK1/2 pathway on proliferation and osteogenic differentiation of rat bone marrow mesenchymal stem cells.METHODS: The 3rd passage of rat bone marrow mesenchymal stem cells were received EMF treatment (15 Hz, 1 mT, sine wave), 20 μmol/L PD98059 + EMF treatment, or only PD98059 treatment. Simultaneously, a normal control group was established. Western blotting was applied to detect the activation of ERK signal pathway after EMF exposure. MTT assay was used to determine the activation of proliferation of cells. And alkaline phosphatase (ALP) activity in cells was detected by an ALP kit. RESULTS AND CONCLUSION: The ERK1/2 phosphorylation, proliferation and ALP activity of rat bone marrow mesenchymal stem cells were remarkably increased after exposure to EMF (P < 0.01). PD98059 could effectively block the increasing of ERK1/2 phosphorylation and cell proliferation (P < 0.01), but elevate ALP activity in a certain level (P < 0.01). EMF stimulation can fast activate ERK1/2 signal pathway and then promote the proliferation of rat bone marrow mesenchymal stem cells, however, ERK1/2 signal pathway activation has a less effect on osteogenic differentiation of bone marrow mesenchymal stem cells.

Antisense oligodeoxynucleotide (ASODN) has been used to inhibit the expression of antiapoptotic proteins(f.g bcl 2, bcl x L , survivin),aimed to induce apoptosis or enhance chemosensitivity.The combination of ASODN with conventional treatment is possibly a reasonable approach in furture cancer therapy. 5.Costimulatory blockade with anti-inducible costimulator antibody in combination with CTLA4Ig on prevention of islet allograft rejection

Objective To study the effects of costimulatory blockade with anti-inducible costim- ulator antibody(ICOS mAb)in combination with CTLA4Ig on prevention of islet allograft rejection. Methods Experimental animals were randomly divided into 4 groups(10 rats in each group).CT- LA4Ig + ICOS mAb group(group A):intraperitoneal injection of CTLA4Ig on day 0,2,4 and ICOS mAb on day 1,3,5 after islet transplantation;ICOSmAb group(group B):intraperitoneal injection of ICOS mAb on day 1,3,5 after islet transplantation;CTLA4Ig group(group C):intraperitoneal injection of CTLA4Ig on day 0,2,4 after islet transplantation;control group(group D):simple islet transplantation.The islet allograft survival and pathological changes in the transplanted islets after transplantation were observed.By using RT-PCR,the expression of IL-2 and IL-10 mRNA in the transplanted islets was detected.The expression of CD4~+ and CD8~+ T cell was detected by flow cy- tometry.Results In group A,the survival time was obviously prolonged as compared with other three groups and the transplanted islets were near normal under a light microscope.As compared with other three groups,the expression of IL-2 mRNA was significantly decreased in group A(P0.05).The expression of CD4~+ and CD8~+ T cell was not obviously up-regulated on the day 21 after transplantation.Conclusion The blockade of costimulatory signals with ICOS mAb in combination with CTLA4Ig has a favorable effects to restrain the rejection of islet transplantation.

Objective To observe the effects of complex prescription with the effects of replenishing Qi, activating blood and resolving phlegm on the levels of LN and PC-Ⅲ in BALF and the changes of lung tissue and figure in rat with pulmonary fibrosis. Method The rat model of pulmonary fibrosis was established by injecting pinyamycin, then the changes of the levels of LN and PC-Ⅲ in every group (TCM group, model group and control group) in 7, 28 days respectively were observed. 9.Closed-form Algorithm for Relative Pose Parameters of Spacecraft Based on 3D Reconstruction

Objective To research a new algorithm to get the relative pose parameters of spacecraft during the final approach phase of spacecraft rendezvous and docking(RVD).Methods The method was based on the shape of five non-coplanar points and the model of the pinhole,and the conception of 3D reconstruction was employed.For the shape of the mark was known,the 3-D shape of the cross-mark was reconstructed based on the single visual system,and the closed-form algorithm for relative pose parameters of spacecraft was given.Results The algorithm had been validated by the mathematical simulation,and the precise results of the relative pose parameters were calculated.Conclusion The simulation results show that the algorithm is rather simple and with relative accuracy.Furthermore it is meets exact the need in real time.
10.The possible mechanism of the acute lung injury induced by S protein of SARS-CoV

Objective: To investigate the role of the recombinant S protein of SARS-CoV in the induction of chemokine IP-10 and other cytokines in airway epithelial cells and immunocytes. Methods: Using insect-baculovirus expression system and Nickel affinity Magnet Beads, S protein of SARS-CoV was produced and then used to stimulate cultured human bronchial epithelial cells (16HBE), human peripheral blood mononuclear cells(PBMC), human peripheral blood monocytes and alveolar macrophages. The levels of IP-10 and the cytokines involved in immunoreaction in response to virus infection were detected in the supernatants of those cells cultured with the S protein by liquid chip system. Results: Under normal condition, no detectable IP-10 was found in 16HBE. A high level of IP-10(79.97? 13.81) pg/ml was detected in the 16HBE 12 hrs after being treated with the S protein, and the induction of IP-10 by S protein displayed at a significant quantity-effect reaction, but not in PBMC, monocytes and alveolar macrophages. In contrast, IFN-? was able to induce the production of IP-10 in either 16HBE or the immunocytes. Conclusion: 1.S protein of SARS-CoV can induce a high level of IP-10 in lung epithelial cells at early stage after the virus infection, which may initiate the process of the immune damage in the lung. 2. S protein of SARS-CoV induces the production of IP-10 by a way of IFN-? independent.
