The increasing prevalence of diabetes worldwide has attracted more and more attention;consequently,diabetes literatures have been on a steady rise over the last decade as sustaining efforts were made to control the disease.This paper,through bibliometric analysis of the 1995-2004 literatures(with a defined amount) from ISI Web of Science and other databases,revealed the amount of literatures,citations,and research focuses of diabetes of different countries,institutions,individuals and journals,reflecting the development in diabetes research over the last 10 years from a bibliometric approach.

BACKGROUND:At present,application revascularization and drug treatment may arrest aggravation of heart function and arrhythmic development,but effectiveness is limited.Infarcted myocardial cells are still impossible to regenerate.How to make necrotic myocardial cell regeneration and diastolic myocardial function is the problem to solve.OBJECTIVE:To analyze the study progress of bone marrow stem cells in treatment of myocardial infarction.RETRIEVAL STRATEGY:A computerized online database of Pubmed was searched to identify articles published from May 2000 to July 2006 with the key words of "the marrow stem-cell,cell transplant,myocardial infarction" in English.Simultaneously,articles published between October 1997 and December 2006 were identified in China Journal Full-text Database with the same key word in Chinese.Sixty-seven articles were retrieved and data were selected primarily.Inclusive criteria:①articles related to the transplantation of bone marrow stem cells for the treatment of myocardial infarction,②in the same field,articles recently published or in authoritative journals.Exclusive criteria:①articles of repetitive study,and ②Meta analysis.LITERATURE EVALUATION:Articles mainly analyzes the transplantation of stem cells for the treatment of myocardial infarction.Of the 30 included articles,10 were reviews and the rest was clinical or basic studies.DATA SYNTHESIS:①Bone marrow stem cells are composed of bone marrow mononuclear cells,hematopoietic stem cells,bone marrow mesenchymal stem cells,endothelial stem/progenitor cells,multipotent adult progenitor cells and so on.②After the transplantation,bone marrow stem cells can differentiate into myocardiocytes,and do effectively electromechanical coupling with receptors.Bone marrow stem cells can promote the angiogenesis in ischemic region,avoid ethical dispute and immunological rejection,and these cells are easy to collect.Bone marrow stem cells are adult stem cells,and its transdifferentiation deserves further study.Many clinical studies showed that autogeneic bone marrow stem cell transplantation for the treatment of acute myocardial infarction and heart failure could be achieved.③Matrix metalloproteinase,stem cell factor and stromal cell chemokine from bone marrow stem cells play important roles in stem cell mobilization,homing and myocardial repair.Tissue injury and high-level multipotential stem cells are two key factors of the differentiation of bone marrow stem cells in myocardial microenvironment.④ Problems in bone marrow stem cell transplantation included cell type,regulatory mechanism of transdifferentiation,optimal time,manner and safety of transplantation,etc.CONCLUSION:Through establishing complete animal model,the possibility of each technique in clinical procedures is explored.Bone marrow stem cells in treatment of acute myocardial infarction may become a new method,which has different from traditional drug treatment,interventional therapy of cardiovascular disease and surgical intervention.

Objective To obtain abundant human betacellulin(BTC) with biological activity. Methods The whole mature protein coding sequence of BTC gene was amplified by polymerase chain reaction(PCR) method applied to human pancreatic ?-cell tumors cDNA.The fragment was cloned into prokaryotic expression vector pET32a(+) plasmid.The recombinant plasmid was transformed into E.coli BL21 and the fusion protein was expressed under isopropyl-beta-D-thiogalactopyranoside(IPTG).The fusion protein was purified by Ni2+ affinity chromatography.SDS-PAGE and Western blot were employed to determine the expression and purification of the expected protein.BTC was added to culture NIH3T3 cells for 5 days,and cell proliferation was detected by MTT. Results Lots of fusion protein were produced,and the purified protein can stimulate the proliferation of NIH3T3 cells. Conclusion The human BTC can be successfully obtained from the pET32a(+) system with the biological activity of stimulating the proliferation of NIH3T3 cells.

Objective To analyze the characteristics of ocular symptoms of Sjgren syndrome and to examine the relation between signs and symptoms.Design Prospective case series.Participants 36 patients with Sjgren syndrome diagnosed by rheumatisms.Methods A self-designed questionnaire was used to investigate 4 symptoms of patients including dryness,grittiness,soreness or irritation,burning or watering.A series of clinic tests were completed as follows:break-up time,fluorescein corneal staining,lissamine green conjunctival staining,Schirmer I test.The positive rate of such symptoms was calculated.The consistency of the symptoms and signs were compared.Main Outcome Measures Break-up time,fluorescein corneal staining,lissamine green conjunctival staining,Schirmer I test.Results Patients with Sjgren syndrome were marked by ocular symptoms.There were efficient statistically significant correlations between grittiness and fluorescein corneal staining,lissamine green conjunctival staining,break-up time(P

Background Identifying and testing of pain is very necessary for the care and decrease of the suffering of experimental animal in medical experiment.Effective method for testing the pain and distress status of experimental animal with eye disease is still absent in China.Objective This pilot study was to establish an evaluating system for assessing the pain and distress status of bullous keratopathy rabbit model.Methods This study was approved by the Ethic Committee of Beijing University First Hospital.Twelve healthy New Zealand rabbits were selected in this experiment.Bullous keratopathy model was established in the left eyes of 9 rabbits by scraping corneal endothelium as the experimental team,and other 3 rabbits were served as the control team.The cornea lesion was examined by manipulate slit lamp,and the central cornea thickness (CCT) was measured by ultrasound biomicroscopy (UBM).Weight+20 Indexes For Pain and Distress Status Referring Guidelines for Pain and Distress in Laboratory Animals made by International Animal Care and Use Committee (IACUC) were assessed and measured as well.Results Corneal edema and opacity were obvious 1 day after surgery.Corneal bullous appeared from the third day after surgery,and cornea erosion was seen at the location of bullous breakage.The corneal lesions remained until 14 days after surgery.CCT value was (1468±100),(2313±588),(2391±271) and (2362±151) μm,respectively in day 1,3,7 and 14 after the establishment of models,which showed significant increase in comparison with the preoperative CCT (390±6)μm (all P=0.000).However,no significant difference was seen in the CCT between day 3,7 and 14 (P＞0.05).Body weight of the rabbits was (3.29±0.20),(3.20-0.17),(2.77±0.25) and (3.10±0.30)kg respectively in day 1,3,7 and 14 after operation,with significant decrease in comparison with the pre-operative weight (3.52-0.18)kg in the experimental team (P=0.008,0.007,0.003,0.004).The scores for pain and distress status of all rabbits in pre-operation of the experimental team and in the control team were zero,and the score was 7 (7,7),11 (10,12),9 (8,10),9 (9,9)in day 1,3,7 and 14 in the experimental team after surgery,with the highest score in day 3,which was bullous and bullous breakage duration.Seven of twenty indexes,including the reduce of diet and drinking,self-imposed isolation/hiding,grinding teeth,aggression,deceased movement,abnormal posture,vocalization occurred in the model animals after surgery.Conclusions Weight+20 Indexes For Pain and Distress Status is an effective,impersonal and quantitative method for observing and evaluating the pain and stress status in bullous keratopathy rabbit.

Background The orbital fibroblasts (OFs) in thyroid associated ophthalmopathy (TAO) play important roles in the proliferative and inflammatory response.Seeking the drug which inhibit OFs growth is of a vital significance for the prevention and treatment of TAO.Research documented that colchicine has an anti-fibrosis effect.But its influence on OFs of TAO patient is few known.Objective This study was to investigate the effect of colchicine on growth and apoptosis of OFs in vitro.Methods The retroobital connective tissue was obtained form 3 TAO patients and cultured using explant method.OFs were passaged and identified by immunochemistry,and 3-8 genetaions of cells were used in the study.Colchicine at the concentrations of 1 × 10-8,1 × 10-7,1 × 10-6,1 × 10-5,1 ×10-4 mol/L was added into the RPMI 1640 with 10％ fetal bovine serum(FBS) to incubated the cells for 24,48 and 72 hours respectively,and only RPMI 1640 was used to culture the cells as the control group.Cell counting kit-8 (CCK-8)was used to detect the absorbance value (A450) of OFs for the evaluatuion of OFs and the inhibitory rate of colchicine to OFs.The colchicine of 1 ×10-6,1 ×10-5,1 × 10-4 mol/L was added into the culture medium for 48 hours,and then the apoptotic rate of the cells and the cell percentage in various cellular cycle was assayed by flow cytometry(FCM).The expression of transforming growth factor-β (TGF-β)in the cells was detected by immunochemistry to assess the influence of colchicine on the serection of the cells.Results Cultured cells showed the spindle-like in shape and the cell number was significantly increased with the incubation time.After incubated with 1 × 10-4,1 × 10 5,1 × 10-6,1 ×10-7,1 × 10-8 mol/L colchicines,the A450 values were gradually reduced with the increase of the concentrations of colchicine(F ion =62.004,P＜0.05),and significant differences were found between different contrations of colchicine groups(all P＜0.05).Aslo,gradually declined A450 values of the cells were seen with the lapse of culture time among the groups(Ftime =459.582,P＜0.05).The inbitory rate of colchicine to the cells was elevated with the increase of concentrations.The apoptotic rates of the cells were (1.73 ± 0.15) ％,(21.04 ± 4.56) ％,(31.84 ±6.21)％and(35.32±5.56)％ in the control group and 1 × 10-6,1 × 10-5,1 × 10 4 mol/L colchicine groups respectively,with statistically significant difference among the 4 groups (F =83.905,P＜0.05).With the increase of concentrations of colchicines,the cell percentage in G2 +M phase lessened gradually,showing significant difference among the control group and the 1 × 10-6,1 × 10-5,1 × 10-4 mol/L colchicine groups (F =20.443,P＜0.05).The expression of the TGF-β in the cells was (97.60± 2.09) ％ in the control group,and that in the 1 × 10-4 mol/L colchicine group was (44.43 ± 3.96) ％,presenting a significant difference between them (t =65.330,P ＜ 0.05).Conclusions Colchicine can induce apoptosis of OFs and inhibit the prolilferation of OFs in a time-and dose-dependent manner probably by decreasing the TGF-β secretion

Objective To construct eukaryotic expression vector of human pancreatic duodenal homeobox 1(PDX-1) gene,and to detect its expression in NIH3T3 cell lines. Methods The whole coding sequence of PDX-1 gene was amplified by polymerase chain reaction(PCR) from human pancreatic-cell tumors cDNA.The fragment was inserted into eukaryotic expression vector pcDNA3.1 plasmid.The recombinant plasmid was verified by double digestion and DNA sequencing.The expression of PDX-1 gene in NIH3T3 cells was assayed by Western blot. Results The length of specific fragment amplified by PCR was 852 bp,and the recombinant plasmid pcDNA3.1-PDX-1 showed two bands of 5.5 kb and 852 bp by digestion using respective restriction enzymes BamHⅠand EcoRⅠ.The sequence of PDX-1 gene was approved or confirmed by blasting to GenBank.It was suggested that PDX-1 gene had been cloned into pcDNA3.1 vector correctly.Western blot showed that PDX-1 gene was expressed,which was detected 24 h after pcDNA3.1-PDX-1 plasmid was transfected into NIH3T3 cells. Conclusion The recombinant eukaryotic expression vector pcDNA3.1-PDX-1 was successfully constructed and expressed in NIH3T3 cell lines.

Objective To explore the inhibitory effect of the ethanol extract from peanut root on the non-castration adult rat prostate hyperplasia induced by testosterone propionate and its mechanism .Methods 60 SD rats were randomly divided into the control ,model and high ,middle and low dose of peanut root ethanol extract treatment groups ,among them ,the control group was normally fed without medication ;the model group was subcutaneously injected by testosterone propionate (TP ,5 mL/kg/d) and simultaneously gavaged subcutaneous injection with peanut root ethanol extract (10 mL/kg/d ,5 mLg/kg/d or 1 mL/kg/d) for suc-cessive 14 d .The rats in various groups were killed on 15 d and their prostate ,spermatophore and testicle tissues were separated and weighed .The ratio of gland/body-weight(mg/g) was calculated .The expressions of Bcl-2 and Bax proteins in the prostate tissues were detected by immunohistochemistry .Results There was statistical difference in the indexes of prostate between the treatment groups(high and middle dose) and the model group(P<0 .01) ,while there was no statistical difference between the low dose group and the model group(P>0 .05) .This result showed that there was a dose-effect relationship between the inhibition effect on rat prostate hyperplasia and the ethanol extracts from peanut root .There was no statistical differences between the model group and the treatment groups in the indexes of testicle ,spermatophore or the change of weight (P>0 .05) .There was statistical difference be-tween the control group or model group and the treatment group in the expression of Bcl-2 or Bax protein(P<0 .05) ,while no sta-tistical difference between the treatment group and the control group (P>0 .05) .Conclusion Ethanol extract from peanut root has good inhibiting effect on prostate hyperplasia and its mechanism could reach the curative effect by regulating the balance relation be-tween apoptosis gene Bcl-2 and Bax protein proportion and promoting apopitosis of benign prostatic hyperplasia .
