1.Is glaucoma a central never system disease?

Studies on glaucoma have been focused only on optic nerve damage and aqueous humor dynamic for many years. In recent years, with the advances in understanding of glaucoma and visual sciences, and the development of a neurological cross-discipline, new interdisciplinary problems have been raised. Notably, the question of whether glaucoma is only an optic nerve disease. Is it a disease that begins from the eye, and then involves the whole visual pathway? Or is it a particular central nervous system disease, which manifests itself in the eye? The answers to these problems are inconclusive so far, but there are reasons to believe that glaucoma is a disease with multi-level, multi-factor damage to the entire visual pathway involved. Its mechanisms are complex and involve cross-synaptic damage, visual pathway blood disorders, blood-brain barrier disorders, and so on. As we come to understand that glaucoma is an ocular and central never system neurodegenerative disease, we may better understand the pathogenesis of glaucoma and therefore establish comprehensive treatment strategies for this disease.
2.Study on the differentiation and apoptosis of HL-60 cell line induced by Puerarin

Objective To investigate the differentiation of HL-60 cells induced by different doses of Purerarin(PR) comparing with all-trans retinoic acid(ATRA)and to see if PR can induce apoptosis of HL-60 cells. Methods Cell differentiation was analyzed by NBT reduction, the ratio of NBT/MTT and CD11b ,apoptosis by morphology, DNA electrophoresis, and flow cytometry(FCM). Results 80 μg.ml-1, 160 μg.ml-1, 320 μg.ml-1 PR could induce differentiation of HL-60 cells, no significant difference was observed between the cells treated with 1 μmol.L-1 ATRA and 320 μg.ml-1 PR. Treated with 320 μg.ml-1, 640 μg.ml-1PR, HL-60 cells exhibited a morphological characteristic of apoptosis and typical DNA ladder on gel electrophoresis. FCM analysis showed that PR could interfere with cell cycle in HL-60 cells, with a increased ratio of sub-G1 in HL-60 cells. Conclusion PR exerts effect on differentiation and induces apoptosis in HL-60 cells.
3.Value of 18F-FDG PET/CT in the post-treatment evaluation of cervical cancer

Objective To evaluate the value of whole body 18F-FDG PET/CT in the detection of recurrence/metastasis of cervical cancer.Methods A retrospective study was performed on 95 patients with cervical cancer who underwent 18F-FDG PET/CT after treatment.The lesion characteristics on 18F-FDG PET/CT were assessed visually and semi-quantitatively.A final diagnosis was confirmed by histopathology,diagnostic treatment and clinical follow-up imaging.The data were analyzed by Kappa test.Results 18 F-FDG PET/CT was positive in 54 patients,including 24 with local recurrence and 30 with distant metastases.The sensitivity,specificity and accuracy of 18F-FDG PET/CT for the detection of recurrence/metastasis of cervical cancer were 98.1% (52/53),95.2% (40/42) and 96.8% (92/95),respectively.The positive predictive and negative predictive value were 96.3% (52/54) and 97.6% (40/41),respectively.18F-FDG PET/CT showed concordant results with pathological/clinical follow-up findings (Kappa =0.936,P<0.05).Conclusion 18F-FDG PET/CT is a sensitive and specific modality for the detection of recurrence/metastasis of cervical cancer and might be useful for further treatment plan.
4.Study on the Sustained Release of BSA from Chitosan-OREC/BSA Films Coated Mats in Vitro

Objective To investigate the sustained release of BSA from chitosan-OREC/BSA films coated mats in vitro.Methods The negatively charged cellulose acetate (CA) fibrous mats were modified with multilayers of the positively charged chitosan or chitosan-OREC intercalated composites and the negatively charged bovine serum albumin (BSA) via electrostatic layer-by-layer (LBL) self-assembly technique.The adsorption and rinsing steps were repeated until the desired number of deposition bilayers was obtained.The in vitro BSA encapsulation and release experiments demonstrated that OREC could affect the degree of protein loading capacity and release ficiency of the LBL films coating.Results In the pH-gradient release assay,only a small amount of BSA was released from the mats in 1 h.As the time increased,the release rate of BSA of all the samples gradually went up to the maximum data within 8 h.For the samples with identical number of bilayers and record time,obvious increasing of the release amount could be seen in pH 7.4,in comparison with pH 1.2.Besides,doubling bilayers film-coated mats generally.Meanwhile,it was slightly distinguishable between 5 and 5.5 as well as 10 and 10.5 bilayers (t=0.651~ 1.324,P>0.05).Interestingly,it could be seen that protein release of the chitosan-OREC/BSA films coated mats remarkably increased compared with that of chitosan/BSA films coated mats(t=2.264~ 2.305,P<0.05).Conclusion The release of protein in the initial time could be controlled by adjusting the number of deposition bilayers,the outmost layer and the composition of coating bilayers.
5.The changes of phosphodiesterase-4 activity in rat acute lung injury

Aim To investigate the changes of phosphodiesterase4(PDE4,type 4 cAMP-specific PDE) activity,TNF-? and neutrophil recruitment in experimental rat lung injury(ALI).Methods ALI in the rat was induced by lipopolysacdharide(LPS).PDE4 activity was measured with HPLC,and the level of TNF-? was detected with ELISA,neutrophil infiltration in bronchoalveolar lavage fluid(BALF) and lung tissues was detected by cell count and morphological analysis.Result Lung tissue PDE4 activity significantly increased as early as 1 h,peaked 6 h,and then markedly lowered at 24 h after intratracheal administration of LPS,while there was a same time-course change of total white cell and neutrophil in the BALF(r=0.83,P
6.Advances in Study on Relationship between Heat Shock Protein 70 and Gastric Cancer

Gastric cancer is one of the most commonly seen malignant tumors,and is the fourth leading cause of morbidity and second leading cause of mortality among malignancies worldwide. The genesis of gastric cancer is the result of interaction between genetic and environmental factors,and is a multi-factor and multi-step carcinogenesis. As one of the most important members in the heat shock protein(HSP)family,HSP70 plays a molecular chaperone role,and is involved in body specific immunity and innate immunity. Studies have demonstrated that over expression of HSP70 often correlates with the genesis and development of gastric cancer. This article reviewed the advances in study on relationship between HSP70 and gastric cancer.
7.Automated synthesis and quality analysis of 18F-FMISO based on CFN-MPS-100 module

Objective To synthesize 18F-FMISO and analyze the quality of the product.Methods 1-(2'-nitro-l'-imidazolyl)-2-O-tetrahydropyranyl-O-trluendulfonylpropanediol (NITIT) was taken as the precursor and simple one pot method was used.CFN-MPS-100 fluorine multifunction radiopharmaceutical chemical synthesis module was adopted to complete the radioactive fluorination reaction in a closed flat flask,and the crude product was purified by semi-preparative HPLC,the solvent was removed by rotary evaporation.Then 15 ml saline was added into the product to get 18F-FMISO injection.Radio-HPLC and radio-TLC were applied for quality control.Results 18F-FMISO was obtained in 60 min with the radiochemical yield of (32±5.0)% (no decay corrected,n=25).The radiochemical purity was above 99.0% and still above 98.5% after 6 h.The radioactive concentration was above 1.11 × 1012 Bq/L.The product was colorless solution,with pH value of 7.0.The radioactive nuclear purity was more than 99%.The K222 was less than 25 μg/ml.Conclusion 18 F-FMISO could be synthesized with automatic synthesis method based on the CFN-MPS-100 fluorine multifunction module.The labeling rate,stability and chemical purity are high.
8.Screening of efficient siRNA target sites directed against gatekeeper genes for DNA repair.

To investigate the RNA interference (RNAi) effect induced by vector-derived small interfering RNA (siRNA) targeting the three gatekeeper genes (Rad52, Ku70, Ku80) and screen the more effective target sites from candidates for further research, by using siRNA design tools online, we selected 2 candidate sequences directed to every gatekeeper gene. According to the sequences, six vector-derived siRNAs (denoted psiRNA1-6) and one mocking psiRNA7 were constructed. Among them, psiRNA1 and psiRNA2 targeted Rad52, psiRNA3 and psiRNA4 to Ku70, psiRNA5 and psiRNA6 to Ku80. The mocking psiRNA7 was used as control. After sequence identification, the seven plasmids were transfected into HepG2 cell line. siRNA-induced silencing of gatekeeper genes was determined by using RT-PCR at RNA level and Western Blot at protein level. The results showed that the six plasmids specifically targeting the coding region of gatekeeper genes were successfully designed and constructed. To some extent, the six plasmids could reduce the expression of target gene. Comparatively, the plasmid-derived siRNA psiRNA1, psiRNA4 and psiRNA5 were more effective than their counterparts. The results suggest that the gene silencing efficiency of siRNA is different, depending on their targeted region, and siRNA may provide us with practical tools for further study on the three gatekeeper genes, i.e. Rad52, Ku70, Ku80.
9.CT manifestations of pneumocystis jirovecii pneumonia and cytomegalovirus pneumonia after allogeneic hematopoietic stem cell transplantation

Objective To compare CT manifestations of pneumocystis jirovecii pneumonia(PCP)and cytomegalovirus pneumonia(CMVP)after allogeneic hematopoietic stem cell transplantation(allo-HSCT).Methods Totally 21 PCP patients(PCP group)and 26 CMVP patients(CMVP group)after allo-HSCT were retrospectively enrolled,and CT findings were compared between groups.Results Significant differences of distribution patterns of ground glass opacity(GGO)and nodule lesions,lung cysts,the time of condition reached most severe,the proportion of cases with lesions absorption more than 50% within 30 days,of cases with lesions completely absorbed and the first total severity score(TSS)of GGO were found between groups(all P<0.05).Conclusion CT manifestations of PCP and CMVP after allo-HSCT were somehow similar,but GGO distributed widerly,with more lung cysts and better prognosis in the former,while GGO distrubuted relatively limitted with diffuse nodules and poor prognosis in the latter.
10.Preliminary exploration of 18 F-FMISO PET/CT SUVmax with relevant factors for nasopharyngeal carcinoma

Objective To explore the relationship between the maximum standardized uptake value ( SUVmax ) of 18 F-fluoromisonidazole ( FMISO) PET/CT and the pathological classification, differentiation, T stage and primary tumor volume of nasopharyngeal carcinoma ( NPC) . Methods A retrospective analysis was performed on 41 patients with NPC (31 males, age 18-74 years;10 females, age 35-67 years) who underwent head and neck 18 F-FMISO PET/CT from 2012 to 2015. The relationship between the clinicopath-ological parameters (pathological classification, differentiation, T stage, tumor volume) of primary lesion and SUVmax were analyzed. Mann-Whitney u test, approximate t test and Spearman correlation were used for data analysis. Results There was no significant difference in SUVmax between non-keratinizing carcinoma and squamous cell carcinoma ( u=183.5, P>0.05) , nor between the differentiated carcinoma and undiffer-entiated carcinoma( t'=-1.23, P>0.05) . SUVmax of T1-T2 primary tumor was significantly lower than that of T3-T4 tumor (1.52±0.43 vs 2.05±0.85; t'=-2.60, P<0.05), and SUVmax was correlated with primary tumor volume ( rs=0.488, P<0.05) . Conclusions The hypoxic degree is related with T stage and primary tumor volume in NPC. The combination analysis of T stage and tumor size will contribute to the assessment of oxygen level and prognosis of primary NPC.
