1.Application of double pipe for negative pressure drainage in reducing intra-abdominal infections after gastrointestinal operation

Objective To discuss application of double pipe for negative pressure drainage in reducing intra-abdominal infections after gastrointestinal operation.Methods 85 patients with intra-ahdominal drainage after gastrointestinal operation were randomly divided into the control group (43 patients) and the observation group(42 patients).The control group used traditional latex drainage,the observation group took the double cannula negative pressure continued drainage.The intra-abdominal infection and bacterial number distribution,patients drainage time,drainage volume and drainage tube occlusion were observed.Results 25 bacterial cultures were positive in two groups of patients.Compared between the two groups,the bacterial number detection rate were significant.31 pathogenic bacteria were detected in 25 specimens.Compared between these two groups,staphylococcus aureus,clearing time of drainage,drawing tube time after drainage liquid being clear,the total catheterization time,average daily drainage volume were all significantly different.Conchrsious Double pipe negative pressure drainage for postoperative abdominal drainage can effectively reduce abdominal infection,shorten drainage time,and its effect is very good.
2.Cervical cancer screening in physical examination

Objective To investigate cervical screening procedure in routine physical examinations.MethodsA retrospective study was carried out to assess the procedures and outcomes of cervical cancer screening among women receiving physical examinations in our hospital from October 2008 to September 2009.ResultsA total of 11 542 women underwent gynecological examinations .Of them,13.55% only received TCT,6.60% finished TCT+HPV screening.The positive detection rate of TCT+HPV screening was higher than TCT alone(x2=15.87,P<0.05).Conclusion Cervical cancer screening rate may be low.
3.Application of ritodrine hydrochloride combined with magnesium sulfate in gestational diabetes mellitus pregnant women with threatened premature labor

Objective To explore the effect of ritodrine hydrochloride combined with magnesium sulfate in gestational diabetes mellitus pregnant women with threatened premature labor.Methods 74 gestational diabetes mellitus pregnant women with threatened premature labor were selected as the research subjects.According to the digital table,they were randomly divided into observation group and control group,37 cases in each group.The control group was treated with magnesium sulfate,the observation group was treated with ritodrine hydrochloride combined with magnesium sulfate.During treatment and after drug withdrawal 3 days,the fasting blood glucose levels and postprandial 2h blood glucose levels during pregnancy,prolonged time,complications and neonatal conditions in the two groups were recorded.Results The total effective rate of the observation group was 97.30%,which was significantly higher than 83.78% of the control group,the difference was statistically significant between the two groups(u =2.336,P =0.019).The fasting blood glucose and 2h postprandial blood glucose levels were higher than before treatment in the two groups,but there were no statistically significant differences between the two groups (t =1.904,P =0.060;t =1.698,P =0.093).After stopping the drug,the fasting blood glucose and 2h postprandial blood glucose of the two groups were reduced to the pre-treatment levels,but there were no statistically significant differences between the two groups (t =0.953,P =0.343;t =0.749,P =0.456).The average gestation duration prolonged time of the observation group was (22.59 ± 2.84)d,which was significantly longer than that of the control group(t =11.737,P =0.000).The neonatal birth weight and Apgar score of the observation group were significantly higher than those of the control group (t =12.746,P =0.000;t =10.159,P =0.000).The neonatal survival rate of the observation group was 97.30%,which of the control group was 93.78%,the difference was statistically significant between the two groups (x2 =3.944,P =0.047).Conclusion Ritodrine hydrochloride combined with magnesium sulfate in the treatment of gestational diabetes mellitus pregnant women with threatened premature labor has good results,and can prolong pregnancy,and has less complications.
4.Inhibition effect of small hairpin RNA on expression of Topo Ⅱ gene in K562/AS2 cell line resistant to arsenic trioxide

Objective To investigate the reversal effect of Topo Ⅱα-shRNA and Topo Ⅱβ-shRNA on Topo Ⅱ gene in K562/AS2 cells. Methods Three pieces of Topo Ⅱα-shRNA and three pieces of Topo Ⅱβ-shRNA were designed,synthesized and transfected into K562/AS2 cells by liposome. Expression level of Topo Ⅱα and Topo Ⅱβ mRNA were determined by real time fluorescence quantitative PCR. The expressions of Topo Ⅱα and Topo Ⅱβ protein were assayed with flow cytometer. Results After treated with Topo Ⅱα-shRNA or Topo Ⅱβ-shRN A for 24 hours,the expression level of Topo Ⅱα mRNA and Topo Ⅱβ mRNA protein in K562/AS2 cells decreased at most (78.22±0.01) %,(31.17±1.27) % (P <0.05),and (57.36±0.01)%,(23.98±1.22) % (P <0.05) respectively. Conclusion The expression of Topo Ⅱ gene can be down-regulated after infected with Topo Ⅱ-shRNA in K562/ AS2 cell line.
5.Analysis of mutations of 14 genes among 87 patients with myelodysplastic syndrome.

OBJECTIVE:
To explore the correlation of genetic mutations and clinical features of myelodysplastic syndromes (MDS) with scores of Revised International Prognostic Scoring System (IPSS-R).
METHODS:
Eighty-seven patients with de novo MDS were enrolled. Mutations of MDS-related genes and clinical features were used to determine the incidence and subtype of mutations. Clinical features and IPSS-R scores of the patients with high frequency mutations involving TET2, TP53, ASXL1, RUNX1 and SF3B1 genes were compared.
RESULTS:
Fifty-four patients (62.1%) harbored at least one point mutation. The incidences of various mutations were significantly different, with the incidence of MDS-EB-2 being 100% and MDS-SLD being only 38.9%. Compared with the wild types, patients harboring mutations had higher lactate dehydrogenase, higher β2 microglobulin, higher percentage of bone marrow blast cells and lower hemoglobin levels (P=0.027, <0.01, <0.01, 0.046, respectively). The IPSS-R scores of MDS patients with mutations were significantly higher than the wild types (P<0.01). The IPSS-R scores of the TP53 mutation groups were 7.82±1.83, which was significantly higher than the control group (3.77±1.66, P<0.01). No difference was found between the IPSS-R between patients carrying TET2, ASXL1, RUNX1, and SF3B1 mutations or the wild types (P>0.05).
CONCLUSION
Genetic mutations are commonly found in MDS. MDS patients with mutations have unique clinical laboratory characteristics. Although the prognostic value of most genes is controversial, TP53 is an definite indicator of poor prognosis.

6.Spatiotemporal Profile and Morphological Changes of NG2 Glia in the CA1 Region of the Rat Hippocampus after Transient Forebrain Ischemia

Neuron-glial antigen-2 (NG2) glia undergo proliferation and morphological changes following brain insults. Here, we show that NG2 glia is activated in a characteristic time- and layer-specific manner in the ischemia-vulnerable CA1 region of the rat hippocampus. Resting NG2 glia of the pyramidal cell layer (somatic region) shared morphological features with those of the neighboring dendritic stratum radiatum. During the postischemic period, reactive NG2 glia of the pyramidal cell layer exhibited shortened, scarcely branched processes, while those of the stratum radiatum had multiple branching processes with their arborization being almost indiscernible 7~14 days after reperfusion. Immunoelectron microscopy demonstrated that NG2 immunoreactivity was specifically associated with the plasma membrane and the adjacent extracellular matrix of NG2 glia in the stratum radiatum at 14 days. NG2 glia also exhibited differences in their numbers and proliferation profiles in the two examined hippocampal strata after ischemia. In addition, induced NG2 expression in activated microglia/macrophages exhibited a characteristic strata-dependent pattern in the ischemic CA1 hippocampus. NG2 induction was prominent in macrophage-like phenotypes which were predominantly localized in the pyramidal cell layer, compared with activated stellate microglial cells in the stratum radiatum. Thus, our data demonstrate that activation of NG2 glia and the induction of NG2 expression in activated microglia/macrophages occur in a distinct time- and layer-specific manner in the ischemic CA1 hippocampus. These characteristic profiles of reactive NG2 glia could be secondary to the degeneration processes occurring in the cell bodies or dendritic domains of hippocampal CA1 pyramidal neurons after ischemic insults.

Zinc levels are high in pancreatic β-cells, and zinc is involved in the synthesis, processing and secretion of insulin in these cells. However, precisely how cellular zinc homeostasis is regulated in pancreatic β-cells is poorly understood. By screening the expression of 14 Slc39a metal importer family member genes, we found that the zinc transporter Slc39a5 is significantly down-regulated in pancreatic β-cells in diabetic db/db mice, obese ob/ob mice and high-fat diet-fed mice. Moreover, β-cell-specific Slc39a5 knockout mice have impaired insulin secretion. In addition, Slc39a5-deficient pancreatic islets have reduced glucose tolerance accompanied by reduced expression of Pgc-1α and its downstream target gene Glut2. The down-regulation of Glut2 in Slc39a5-deficient islets was rescued using agonists of Sirt1, Pgc-1α and Ppar-γ. At the mechanistic level, we found that Slc39a5-mediated zinc influx induces Glut2 expression via Sirt1-mediated Pgc-1α activation. These findings suggest that Slc39a5 may serve as a possible therapeutic target for diabetes-related conditions.
