1.Evaluation of nitrative stress on retinal capillary by detection of trans-arachidonic acid with GC/MS in diabetic rats

Background Trans-arachidonic acids (TAAs) are specific lipid markers of nitrative stress and the major products of NO2·-mediated ismoerization of aracchidonic acid(AA),and they represent a possible new mechanism of NO2·-induced toxicity in ischemic retinopathy.Our previous study demonstrated that high glucose induce nitrative stress response. Objective This study aimed to evaluate the effect of nitrative stress on retinal capillary in diabetic rats by detecting the TAAs with gas chromatography and mass spectrometry(GC/MS).Methods One hundred 2-week-old clean male SD rats were randomly divided into control group and diabetic group.Diabetic rat models were induced by intraperitoneal injection of streptozoticin of 60 mg/kg,and the successful models were identified as the blood glucose level>16.7 mmol/L.The synthesis method was modified in this study andaraehidonic acid w8$utilized as starting material.14E-AA was obtained as a standard sample via expoxidation and deoxidation in a safe and practical pattern.Serum TAAs and AA in diabetic rats were detected by GC/MS in 2,4,8,12 and 16 weeks after injection of streptozoticin.The ratio of peak area of TAAs to AA with selected ion of 79 was estimated and compared with normal rats. Results Fifty rats had been in Diabetic models were established in 50 rats with the successful rate 100%,and the high blood glucose level in model rats retained throughout the experimental duration.GC/MS results showed that the 14E-AA was obtained with the purity of 97.3%,which provided a standard sample to nitrative stress-related research as a premise. No significant differences were found in serum TAAs/AA values at 2 weeks and 4 weeks between model group and control group (t =-0. 376, t =-0. 642,P>0. 05). However,serum TAAs/AA values in at 8,12 and 16 weeks after streptozoticin injection were considerably elevated in comparison with those of control group (0. 0832 vs 0. 1042,0. 0910 vs 0. 1568, 0. 1100 vs 0. 1895;t=-36.409,t=-166.714,t=-148.212,P<0. 05). Conclusion Elevation of serum TAAs/AA is associated with diabetic duration,implying that damage of nitrative stress to retinal capillary is a parallel procedure to diabetic course.Establishment of GC/MS detection system offers a new evaluating indicator in the research on microvascular ischemic disease and could be implemented in clinical testing.

3.Expression levels and clinical significance of FOXQ1 and E-cadherin in esophageal squamous cell carcinoma

Objective To investigate the expression levels and clinical significance of (forkhead box Q1) FOXQ1 and E-cadherin in esophageal squamous cell carcinoma (ESCC). Methods Expression levels of FOXQ1 and E-cadherin were in ESCC tissues (ESCC group, n=42) and adjacent normal esophageal tissues (control group, n=42) were detected using im?munohistochemistry. Correlations of FOXQ1 and E-cadherin expressions with clinical pathological parameters and progno?sis were analyzed between two groups. Results The expression level of FOXQ1 was significantly higher in ESCC group than that in control group(64.29% vs 28.57%,χ2=5.384,P<0.05). The expression level of E-cadherin was significantly lower in ESCC group than that incontrol group(52.38%vs 90.48%,χ2=7.691,P<0.05). There were significant differences in FOXQ1 expressions between different TNM stages and whether lymph node metastasis is involved within ESCC group. There were significant differences in expression of E-cadherin between different tumor differentiation, depth of invasion, TNM stage and whether lymph node metastasis is involved within ESCC group. The expression of FOXQ1 was negatively cor?related with E-cadherin in ESCC (r=-0.412, P<0.05). The 5-year survival rates were significantly lower with high expres?sion of FOXQ1 or with low expression of FOXQ1(18.52%vs 66.67%,χ2=9.737,P<0.05). The 5-year survival rates were significantly higher with high expression of E-cadherinor low expression of E-cadherin(59.09%vs 10.00%,χ2=10.996,P<0.05). A multivariate Cox's proportional hazard regression analysis indicated that high FOXQ1 expression, low E-cadherin expression and lymph node metastasis were independent prognostic factors for ESCC. Conclusion The expression of FOXQ1 and E-cadherin showed a good correlation with ESCC. And examining expressions of both FOXQ1 and E-cadherin in ESCC may have practical values in estimating the prognosis of ESCC and directing future treatment .
4.Qinzhu Liangxue Decoction in treatment of blood-heat type psoriasis vulgaris: a randomized controlled trial

OBJECTIVE: To explore the clinical efficacy of Qinzhu Liangxue Decoction (QZLXD), a compound traditional Chinese herbal medicine, in patients with blood-heat type psoriasis vulgaris. METHODS: Fifty-eight patients diagnosed with blood-heat type psoriasis vulgaris were randomly divided into two groups: QZLXD group (30 cases) and ampeptide group (28 cases). Patients in both groups were treated for 4 weeks. Psoriasis Area and Severity Index (PASI) score, Dermatology Life Quality Index (DLQI) score and the level of serum vascular endothelial growth factor (VEGF) were obtained to evaluate the efficacy of the two treatments. RESULTS: There was a better curative result in QZLXD group than in ampeptide group (P<0.05). The response rates in QZLXD and ampeptide groups were 83.33% and 64.28% respectively. DLQI score was also significantly improved during 4-week treatment in QZLXD and ampeptide groups (P<0.05). There was no significant difference in VEGF level between QZLXD and ampeptide groups. CONCLUSION: QZLXD is remarkably advantageous in treatment of psoriasis vulgaris of blood-heat syndrome and improvement of patient's quality of life. The mechanism may be related to decrease in serum VEGF level and action against neogenesis of blood vessels.

Nutrition investigations on the workers of drilling rig “Bohai 7” belong ing to Bohai Oil Company were conducted in November (winter) 1990 and June (summer) 1991, Energy consumptions of seven major and nine special actions of the offshore personnels were studied. Based on this, recommended amount of food to be taken and that of nutrients to be supplied in the daily dietary intake have been proposed for the workers on offshore drilling platforms, to improve the present improper food pattern which was characterized by high fat and low vitamin to ensure the workers health and to increase their potential adaptability to the special environment.
6.Study on induction of immunotolerance against Graves' disease in neonatal mice

Objective To investigate the feasibility of inducing neonatal immunotolerance against Graves'disease by gene TSH receptor (TSHR) 289 and its possible mechanism.Methods Neonatal (0-24 h) female BALB/c mice were divided into intraperitoneal injection group,intramuscular injection group,model group,and normal control group.The intraperitoneal group and the intramuscular group were further divided into low-dosage,middle-dosage,high-dosage tolerance groups,and the coresponding control groups.The tolerance groups and the controls were intraperitoneally or intramuscularly pretreated with low-dosage( 1×106 particles),middle-dosage( 1 × 108particles),high-dosage( 1 × 1010 particles)of Ad-TSHR 289 or Ad-lacz respectively.6 to 7 weeks later,the normal control group received intramuscular injection with Ad-lacz; the other groups were immunized with Ad-TSHR289,three times at 3 weeks interval.10 days after the first immunization,serum TRAb was detected.4 weeks after the last immunization,serum TRAb,TT4,splenic CD4 + CD25 + Foxp3/CD4 + were tested,and the thyroid tissues were examinated histologically.Results Ten days after the first immunization,no antibody response against TSHR was detected in the two high-dose tolerance groups,but the TRAb titer in respective controls was significantly higher( P<0.05 ).4 weeks after the last injection,in high-dose tolerance groups,only 1/10 of mice immunized by intraperitoneal or intramuscular injection elicited anti-TSHR antibody,and no mice immunized intraperitoneally had elevated serum TT4.Two of ten mice challenged intramuscularly showed slightly increased TT4 levels,but the respective controls displayed a strong antibody response( P<0.01 ) and elevated TT4 level ( P<0.05 ).The similar percentages of high TT4 and thyroid hyperplasia were found in all groups.Additionally,the frequencies of CD4+CD25 +Foxp3/CD4+in two high-dose tolerance groups were significantly increased as compared to those in controls( P<0.05 ).The incidence of Graves' disease in the other groups by intraperitoneal or intranuscular injections was not statistically different from those in the corresponding control groups and the model group.Conclusions The immune tolerance against Graves'disease is induced in neonatal mice by either intraperitoneal or intramuscular pathway with specific antigen of TSHR 289,carried by adenovirus vector,and then inhibits Graves' disease in adults. Stimulation with the high-dosage antigen is liable to induce immune unresponsiveness.CD4 + CD25 + Foxp3 +T cells may play an important role in the induction and maintenance of tolerance.
7.Gender difference in the establishment of animal model of Graves′ disease

Objective To investigate the effect of mice gender on the TSH receptor antibody(TRAb)titers, the levels of TT4,and the degree of thyroid hyperplasia by establishing an animal model of Graves′ disease in male and female BALB/c mice. Methods Male and female BALB/c mice were immunized with recombinant adenovirus expressing TSHRA subunit(Ad-TSHR289)to induce Graves′ disease. Animals were injected 3 times at intervals of 3 weeks. All mice were sacrificed 4 weeks after the last injection to obtain blood for measurement of TSHR antibody titers and TT4evels, and thyroid glands for histological examination. Results TRAb positive rates were 100% both in female or male mice. No significant difference was observed in titers of TRAb between them. The incidence of hyperthyroidism in female mice was higher than that in male mice, being 75.0% and 41.7% respectively. There was statistical difference in levels of TT4between females and males(P<0.01). Mice with high TT4exihibited marked thyroid hyperplasia. Conclusion Despite TSHR antibodies were similar between female and male mice, the incidence and degree of hyperthyroidism showed sex bias in Graves′s animal model. The results indicated that it was easier to induce model in females than in males by immunizing BALB/c mice with Ad-TSHR289.
8.Clinical observation on spleen-invigorating and qi-benefiting pediatric massage for treating recurrent respiratory tract infection in children with cerebral palsy due to qi deficiency of spleen and lung

Objective: To observe the clinical efficacy of spleen-invigorating and qi-benefiting pediatric massage (tuina) for treating recurrent respiratory tract infection in children with cerebral palsy due to qi deficiency of spleen and lung. Methods: A total of 70 children with cerebral palsy who suffered from recurrent respiratory tract infection due to qi deficiency of spleen and lung were randomized into an observation group and a control group by the random number table method, with 35 cases in each group. Both groups were treated with conventional rehabilitation training, while the observation group was given additional spleen-invigorating and qi-benefiting pediatric massage, and the control group additionally took oral Yu Ping Feng granule. The clinical efficacy was evaluated by the total effective rate, traditional Chinese medicine (TCM) symptom score, and serum levels of immunoglobulin (Ig) A, IgM and IgG. Results: The difference in total effective rate between the two groups after treatment was statistically significant (P<0.05). After treatment, the TCM symptom scores in both groups decreased to varying degrees than those before treatment, and the intra-group differences were statistically significant (all P<0.01); the differences in the scores of various TCM symptoms between the two groups were statistically significant (all P<0.05). After treatment, the levels of IgA, IgM and IgG of the children in both groups increased to varying degrees, and the intra-group differences were statistically significant (all P<0.05). The between-group differences in the IgA, IgM and IgG levels after treatment were statistically significant (all P<0.05). Conclusion: Spleen-invigorating and qi-benefiting pediatric massage can effectively treat recurrent respiratory tract infection due to qi deficiency of spleen and lung in children with cerebral palsy, relieve the clinical symptoms and improve immune function, and thus is worthy of clinical promotion and application.
9. Transplantation of autologous angiogenin-overexpressing marrow stromal cells in treatment of a porcine chronic ischemia model

Objective: To investigate the effect of autologous angiogenin(ANG)-overexpressing marrow stromal cells transplantation on the neoangiogenesis and cardiac function in a porcine model of chronic ischemia. Methods: Porcine models of chronic ischemia were established and were randomized into the following 3 groups: autologous marrow stromal cells transfected with Ad-ANG (Group I, n=11), autologous marrow stromal cells without transfection (Group II, n=10), and controls injected with serum-free DMEM medium (Group III, n=10). The autologous marrow stromal cells were cultured and were transfected with Ad-ANG, and the cells were labeled by CM-Dil. The cels were transplanted to the ischemia site of porcine heart. The parameters we observed included Rentrop score, ejection fraction, percentage of infracted area, CM-Dil labeled cells under fluorescence microscope, number of blood vessels and expression of ANG protein. Results: Compared with group H and IE, Group I had significantly increased Rentrop score, ejection fraction, numbers of blood vessels in the infarcted areas, CM-DiI labeled cells, and ANG protein expression, but significantly decreased infarcted area (P < 0. 05 or P < 0. 01). Conclusion: Transplantation of autologous angiogenin-overexpressing marrow stromal cells can yield a high survival rate of cells, greatly improving the blood supply of the infarcted cardiac area and cardiac function in porcine model.
10. Thoracoscopic and laparoscopic esophagectomy versus open procedure for early stage esophageal carcinoma: Comparison of clinical outcomes

Objective To evaluate the clinical value of thoracoscopy combined with laparoscopy for the treatment of early stage esophageal carcinoma. Methods From June 2010 to December 2011, 56 patients with esophageal carcinoma receiving thoracoscopic and laparoscopic esophagectomy (TLE) in our hospital and 53 receiving conventional open esophagectomy (OE) were included in the present study. The operative procedures, postoperative complications, recurrence or metastasis rates, mortality and follow-up outcomes were compared between the two groups. Results Compared with the OE group, TLE group had significantly less blood loss, shorter hospital stay, fewer ICU cases, shorter chest tube duration, less chest drainage amount and longer operation time(P<0. 05). The number of harvested lymph nodes and positive lymph node rate were not significantly different between the two groups. The incidence rate of postoperative complication was 8. 9% in TLE group and 11. 3% in OE group (P>0. 05). The two groups were followed up for 3 to 21 months. TLE group had a recurrence or metastasis rate of 5. 7%, similar to that in the OE group (8. 3%, P>0. 05). The total survival rate of TLE group was 98. 1%, which was similar to that in the OE group (97. 9%, P>0. 05). Conclusion Thoracoscopic and laparoscopic esophagectomy is a safe,feasible, effective and minimally invasive method for treatment of esophageal cancer; it has less complications and quick recovery and has a similar short-term outcome to conventional open esophagectomy.
