2.Application of nursing quality of bull’s-eye pattern on caring cancer patients with religious beliefs

ObjectiveTo study the effect of nursing quality of bull’s eye pattern on caring cancer patients with religious belief. Methods Three hundred and thirty-six cancer patients with religious beliefs were cared according to nursing quality of bull’s-eye pattern.All patients were evaluated by European organization for research and treatment of cancer quality of life questionnaire core 30(EORTC-QLQ-C30)before and after applications of nursing quality of bull’s-eye pattern and the results were compared.Result The scores of the dimensions of overall quality of life and function in EORTC-QLQ C30 questionnaire among those patients with implementation of nursing quality of bull’s eye patterns were higher than those before implementation of the pattern and the symptom score after the use of the pattern was statistically lower than that before its use in respect of dimensions of cancer symptoms(P<0.01). Conclusion The applications of nursing quality of bull’s-eye pattern on caring cancer patients with religious beliefs can improve the patients’ quality of life.
3.Effect of high volume hemofiltration on inflammatory cytokine clearance and hemodynamics in children with sepsis

Objective To evaluate the effect of high volume hemofiltration (HVHF) treatment on inflammatory cytokine clearance and hemodynamics in children with sepsis.Methods Thirty-four cases of septic children were randomly divided into two groups:the control group(17 cases) was administrated with the routine therapy,while the observation group(17 cases) was underwent with HVHF treatment on the basis of routine therapy.The inflammatory cytokines and hemodynamics were analyzed before and after the treatment.Results (1) Before treatment,the levels of tumor necrosis factor (TNF)-α in the control group and observation group were (103.6 ±26.1) ng/L and (106.6 ±31.6) ng/L,and the levels of interleukin(IL)-6 were (772.8 ± 92.4) ng/L and (761.3 ± 89.2) ng/L,respectively.The concentration of TNF-α and IL-6 were markedly reduced in both two groups after treatment (P < 0.05),with TNF-α and IL-6 reducing to (85.6±23.7) ng/L,(63.6 ±26.4) ng/L and(109.7 ±38.5) ng/L,(78.1 ±24.7) ng/L,respectively.Most importantly,the reduction in the observation group were significantly higher than that in the control group(P < 0.05).(2) Before treatment,no significant differences between the observation group and the control group were observed in hemodynamics,including mean arterial pressure,saturation oxygen,and heart rate(P > 0.05),whereas after the treatment,mean arterial pressure and saturation oxygen in the observation group were significantly elevated compared with those of the control group [(100.4 ± 12.1) mmHg vs.(82.2±11.7) mmHg(l mmHg=0.133kPa),(94.2±5.3)% vs.(91.4±3.2)%,P<0.05],andheart rate in the observation group[(85.1 ± 17.3) times/min] was significantly lower than that of the control group [(98.7 ± 18.5) times/min] (P < 0.05).Conclusion HVHF treatment on septic children efficiently promotes the clearance of inflammatory cytokines and improves hemodynamic property,and thereby leads to protection of organ function and the better prognosis for septic children.
4.Clinical Significance of Urinary Microprotein Examination on Early Diagnosis of Obesity - Related Glomerulopathy in Children

Objective To study the clinical significance of urinary mtcroprotems examination on early diagnosis of kidney damage of children with obesity. Method Urinary microproteins were tested in 40 obese children by enzyme - linked immunosorbent assay and 50 healthy children as controls. Results Urinary microprotem examination were abnormal in 8 obese children. Albumin in 5 cases increased, and transferrin(TRF) with retinol-binding protein in 1 case increased meanwhile. TRF in 3 cases increased. One case was abnormal by kidney biopsy.Conclusion Urinary microprotem examination was the sensitive index in early diagnosis of obesity-related glomerulopathy in children.
5.Questionnaire survey of the post-graduate thesis quality of Shanghai University of TCM

The quality of thesis is one of the key indicators of post-graduate education. To get the general current situation of the students’thesis quality in our University(Shanghai University of TCM),we designed a questionaire which mainly focus on topic selection,instructions from adviser, scientific research training,opening report,intermediate stage examination,thesis evaluation and presentation,etc. And we distributed it to post-graduate students and their advisers in TCM University widely. We got a general view of our current post-graduate thesis,made some strategies and suggestion for thesis improvement.
6.Application of SP and OSCE in the Clinical Skill Evaluation to Medical Students

To induce the technology into TCM clinical evaluation is in accordance with the requirements of TCM educational development,which can make students better grasp clinical various basic skills within limited studying periods.
7.Preliminary study on the distal femur rotational alignment

Objective To explore three dimensional relationships of commonly used bony marks of the distal femur, and to investigate its clinical application of distal femur rotational alignment. Methods One hundred and fifteen normal distal femurs of 62 volunteers were studied. With MR sagittal imaging and three dimensional reconstruction, correlative parameters, including PCA (posterior condylar angle), angle between surgical transepicondylar axis (STEA) and posterior condylar line (PCL); CTA(condylar twist angle), angle between clinical transepicondylar axis (CTEA) and PCL; APA, angle between the perpendicular of antero-posterior line (APL) and PCL; ATA, angle between the perpendicular lines of APL and STEA; CSA, angle between CTEA and STEA. All these angles were measured. The parameters of the two groups both in males and females, as well as in right and left knees were analysed. Results PCA: 3.44??1.08? (range, 0.62? to 6.75?); CTA: 6.03??0.86? (range, 2.93? to 8.92?); APA: 4.31??0.64? (range, 1.92? to 7.05?); ATA: 0.87??0.78? (range, -1.72? to 3.84?); CSA: 2.58??0.57? (range, 0.22? to 4.51?). There was no significant difference between left and right knee in terms of all the above mentioned parameters, yet there was significant difference between male and female groups in terms of PCA, CTA and ATA. PCA and CTA were larger in male groups than that of female group, while ATA in female group was larger than that of male group. Conclusion Three dimensional reconstruction based on imagings of MR is a reliable method to study the distal femur rotational alignment. In the normal knees, there is constant relationship between these parameters. Operators should use distal femur rotational alignment according to practical conditions in performing the total knee arthroplasty.
8.Experimental study of transplantion human embryonic OECs on transected spinal cord of rat

[Objective]To investigate the therapy of human enbryonic olfactory ensheathing cells(hOECs) transplantation on adult rats' transected spinal cord.[Method]Separated,cultured hOECS from abortus were obtained and percent age of P75 positive cells was calculated.Twenty-four Wistar rats were choosen and divided into experimental group and control group randomly.All rats of two goups were transected at the T_(10) segment of spinal cord.Nine to ten days later 5 ul(2.5?10~5) hOECs(labled by Hoechst 33342) were injected in the contusion area of lesion site in transplantation group.In control group same volume of DMEM-F12 were transplanted.1,2,4,6,8 and 10 weeks later all rats were estimated by system of the BBB locomotion score respectively.Then part of them were killed.The tissue section of spinal cords were separated and inspected by fluorescence microscope or the methods of immunohistochemical staining.Data were processed with statistical tools in double-blinded condition.[Result]Transplanted hOECs can survive for at least ten weeks and even migrated from the injuried area in spinal cord.From 4 weeks to 10 weeks the BBB locomotion scores of experimental group were improved significantly contrasting to that of control group(P
9.Evaluation of nitrative stress on retinal capillary by detection of trans-arachidonic acid with GC/MS in diabetic rats

Background Trans-arachidonic acids (TAAs) are specific lipid markers of nitrative stress and the major products of NO2·-mediated ismoerization of aracchidonic acid(AA),and they represent a possible new mechanism of NO2·-induced toxicity in ischemic retinopathy.Our previous study demonstrated that high glucose induce nitrative stress response. Objective This study aimed to evaluate the effect of nitrative stress on retinal capillary in diabetic rats by detecting the TAAs with gas chromatography and mass spectrometry(GC/MS).Methods One hundred 2-week-old clean male SD rats were randomly divided into control group and diabetic group.Diabetic rat models were induced by intraperitoneal injection of streptozoticin of 60 mg/kg,and the successful models were identified as the blood glucose level>16.7 mmol/L.The synthesis method was modified in this study andaraehidonic acid w8$utilized as starting material.14E-AA was obtained as a standard sample via expoxidation and deoxidation in a safe and practical pattern.Serum TAAs and AA in diabetic rats were detected by GC/MS in 2,4,8,12 and 16 weeks after injection of streptozoticin.The ratio of peak area of TAAs to AA with selected ion of 79 was estimated and compared with normal rats. Results Fifty rats had been in Diabetic models were established in 50 rats with the successful rate 100%,and the high blood glucose level in model rats retained throughout the experimental duration.GC/MS results showed that the 14E-AA was obtained with the purity of 97.3%,which provided a standard sample to nitrative stress-related research as a premise. No significant differences were found in serum TAAs/AA values at 2 weeks and 4 weeks between model group and control group (t =-0. 376, t =-0. 642,P>0. 05). However,serum TAAs/AA values in at 8,12 and 16 weeks after streptozoticin injection were considerably elevated in comparison with those of control group (0. 0832 vs 0. 1042,0. 0910 vs 0. 1568, 0. 1100 vs 0. 1895;t=-36.409,t=-166.714,t=-148.212,P<0. 05). Conclusion Elevation of serum TAAs/AA is associated with diabetic duration,implying that damage of nitrative stress to retinal capillary is a parallel procedure to diabetic course.Establishment of GC/MS detection system offers a new evaluating indicator in the research on microvascular ischemic disease and could be implemented in clinical testing.
10.Cytological Study in vitro on Co-delivery of siRNA and Paclitaxel within Solid Lipid Nanoparticles to Overcome Multidrug Resistance in Tumors.

Multidrug resistance (MDR) remains the major obstacle to the success of clinical cancer chemotherapy. P-glycoprotein (P-gp), encoded by the MDR1, is an important part with complex mechanisms associated with the MDR. In order to overcome the MDR of tumors, we in the present experimental design incorporated small interfering RNA (siRNA) targeting MDR1 gene and anticancer drug paclitaxel (PTX) into the solid lipid nanoparticles (SLNs) to achieve the combinational therapeutic effects of genetherapy and chemotherapy. In this study, siRNA-PTX-SLNs were successfully prepared. The cytotoxicity of blank SLNs and siRNA-PTX-SLNs in MCF-7 cells and MCF-7/ADR cells were detected by MTT; and the uptake efficiency of PTX in MCF-7/ADR cells were detected via HPLC method; quantitative real-time PCR and flow cytometry were performed to investigate the silencing effect of siRNA-PTX- SLNs on MDR1 gene in MCF-7/ADR cells. The results showed that PTX loaded SLNs could significantly inhibit the growth of tumor cells, and more importantly, the MDR tumor cells treated with siRNA-PTX-SLNs showed the lowest viability. HPLC study showed that SLNs could enhance the cellular uptake for PTX. Meanwhile, siRNA delivered by SLNs significantly decreased the P-gp expression in MDR tumor cells, thus increased the cellular accumulation of rhodamine123 as a P-gp substrate. In conclusion, the MDR1 gene could be silenced by siRNA-PTX-SLNs, which could promote the growth inhibition efficiency of PTX on tumor cells, leading to synergetic effect on MDR tumor therapy.
