1.Study on the injury effects and mechanisms of circulating histones on the hepatocytes of patients with hepatitis B virus-related liver failure

Objective To investigate the injury effects and mechanisms of circulating histones on the hepatocytes in patients with hepatitis B virus (HBV )-related liver failure (HBV-LF) .Methods Serum samples from patients with HBV-LF were collected . The levels of serum histone H3 , histone H4 , prothrombin activity (PTA ) ,total bilirubin (TBil) ,creatinine (Cr) and international normalized ratio (INR) of the patients were measured .Model for end-stage liver disease (MELD) score was calculated in the patients .The serum levels of histones were compared between patients and the healthy volunteers . The correlation of histones with the MELD score and PTA was analyzed .The human liver cell line L-02 cells were cultured and treated with the serum of patients or L-02 cellular lysate supernatant preincubated with or without anti-histone H3 and H4 antibodies .The cellular morphology and rate of apoptosis were observed .Intracellular calcium ion concentration and Caspase-3 activity were detected in the cultured L-02 cells treated with histones .Mean of two independent samples was compared using t tests .Relationship between histones and the MELD score or PTA was conducted using Spearman correlation analysis .Results The levels of serum histones in the patients with HBV-LF were much higher than those in the healthy volunteers (H3 :[5 390 .3 ± 1 032 .0] μg/mL vs [42 .7 ± 12 .8] μg/mL , t = 32 .76 , P < 0 .01 ; H4 :[4 205 .1 ± 662 .3] μg/mL vs [40 .3 ± 14 .6] μg/mL ,t = 39 .74 , P< 0 .01) .In addition ,serum histones (H3/H4) levels in patients were negatively correlated with serum PTA (r= - 0 .325 ,P= 0 .038 and r =- 0 .572 ,P= 0 .028 ,respectively) ,but positively correlated with the MELD score (r= 0 .359 ,P= 0 .021 and r = 0 .568 , P = 0 .007 , respectively ) . Both serum of patients with HBV-LF and L-02 lysate supernatant were toxic to cultured L-02 cells .The injury effect was inhibited by anti-histone antibodies ([9 .3 ± 1 .5]% vs [14 .3 ± 0 .6]% , t = 4 .259 , P= 0 .02) .L-02 cells treated with calf thymus histone were cultured for 4 h . Cellular toxicity of histones resulted in Caspase-3 activation . The effect was inhibited by anti-histone antibodies ([3 .5 ± 0 .5]% vs [5 .2 ± 0 .6]% ,t= 4 .243 ,P= 0 .02) .Conclusion The elevated circulating histones in the patients with HBV-LF may aggravate the liver damage .
2.Changes and correlation between platelet activation and inflammatory response in patients with essential hypertension

Objective To investigate the changes and correlation between platelet activation and inflammatory response in patients with essential hypertension(EH). Methods Sixty-one patients with essential hypertension and sixty-four patients with paroxysmal supraventricular tachycardia as control group were enrolled in the study. The levels of high sensitivity C reactive protein ( hs-CRP ) and mean platelet volume ( MPV ) were measured and compared between the control group and the essential hypertension group. Results Compared with the control group,the levels of hs-CRP and MPV in the EH group were significantly higher( [3.51 ± 1.95]mg/L vs [ 1.15 ± 0. 77 ] mg/L, P < 0. 05 ). The MPV in the EH group was significantly higher than that in the control group ( [ 11.29 ± 2. 18 ] flvs [ 9. 20 ± 2. 31 ] fl, P < 0. 05 ). The level of hs-CRP positively correlated to the level of M PV (r = 0. 452, P = 0. 003 ). Conlusions There were inflammatory response and platelet activation in patients with EH and the serum levels of hs-CRP and MPV is correlated with the development of essential hypertension.

SWI/SNF is an ATP-dependent chromatin remodeling complex .ARID1 A gene is an important subunit of SWI/SNF complex and its dysfunction can cause abnormal chromatin remodeling , resulting in tumorigenesis .AR-ID1A occurs frequently low expression or deletion mutation in a variety of malignant tumors , such as ovarian canc-er, liver cancer, breast cancer, stomach cancer, lung cancer and so on, which indicates that ARID1A is an impor-tant tumor suppressor gene .
4.Clinical significance of serum prealbumin and bilirubin in patients with coronary heart disease

Objective To investigate the changes and clinical significance of serum prealbumin (PAB) and bilirubin in patients with coronary heart disease (CHD). Methods The levels of serum PAB and bilirubin were measured respectively in 234 patients with CHD (CHD group) and 77 patients with paroxysmal supraventricular tachycardia (PSVT, control group), and the former group was divided into stable angina pectoris (SAP) group with 80 patients,unstable angina pectoris (UAP) group with 84 patients and acute myocardial infarction (AMI) group with 70 patients,according to the chnical manifestation. Results The levels of total bilirubin (TBIL),direct bilirubin (DBIL) and indirect bilirubin (IBIL) in CHD group [ (11.8 ± 6.1 ), (4.8 ± 1.0) and (7.7 ± 2.7) μ mol/L] were significantly lower than those in control group [(14.6 ±5.6), (6.2 ±1.3) and (9.5 ±6.1)μmol/L] (P <0.05). The levels of TBIL, DBIL and IBIL decreased gradually from SAP group, UAP group to AMI group, but there was no significant difference among three groups (P > 0.05 ). The levels of serum PAB in CHD group were also significantly lower than those in control group [(205.55 ±5.68) mg/L vs.(229.78 ± 9.62) mg/L] (P < 0.05),and decreased gradually from SAP group,UAP group to AMI group [(215.73 ± 7.98), (214.12 ± 3.79) and (195.75 ± 7.07) mg/L],but there was no significant difference among three groups (P > 0.05 ). Conclusion The levels of serum PAB and bilirubin are related to the severity of CHD, and can be used as important biochemical indicators in monitoring the attack and severity in patients with CHD.
5.Iron-deficiency anemia complicated by cerebral thrombosis an analysis of 4 cases

Objective To investigate the relationship between cerebral thrombosis and iron-deficiency anemia, and their clinical manifestations and therapy. Methods The clinical data of 4 patients with iron deficiency anemia complicated by cerebral thrombosis were analyzed retrospectively. Results Four patients were moderate-to-sexere anemia with thrombocytosis, and their blood pressure was relatively low. Two had chronic infection foci. All patients had cerebral infarction in the internal carotid system, of those, 2 were weatershed infarctions, and 2 were basal ganglia infarctions. Oxygen inhalation, correcting anemia, adjusting blood presssure to an appropriate level, and other conventional treatment were effective. Conclusions The patients with moderate-to-severe anemia, such as accompanied by thrombocytosis and lower blood pressure, are easy to came thrombosis. While in the routine treatment of cerebral thrombosis, attention should be paid to finding the causes of anemia and correcting them.
6.Investigation on the Mode of Double Testing of Blood for Virus Markers

Objective To probe into the significance and role of double testing of blood for virus markers.Methods The data on the results of routine blood testing at some blood centers in China from July to December,1999 were gathered and analysed.The blood donors with inconsistent results during the double testing were followed up and their blood samples were collected at least half a year later.Nucleic acid amplification testing(NAT) was performed on the samples.Results Only 0.63% of the donors showed inconsistent results during the double testing.Most of the positive results were false positive.Conclusion The double testing currently implemented does not help significantly improve the quality of the blood supply.It is important to enhance blood safety further by improvement of quality of the reagents and introduction of NAT.
9.Visual sexual stimuli induced asymmetrical activation on Broca's area on right hemisphere:an fMRI research

Objective:To investigate the brain activation patten associated with visual sexual stimuli.Method:Thirteen healthy right-handed women were enrolled through advertisements on campus.They were scanned under functional magnetic resonance imaging(fMRI) when they watched emotional neutral films excepts and erotic films excepts.Statistic on theses data reveals the difference of brain activation.Result:The results showed that,compared to emotional neutral films,sexual visual stimuli were associated with the activation of a set of bilateral brain areas including lateral occipital cortex,postcentral gyrus,superior parietal lobule,occipital fusiform gyrus, hippocampus,and so on.Most activated brain areas are bilateral.Only in inferior frontal gyrus pars opercularis(Z=3.64,P<0.0001) and pars triangular(Z=3.42,P=0.003) there were significant right hemispheric activation.Conclusion:fMRI results indicate that inferior frontal gyrus is related to the visual sexual stimuli of women subjects,and is specified on the right hemisphere rather than left one.This sphere-special area may play an important role on sexual message processing.
10.Influence of intraspinal implantation of pSVPoMcat genetically modified schwann cell on regeneration of injured spinal cord

Objective In order to observe the role of genetically modified Schwann cell (SC) with pSVPoMcat in the regeneration of injured spinal cord.Method The cells were implanted into the spinal cord.Ninety SD rats were used to establish a model of hemi- transection of spinal cord at the level of T8,and were divided into three groups,randomly, that is,pSVPoMcat modified SC implantation(Group A), SC implantation(Group B),and without cell implantation as control(Group C).After three months the presence of axonal regeneration of the injured spinal cord was examined by means of horseradish peroxidase(HRP)retrograde labeling technique and stereography.Result The results indicated that HRP labeled cells in Group A and B could be found in the superior region of injured spinal cord and the brain stem such as the red nuclei and oculomotor nuclei. The density of ventral horn neurons of the spinal cord and the number of myelinated axons in 100 μ m of the white matter was A >B >C group.Conclusion In brief,the pSVPoMcat modified SC intraspinal implantation could promote regeneration of the injured spinal cord.
