Objective To explore NF-κB(P65)expression in breast cancer and its clinical significance.Methods Immunohistochemical staining was used to examine the expression of NF-κB(P65)and its relationship with related elinicopathological factors were analyzed in eighty-seven IDC(invasive ductal cancinoma).Results NFκB(P65)expression was significantly correlated with lower tumor grading,maxilary lymph node metastasis,later TNM styng and HER-2 positive(all P<0.05),but not with tumor size,age of the patient and the state of ER、PR.Conclusion NF-κB(P65)should be risk factors of poor prognosis in IDC.
2.Study on the Effects of Prenatal Exposure to Low Level Lead on the Physical and Neuro-behavioral Development of Mice and the Interaction Between Lead and Zinc

Objective To study the effects of prenatal exposure to low level lead on the physical and neuro-behavioral development of mice and the interaction between lead and zinc. Methods Kunming pregnant mice were provided with drinking water containing lead, zinc and lead-zinc jointly and respectively. The body weight of maternal mice, offspring's survival rate and body weight were observed. Neuro-behavioral development of offspring was assessed by observation of offspring's behavior performance. The levels of dopamine (DA), 5-hydroxytryptamine (5-HT) and malonaldehyde (MDA) in offspring's brains were measured. Results At the doses of lead and zinc which did not effect the increase of maternal body weight during pregnancy, the lead exposure group showed low survival rate, growth retardation and delay of physiological development and neonatal reflexes in the offspring, the levels of DA and MDA in offspring's brains increased compared with the control group. Offspring's low survival rate and growth retardation couldn't be antagonized by zinc, but equal mole as lead of zinc could inhibit the harmful effects on neuro-reflex development. Zinc could also inhibit the increase of DA and MDA levels in offspring's brains induced by lead . However joint exposure to lead and half dosage as much as lead of zinc could deteriorate the effects on delay of physical, neuro-behavioral development and accelerate the DA and 5-HT decompose compared with that in the lead exposure group. Conclusion The harmful effects on offspring's growth and neuro-behavioral development could be induced by prenatal exposure to low level of lead. The types of joint-effect of lead and zinc were different according to the concentration of zinc, which indicated that the zinc-lead interaction was very complicated and it should be very prudent to antagonize lead toxicity using zinc.
4.Anti-oxygen free radicals and anti-lipoperoxidation of nicotinic acid on hyperlipodemia quail

Aim To study the oxygen free radical scavenging activities and antilipid peroxidation of nicotinic acid. Methods High lipid models of quail were induced by high fat diet. Five groups were divided, namely control group, high lipid group, NA group(150 mg?kg-1 and 75 mg?kg-1), and Vit E group (100 mg?kg-1). The contents of TC,TG,MDA and activities of SOD were examined for 3 times in the plasma and liver after administering NA for 9 weeks. Results The results showed the increase of TC,TG and MDA contents and decrease of SOD activities in high lipid groups by 3 weeks high fat diet. After administering NA for 3 weeks, NA reduced TC,TG and MDA contents and increased SOD activities, revealing the relationship to dose-dependence. Conclusion NA is most likely to possess the capabilities of anti-oxygen free radicals and anti-lipoperoxidation to hyperlipodemia quail.
5.Viral shedding in day-care children and students infected with norovirus in outbreaks

Objective:This study aimed to investigate the time duration of norovirus shedding among day-care children and students during norovirus outbreaks, as well as influencing factors affecting the viral shedding. Methods:Suspected cases of norovirus infection and their close contacts were collected from child care and school settings during norovirus outbreaks in Xuhui District, Shanghai, from 2017 through 2019. Specimens were detected using real-time RT-PCR to determine whether children had been infected with norovirus. Subsequently, further specimens were collected every 3-7 days from infected children until specimens tested negative for norovirus. Results:A total of 76 outbreaks were reported involving 1 014 suspected cases. In the 421 suspected cases, 311 confirmed cases were diagnosed after examination. Furthermore, a total of 58 confirmed cases participated in this study with informed consent, with a participation rate of 18.65%. The average time duration of norovirus shedding was (16.24±13.80) days, in which 79.31% had viral shedding more than 7 days, 37.93% more than 14 days and 17.24% more than 21 days. ObjectiveTo analyze the epidemiological characteristics and incidence trend of influenza-like illness in Xuhui District from 2010 to 2019, in order to understand the patterns of influenza epidemic of Xuhui District,and to provide evidence for the prevention and control of influenza in the future. MethodsSurveillance data of influenza-like illness (ILI) in Xuhui District from 2010 to 2019 were collected from the National Influenza Surveillance System for statistical description and trend analysis. ResultsFrom 2010 to 2019, the overall proportion of influenza-like illness was 0.54%. The annual proportions of ILI were between 0.35%~1.14%.The majority of cases were reported in age group of 25~59, accounted for 54.50% of the total ILI. A total of 9 053 throat swab specimens from ILI patients were collected and tested, from which 2 137 specimens were positive, with a positive rate of 23.61%. The most frequent subtype of influenza virus detected was influenza A virus (accounting for 67.62%). The proportion of ILI and the positive rate of influenza virus nucleic acid detection reached the peak in summer and winter over time. Objective To observe the effect of Bingdouting capsule(BC)on rabbit model of sick sinus syndrome.Methods Rabbit models of sick sinus syndrome were established by external application of 20 % methanal on the region of sinoatrial node.The effect of BC on heart rate and electrophysicological parameters of sinoatrial node was observed.Results BC markedly increased the heart rate,shortened the sinoatrial conduction time(SACT)and corrected sinoatrial node recovery time(CSNRT).Conclusion BC has an obvious effect for the treatment of sick sinus syndrome in rabbits.
8.RGD4C modified ferritin nanocages for rat hepatic stellate cells-targeted drug delivery

Objective The purpose of this study was to prepare RGD4C modified ferritin nanocages (RGD4C-FRT) for targeted drug delivery to rat hepatic stellate cells (HSC-T6). Methods RGD4C modified human H-chain ferritin was expressed and purified. Doxorubicin (Dox) was encapsulated into the cavity of RGD4C-FRT through ion channels, which resulted in RGD4C-FRT-Dox. The target of RGD4C-FRT-Dox to HSC-T6 was detected using fluorescence microscopy. Results Transmission electron microscopy showed that RGD4C-FRT was hollow spherical-structured with uniform size and good dispersion. The average particle diameters of RGD4C-FRT and RGD4C-FRT-Dox were 12.57 nm and 20.13 nm , respectively. The drug encapsulation efficiency and loading percentage of RGD4C-FRT-Dox were 77.32% and 15.88% respectively. RGD4C-FRT-Dox was significantly uptaken by HSC-T6, and the uptake could be blocked by the empty carrier RGD4C-FRT. Conclusion RGD4C-FRT-Dox can specifically target HSC-T6. Further animal experiments are needed to inspect its therapeutic effect on liver fibrosis.
9.Clinical, myopathological and genetic research of a patient with mitochondrial encephalomyopathy, lactic acidosis and stroke-like episodes/Leigh syndrome overlap syndrome caused by m.10158 T>C mutation

Objective To report the clinical, myopathological and genetic features of a patient with mitochondrial encephalomyopathy, lactic acidosis and stroke-like episodes (MELAS)/Leigh syndrome (LS) overlap syndrome who carried m.10158 T>C mutation. Methods The patient′s clinical and imaging materials were collected. An open biopsy of right biceps brachii was performed. DNA samples were prepared from the patient and her mother′s blood. Direct sequencing of the complete mitochondrial genome was performed to detect the mtDNA mutation.Western blotting was used to estimate the content of respiratory complexes in the patient′s muscle. Results The patient was a 40-year-old female. She had seizures and lost consciousness for 9 months. Brain MRI findings consisted of asymmetrical lesions in the cerebral cortex of the frontal and temporal lobes, as well as symmetrical lesions bilaterally in the basal ganglia. Muscle biopsy showed typical ragged red fibers. Direct sequencing of the complete mitochondrial genome from blood and muscle of the patient revealed the T-to-C transition at nucleotide position 10158 in the MT-ND3 gene.The mutation rate was 9.31% and 70.0%, respectively.Western blotting demonstrated that the contents of complexes Ⅰ and Ⅳ were significantly lower in the patient′s muscle mitochondria compared with the normal controls (53.1%±1.2% vs 88.6%±1.7%, t=4.08, P<0.05;57.3%±2.4% vs 80.1%±2.1%, t=3.39,P<0.05).Conclusion We reported a case of MELAS/LS overlap syndrome who carried m.10158 T>C mutation in MT-ND3 gene and DNA test is very important for the diagnosis of the disease.
10.Expression of FGFR3 in acute lymphoblastic leukemia patients and its contribution to proliferation of circulating endothelial cells

AIM: To evaluate the expression of fibroblast growth factor receptor 3 (FGFR3) in acute lymphocytic leukemia (ALL) patients and its contribution to the proliferation of circulating endothelial cells (CECs).METHODS: The mRNA expression levels of FGFR3 in 44 patients with ALL were assayed by RT-PCR.Overall survival (OS) rates of the patients in FGFR3+ group and FGFR3-group were estimated by Kaplan-Meier analysis.The CECs were sorted from peripheral blood by magnetic-activated cell sorting and then counted by 3-color flow cytometry.The cell counts, antigen expression, growth curve and colony forming rate of the CECs in the 2 groups were determined.The FGFR3 expression of CECs was identified by immunofluorescence staining.RESULTS: The positive rate of FGFR3 mRNA expression was 43.2% in 44 ALL patients with normal karyotype.T-ALL expressed higher level of FGFR3 than B-ALL (P<0.05).FGFR3 was over-expressed in ALL patients with bone marrow blast proportion ≥5% (P<0.05).The probability of OS was significantly lower in FGFR3+ group than that in FGFR3-group (P<0.05).The sorted CECs highly expressed CD31, CD144, VEGFR-2 and CD146, and rarely expressed CD45.The counts of CECs and expression level of CD133 significantly increased in FGFR3+ group compared with FGFR3-group.The same result of the amount of colony formation was observed (P<0.05).There was significant difference at 3 time points of cultured CECs count in vitro between FGFR3+ group and FGFR3-group (P<0.05).The positive rate of FGFR3 expression of CECs from 19 ALL-FGFR3+ patients was (29.00±15.71)%.CONCLUSION: The over-expression of FGFR3 gene in ALL may be helpful to evaluate the prolife-ration of CECs, and become a double target with anti-tumor and anti-angiogenesis effects to offer more choice for molecular therapy in the future.
