1.The effects of Slit2 protein on proliferation and migration of rats vascular smooth muscle cells stimulated by TNF-α

Objective To observe the effects of Slit2 protein on the proliferation and migration of VSMCs .Methods The VSMCs was cultured in our laboratory .The experiment was divided into two parts ,part one:VSMCs were divided into normal con‐trol group and experimental groups(culture with 50 ,75 ,100 ,125 and 150 ng/mL Slit2 respectively);part two :VSMCs were divided into normal control group ,positive control group(culture with TNF‐α10 ng/mL) and experimental groups(culture with TNF‐α10 ng/mL+Slit2 50 ng/mL ,TNF‐α10 ng/mL+Slit2 75ng/mL ,TNF‐α10 ng/mL+ Slit2 100 ng/mL ,TNF‐α10 ng/mL+ Slit2 125 ng/mL and TNF‐α10 ng/mL+Slit2 150 ng/mL respectively) .To detect proliferation and migration of VSMCs by CCK‐8 and tran‐swell experiment .Results The difference of OD value and numbers of VSMCs has no statistical significance in the presence of Slit 2 (P=0 .516 ,P=0 .52) .The numbers of VSMCs has statistical significance between control and positive control groups (P=0 .00) . The numbers of VSMCs in experimental groups were fewer than positive control group (P<0 .05) ,whereas the difference of OD value still has no statistical significance between experimental and positive groups (P= 0 .173) .Conclusion Recombinant Slit2 could inhibits migration in VSMCs induced by TNF‐α,whereas it has no effect on proliferation of VSMCs .

Alzheimer’s disease (AD) has become one of the worldwide critical diseases which seriously threaten the health of the elderly. Exploring and developing medicine with high efficiency and low toxicity for AD patients is one of the vital medicine issues. Traditional Chinese medicine has high research and development value in the prevention and treatment of AD. Currently, increasing researches have proved that Ginseng Radix et Rhizoma, as a kind of traditional and valuable Chinese herbal medicine, shows effects on improving learning and memorizing ability and prevention and treatment of AD. This article reviewed Ginseng Radix et Rhizoma extract improving symptoms of AD and its mechanism of action in detail, with a purpose to provide references for Ginseng Radix et Rhizoma extract improving learning and memorizing ability and prevention and treatment of AD in clinic.
3.Study on biological activity of thrombopoietinⅡ in vivo

AIM: To investigate the biological activity of thrombopoietin Ⅱ(TPOⅡ) in vivo , which consists of two new kinds of ligand binding with thrombopoietin receptor. METHODS: Purified ligandⅠof TPOⅡ, artificial compound ligandⅡ of TPOⅡand rhTPO were injected into purebred Babl/c mice respectively in 7 days by intraperitoneal injection once for a day. Then the biological activity of TPOⅡ was analyzed by measuring peripheral platelet counts by the end of the seventh day. RESULTS: On the seventh day, the platelet counts of mice treated by ligandⅠof TPOⅡ were higher than that in the negative control group( P 0.05). On the fourteenth day, the platelet counts increased in two all experimental groups of TPOⅡcompared with negative control group( P 0.05). Moreover the platelet counts of mice in two experimental groups of TPOⅡ and the positive group showed increase with experimental days. CONCLUSION: The purified ligandⅠof TPOⅡ had obvious activity in increasing platelet production, which is not different from the effect of rhTPO.
4.Effects of Compound Kushen Tang on Ulcerative Colitis in Rats and the Underlying Mechanism

Objective: To investigate the therapeutic effects of compound Kushen Tang and its relevant mechanism in TNBS-in-duced ulcerative colitis ( UC) rats. Methods:UC was induced by TNBS in rats. After compound Kushen Tang was given orally, the levels of MDA, iNOS, and NO and the activity of MPO, SOD, and GSH-Px were measured. The general condition of rats and colon tissue morphology were observed. Results:The levels of MDA (P<0. 05), iNOS (P<0. 01) and NO (P<0. 01) and the activity of MPO (P<0. 01) in tissues of UC rats were significantly higher than the control group. The activity of SOD (P<0. 01) and GSH-Px (P<0. 05) were significantly lower than those in the control group. After the treatment with high doses of compound Kushen Tang, the levels of MPO (P<0. 01), MDA (P<0. 05), iNOS (P<0. 05) and NO (P<0. 01) were significantly decreased, and the activity of SOD (P<0. 01) and GSH-Px (P<0. 05) significantly increased. The therapeutic effect was dose-dependent and the general con-dition of rats and colon tissue morphology were also significantly improved. Conclusion:Compound Kushen Tang is considered as a no-vel therapeutic alternatives for the treatment of UC, which can reduce coloni inflammatory injury and ameliorate the colitis.
5. The impact of urination behavior on overactive bladder of female operating room nurses

Objective:
To determine the impact of urination behavior of overactive bladder in female operating room nurses.
Methods:
A total of 381 female operating room nurses were recruited from three first-class hospitals in Nanjing by convenience sampling method. They were investigated with Chinese version of OAB symptom score scale and Chinese version of urinary behavior scale. The relationship between urination behavior and overactive bladder was analyzed.
Results:
The total score of overactive bladder female in operating room nurses was 10.05±3.28. There were significant positive correlations between score of urination behavior and score of overactive bladder. Regression analysis showed that the factor scores of urination behavior such as urinary behavior, no urine urination behavior could independently explain 41.3% of the variance for overactive bladder after controlling demographic variables.
Conclusion
Clinical managers should focus on the shortcomings of urination behavior of female operating room nurses, and take effective measures to to improve the symptoms of overactive bladder in female operating room nurses.
6.The mechanism of overeXpression of retinoic acid receptor alpha in attenuating renal interstitial fibrosis in rats

Objective To study the mechanism of overexpression of retinoic acid receptor alpha( RARα)in attenuating renal interstitial fibrosis(RIP)in rats. Methods Porty 6_week_old male SD rats were randomly divided into 4 groups:sham operation group,model group,negative control group and transfection group,with 10 rats in each group. Rats in model group were separated and double ligated with left ureter;rats in sham operation group were not li_gated with ureter;rats in transfection group and negative control group were transfected with adeno_associated virus and negative control virus carrying RARα gene on the basis of model group,respectively. All rats were sacrificed 2 weeks later. Left kidney tissues were taken for pathological examination and RIP index was calculated. The expression of colla_genⅣ(Col_Ⅳ)and fibronectin(PN)in renal tissue was detected by using immunohistochemistry. The expressions of RARα,prohibitin(DHB)and transforming growth factor_beta 1(TGP_β1)in renal tissue were detected by using real_time fluorescence quantitative polymerase chain reaction( RT _qDCR)and Western blot. This commentary was based on the paper "Calorie Restriction with or without Time-Restricted Eating in Weight Loss" (TREATY trial) published recently in N Engl J Med, with the aim to introduce the study design of TREATY trial and discuss the main findings along with its clinical applicability. This trial offers additional evidence on the time-restricted eating and provides insights for future research.
8.Combination of neutrophil to lymphocyte ratio, NIHSS score and collateral circulation Tan score predicts the outcome of intravenous thrombolysis in patients with anterior circulation large vessel occlusive stroke

Objective:To investigate the predictive value of baseline peripheral blood neutrophil to lymphocyte ratio (NLR), National Institutes of Health Stroke Scale (NIHSS) score combined with collateral circulation Tan score for the outcome of intravenous thrombolysis in patients with acute anterior circulation large vessel occlusive stroke.Methods:Patients with acute anterior circulation large vessel occlusive stroke received alteplase intravenous thrombolysis in the Departmet of Neurology, the Second Affiliated Hospital of Anhui Medical University from January 2019 to May 2021 were enrolled retrospectively. The modified Rankin Scale score was used to evaluate the outcomes at 90 d after thrombolysis. 0-2 was defined as good outcome and 3-6 were defined as poor outcome. The demographics, clinical data and imaging data between the good outcome group and the poor outcome group were compared. Multivariate logistic regression analysis was used to determine the independent influencing factors of the outcome of thrombolysis, and the receiver operating characteristic (ROC) curve was used to analyze the predictive value of NLR, NIHSS score, Tan score and their combination on the outcome of intravenous thrombolysis. Results:A total of 118 patients with acute anterior circulation large vessel occlusive stroke were enrolled during the study. Their age was 67.73±11.91 years, and there were 71 males (60.2%). Fifty-eight patients (49.2%) were in the good outcome group and 60 (50.8%) were in the poor outcome group. Univariate analysis showed that there were significant differences in NLR, collateral circulation Tan score and baseline NIHSS score between the good outcome group and the poor outcome group (all P<0.05). Multivariate logistic regression analysis showed that NLR (odds ratio [ OR] 1.299, 95% confidence interval [ CI] 1.005-1.679; P=0.046), baseline NIHSS score ( OR 1.150, 95% CI 1.037-1.275; P=0.008), collateral circulation Tan score ( OR 0.298, 95% CI 0.160-0.556; P<0.001) were independently associated with the outcomes. ROC curve analysis showed that the areas under the curve of NLR, baseline NIHSS score, Tan score and their combination for predicting poor outcome were 0.640, 0.752, 0.823 and 0.870, respectively. Conclusions:Peripheral blood NLR before thrombolysis, baseline NIHSS score and collateral circulation Tan score could predict the outcomes after intravenous thrombolysis in patients with acute anterior circulation large vessel occlusive stroke, and the combination of the three had a higher predictive value.
9.The best evidence summary for the prevention of postural nerve injury in adult patients undergoing operation with general anesthesia

Objective:To comprehensively retrieve and summarize the best evidence on the prevention of position-related nerve injury in adult patients undergoing general anesthesia, in order to provide evidence-based guidance for standardized position management during general anesthesia surgery in adults.Methods:Clinical decision-making, guidelines, evidence summaries, best practice, practice advisories, systematic reviews, expert consensuses were systematically search in UpToDate, BMJ Best Practice, Guidelines International Network (GIN), Canadian Medical Association: Clinical Practice Guideline(CMA Infobase), National Guideline Clearinghouse (NGC), National Institute for Health and Care Excellence(NICE), Cochrane Library, PubMed, CNKI, Wanfang, and related websites. The literature retrieval period was from the database construction to September 30,2022. The guidelines were independently assessed by 4 researchers, and the remaining literature was independently evaluated by 2 researchers. The literature that met the criteria was extracted. Finally, the expert meeting integrated the evidence and summarized the evidence topics.Results:A total of 17 articles were included, including 6 clinical decision-making, 3 guidelines, 2 practice advisories,5 systematic reviews, and 1 expert consensus. A total of 32 pieces of best evidence and 5 evidence topics were formed: personnel placement, perioperative evaluation, points of surgical position, key points in special surgery or situation, other general principles.Conclusion:This study summarized the best evidence for the prevention and management of surgical position related nerve injuries, and provides a scientific theoretical reference for postural management of adult patients undergoing operation with general anesthesia, to reduce the incidence of nerve injuries related position.
10.The association between the DYS527a/b and DYF387S1a/b multi-allele pattern and Y-SNP haplogroups

Objective To investigate the association of the DYS527a/b and DYF387S1a/b multi-allele pattern with Y-SNP haplogroups.Methods Samples from 295 unrelated males who carrying the DYS527a/b multi-allele pattern were amplified by the YFilerPlus? kit.The genotypes of their frequency distributions,including three multi-copy loci(DYS527a/b,DYF387S1a/b,DYS385a/b)and other single-copy loci were obtained.The DYS527a/b multi-allele pattern and their haplotypes were examined for the associations with Y-chromosome haplogroups using the AIYSNP42 kit,which contains 42 Y-SNP loci.Based on the above results,the association between the DYS527a/b multi-allele patter and its constituent Y-STR haplotypes and related haplogroups was discussed.Results Among the 295 samples,the DYS527a/b tri-allele pattern and tetra-allele pattern accounted for 97.29%and 2.71%respectively,while the DYF387S1a/b tri-allele pattern and tetra-allele encompassed 54.24%and 4.75%.Null allele was detected in DYS448 in 13.22%of the samples.Here,7 Y-SNPs were deticted such as O-M175 and C-M131 which encompassed 45.76%and 45.08%.The haplogroups of R1-M173,N-M231,D1-M174,J-M304 and F-M89 were less than 13 cases,with frequencies ranging from 4.41%~0.34%.There were Y-STR genotypes differences among haplogroups,as haplogroup O-M175 was represented by 4 genotypes of Y-STR profiles characterized by DYS385a/b(12/12,as well as 12/17,12/18,12/19),DYS392(13),DYS593(16)and DYS393(12),and haplogroup C-M130 was characterized by DYS527a/b(19/20/21),DYS385a/b(11),DYS593(17),DYS390(23),Y_GATA_H4(11),and DYS444(13)and so on.Conclusion The DYS527a/b multi-allele pattern is frequently observed in the Kunming population with haplogroup C-M130.In the samples from haplogroups O,C,R1 and N,the DYS527a/b and DYF387S1a/b haplotypes frequently exhibit the multi-allele pattern.Given the frequencies of different haplogroups and the association between Y-SNP haplogroups and Y-STR loci,it could be helpful to look for more details in the paternal lineage search.
