PTEN is a candidate tumor suppressor which has sequence homology with dual-specificity phosphatase. PTEN is a multifunctional protein endowed with a phosphatase activity capable of dephosphorylating both tyrosine phosphate, serine/threonine phosphate residues on proteins and phospholipids of the phosphatidylinositol pathway. PTEN appears to be mutated at considerable frequency in several types of human tumors, including those from brain, breast, endometrium, and prostate. PTEN play an important role in pathogenesis of tumor, tumor cell invasion and metastasis. In this review, we will discuss the chemical structure of PTEN, its phosphatase activity, the ability of affecting signal transduction, and its mutational status in cancer.
2.The Effect of Sepia on the Activity of TPK,PKC and PKA in H_(22) Cancer Cells

The effect of Sepia on the activity of tyrosine protein kinase (TPK) protein kinase C (PKC) and cAMP-dependent protein kinase (PKA) was studied in H 22 cancer cells by using the incorporation of ? 32 P ATP into exogenous substract.The results showed that membranous TPK activity of membrane,cytosol and mitochondria was elevated significantly,while the membranous TPK activity was increased obviously;while the cytosolic PKC activity was increased most.The cytosolic PKA activity was increased.These results suggested that Sepia can changeover the activity of TPK,PKC and PKA,which was abnormally changed by cancer cells,then caused that the up regulation was diminished and the down regulation was enhanced towards the Ras MAPK signal transduction pathways.This inhibited or interrupted the cascade transduction of the Ras MAPK pathway,thus produced the effect of anticancer and accelerated the differentiation by transforming cancer cells from undifferentiated proliferation to differentiated proliferation.
3.Study of apoptotic thershold of cis-diamminedichloroplatinum and adriamycin on hepatocellular carcinoma

AIM:To study the thershold of hepatocellular carcinoma (HCC) apoptosis induced by adriamycin (ADM) and cis-diamminedichloroplatinum(CDDP). METHODS: Using primary human hepatocellular carcinoma culture, immunofluorescence staining of Hoechst 33 258 in HCC cells ,and flow cytometric assay. RESULTS:24 h after HCC cells were cultured with ADM or CDDP, it were found there were dispersive fluorescences in normal cells nuclei, and compact particulate fluorescences in apoptosis cells nuclei by immunofluorescence staiming of Hoechst 33 258. The rate of HCC cells apoptosis was dependent on doses of ADM and CDDP. HCC cells apoptotic threshold were determined, i.e.ADM was 1.0 ?g/mL and CDDP was 1.5?g/mL (The clinical apoptotic sensitive dosages were ADM 20 mg/m2 and CDDP 30 mg/m2. CONCLUSION: HCC cells apototic threshold of ADM and CDDP were efficient in clinical chemotherapy.
4.Expression of PTEN, p53, Ki-67 and its relationship with tumor histopathological grade and proliferation in astrocytoma

Objective To study the incidence of PTEN, p53 and Ki-67 expression in astrocytoma and show the relationship between PTEN, p53 expression and the proliferation activity. Methods The surgical specimens from 68 brain astrocytoma patients were analysed to detect PTEN, p53 and Ki-67 expression with immunohistochemical method. Results The incidence of PTEN, p53 and Ki-67 expression was 54.4 %,45.6 % and 48.5 % respectively in astrocytoma. With the grade of astrocytoma increasing the levels of PTEN protein decreased, on the other hand the levels of p53, Ki-67 increased. There was a negative correlation between PTEN expression and grade of astrocytoma while there was a positive correlation between p53, Ki-67 expression and grade of astrocytoma by using the Spearman Correlation test to analyse the data. The incidence of Ki-67 positive expression was 24.3 % in 37 cases exhibiting PTEN positive staining, whereas the incidence of Ki-67 positive expression was 77.4 % in 31 cases exhibiting PTEN negative staining. In statistics, there was an inverse correlation between PTEN and Ki-67 expression. Conclusion There is an inverse correlation between histopathological grades of astrocytoma and PTEN expression. A positive correlation is found between p53, Ki-67 expression and histopathological grades of astrocytoma. PTEN can inhibit tumor cell proliferation in astrocytoma.
5.Clinical evaluation of Decitabine on the treatment of elderly patients with acute myeloid leukemia

Objective To evaluate the clinical effect of Decitabine as the basis of the chemotherapy regimen on the treatment of elderly patients with acute myeloid leukemia( AML)?Methods The clinical data of elderly patients with AML admitted to the Third People ’ s Hospital of Yancheng from January 2013 to November 2015 were retrospective analyzed?The patients were divided into Decitabine group ( cases ) and the traditional group(19 cases) according to whether or not useed Decitabine,then evaluated the efficacy?Results The overall response rate( ORR) was 70?0% in 10 patients received decitabine,47?4% in 9 patents of the traditional group, and the difference was significant( P=0?03)?At the same time,the median survival time of the two groups was 44?87 months and 13?40 months respectively,the difference was statistically significant between the two groups (P=0?04)?There was no significant difference in the incidence of adverse reactions between the two groups ( granulocyte reduction, PLT reduction, fatigue, cardiovascular events, and respiratory tract infections ) ( P>0?05)?Conclusion The effect of Decitabine as the basis of the chemotherapy regimen on the treatment of AML is better,can prolong the survival time.
6.Change of ET-1 mRNA and ET AR mRNA expression in some organs in sepsis and septic shock rats

Objective: To investigate changes of the endothelin-1(ET-1) and its receptor (endothelin receptor subtype A, ET AR ) mRNA expression in some organs(kidney, lung and small intestinal mucous membrane) in the sepsis and septic shock rats. Methods: Twenty-four male rats randomized into sepsis group, septic shock group, control and normal group was infused with endotoxin(LPS) via indwelling right atria catheters except normal and control group. RT-PCR was used to detect kidney, lung and small intestinal mucous membrane tissue mRNA expression of the ET-1 , ET AR and glucose-6-phospho dehydrogenase(G-6-PD) in every group.Serum BUN, Cr, ALT and A were determined. The arterial oxygen tension (PaO 2) and arterial carbon dioxide partial pressure(PaCO 2)were measured in every group. Results: ET-1 mRNA and ET AR mRNA expression in the sepsis group and septic shock group were significantly higher than in normal group. There were significant differences between the normal/control group and sepsis/shock group in BUN,Cr,ALT,PaO 2 and PaCO 2. Conclusion: A higher expression of ET-1 mRNA and ET AR mRNA may be one of the startup factors on multiple organ dysfunction syndrome (MODS) and may play an important role on pathogenesis in sepsis and septic shock.
7.Whole cell screening of phage-display peptide library for mimicry peptides of glioma SWO-38

AIM and METHODS:The Ph.D.-7 phage display library was used to isolate peptides specific for glioma SWO-38 cell by whole cell screening.Moreover,binding efficiency analysis was carried out to test the binding specificity of the clones obtained. RESULTS: After three rounds of biopanning,a high concentration of phage clones was obtained and two of them were found to be highly specific to glioma SWO-38. CONCLUSION: Highly specific clones against neurtral glioma cells can be obtained from a phage display library by simple procedures.

Objective To study of zoledronic acid in the treatment of multiple myeloma bone dis-ease clinical effect and detection of serum macrophage inflammatory protein (MIP)changes of primary mye-loma (mm)in patients with serum macrophage inflammatory protein levels and multiple myeloma bone dis-ease curative effect.Methods 48 cases of multiple myeloma bone disease patients were treated with VTD regimen chemotherapy were randomly and equally divided into two groups,one group (group A)chemother-apy intermission applied zoledronic acid 4 mg per month 1 time,treatment 2 course of treatment,observa-tion of curative effect and adverse reaction,another group (B group)declined to azole phosphonic acid treatment.Results Group of pain Solution of 16 cases were markedly effective,effective in 4 cases,4 ca-ses were ineffective,efficiency 83.3%.B group bone pain relieved markedly effective in 12 cases,effective in 4 cases,8 cases were ineffective,have efficiency 66.7%.A compared to the B,the curative effect was obvious (P <0.05).By enzyme linked immunosorbent assay for the detection of the patients with a,levels of peripheral serum MIP-1a and MIP-1 beta B two groups before and after treatment.Conclusions zole-dronic acid in the treatment of multiple myeloma bone disease effectively,can significantly improve the qual-ity of life in patients with MM patients serum MIP-1a and MIP-1 beta level and multiple myeloma tumor bone disease curative effect is negative correlation,used for evaluating the effect The reference index.
9.Effects of Compound Kushen Tang on Ulcerative Colitis in Rats and the Underlying Mechanism

Objective: To investigate the therapeutic effects of compound Kushen Tang and its relevant mechanism in TNBS-in-duced ulcerative colitis ( UC) rats. Methods:UC was induced by TNBS in rats. After compound Kushen Tang was given orally, the levels of MDA, iNOS, and NO and the activity of MPO, SOD, and GSH-Px were measured. The general condition of rats and colon tissue morphology were observed. Results:The levels of MDA (P<0. 05), iNOS (P<0. 01) and NO (P<0. 01) and the activity of MPO (P<0. 01) in tissues of UC rats were significantly higher than the control group. The activity of SOD (P<0. 01) and GSH-Px (P<0. 05) were significantly lower than those in the control group. After the treatment with high doses of compound Kushen Tang, the levels of MPO (P<0. 01), MDA (P<0. 05), iNOS (P<0. 05) and NO (P<0. 01) were significantly decreased, and the activity of SOD (P<0. 01) and GSH-Px (P<0. 05) significantly increased. The therapeutic effect was dose-dependent and the general con-dition of rats and colon tissue morphology were also significantly improved. Conclusion:Compound Kushen Tang is considered as a no-vel therapeutic alternatives for the treatment of UC, which can reduce coloni inflammatory injury and ameliorate the colitis.
10. Clinical observation of priming regimen with pegylated recombinant human granulocyte colony-stimulating factor for treatment of initial treatment elderly patients with acute myeloid leukemia

Objective:
To evaluate the efficacy and side effects of priming regimen with pegylated recombinant human granulocyte colony-stimulating factor (PEG-rhG-CSF) in the treatment of initial treatment elderly patients with acute myeloid leukemia (AML).
Methods:
Thirty-five elderly patients with early-stage AML (non-M3) who received pre-excitation chemotherapy in Yancheng Third People's Hospital from February 2015 to January 2019 were retrospectively analyzed. According to the different granulocyte colony-stimulating factor (G-CSF) in the chemotherapy regimen, 15 cases were in PEG-rhG-CSF group, 6 mg PEG-rhG-CSF was used alone on day 0 by subcutaneous injection; 20 cases were in recombinant human granulocyte colony-stimulating factor (rhG-CSF) group, 200 μg/m2 rhG-CSF was used per day from day 0 to day 13 by subcutaneous injection, rhG-CSF was suspended or continued according to the number of white blood cells. In addition, both groups were given priming regimen with cytarabine and arubicin, or cytarabine and harringtonine. The efficacy and adverse reactions of the two groups were compared.
Results:
In the PEG-rhG-CSF group, there were 5 cases of complete remission, 6 cases of partial remission, 4 cases of non-remission, and 11 cases were effective. In the rhG-CSF group, there were 8 cases of complete remission, 7 cases of partial remission, 5 cases of non-remission, and 15 cases were effective.
