Objective To explore the relationship between the dynamic change of plasma tissue-type plasminogen activator(t-PA)and its inhibitor-1(PAI-1)levels and infarct size in patients with acute cerebral infarction. Methods There were 100 patients with acute cerebral infarction,22 cases of large area infarction,36 cases of small area infarction,42 cases of lacunar infarction. The levels of plasma t-PA or PAI-1 in 24 h,2 d,14 d,21 d after cerebral infarction were detected by chromogenic substrate method, compared with normal control and the level of blood plasma t-PA or PAI-1 in patients with different size of cerebral infarction. Results Compared with normal control, levels of plasma t-PA at 24 h,2 d,14 d after acute cerebral infarction decreased significantly, and plasma PAI-1 level increased significantly(all P0.05).The plasma t-PA level in patients with large size of infarction was lower obviously than that in patients with small area infarction and lacunar infarction, small size of infarction was lower than that in lacunar infarction(all P0.05).Conclusion The plasma t-PA level decreased and PAI level increased in patients with acute cerebral infarction. The the size of infarction area is larger, the plasma t-PA level is lower, but there is no relationship between the plasma PAI-1 level and infarction area.

To explore the effects of different organic solvents including trimethylglycine on the PCR amplification of G+C rich DNA templates, two high G+C containing DNA templates (cyclin dependent kinase inhibitor gene ARF and IGFR II) were amplified using standard PCR and modified PCR with the addition of glycerin, dimethylsulfoxide (DMSO), formamide and trimethylglycine respectively. There were no specific amplified products, when standard PCR and modified PCR with the addition of glycerin, DMSO and formamide were used to amplify the G+C rich ARF and IGFR II genes. But the two genes could be specifically amplified with the addition of optimal concentration of trimethylglycine in the PCR mixture. These results indicate that trimethylglycine can abolish or reduce the formation of second structures in G+C rich DNA regions and promote the amplification of high G+C containing DNA templates.

Objective To investigate the effects of Na +-dependent high-affinity dicarboxylate cotransporter 3(NaDC3) and glucose on the transport of succinate in human proximal tubular epithelial cells (HKC) . Methods Western blot was used to detect the expression of hNaDC3 protein in the pcDNA3-hNaDC3, pcDNA3-AhNaDC3 and pcDNA3, transfected HKC, and normal HKC cell. Analysis of succinate contents in the intaking-buffer at the various time points and in the variouse concentrations of glucose were determined by capillary zone electrophoresis. Results The cell transfected with pcDNA3- hNaDC3 was found to have over-expression of hNaDC3 protein for about 1.5 times that of the normal HKC, while succinate content in intaking-buffer was decreased. Although the cells transfected with pcDNA3-AhNaDC3 showed lower expression of hNaDC3 protein, being about 0.6 times that of normal HKC cells, the decrease of succinate in intaking buffer was not obviously effected. There were no differences in hNaDC3 protein content and succinate intaking between the cells transfected with pcDNA3 and normal HKC cells. With the increase in glucose concentration, the decrease of succinate in intaking-buffer was accelerated. Conlusion The results suggested that the overexpression of hNaDC3 protein can cause the succinate-intake more quickly in HKC.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2014;28(11):785-789
OBJECTIVE:
The purpose of this investigation was to observe the effects of the Acetaminophen on auditory in mice.
METHOD:
Regardless of male and female mice, healthy seven-year-old C57 mice were randomly divided into Acetaminophen low-dose group (150 mg/kg), Acetaminophen medium-dose group (300 mg/kg), Acetaminophen high-dose group(600 mg/kg) and control group. Then mice were tested for ABR to observe the changes of ABR's threshold in the zero, second, fourth and ninth day separately. The change of cochlea hair cells morphology was studied by immunofluorescent labeling. And HPLC detects the concentration of Acetaminophen in endolymph of mice cochlea.
RESULT:
After 30 minutes following administration, the Acetaminophen in endolymph of mice cochlear can be assayed by high performance liquid chromatography (HPLC). Acetaminophen increased the hearing thresholds compared to the control group. Hearing thresholds increased significantly in the Acetaminophen at 9 d,compared to the control group (P < 0.05). After administered medium-dose and high-dose of Acetaminophen, on the 9th day with the time of giving medicine to all the groups strengthens, the ABR thresholds heightens. And the average threshold is (44.75 +/- 16.00) dB, (50.00 +/- 11.00) dB respectively. Hair cells damages could be observed in experimental group after operations.
CONCLUSION
Acetaminophen can pass through blood-labyrinth barrier to the inner ear. These data demonstrate that taking certain acetaminophen can induce hearing impairment in mice.

Chinese Journal of Nosocomiology 2006;0(06):-
OBJECTIVE To investigate and control the risk factors about nosocomial infection in cerebral hemorrhage patients.METHODS Totally 986 hospitalized patients with cerebral hemorrhage in our hospital from Jan 2002 to Dec 2005 were studied retrospectively.RESULTS It showed that the incidence of nosocomial infection rate was 32.05%,case infection rate was 44.62%.The nosocomial respiratory tract infection was the highest(66.36%),the second one was the urinary tract(20.68%).We also found that the risk factors prone to nosocomial infection were aging, long hospitalization, respiratory machine application, respiratory tract inicison, indwelling catheter and coma.CONCLUSIONS Nosocomial infection is still a high frequent complication in cerebral hemorrhage. It is suggested that there be urgent need for treating the underlying disease and reducing the risk factors, then can reduce the development of infection in patients with cerebral hemorrhage.
