China Pharmacist 2015;(1):89-90,91
Objective:To observe the curative effect of local application of minocycline-HCl ointments and the influence on high sensitivity C reaction protein (hs-CRP) and interleukin-10 (IL-10) levels in gingival crevicular fluid of patients with chronic periodon-titis. Methods:Totally 72 cases of patients with chronic periodontitis (96 teeth) were selected and divided into the observation group (36 cases, 47 teeth) and the control group (36 cases, 49 teeth) by a random number table. The patients in the two groups were given the routine periodental non-surgical treatment, such as supragingival scaling, subgingival scaling, scaling and root planning etc. The patients in the observation group were given minocycline-HCl ointments filled in the periodontal pockets, and the patients in the control group were given compound iodine glycerol liquids filled in the periodontal pockets once a week for 4 weeks. The changes in hs-CRP and IL-10 levels in gingival crevicular fluid of the patients before and after the medical treatment were observed, and the clinical cura-tive effect was compared as well. Results:After the 4-week medical treatment, the hs-CRP levels in gingival crevicular fluid of the pa-tients in the two groups were obviously declined, while the IL-10 levels were obviously increased(P<0. 01 or P<0. 05), and the de-clining or increasing rate in the observation group was much higher than that in the control group (P<0. 05). The total clinical effi-ciency in the observation group was 94. 44%, which was much higher than that in the control group (77. 78%, P<0. 05). Conclu-sion:The local application of minocycline-HCl has favorable curative effect on chronic periodontitis, which can reduce the hs-CRP lev-els in gingival crevicular fluid, increase the IL-10 levels, control the inflammatory damage in periodontium effectively and improve the local inflammatory reaction in periodontium.

Objective To investigate the reasonable anticoagulant therapy for patients over sixty five years old in Yunnan-Guizhou plateau after heart biological valve replacement , and study the standard of reasonable anticoagulant therapy. Methods We collected the recent cases by retrospective way . Low-intensity anticoagulation standard INR (1.5~2.5) was used on 130 patients over sixty five years old. The warfarin dose and anticoagulation INR values in different surgery operation were compared when the patients reached the stable anticoagulation status. The frequency of INR values and warfarin dosage were studied. The differences between the patients with high risk factors and the ordinary patients without the high risk factors in INR values and warfarin dosage were investigated. Results The INR values and warfarin doses showed no significant difference (P > 0.05) in different operation groups when they reached stable anticoagulation status. The INR values was 1.94 ± 0.50 and the daily warfarin dose was (2.45 ± 0.82)mg/d. No significant differences in INR values and warfarin doses were found between the high-risk group and the general group (P>0.05) in steady-state. During follow-up, both the incidence rate of thrombosis and general bleeding were 0.77%. Conclusion The precent study showed that the low-intensity anticoagulation INR values (1.5~2.5) can be the standard of reasonable anticoagulation in Yunnan-Guizhou plateau after heart biological valve replacement with satisfactory anticoagulant effect.

Chinese Mental Health Journal 2018;32(3):252-258
Objective: To explore risk factors for cognitive impairment among elderly patients. Methods: There were 257 qualified subjects recruited, a case-control study was performed by collecting data of Mini Mental State Examination, carotid artery color Doppler flow imaging, biochemical tests, personal information on ages, medical histories, smoking as well as physical exercise among the recruited elderly in-patients/out-patients. A logistic regression was conducted to screen main effects of variables on occurrences of cognitive impairment. Results: Using logistic regression, the carotid intima-media thickness was found to be a significant risk factor (P<0.001) with larger regression coefficients (15.105 - 22.065) for the occurrence of cognitive impairment among elderly; about 71% -78% and 4% decrease in risks of dementia occurrence for every 1 pg/mL serum free triiodothyronine and lkg body weight increase. Conclusion: The increase in carotid intima-media thickness is a significant risk factor for the occurrence of cognitive impairment among elderly patients. The decrease in serum free triiodothyronine and body weight are the related factors to increase risk for dementia.

Objective To explore the association between death receptor (DR) 4 gene polymorphism and the predis-position of ulcerative colitis(UC). Methods DR4(rs20575)genotypes were determined by direct sequencing in a total of 147 UC patients and 244 healthy controls. Logistic regression analysis was employed to assess the relationship be-tween DR4 (rs20575)gene polymorphism and UC. Results Compared to the controls, the frequencies of variant allele(G)and genotype(CG+GG) of DR4(rs20575)were significantly increased in UC patients(5.78% vs 1.23%,P=0.001,OR=4.930,95%CI1.192-12.651;7.48% vs 2.46%,P=0.025,OR=3.208,95%CI1.161-8.869). According to the Tru-elove &Witt Activity Index, the severity of UC was further stratified into the mild, moderate and severe disease. In re-sult, variant allele (G)and genotype(CG+GG)in DR4(rs20575)were significantly higher in patients with moderate and severe UC than those with mild UC (10.64% vs 3.50%,P=0.020,OR=3.282,95%CI1.208-8.916; 14.89% vs 4.00%,P=0.028,OR=4.200,95%CI1.165-15.146). DR4(rs20575) mutant allele and genotype frequencies between extensive colitis and distal colitis were no statistical difference(P>0.05). Conclusion The genetic polymorphism of DR4(rs20575)is significantly related with the susceptibility to UC, as well as severity of the disease in this cohort of Chinese pa-tients.

Objective The aim of this study was to screen and verify the proteins interacting with Vimentin, investigate the regulatory relationship between FABP5 and candidate proteins, and further explore the mechanism of FABP5 in hepatocellular carcinoma. Methods Immunoprecipitation combined with tandem mass spectrometry(IP-MS)was used to screen the proteins that bind to FABP5.The binding relationship between FABP5 and candi-date interacting proteins was verified from the exogenous and endogenous levels by Co-immune precipitation assay(Co-IP).RT-qPCR,Western blot and immunofluorescence were used to observe the effect of knockdown FABP5 on the transcription and translation of Vimentin in HCC cells. The effect of overexpressing FABP5 on the cytoskeleton of HCC cell was observed by phalloidin staining. Results 336 potential target proteins that bind to FABP5 were identi-fied through IP-MS.Based on literature, five candidate proteins related to tumors were selected, namely PRDX1,PRSS3,PKM,HSP90AA1,and Vimentin. The binding relationship between FABP5 and Vimentin protein was con-firmed through both exogenous and endogenous Co-IP.Knockdown FABP5 has no significant effect on the expression of Vimentin mRNA,but it can inhibit the expression of Vimentin protein, and overexpression of FABP5 can affect the cytoskeleton of HCC cell. Conclusions FABP5 promotes the migration and invasion of HCC cells by the regula-tion of Vimentin and the influence of cytoskeletal remodeling, and thus it is expected to be a potential target for anti-HCC and provide new ideas for the treatment of HCC.

The pathogenesis of bipolar disorder(BD)is unknown, and objective biomarkers with clinical guidance are lacking.Extracellular vesicles(EV)are lipid bilayer-enclosed vesicles that carry cargo from originating cells, influencing the processes of recipient cells. They are capable of crossing the blood-brain barrier and reflecting the ongoing processes in the central nervous system.Compelling data indicates that EV could mediate BD pathophysiological processes such as neurocognitive impairment, neuroinflammatory diffusion, and metabolic dysfunction. Therefore,EV has the potential to become a reliable biomarker and therapy for BD,providing new ideas for revealing the pathogenesis and clinical diagnosis and treatment of BD.
