1.MicroRNA-181a-5p Curbs Osteogenic Differentiation and Bone Formation Partially Through Impairing Runx1-Dependent Inhibition of AIF-1 Transcription

Background:
Evidence has revealed the involvement of microRNAs (miRNAs) in modulating osteogenic differentiation, implying the promise of miRNA-based therapies for treating osteoporosis. This study investigated whether miR-181a-5p influences osteogenic differentiation and bone formation and aimed to establish the mechanisms in depth.
Methods:
Clinical serum samples were obtained from osteoporosis patients, and MC3T3-E1 cells were treated with osteogenic induction medium (OIM) to induce osteogenic differentiation. miR-181a-5p-, Runt-related transcription factor 1 (Runx1)-, and/or allograft inflammatory factor-1 (AIF-1)-associated oligonucleotides or vectors were transfected into MC3T3-E1 cells to explore their function in relation to the number of calcified nodules, alkaline phosphatase (ALP) staining and activity, expression levels of osteogenesis-related proteins, and apoptosis. Luciferase activity, RNA immunoprecipitation, and chromatin immunoprecipitation assays were employed to validate the binding relationship between miR-181a-5p and Runx1, and the transcriptional regulatory relationship between Runx1 and AIF-1. Ovariectomy (OVX)-induced mice were injected with a miR-181a-5p antagonist for in vivo verification.
Results:
miR-181a-5p was highly expressed in the serum of osteoporosis patients. OIM treatment decreased miR-181a-5p and AIF-1 expression, but promoted Runx1 expression in MC3T-E1 cells. Meanwhile, upregulated miR-181a-5p suppressed OIM-induced increases in calcified nodules, ALP content, and osteogenesis-related protein expression. Mechanically, miR-181a-5p targeted Runx1, which acted as a transcription factor to negatively modulate AIF-1 expression. Downregulated Runx1 suppressed the miR-181a-5p inhibitor-mediated promotion of osteogenic differentiation, and downregulated AIF-1 reversed the miR-181a-5p mimic-induced inhibition of osteogenic differentiation. Tail vein injection of a miR-181a-5p antagonist induced bone formation in OVX-induced osteoporotic mice.
Conclusion
In conclusion, miR-181a-5p affects osteogenic differentiation and bone formation partially via the modulation of the Runx1/AIF-1 axis.
2.Allergic diseases, immunoglobulin E, and autoimmune pancreatitis: a retrospective study of 22 patients

Background Autoimmune pancreatitis (ALP) is a chronic inflammatory disease of pancreas.We evaluated the clinical manifestations,imaging,and histological presentations of AlP in Chinese patients,and investigated the roles of immunoglobulin E (IgE) and allergic diseases in the diagnosis and pathogenesis of AIP.Methods The clinical records of 22 patients diagnosed with AlP were reviewed and analyzed.All patients with AlP fulfilled the 2006 revised diagnostic criteria proposed by Japan Pancreas Society or the Korean Criteria for AIP.Results Half (11/22) of AlP patients had allergic diseases.Twenty-one patients had elevated serum IgE levels,and 14 patients had IgE levels more than 3 times that of normal.There were no significant differences between the patients with higher or lower IgE,with or without allergic disease,in clinical features,laboratory tests,diffuse or focal lesions,or the choice of treatment methods; however,more complaints of body weight loss were observed in patients with higher IgE levels.Patients with higher IgE levels and with allergic diseases were more likely to have onset in March,April,May,August,September,or October.IgE levels decreased after therapy,but increased again during recurrence.Increased number of mast cells was found in the pancreatic tissue in AIP.Conclusions IgE maybe a useful marker for monitoring therapeutic response and recurrence of AlP.Allergic processes may play an important role in the pathogenesis of AIP.
3.Determination of the polysaccharide in Verdihong paint

Objective To establish the determination method of the polysaccharide in Verdihong paint .Methods The anhy-drous dextrose was used as the control , 0.2%anthrone-sulfuric acid as the stain , the content of polysaccharide was determined by ul-traviolet spectrophotometer at the wavelength of 620 nm.Results The polysaccharide curve was linear within a range of 1.9~34.2μg/ml, linear equation was Y=0.033 X-0.006 7 (r=0.999 9), the average recovery was 98.6% with RSD of 1.06%(n=6). Conclusion This method was convenient , stable and coud be used to assay content of the polysaccharide in Verdihong paint .
4.Advances in the research of predicting the time since deposition of evidentiary stains

In addition to obtaining DNA-STR typing of an evidentiary stain for individual identification and paternity tests, knowing the time since deposition (TSD) is also highly desired in forensics. To provide a reference for the research of predicting the TSD, this article reviews the reported optical, cell biological and molecular biological methods of determining the age of bloodstains domestic and overseas, and also introduces the application of microbial forensics, a new field of forensic science, to provide space-time clues of evidentiary stains.
5.Depression symtoms and related factors of fire fighters

Objective:To analysis the status of the mental health and related factors of fire fighters in Tianjin Binhai New District, and to provide suggestions for their psychological health protection.Methods:399 fire fighters in Tianjin Binhai New District were selected as study subjects in Jan to April 2019. Depression symptoms were measured by the depression module of the Patient Health Questionnaire (PHQ-9) . The Chinese version of Efrort Reword Imbalance (ERI) Questionnnaire were used to investigate and evaluate their occupational stress. Chi-Square test was used to analysis Categorical data. Binary logistic regression model was used to analysis the ralated factors of depression.Results:Among the 399 fire fighters, 71.1% (280/394) were found high level of depression symptom. The detection rates of depression symptoms in the related influceing factors ERI、station、disease、life pressure、eating habits and sleep disorder occupational stress were difierent ( P<0.05) . Sleep disorder, life pressure and ERI occupational stress were risk factors for depressive symptoms ( OR=1.921, 95% CI=1.002-3.682; OR=2.852, 95% CI=1.561-5.212; OR=2.367, 95% CI=1.163-4.818, P<0.05) . Conclusion:The rate of depression of fire fighters is relatively higher. Government should pay attention to and take measures to improve the psychological condition of fire fighters.

7.Adult-onset generalized autoimmune enteropathy involving small intestine and colon: report of a case and review of literature.

OBJECTIVETo investigate the clinicopathologic features of adult-onset autoimmune enteropathy (AIE).
METHODSA case of adult-onset AIE was described along with a literature review.
RESULTSA 41-year-old male patient was admitted for intractable diarrhea for more than three months despite of any dietary restriction or anti-inflammatory therapy. Fat globule was observed by stool examination and Sudan III staining of the stool was positive. Enteroclysis showed weak movement and few plica of small intestine, while colonoscopy appeared normal. Small bowel biopsies revealed villus atrophy and increased crypt apoptotic bodies and lymphocytic infiltration in deep crypt. Although without significant surface intro-epithelial lymphocytosis, there were a large number of monocytes, lymphocytes, plasmacytes and neutrophilic granulocytes infiltrating in the lamina propria. Morphologically, the colonic mucous was similar to the small intestine although cryptitis and crypt abscess were significant in the former. Serum IgG anti-goblet cell antibody was demonstrated by indirect immunofluorescence. Other causes of diarrhea were excluded on the base of medical history, histopathology and other accessory examinations before the diagnosis of AIE was made. The patient had a complete remission after steroid treatment without recurrence for eight months during the follow-up even after steroid withdrawal for five months.
CONCLUSIONSAIE is exceedingly rare and timely diagnosis is important for successful therapy. Histological differential diagnoses should include ulcerative colitis, celiac disease, lymphocytic colitis, etc. The final diagnosis should be based on histological examination combined with the patient history, clinical manifestation, endoscopy finding and serological testing.

8.Fetal STR typing and paternity identification of early pregnancy aborted tissue based on next-generation sequencing technology

Fetal STR typing of aborted tissue has long been a major problem in forensic DNA.Especially for the first trimester abortion tissue,it is difficult to isolate the embryonic components by histomorphological means,resulting in the inability to accurately obtain the STR typing of the fetus.The mixed STR typing results of mother and fetus can provide a key basis for the identification of suspects in cases of rape-induced pregnancy.In this study,next generation sequencing was used to successfully detect mixed STR typing of mother and suspected fetus or single STR typing of suspected fetus in 4 rape-induced early pregnancy abortion tissues.Combined with Y-STR and flank sequence information,it provides a more comprehensive and reliable genetic basis for the identification of suspects.
9.Study on Tissue Distribution of Apigenin Nanosuspension in Mice

OBJECTIVE:To study the distri bution and targeting characteristics of Apigenin nanosuspension (AP-NPs)in mice. METHODS:AP-NPs was prepared with ultrasound microprecipitation. Kunming mice were randomly divided into apigenin (AP) solution group and AP-NPs suspension group ,with 45 mice in each group. The mice were given relevant medicine intragastrically (80 mg/kg);blood sample of eyeball 500 μL were collected before medication(0 h)and 0.25,0.5,1,2,4,6,8,10 h after medication. After the last blood collection ,the mice were sacrificed and their heart ,liver,spleen,lung,kidney and brain tissues were taken. After protein precipitation with methanol ,HPLC method was adopted for determining plasma and tissues. The determination was performed on Shimadzu ODS-SP column with mobile phase consisted of methanol-water (70 ∶ 30,V/V)at the flow rate of 1.0 mL/min. The detection wavelength was set at 340 nm,and column temperature was 35 ℃. The sample size was 20 μL. The concentration of AP in different samples was calculated according to standard curve,main pharmacokinetic parameters (AUC,cmax)of AP and the ratio of peak concentration (ce),relative uptake rate (RUE),uptake ratio and its change value were calculated with DAS 2.0 software and Excel 2010 software;the tissue distribution and targeting characteristics of AP were analyzed. RESULTS:The linear range of AP in plasma and tissue s were 0.1-25.0 μg/mL(all r>0.99);the lower limits of quantification were 0.1 μg/mL. RSDs of intra-day and inter-day were all lower than 15%,and the accuracy were 94.37%-117.48%. The extraction recovery rates were all more than 80%. Compared with AP solution group ,the concentrations of AP in plasma sample (during 0.5-6 h),liver tissue (during 0.25-8 h),spleen tissue (during 0.25-8 h)and cerebral tissue (during 0.25-4 h)were increased significantly in AP-NPs suspension group (P<0.05 or P<0.01),and the highest in liver tissue. The concentrations of AP in heart tiusse (6 h),liver tissue (10 h),lung tissue (0.5 h),spleen tissue (during 0.25-10 h)were decreased significantly (P< 0.05 or P<0.01). There was statistical significance in AUC and cmax of AP in plasma and tissue samples between 2 groups(P< 0.05). The ce,RUE,uptake ratio and its change value of liver tissue were the highest ,being 1.34±0.40,1.99±0.29,48.49% and 15.71% . CONCLUSIONS :After AP is made into nanosus- pension,the distribution of drug tissue is changed ,especially targeting effect on liver tissue is improved.
