1.Positive inotropic effect of methyl polyglycoside on isolated guinea pig atria

AIM To study the positive inotropic effect of methyl polyglycoside (Mpg) on isolated guinea pig atria and its mechanism. METHODS The effects of Mpg on contractility, beat rate of right atria, post-rest contraction and positive staircase phenomenon were observed in isolated guinea pig left and right atria. Na+-K+-ATPase activity of rat myocardial cell membrane was measured. RESULTS Mpg (0.01~3 mmol*L-1) produced a concentration-dependent increase in myocardial contractility of atria and decrease in beat rate of right atria. Mpg markedly enhanced the post-rest contraction and the positive staircase phenomenon induced by increasing stimulation frequency in left atria, and inhibited Na+-K+-ATPase activity of rat myocardial cell membrane. CONCLUSION The results suggest that Mpg can strengthen myocardial contractility of atria and decrease beat rate of right atria. Its positive inotropic effect may be related to inhibiting Na+-K+-ATPase activity of myocardial cell membrane, increasing the transmembrane influx of calcium and the amount of calcium released from intracellular stores.
2.Preparation and characterization of anti-hBLyS monoclonal antibody by DNA immunization

AIM: To establish the monoclonal antibody against human B lymphocyte stimulator (hBLyS) by DNA immunization and analyse its characterization. METHODS: The 858 bp DNA fragment of hBLyS was cloned into pcDNA3 plasmids. The cloned insert was identified by both sequence analysis and double digestion of the recombinant plasmid with restriction enzymes Xho I and EcoR I . After the splenocytes from BALB/c mice immunized with the recombinant plasmid of pcDNA3/hBLyS were fused with myeloma cells SP2/0,the hybridoma which can produce monoclonal antibodies against hBLyS were obtained. The specificity of anti-BLyS monoclonal antibody from hybridoma was verified by ELISA, Western blot and flow cytometry. RESULTS: The recombinant mammalian cell expression vector of pcDNA3/hBLyS was constructed, the sequence of the insert gene was identified to be the sequence encoding hBLy S antigen. The culture supernatants of hybridoma 9c10 were tested to be the monoclonal antibody with specificity against hBLyS on human peripheral blood CD3 + T cell activated by hIFN-? by ELISA, Western blot and flow cytometry. CONCLUSION: The monoclonal antibodies against hBLyS with high activity and specificity have been established successfully, and will be an useful tool in the studies of relationship between hBLyS and human autoimmunity diseases.

5.The effect of comprehensive nursing intervention in children with diabetic ketoacidosis

Objective To evaluate the effect of comprehensive nursing intervention in children with diabetic ketoaci-dosis. Methods A total of 24 cases of children with diabetic ketoacidosis were randomly divided into two groups, with 12 cases of comprehensive nursing intervention in the observation group and 12 cases of routine nursing in the control group. Hospitalization stay, hospitalization costs and glycemic control after hospitalization of the two groups were com-pared. Results Hospitalization stay and hospitalization costs of the observation group were less than those of the control group (P<0.05). Glycemic control in the observation group were better than that of the control group (P<0.05). Conclu-sion The comprehensive nursing intervention has some effects in children with diabetic ketoacidosis, and has shorter hospitalization stay, less cost of hospitalization and better glycemic control. It is worthy of promotion in nursing.
6.Incidence of abdominal obesity among primary and middle school students and its association with parental weight related knowledge, attitude and practice

Objective:
To investigate the association of parental weight related knowledge, attitude and practice (KAP) with abdominal obesity among primary and middle school students and to provide evidence for related intervention measures.
Methods:
In September 2013, the multistage random sampling method was used to select 17 307 primary and secondary school students with normal waist circumference in seven provinces/municipalities in China. After nine month follow up, 14 118 students aged 6-18 years were included in the final analysis. Questionnaires were used to assess parental weight related KAP, and students’ waist circumference was measured objectively at baseline and follow up. Multiple linear regression and multivariate Logistic regression were conducted to analyze the relationship between parents’ KAP with the change of students waist circumference and abdominal obesity, respectively.
Results:
The nine month cumulative incidence of pre and abdominal obesity among students was 6.67% and 0.85%, respectively. The scoring rates of parents knowledge, attitude and practice were 80.17%, 78.17%, 50.50%, respectively. The score of parents weight related attitude was inversely associated with the changes of waist circumference ( β=-0.18, 95%CI = -0.26- -0.10) and cumulative incidence of pre and abdominal obesity ( OR=0.90, 95%CI =0.82-0.98)( P <0.05).
Conclusion
Parental positive and correct weight related attitude is inversely associated with the incidence of abdominal obesity among children and adolescents. Interventions targeting establishing parents correct beliefs may be an effective way to prevent abdominal obesity in children and adolescents.
7. Research progress of monoclonal antibodies in pharmacokinetic characteristics, analysis methods and method validation

In recent years, monoclonal antibodies (mAbs) have been developing rapidly and widely used in fields of tumor, immunity, blood and other systemic diseases. Global share of mAbs in prescription drug market had reached up to $ 140 billion, 15.3% by 2019. As macromolecule proteins, with special structures and physiological properties, mAbs have great differences in absorption, distribution, metabolism and excretion in vivo compared with small molecule drugs, including characteristics of relatively large molecular mass, high specificity and selectivity in target combination, non-linear pharmacokinetics, time dependence, long half-life and so on. Fully understanding of these special pharmacokinetic characteristics shall effectively guide analysis of mAbs. Meanwhile, the particularity and complexity of disposal mechanism in organisms greatly increase the difficulty of biological detection. As a result, it is necessary to establish exclusive, sensitive, accurate and repeatable quantification methods in biological samples. This article focuses on pharmacokinetic characteristics, main analytical methods, and methodology validation of quantification for mAbs in biological samples, expounding with comparisons of small molecular drugs, so as to prompt development of pharmacokinetic study of this kind of drugs.
8.Microneedles in diagnosis and treatment: a review.

Microneedles have been developed rapidly in the field of transdermal administration in the past few decades. In recent years, the development of microelectronics technology has expanded the applications of microneedles by combining with microelectronic systems, especially in biological diagnosis and treatment. Different types of microneedles have been designed to extract blood and tissue fluids for detection, or as electrodes to directly detect blood sugar, melanoma and pH in real-time in vivo, both show good prospects for real-time detection applications. In this paper, we review the design of materials and structure of microelectronic-based microneedles, and discuss their advances in biological diagnosis.
