1.ULTRASTRUCTURAL CHANGES OF HUMAN CHORIONIC VILLI AFTER EXPOSURE TO DIAGNOSTIC ULTRASOUND

Chorionic villi of 6-9 week normal pregnant women were obtained twenty-four hours after exposure to diagnostic ultrasound and observed under transmission electron microscope. Trophoblastic syncytium and trophoblastic cell showed remarkable ultrastructural changes which included:① cellular membrane lost its integrity, ② cytoplasm escaped into the interstitial arca, ③ endoplasmic reticulum dilated and ④ mitochondria swelled, cirstane reduced with small vesticle and irregular morphology.
2.Roles of histone acetyltransferases and deacetylases in the process of cell proliferation,differentiation and tumorogenesis

Acetylation of lysine residues of core histone N-terminal domains has been found correlatively associated with transcriptional activation in eukaryotes. Recent studies showed that some transcriptional regulators with histone acetyltransferase(HAT) and deacetylase(HDAC) activities may play a causative role in regulating gene expression. The link between tumorigenic processes and misregulated or mistargeted HAT and HDAC activities has been reported.
3.Study of Permeation Enhancers of Shuxiong Cataplasm

Objective To investigate the effect of two kinds of permeation enhancers,azone and clove bud oil,on transdermal permeation of Shuxiong cataplasm. Methods Cataplasm containing azone and clove bud oil,at different concentrations were prepared,with ferulic acid as the index,the role of enhancing permeation of azone and clove bud oil for the main active component were studied by in vitro permeation experiments. Results Both azone and clove bud oil at different concentrations could increase permeation. Clove bud oil was better than azone and the best concentration of Clove bud oil is 3%. Meanwhile,the permeation enhancing multiple of clove bud oil group was the highest which was 3.68. Conclusion With the cumulative permeation quantity of ferulic acid as the index,3% clove bud oil is more suitable to act as a permeation enhancer of Shuxiong cataplasm.
4.The association of 5-HTTLPR with depression and its clinical symptoms

Objective To investigate the association of the polymorphism of serotonin transporter gene linked polymorphic(5-HTTLPR) region with depression and its clinical symptoms in the Han Chinese.Methods PCR-based technique was used to genotype 5-HTTLPR in 64 patients with major depression and 67 unrelated controls,and the association between the polymorphism and clinical symptoms of depression was explored according to HAMD score.Results Fewer patients tended to be of L/L genotype(9.4%) than controls(19.4%),but there was no significant difference.Among women,fewer patients tended to be of L/L genotype(2.9%) than controls(13.9%),but there was no significant difference.The symptom analysis showed that depressive symptoms were significantly associated with 5-HTTLPR genotypes or alleles in the patients of L/L genotype,and that L allele had significantly lower depressive scores compared with those of other genotypes or alleles(P
5.A Dietary Investigation of pupils in Mei County in Shaanxi

We carried out a dietary investigation of pupils aged 10 - 11 in Mei county in Shaanxi with level-based community random sample method. The result showedthat the average intake of daily food was mainly grain. That of energy was less than 70% of RDA and 60% of WHO. That of the protein was less than 70% of RDA in male and 60% of RDA in female. That of the other nutrients was less than 60% of RDA except iron, nicotinic acid and thiamine. Energy allocation among three mels was not adequate in male pupils. There waas higher energy in breakfast and supper, not sufficent at lunch.

Objective To explore methylprednisolone and conventional dose prednisone treatment for hematological damage in systemic lupus erythematosus(SLE) in the near future response. Methods Hemocytopenia in 147 patients with SLE were treated by intravenous injecting methylprednisolone and conventional dose prednisone and therapy response were observed in the tenth day after treatment. BACKGROUND: Evidence exists that inhibition of matrix metanoproteinase-2(MMP-2) secretion in the proliferating hernangioma tissue by transfection of adenovirus-active MMP-2(Ad-aMMP-2) cDNA would become an important means for treatment of proliferating hemangioma.OBJECTIVE: To investigate the influences of Ad-aMMP-2 cDNA transfection on human proliferating hemangioma growth in nude mice.DESIGN, TIME AND SETTING: A randomized, grouping, and controlled observation was performed in West China Hospital of Sichuan University between August 2003 and September 2004.MATERIALS: Eighteen BALB/c-nu/nu nude mice, weighing approximately 20 g, were included. Cavernous hemangioma specimen pathologically confirmed as proliferating hemangioma was resected from one 52-day-old female child patient.METHODS: The freshly reseoted human proliferating hemangioma specimen was sliced into small pieces with a size of 5 mm×4 mm×3 mm and subcutaneously implanted into the back of 18 nude mice within 1 hour to develop mouse models of hemangioma.Forty-five days after hemangioma implantation, 15 successful hemangioma nude mice were treated by intratumoral administration of adenovirus green fluorescent protein (Ad-GFP1 n = 51 Ad-GFP group), adenovirus-active MMP-2 (n = 5, Ad-aMMP-2 group), or the same amount of phosphate buffered saline (PBS1 n = 51 control group). Intratumoral administration was performed once every other day, for a total of 4 times.MAIN OUTCOME MEASURES: Observation of tumor volume and compadson of tumor necrosis area among 3 groups; detection of GFP expression in nude mouse; gross, hematoxylin-eosin staining, and transmission etectron microscope observation of tumor tissue morphology; determination of MMP-2 cDNA expression and microvascular density by immunohistochemistry; and detection of growth cycle and apoptosis of tumor cells by flow cytometry.RESULTS:①Ad-aMMP-2 could inhibit hemangioma growth in vivo, without marked adverse reactions. Tumor necrosis of different degrees was found in each group, and tumor necrosis area was significantly greater in the Ad-aMMP-2 group than in the control and Ad-GFP groups (P < 0.01). ②Histological sections displayed GFP gene expression in the Ad-GFP group. ③Gross observation results revealed relatively large tumor tissue in the control and Ad-GFP groups and relatively small tumor tissue in the Ad-aMMP-2 group. Hernatoxylin-eosin staining results showed that in the control and Ad-GFP groups, endothelial cells aggregated together in strip-shaped or lump-shaped appearance, and in the Ad-aMMP-2 group, there were many necrotic loci arranging in lamellar-shape appearance. Transmission electron microscope results revealed vascular endothelial cells with normal morphology in the control group and tumor cells with apparent nucleoli in the Ad-GFP group, while in the Ad-aMMP-2 group, some vascular endothelial cells exhibited chromatin pycnosis in the nucleus, forming apoptotic bodies.④ MMP-2 expression and microvascular density were significantly reduced in the Ad-aMMP-2 group than in the Ad-GFP and control groups (P < 0.05). ⑤The percentage of tumor cells in G0/G1 phase was significantly higher (P < 0.05), while the proliferating index was significantly decreased, in the Ad-aMMP-2 group than in the Ad-GFP and control groups. The Ad-aMMP-2 group exhibited higher apoptosis rate of tumor cells (P < 0.05), as well as more markedly increasing apoptosis index, than the control and Ad-GFP groups.CONCLUSION: It is feasible to block human proliferating hemangioma growth by transfeotion of Ad-aMMP-2 cDNA. The included mechanisms are to inhibit vascular endothelial cells to secrete MMP-21 thereby leading to local ischemia.
10.Highly Sensitive Fluorescent Aptasensor for Thrombin Detection Based on Competition Triggered Rolling Circle Amplification

Based on the competition reaction of target protein, aptamer probe, padlock probe and complementary sequence, a highly sensitive fluorescent aptasensor was developed in this study in combination with rolling circle amplification. In the absence of target protein, the ligation-rolling circle amplification reaction was repressed because the complementary sequence hybridized with aptamer probe to form double-stranded duplex. While in the presence of target protein, the target molecules bound specifically with aptamer probe, inducing displacement of the complementary sequence and hybridization with padlock probe. The padlock probe was circularized with the assistance of E. coli DNA ligase, and the rolling circle amplification process could be accomplished by Phi 29 DNA polymerase. The amplification product contained thousands of repeated sequences which could hybridize with the loop of molecular beacon ( the detection probes) , resulting in a significant fluorescence signal. The effects of length of complementary DNA ( CDNA ) sequence and concentration of padlock probe were investigated. Under the optimized experimental conditions, the model target protein thrombin could be highly sensitively detected by the proposed aptasensing system in a linear range of 0 . 067-32 . 4 nmol/L with a detection limit of 0 . 03 nmol/L ( approximately 90 amol target molecules). Moreover, the presented sensing method was universal for other target analysis by skillfully design of the sequence of aptamer probe and related oligonucleotides.
