1.Optimization of expression and purification conditions of recombinant Hq001 protein in salivary glands of Haemaphysalis qinghaiensis

Objective To optimize the expression conditions(expression and induction conditions)and purification methods(non-denaturing and denaturing purification)of recombinant Hq001 protein in salivary glands of Haemaphysalis qinghaiensis.Methods The recombinant plasmid pET-30a-Hq001 was transformed into competent cells E.coil BL21(DE3),E.coil Rosetta(DE3)and E.coil ArcticExpress(DE3)pRARE2 respectively for the selection of an optimal expression strain.The final concentration of IPTG(0,0.5,1.0 mmol/L),induction temperature(20,25 ℃)and induction time(0,2,4,6,8 h)were optimized.The recombinant bacteria expressed under the ideal induction condition were homogenized by French press and the target protein was purified by passing through a Ni-NTA affinity chromatography column under either native(denaturationrenaturation-column chromatography)or denatured conditions(denaturation-column chromatography-renaturation).The purified products were analyzed by 12% SDS-PAGE.Results E.coil BL21(DE3)was proved to be the most suitable strain for the expression of recombinant Hq001 protein.The optimum induction condition was induction with 0.5 mmol/L IPTG for 4 h at 25 ℃.The target protein with a relative molecular mass of approximately 18 800 was obtained by non-denaturing purification method,and the size was consistent with the expectation.Conclusion The recombinant protein rHq001 in salivary glands of Haemaphysalis qinghaiensis can be obtained by the optimized expression conditions and purification methods.
2.Experience in the Management of the Facilities and Equipment of Pharmacy Intravenous Admixture Ser-vice in Our Hospital

OBJECTIVE:To provide a reference for the management of the facilities and equipment of the pharmacy intrave-nous admixture service(PIVAS). METHODS:The experience in the management of the facilities and equipment of PIVAS in our hospital was introduced in respect of system establishment,archives management and the maintenance of important equipment,and the performance of core purification equipment was tested to evaluate management effect. RESULTS & CONCLUSIONS:The PIV-AS in our hospital established a practical system by developing and improving 7 management systems,9 sets of operational proce-dures and 5 contingency plans,increased the awareness of archives management by perfecting 27 archive files and standardized and conducted the maintenance of key equipment. In 2014 and 2015,after implementing management,the passing rates of all test items of the purification equipment were 100%. It is suggested that facilities management should focus on active preparation of an intelli-gent monitoring system,the improvement of the information system and the training of professional management personnel.
3.Study on Genetic Toxicity of Detergent to Root Tip Cells of Vicia faba

Objective To study the genetic toxicity of 3 brands of detergents to the root tip cells of Vicia faba in the inducing time and the inducing way(continual mutagenesis,accumulative mutagenesis) with micronucleus and the chromosomal aberration Methods The root tip cells of Vicia faba were taken as the subjects,the genetic safety tests were conducted on those materials treated with detergent of a certain concentration(0.25%) for 2、4、6 h of continual mutagenesis and accumulative mutagenesis respectively.Distilled water was used as the control.Results The continual mutagenesis showed that all the three detergents could induce the root tip cells of Vicia faba to produce micronucleus and chromosomal aberration and as the mutagenic time prolonged,at 6 h,the changes became weak.The genetic toxicity in the accumulative mutagenesis group was greater than that in the continual mutagenesis group.Conclusion According to the results of the present paper,to some extent,detergents may produce genetic toxicity.
4.Indoor Environment and Childhood Asthma in Ningxia:a Case-Control Study

Objective To know the risk factors of indoor environment associated with childhood asthmas. Methods Using 1∶1 matched case-control study design,140 cases and 140 age-sex-matched controls from the Affiliated Hospital of Ningxia Medical University were investigated for the indoor environment risk factors,from July 2004 to May 2008. Conditional Logistic regression model was used for data analysis. 5.The Views of Cancer Information Communication in Different Groups of People

Objective: To study and compare the views on cancer associated information and communication needs of cancer patients among cancer patients, their relatives, doctors and nurses, and other groups of people. Methods: A total of 501 cancer patients, 169 relatives,86 doctors and nurses, 176 healthy elderly people and 505 first grade college stu- dents were asked to complete the Information Preference Questionnaire of Cancer Patients and 2 questions regarding gen- eral information amount and content (IPQCP+2), and the Information Need Questionnaire (INQ). The elderly and the stu- dents were asked to complete two sets of IPQCP+2 and INQ in two roles: one as themselves and the other as patients. All data were analyzed in detail. Results: The views of different groups are significantly different. The differences include the viewpoints on whether to disclose the diagnosis of cancer and to inform the life expectancy to cancer patients, and whether the patients should participate in decisions about their medical care and treatments. The mean score of IPQCP on the over- all items and the 4 dimensions of all groups, and their opinions on general information amount and content for patients are significantly different. Conclusion: The views of different groups of people on cancer associated information and commu- nication needs of cancer patients are different. The views and requests of cancer patients themselves are the basis of doc- tor- patient communication. The prevention of cancer should be based on the characteristics of different groups of people and the patients’views should be considered.
6.Information Needs of Cancer Patients (4) Satisfaction with Doctor-patient Communication and Associated Factors

Objectives:To study the satisfaction with doctor-patient communication among cancer patients, their relatives and medical staffs, and its associated factors.Methods:A total of 423 cancer patients were asked to rate their satisfaction with information communication, to complete the Information Need Questionnaire (INQ), the Demographic Questionnaire (DQ), and 4 questions regarding general information amount and content the patients got and wanted respectively. 82 relatives, 86 medical staffs were also asked to rate their satisfaction with information communication.Results:A certain proportion of cancer patients, their relatives and medical staffs were not very satisfied with their information communication. There are significant differences among them. The satisfaction of medical staffs was less. It suggests that they want to improve their communication level. Satisfaction with information communication of cancer patients was affected by the contents and styles of information communication, and whether their communication experiences met their preferences.Conclusions:Before providing information to cancer patients, we need to consider the style they want and to find out their needs and preferences.
7.Development and Evaluation of the Psychological Adjustment Scale for Cancer Patients

Objectives: To develop and evaluate the Psychological Adjustment Scale for Cancer Patients (PASCP).Methods: A self-report rating scale was developed with structured methods. A total of 557 cancer patients were asked to complete the scale and evaluate the importance of each item. And 183 relatives and 96 doctors and nurses were also asked to evaluate the importance of each item. The scale was refined and its reliability, validity and responsiveness were evaluated. Results: (1)PASCP was finally refined as a scale with 36-item, 5 dimensions including the impact on emotion/self-esteem, self-perception, daily life, relationship with others/social life, others, and an item to value the general psychological adjustment after the patients were diagnosed with cancer. (2)The scale had good content validity. Its construct was consistent with the concept designed. The factor loading spectrum and ANOVA are 0.63-0.79 and 7.44, 0.67-0.76 and 5.42, 0.69-0.88 and 4.77, 0.66-0.91 and 4.77, 0.63-0.80 and 4.38 respectively. The criterion validity with HAD and FLIC were 0.78 and 0.69 respectively. (3)The test-retest coefficients for the 5 dimensions and the total score were 0.83、0.82、0.86、0.78、0.76 and 0.89 respectively;Cronbach ? for them were 0.83, 0.86, 0.80, 0.78, 0.69 and 0.89 respectively. The split-half reliability for the scale was 0.93. 4.The scale could reflect the changes of the patients' psychological adjustment before and after treatment and recovery. Conclusion:PASCP has high validity, reliability and responsiveness. It is applicable in clinical practice in China.
8.Diagnosis and surgical treatment of solid pseudopapillary tumor of the pancreas

Objective To evaluate the diagnosis and therapy of solid pseudopapillary tumor of the pancreas (SPTP).Methods From January 2006 to October 2011,the clinicopathological data of 24 SPTP patients diagnosed and surgically treated at Tianjin Medical University Cancer Hospital were analyzed retrospectively.Results There were 22 women and 2 men,the median age was 27.5 years old,with a range from 12 to 51 years.The most common tumor localization were at the head of the pancreas ( 11 patients,45.7% ),followed by the body (5 patients,20.8% ),the tail (4 patients,16.7% ),the body and the tail (3 patients,12.5% ) and the neck (one patient,4.2% ).Clinical symptoms were nonspecific and the preoperative diagnosis of SPTP depended on imaging study such as ultrasonography,CT and MR.All patients were treated surgically and the correct diagnosis was made according to pathology after surgery.In a follow-up ranging from 2 to 55 months( median 27.2 months),23 cases were alive with no evidence of disease recurrence,one patient was alive with tumor recurrence.Conclusions SPTP was a less common neoplasm with low-grade malignancy,occurring mainly in young women.Complete tumor resection results in satisfactory prognosis.
9.Significance and expression of S100A9 and NMP238 in cervical carcinoma tissues with different concurrent chemoradiotherapy sensitivities

Objective To determine the significance and expression of S100A9 and NMP238 in cervical carcinoma with different concurrent chemoradiotherapy sensitivities. Methods Fresh carcinoma tissues were collected from untreated cervical carcinoma patients and preserved at -80 ℃. The tissues were classified into 2 groups:a high sensitivity group (HS) and a low sensitivity group(LS) according to their response to concurrent chemoradiotherapy. Protein was separated by 2-dimensional gel electrophoresis (2-DE). Peptide mass fingerprintings (PMF) were acquired by matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and the proteins were identified by data searching in the Mascot-database. Differential expressed proteins were assayed by Western blot and immunohistochemistry.Results Most of the gels were clear and were successfully and reproductively analyzed. Intensity and rate of S100A9 expression were higher in the HS group than in the LS group,and those of NMP238 expression were higher in the LS group than in the HS group. Conclusion S100A9 and NMP238 expression is associated with concurrent chemoradiotherapy sensitivity in cervical carcinoma.
10.Analysis of risk factors for venous thrombosis associated with the placement of peripherally inserted central catheters in patients with malignant tumors during the course of chemotherapy

Objective To investigate the induced-factors levels of venous thrombosis associated with the placement of peripherally inserted central catheters (PICC) in patients with malignant tumor,and establish intervention model accordingly. Methods 40 patients complicated with venous thrombosis after PICC were set as the thrombosis group, 40 patients without venous thrombosis were selected as the no-thrombosis group, 30 healthy cases were selected as the control group. All cases' serum samples were ob-tained to measure the levels of molecular biomarkers of the coagulation system, platelet parameters and molecular biomarkers of the fibrinolysis system. The results of the three groups were assessed. Results Patients in the thrombosis group were reported significantly higher levels of vWF, GMP140,TAT than those in both two other groups, but lower levels of ATⅢ. Higher levels of MPV and PCT were found in the throm-bosis group than those in both two other groups.Tthe data also showed higher PLG and PAI levels and lower levels of PLM in the thrombosis group. Conclusions From this study it shows that patients with malig-nant tumors during the placement of PICC have evident hypercoagnlahility, high- activated platelet and se-vere pre- thrombosis state caused by PICC. In treatment of malignant tumor patients with PICC, the level of coag-ulation system molecular markers, platelet parameters,fibrinolysis molecular markers shoud be used as a clinical routine and as the index of early intervention and evaluation of the PICC.
