1.The expressions of endothelin-1 and endothelin A/B receptors mRNA in tissue of benign prostatic hyperplasia treated by daily low-dose sildenafil

Objective To observe the mRNA expressions of endothelin-1(ET-1) and endothelin A/B receptors (ETA/B) in tissue of benign prostatic hyperplasia treated by daily low-dose sildenafil.Methods A total of 32 patients with benign prostatic hyperplasia were randomly divided into two groups:treatment(25 mg sildenafi for 12 weeks,n=16) and control (no drug,n=16) groups.Immunohistochemical staining,ELISA and RT-PCR were used to detect the expression levels of ET-1 protein and ET A/B mRNA,respectively.Results The expressions of ET-1 protein and ET A/B mRNA in prostatic tissue were significantly lower in treatment group than in control group[(53.31±18.56) ng/kg vs.(83.34±31.38) ng/kg,0.356±0.056 vs.0.624±0.083,0.721±0.083 vs.0.933±0.905,t=-3.295,10.715,6.937,all P<0.001].Conclusions Daily low-dose sildenafil can reduce the expressions of ET-1 and ET A/B receptors mRNA in benign prostatic hyperplasia.
2.Research Progress of the Relationship between the Expression of TAR DNA-bind-ing Domain Protein 43 and Brain Injury

T A R D N A-binding dom ain protein 43 (T D P-43) is a highly conserved and w idely expressed nuclear protein. N ow adays, the expression of T D P-43 can be found in m ost neurodegenerative diseases such as A lzheim er's disease, w hich m akes it becom e a neurodegenerative disease associated m arker pro-tein. From the current research status at hom eland and abroad, and around the relationship betw een the expression of T D P-43 and brain injury, this article em phatically probes into the specific expression and function of T D P-43 in acute and chronic brain injury based on the know ledge of its biological charac-teristics, w hich aim s to explore the feasibility for determ ining the cause of death and the injury and dis-ability situations by T D P-43 in forensic pathology.
3.Differences in gut microbiota among primary school students with different levels of sugar sweetened beverage consumption

Objective:
To explore the differences in the gut microbiota of primary school students with different levels of sugar sweetened beverage intake, so as to provide scientific evidence for better identification of health risks in children and the development of targeted health policies.
Methods:
In June 2022, a total of 192 healthy primary school students from Chengdu were selected using a stratified cluster random sampling method. The sugar sweetened beverage intake was assessed through a dietary frequency questionnaire. Based on the median daily sugar sweetened beverage intake, primary school students were categorized into a low intake group ( n =96) and a high intake group ( n =96). The gut microbiota in fresh fecal samples from the two groups of primary school students was analyzed using 16S rRNA high throughput sequencing, and the diversity and community structure differences in the gut microbiota were compared.
Results:
Children in the low intake group had a sugar sweetened beverage intake of (21.3±1.6) mL/d, while the high intake group had an intake of (269.6±37.3) mL/d. Diversity analysis results showed that there were no statistically significant differences between the low intake and the high intake group in terms of α diversity metrics: Observed_otus index [298.50 (259.75, 342.25), 305.50 (244.25, 367.75)], Goods_coverage index [1.00 (1.00, 1.00), 1.00 (1.00, 1.00)], Chao index [304.18 (260.75, 348.78), 305.88 (245.68, 370.88)], Shannon index [5.88 (5.29, 6.45), 5.71 (4.89, 6.28)] and Simpson index [0.95 (0.91, 0.97), 0.94 (0.88, 0.97)] ( Z =-0.64, -0.76, -0.54, -1.76, -1.67, P >0.05). Furthermore, no statistically significant difference was observed in β diversity between the two groups ( R 2=0.006, P >0.05). At the genus level, the abundance of Blautia [0.033 (0.018, 0.055)] and Fusicatenibacter [0.009 (0.005, 0.015)] were higher in the low intake group compared to the high intake group [0.024 (0.013, 0.041),0.006 (0.003, 0.011)]and differences were statistically significant ( Z =-2.52, -2.81, P <0.05). LEfSe analysis highlighted intergroup differences primarily in Blautia, Fusicatenibacter and Sarcina( LDA= 3.56,3.12,3.53, P <0.05).
Conclusions
There is no significant difference in the diversity and overall structure of the gut microbiota in primary school students with different levels of sugar sweetened beverage intake. However, there are species variations at the genus level. The information can serve as a scientific basis for identifying health risks in primary school students and formulating targeted health strategies.
4.Development of the modified medication regimen complexity index of Chinese version(mMRCI-C)and its validation in elderly diabetic patients

Objective:To develop the modified medication regimen complexity index of Chinese version (mMRCI-C)and test its reliability and validity.Method:The Chinese version of MRCI was developed by modification,translation and back translation. The MRCI was interculturally adapted by 2-rounds of expert consultation and pilot study to ensure the semantics, content and conceptual equivalence. The validation of the mMRCI-C scale was tested among 420 community-dwelling elderly patients with type 2 diabetes mellitus(T2DM) in Shanghai Changfeng Community Health Service Center from October to December 2020. SPSS 23.0 was used to analyze the reliability and validity of the scale.Results:The mMRCI-C scale included 3 dimensions, namely drug dosage form (14 entries), medication frequency (5 entries), and additional instructions (6 entries), with a total of 25 entries. Among 420 valid questionnaires collected,the respondents were 212 males (50.4%) and 208 females (49.6%) with a mean age of (71.4±8.1) years. The test-retest reliability was 0.999 and internal consistency reliability was 0.849. The content validity exceeded 0.80,the convergent validity was 0.932; and discriminant validity P<0.001. Conclusion:The preliminary testing results show that the reliability and validity of the mMRCI-C scale are satisfactory.
5.Efficacy of aripiprazole combined with olanzapine for hospitalized male patients with schizophrenia and its effect on metabolic syndrome

BackgroundSchizophrenia and the use of antipsychotic medications are identified to be the likely contributors to the development of metabolic syndrome (MS) and cardiovascular disease, and jeopardize the prognosis of schizophrenia. Therefore, effectively preventing or reducing the risk of developing MS in patients with schizophrenia is critical. ObjectiveTo explore the efficacy of aripiprazole combined with olanzapine for male schizophrenia patients and its effect on MS, so as to provide a certain reference for the selection of antipsychotic drugs for schizophrenia patients. MethodsMale patients (n=80) who were hospitalized in The Third People's Hospital of Meizhou from February to June 2023 and fulfilling the International Classification of Diseases, tenth edition (ICD-10) diagnostic criteria for the schizophrenia were enrolled, and grouped using random number table method, each with 40 cases. Study group was treated with aripiprazole combined with olanzapine, while control group was given aripiprazole monotherapy. The treatment lasted for 6 continuous weeks in both groups. At the baseline, Positive and Negative Symptom Scale (PANSS) score, MS-related indices [fasting plasma glucose (FPG), hemoglobin A1c (HbA1c), body mass index (BMI), waist-to-hip ratio (WHR), lipid profile], S100 calcium-binding protein B (S100B) and high sensitivity C-reactive protein (hs-CRP) were recorded. Then the PANSS scores at the end of the 2nd, 4th and 6th week of treatment, the Clinical Global Impression (CGI) scores at the end of the 2nd and 6th week of treatment, as well as the MS-related indices, S100B, hs-CRP, Treatment Emergent Symptom Scale (TESS) score and Rating Scale for Extrapyramidal Side Effects (RSESE) score at the end of the 6th week of treatment were recorded in all participants. ResultsAnalysis on PANSS score revealed a significant group effect, time effect and group×time interaction effect (F=18.092, 634.780, 2.917, P<0.05 or 0.01). Analysis on CGI score revealed a significant group effect and time effect (F=20.492, 99.190, P<0.01). At the end of the 6th week of treatment, study group detected lower serum concentrations of HbA1c and triglyceride (TG) compared with control group (t=-3.495, -3.293, P<0.05). The post-treatment hs-CRP level was lower in study group than that in control group (t=-3.916, P<0.05). Study group scored lower on TESS compared with control group (t=-4.684, P<0.01). ConclusionAripiprazole combined with olanzapine can effectively alleviate psychotic states in male schizophrenia patients, and the combination therapy yields less impact on MS-related indices than olanzapine monotherapy. 6.Construction of tissue engineered cell sheet.

Scaffold-free tissue engineered cell sheet is an emerging technology in biomedical field. It can avoid the adverse effects of scaffold materials, and can be further assembled to form more complex three-dimensional functional tissues. The construction of cell sheet is mainly based on the culture substrate composed of sensitive materials. By changing the stimulation factors such as temperature, enzyme, light, ion, redox, pH and sugar, the adhesion behavior of the substrate to the cells could be changed to make the cells detach naturally, thus generating the cell sheet. Recent years have seen the development of various simple and efficient construction technologies of cell sheet due to the development of a variety of novel sensitive culture substrates. The resulted cell sheets with excellent performance have greatly expanded their applications. This review summarized the construction methods of tissue engineered cell sheet and discussed the challenges and future perspectives in this field.
