Objective To observe the effect of comprehensive community intervention on cardiovascular risk factors in metabolic syndrome (MS) high risk group.Methods One hundred MS high risk patients were divided into intervention group (50 patients) and control group (50 patients) by random digits table.The intervention group accepted intervention on health education,health action,health habit and individual intervention on drug.The control group accepted natural intervention.The changes of waistcircumference,body mass index (BMI),blood pressure,fasting plasma glucose (FPG),lipids and serum uric acid (UA) were observed.Results In intervention group,the number of physical exercise,alimentary control and medication compliance increased 12%,16% and 18%,and the number of smoking and alcohol drinking decreased 8% and 6%.But in control group,the number of physical exercise,alimentary control,medication compliance increased 2%,0 and 4%,and the number of smoking and alcohol drinking decreased 4% and 2%.There was significant difference (P< 0.05).Before treatment,the level of waist circumference,BMI,systolic pressure,diastolic pressure,FPG,total cholesterol,triacylglycerol,high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C) and UA between two groups had no significant difference (P > 0.05).After 1 year's intervention,except HDL-C,the level of other index had significant difference between two groups (P < 0.05 or < 0.01).Conclusion Comprehensive community intervention can improve the status of obesity and dyslipidemia,and reduce blood pressure,plasma glucose and UA.

With the development of mass spectrometer,protein and peptide separation technology,and data analysis tools,the study on protein expression profiling of various tissues,organs or cells has been developed rapidly.The most basic purpose of proteomics is qualitative and quantitative identification of the whole protein in a cell or tissue.Therefore,how to make full use of mass spectrometry data has an important significance in proteome.Recently years,proteomics has begun to be applied in allograft rejection and discovery of rejection related biomarkers in organ transplantation.Through the extensive application of the technology in organ transplantation,graft rejection can be earlier detection and treatment.Therefore,the proteomics research have potential application prospects in the diagnosis of the organ transplant rejection.
4.Diagnostic value of a new high-resolution microendoscopy for early esophageal cancer

Objective To investigate the therapeutic value of high-resolution microendoscopy for early esophageal cancer.Methods A total of 60 patients,who underwent esophageal endoscopy,were included in this study.The therapeutic value of high-resolution microendoscopy was investigated with reference to pathological results as golden standard.Results Diagnostic sensitivity,specificity,positive predictive value,negative predictive value and accuracy of microendoscopy for high grade dysplasia/early cancer were 81.22%,94.12%,81.82%,94.12%,91.11%,respectively.Diagnosis of severe dysplasia/carcinoma with high-resolution microendoscopy was highly consistent with that of pathology (K =0.760).Conclusion The high-resolution microendoscopy is of high diagnostic value for early esophagus cancer.
5.Study on human immunodeficiency virus-1 super-infection among high-risk populations in Myanmar

Objective To investigate the occurrence and the characteristics of human immunodeficiency virus (HIV)-1 co/super-infection among high-risk populations in Myanmar.Methods Forty-six HIV-1 positive plasma in Myanmar were collected. Possible cases with HIV-1 co/super infection were identified by discordant sequence results obtained with different polymerase chain reaction (PCR)/sequencing primers or by ambiguous readings in direct sequencing. HIV-1 quasispecies in plasma were then characterized by clonal sequence analysis of independent PCR-clones generated by TA cloning method. Thereafter, their phylogeny and recombinant structure were investigated. Results Co/super infection was identified in 3 (6.5 %) cases among the 46 screened HIV-1 positive patients.All of these three patients were heterosexuals and were co/super infected with CRF01_AE/subtype B′recombinants. Conclusions HIV-1 co/super infections are relatively common and provide a prerequisite for rapid generation of new recombinant forms in Myanmar.
6.Expression and clinical significance of E3 ubiquitin ligase Iduna in non-small-cell lung cancer

Objective To investigate the mRNA and protein expression of E3 ubiquitin ligase Iduna in non-small-cell lung cancer tissue and para-neoplastic lung tissue,and the correlation of the Iduna expression with clinicopathological factors and prognosis.Methods The expression levels of the Iduna mRNA and protein in non-small-cell lung cancer tissue and para-neoplastic lung tissue were determined by reverse transcriptase-polymerase chain reaction (RT-PCR),Western-Blot and immunohistochemistry respectively,and the correlation of the Iduna expression with clinicopathological factors and prognosis was analyzed.Results RT-PCR and Western-Blot showed the expression levels of the Iduna mRNA and protein in non-small-cell lung cancer tissue (0.468 ± 0.086 and 2.554 ± 0.544) were significantly higher than those in para-neoplastic lung tissue (0.203 ± 0.070 and 1.570 ± 0.316),there were statistical differences (P < 0.05).Immunohistochemistry results showed that Iduna was negative expression in the alveolar epithelium cells,negative or weak positive expression in normal bronchial and positive expression in different degrees in the non-small-cell lung cancer tissue.Iduna high expression rate was negative correlation with tumor differentiation (P =0.002),Iduna high expression rate was positive correlation with large tumor size (P =0.044),TNM staging (P=0.015) and lymph node metastasis (P=0.009).Iduna high expression of I stage non-small-cell lung cancer patients was correlated with poor post-operative survival (P =0.016).Conclusions High expression of Iduna may be related to the process of invasion and metastasis of nonsmall-cell lung cancer.It is possible that Iduna serve as potential markers for predicting prognosis in nonsmall-cell lung cancer.
7.Establishment of a EV71 virus infection model of tree shrew primary renal cells

Objective To establish an enterovirus 71(EV71) infection model of tree shrew primary renal cells.Methods Tree shrew primary renal cells were obtained by trypsin digestion.After subculture and purification,EV71 virus was used to infect these primary cells.The culture supernatant of these EV71-infected cells was collected for virus titer detection at 1,2,4,6 and 8 days post-infection.The cells were collected for detection of EV71 VP1 protein by Western blot assay.Furthermore,the expression and location of VP1 protein in the infected cells were detected by indirect immunofluorescence assay.Vero cells were taken as positive control to evaluate the infectivity of EV71 virus to tree shrew primary renal cells.Results Morphologically,the cultured cells were proved to be majorly consisted of the primary renal cells after subculture and purification.The obtained primary cells were infected by EV71 virus.The virus titer was up to 1.3×106 TCID 50/mL during 48-96 h post-infection,proving that EV71 virus infected and proliferated in the tree shrew primary renal cells.Western blot showed that the viral VP1 protein was detected from infected primary cells at 2 to 8 d post infection.VP1 protein was also observed in the cytoplasm at 2 to 6 d post infection by indirect immunofluorescence.Compared with Vero cells,the infectivity of EV71 virus to tree shrew primary renal cells and its proliferation were confirmed.Conclusions Based on the successful establishment of cell culture of tree shrew primary renal cells,the infectivity to the obtained cells and proliferation of EV71 virus in the cells are confirmed.The model of EV71 virus-infected tree shrew primary renal cells is initially established.
8.Heparanase Expression Correlates with Angiogenesis and Lymphangiogenesis in Human Lung Cancer

Background and objective Heparanase has been thought to be a good molecular marker of tumor, and the heparanase expression level was correlated closely with tumor metastasis. In this study, we investigate the effects of heparanase on angiogenesis and lymphangiogenesis of lung cancer and the relationship between heparanase expression and vascular endothelial growth factor (VEGF), vascular endothelial growth factor-C (VEGF-C).Methods Immunohistochemistry was used to detect the expression of heparanase, VEGF, VEGF-C protein and microvascular density (MVD), lymphatic vessel density (LVD) in 115 cases of non-small cell lung cancer (NSCLC) and 45 cases of adjacent normal tissue samples.Results Our results showed that heparanase expression was significantly increased in 91 (79.13%) of the 115 cases and correlated with lymph node metastasis (node positive rate 87.0%; node negative rate 36.8%; P=0.003). Heparanase positive expression cases have significantly higher concentration of microvascular density (MVD) and lymphatic vessd density (LVD) as compared with heparanase negative expression cases (P<0.01, P<0.01, respectively), heparanase expression was significantly correlated with VEGF, VEGF-C expression in NSCLC.Conclusion Heparanase overexpression was associated with angiogenesis and lymphangiogenesis of lung cancer, targeting ofheparanase may represent a significant therapeutic potential for lung cancer.
9.The expression and mechanism of PTEN and COX-2 in myeloid leukemia cells

Objective To investigate expression and regulatory mechanism of phosphatase and tensin hemology deleted on chromosome ten gene (PTEN) and cyclooxygenase-2 (COX-2) in human myeloid leukemia cells.Methods Thirty patients was collected from the First Hospital of Baoding and Second Affiliated Hospital of Hebei Medical University,including 10 chroni myeloid leukemiac (CML)patients in chronic phase (CML-CP),10 CML patients in blast crises (CML-BC) and 10 normal controls.The recombinated adenovirus containing green fluorescent protein (GFP) and PTEN ( Ad-PTEN-GFP) or empty vector (Ad-GFP) was transfected into human CML K562 cells.The growth of K562 cells and cell adhesion ability was observed by 3-[4,5-dimethyl-2-thiazolyl]-2,5-dip (MTF) assay; PTEN and COX-2 messenger ribonucleic acid (mRNA) levels were detected by real-time fluorescent relative-quantification reverse transcriptional PCR (FQ-PCR).PTEN and p-Akt protein levels were detected by western blot,and COX-2 protein were measured with cytochemical staining.ResultsThe mRNA expression levels of PTEN in CML-BC patients (0.022 ±0.021 ) were lower than CML-CP patients (1.134 ± 1.124) and normal control ( 1.059 ±0.595).The mRNA expression levels of COX-2 in CML-BC patients (0.761 ±0.418) were higher than CML-CP (0.211 ± 0.158) and normal control (0.165 ± 0.152).The growth of K562 cells was suppressed markedly,and the maximum growth inhibition rate was 38.67% ± 4.30% after transfected with PTEN gene,which was higher than Ad-GFP group (5.34% ±0.31%,t =13.39,P <0.01 ).COX-2 mRNA expression levels in Ad-PTEN-GFP(0.013 ± 0.001 )were significantly lower than Ad-GFP group (0.199 ±0.018) and untransfected group (0.217 ± 0.021,F =499.45,P < 0.01 ),and p-Akt as well as COX-2protein were also down-regulated after K562 cells transfected ( MOI =200) with wild type PTEN in three days.ConclusionPTEN may inhibit proliferation and adhesion ability of leukemia cell in myeloid leukemia via down-regulating COX-2 expression.
10.Relationship between metastasis-associated protein 1 expression and metastasis and prognosis in non-small cell lung cancer.

BACKGROUNDMetastasis is the main cause of the death of lung cancer patients. Much attention has been pain to the research of lung cancer metastasis. Metastasis-associated protein 1 (MTA1) is one member of metastasis associated protein family. Its overexpression is correlated with metastasis of esophageal carcinoma and breast cancer, but the generality of its expression in cancer and the significance for judging biological behaviors of tumor and evaluating prognosis of patients is to be investigated. The aim of the study is to study the relationship between MTA1 expression and clinicopathological factor regarding metastasis and prognosis of human non-small cell lung cancer (NSCLC).
METHODSThe expression of MTA1 was detected in 101 parafin-embedded specimens by immunohistochemistry method, as well as in 35 freshly-taken NSCLC tissues by Western blot.
RESULTSThere were 56 cases (55.4%) of NSCLC with yellow or even brown particles in nucleus of tumor cell among 101 cases, and MTA1 protein showed negative expression in epithelia of bronchi or alveoli in neighboring noncancerous tissue. Western blot analysis showed the level of MTA1 in NSCLC tissues was remarkably higher than that in normal tissues (t=3.953, P=0.000). Expression of MTA1 was remarkably higher in tumor with metastasis than that in tumor without metastasis (t=4.057, P=0.000). Expression of MTA1 significantly correlated with differentiation (Chi-square=10.131, P=0.006), lymphatic metastasis (Chi-square=8.535, P=0.003) and p-TNM stage (Chi-square=17.419, P=0.000). The survival time of pa-tients with negative MTA1 expression was (44.866±12.946) months, which was significantly higher than that of patients with positive MTA1 expression [(23.714±7.498) months] (Chi-square=10.006, P=0.002). In multivariate analysis, only lymphatic metastasis and TNM stage could be considered as independent prognostic factors.
CONCLUSIONSMTA1 might play an important role in the development and metastasis of NSCLC. Patients with MTA1 expression have a greater chance of metastasis and a poorer prognosis. However, MTA1 expression is not an independent prognosis factor.
