1.Identification of gene expression patterns in rat model of nonalcoholic fatty liver disease

Objective To compare and analyze gene expression patterns in rat model of nonalcoholic fatty liver disease (NAFLD).Methods Twelve male Sprague-Dawley rats were randomly given either general diet (control group) or a high-fat diet (model group) for 4 weeks.The histopathologic changes of the liver were observed and gene expression patterns were analyzed and compared by cDNA mieroarray.Results Hepatocellular steatosis and inflammatory infiltration were observed in model group after high-fat diet for 4 weeks.Fifty-one differential genes were found in model group,20 of which were up-regulated (sterol regulatory element binding factor 1,stearoyl-coenzyme A desaturase 1 and Bcl 2 modifying factor)and 31 were down-regulated (peroxisomal enoyl coenzyme A hydratase 1,heat shock 70 protein 1A and ATPase inhibitory factor 1). The up-regulating genes were involved in lipogenesis and cell apoptosis,while down-regulated genes were involved in fatty acid oxidation, protein modification and energy metabolism.Conclusions The differential expression of genes may involve in the pathogenesis of NAFLD.
2.Expression and significance of VEGF-D and MMP-2 in pancreatic carcinoma

Objective To investigate the expression of VEGF-D and MMP-2 and explore their relationship with tumor invasion and local metastasis in pancreatic carcinoma.Methods The expression of VEGF-D and MMP-2 in 30 patients with pancreatic carcinomas and 6 cases with normal pancreatic tissues were detected by SABC immunohistochemical method.Results The positive rates of VEGF-D and MMP-2 proteins were 60%(18/30)and 66.7%(20/30)respectively,no expression of VEGF-D and MMP-2 was detected in normal pancreatie tissues.The expression of VEGF-D was significantly correlated with lymph node metastasis(P<0.05),and the expression of MMP-2 was significantly correlated with lymph node metastasis and tumor local invasion(P<0.05).The expression of VEGF-D protein Was positively correlated with MMP-2 protein(rs=0.54,P<0.01).Conclusions There is a significantly positive correlation between high expression of VEGF-D and MMP-2 proteins in pancreatic carcinoma,and they may contribute to the invasive properties of pancreatic cancer.
3.The role of lumbar drainage in symptomatic cerebral vasospasm after aneurysmal subarachnoid hemorrhage

Objective To investigate the risk factors of symptomatic vasospasm after aneurysmal subarachnoid hemorrhage and evaluate the effect of lumbar drainage of cerebrospinal fluid on vasospasm.Methods In this retrospective controlled-cohort study, 175 patients with aneurysmal subarachnoid hemorrhage met our study criteria between January 2012 and December 2013.By multi-factor regression analysis, gender, age, Hunt-Hess grade, modified Fisher grade and lumbar drainage were analyzed.The outcomes were assessed by the presence or absence of symptomatic cerebral vasospasm and vasospasm-related infarction, and the mean days of hospital stay and score of Glasgow Outcome Scale at 1-month follow-up.Results Several factors affected the prognosis of subarachnoid hemorrhage.Lumbar drainage was a protective factor of symptomatic cerebral vasospasm (OR =0.243, 95% CI: 0.119-0.497) and vasospasm-related infarction (OR =0.305, 95% CI: 0.154-0.604).The patients with lumbar drainage had higher score of Glasgow Outcome Scale (P < 0.05).But the patients with lumbar drainage had longer hospital stay (P < 0.05).Conclusions Lumbar drainage of cerebrospinal fluid after aneurysmal subarachnoid hemorrhage markedly reduced the risk of symptomatic cerebral vasospasm and improved outcome.
4.Experimental study of kansui root and LMWH combined therapy for severe acute pancreatitis

Objective To investigate if there is a synergism of kansui root and low molecular weight heparin(LMWH) in combined treatment of SAP in rats.Methods SD rats were randomly divided into 5 groups:Sham group(A group),severe acute pancreatitis group(B group),kansui root treatment group(C group),LMWH treatment group(D group),combined treatment group(E group).The serum TNF-?,IL-6 level and the ratio of TXB2/6-keto-PGF1? in pancreatic tissue,and the histological examination were evaluated.Results The values of TNF-??IL-6 and the ratio of TXB2/6-keto-PGF1? in C,D,E group were significantly lower than those in B group at the time point of 6,12,24h.But there were no significant differences in C,D,E group at all the time points.The histological examination showed that the pancreas was normal in group A;the pathological changes were milder in C,D,E group than that in B group.Conclusions Combined treatment of kansui root and LMWH was effective in SAP,but there was no synergism,and also no side effects.
5.Establishment of Quality Standard for Zhike Qutan Oral liquid

Objective: To establish the quality standard for Zhike Qutan oral liquid.Methods: A TLC method was used for the qualitative identification of Platycodonis Radix and Glycyrrhizae Radix et Rhizoma;the content of belamcandin in Belamcandae Rhizoma was determined by an HPLC method on a Waters Symmetry C 18 column(150 mm× 4.6 mm , 3.5 μm)with the mobile phase consisting of acetonitrile-water (14∶86), the flow rate was 1.0 ml·min-1 , the column temperature was 35℃ and the detection wavelength was 265 nm.Results: The TLC spots were clear without interference from the negative control.The linear range of belamcandin was 0.115-2.880 μg (r=0.999 9),and the average recovery was 92.44% (RSD=1.83% , n =6).Conclusion: The method is simple and rapid with good reproducibility, which can be used for the quality control of Zhike Qutan oral liquid.
6.Peripheral blood CircRNA expression profile analysis in patients with rheumatoid arthritis

Objective To screen the peripheral blood circRNA differently expressed in patients with rheumatoid arthritis (RA) and to explore the pathogenetic role of peripheral blood cicRNA in RA by analyzing the data with bioinformatics.Methods The study was performed in 3 RA cases and 3 healthy controls,using circRNA microarrays to screen the circRNA in peripheral blood of patients with RA.The data were normalized and analyzed by R soft package,screening by fold change and P value and searching the differently expressed circRNAs between the two samples by t test.Bioinformatics was performed to analyze the differently expressed circRNAs.Results The results from circRNA microarrays revealed that 36 circRNAs were significantly al-tered in RA patients (P <0.05) compared with the control group.Among them,22 were significantly up-regulated,and the other 14 were down-regulated.The GO analysis of the genes involved in the circRNA showed that these genes participated in the progress of biological regulation,cell differentiation,and metabolism.We predicted the target miRNA of all the differently expressed circRNAs,among the results there was a miRNA (hsa-miR-125a-3p) targeted by a circRNA(hsa_circ_0005397) experimentally confirmed by other studies.The relationships among circRNA-miRNA-Gene were predicted by Cytoscape software.Conclusion There are many differently expressed cireRNAs in peripheral blood of patients with RA,and the circRNAs maybe involved in the regulatory mechanisms of Rheumatoid Arthritis.
7.AQP4 regulation for cerebral edema.

Water balance is one of the basic regulation mechanisms of homeostasis. There are 13 subtypes of aquaporins in mammals (AQP0-AQP12). In neural system, the AQP4 is mainly distributed in astrocytes. Phosphorylation and expression regulation of AQP4 is involved in the formation of brain edema, particularly in the clearance of vasogenic edema and the formation of cytotoxic edema. This article reviews regulations and functions of AQP4 in vasogenic edema and cytotoxic edema.

8.Gonadotropin-inhibitory hormone in regulation of reproduction and behavior in mammalians.

RF-amide related peptide (RFRP) is the orthologue of gonadotropin-inhibitory hormone (GnIH) in mammals. The bodies of RFRP cell are located in the dorsomedial nucleus of the hypothalamus (DMH) and the fibers project to preoptic area (POA) and median eminence of the hypothalamus. Its receptor mainly distributes in hypothalamus. RFRP fibers project to GnRH cells to regulate mammalian reproduction axis. This paper reviews the progress of current researches on RFRP in regulation of animal behaviors, including reproduction, food intake, anxiety and stress response.

9.Early and non- early surgery for ruptured intracranial aneurysms: analysis of 184 cases

Objective To compare the therapeutic efficacy between emergency and non-emergency operation for ruptured intracranial aneurysms. Method A retrospective analysis of 184 patients with ruptured intracranial aneurysms the Second Affiliated Hospital Zhejiang University College of Medicine, admitted from Dec 2008 to Sep 2009, was carried out to evaluate the efficacy of operation to be done earlier. The patients were divided into 2groups according to the time of surgery. In the early operation group ( n = 102), the patients were operated on within 3 days of rupture of aneurysms, and in the delayed operation group ( n = 82), the patients were operated on after 3 days. The comparison in the rate of rebleeding before surgery, rate of complete occlusion of the ruptured aneurysm and rate of major complications such as cerebral infarction and hydrocephalus between two groups was made. The Glasgow outcome scale (GOS) scores of these patients were also evaluated by 6- 12 months follow-up after operation. Results Preoperative re-bleeding happened in 2 patients of the early operation group and in 7 patients of the delayed operation group. The rates were significantly different ( P < 0.05). The complete occlusion rate in the early operation group was 91.2 % ( 93/102 ), while was 80.5 % ( 66/82 ) in the delayed operation group (P<0.05). There were no statistically significant differences in post-operative cerebral infarction rate, post-operative hydrocephalus rate or GOS scores on follow-up between two groups. Conchusions Early operation can significantly reduce the re-bleeding before surgery, reducing the risk of death and disability. In early operation, the continuous lumbar drainage by cannulation and other methods can be used to reduce intracranial pressure, significantly increasing the rate of complete occlusion, and promoting rehabilitation.
10.Microvascular decompression for hemifacial spasm induced by vertebral artery dissecting aneurysm: one case report.

A 61-year-old female presented with 4 years history of left-sided hemifacial spasm. Head MRI and angiography indicated left vertebral artery dissecting aneurysm which compressed ipsilateral cranial nerves Ⅶ and Ⅷ. Microvascular decompression was performed. The dissecting aneurysm was pushed apart and the distal part of the parent artery was adhered to the dura on the petrosum. The compressed nerves were totally decompressed. The symptom of facial spasm was completely resolved immediately after surgery and did not recur during 6 months of follow up.
