1.Successful pregnancy derived from slowly freezing donated human oocytes by intracytoplasmic sperm injection

Objective To set up a novel clinical method for storage of oocytes by slowly freezing and successful pregnancy by intracytoplasmic sperm injection (ICSI). Methods The frozen oocytes (n=33) in metaphase Ⅱ (28),which were normal in morphology,were denuded of their cumulus-corona complex.The protective freezing solution contained 1,2-propanediol (1.5 mol/L) and sucrose (0.35 mol/L) and the process was slow for freezing and rapid for thawing.The recipients of these oocytes received hormone replacement therapy (HRT). Results A high survival rate of oocytes (32/33) was obtained when the sucrose concentration was 0.35 mol/L.ICSI induced a higher fertility rate (23/28),a good embryo cleavage rate (21/23) and a satisfactory embryo morphology for thawed oocytes.Embryo transfers were performed in 6 cycles/6 cases. Four cases got clinical pregnancy by demonstration of four gestation sacs on B ultrasonic examination 7 weeks after ICSI,1 fetal was first trimester spontaneous abortion,another 3 survival fetal were ongoing pregnancy and the first case gestation week is 17 weeks. Conclusions This study is the first successful application of human oocyte cryopreservation in China and this method can increase the survival rate of freezing oocytes.It’s easy to carry out,low in cost and high in the recovery rate of oocytes after thawing.
2.Experimental Study on the Effect of Fuzhengqingluo(FZQL) Capsule Combined with CTX Against S180 Tu-mor

OBJECTIVE: To observe the antitumor effect of FZQL combined with CTX on S180 tumor and the effect of FZQL on reducing CTX s toxieity .METHODS: The models of S180 tumor bering mice were established .The animals were divided into CTX therapy group, CTX and FZQL combined therapy group, FZQL therapy group and control group.The results were compared among these groups.RESULTS: The weight of tumor in FZQL therapy group was less than that in control group.All of the experimental markers in combined iherapy group were better than those in CTX therapy group.CONCLUSION: FZQL has a marked inhibiting effect on tumor(S180), and it can increase the CTX's inhibiting effect on tumor and reduce the side effect of CTX and increasing the body immunity.
3.Safety and efficacy of very low protein diet in the treatment of patients with severe chronic renal insufficiency

Objective To investigated the effects of long term use of very low protein diet(VLPD,0 3 g?kg-1?d-1) treatment on patients with chronic renal insufficiency without essential amino acids(EAAs) or related ketoacids supplement.Methods Thirty seven patients with established severe chronic renal failure (CRF)[Scr(588 2?123 5)?mol/L, Ccr(9 77?3 48)ml?min-1?(1 73m2)-1]were divided into 2 groups according to their actual protein intake: 20 patients with protein intake of (0 33?0 04)g?kg-1?d-1 were used as VLPD group, while other 17 CRF patients whose protein intake was 0 6 g?kg-1?d-1 were served as low protein diet group (LPD). Results All patients in VLPD group showed good compliance to this very low protein diet,and no one presented signs of protein malnutrition during the observation. The concentrations of serum albumin and transferrin were maintained in normal ranges during the follow up period despite the transferrin levels in both groups gradually decreased as time went on. AIM: To investigate the influence of dialysate on the cell morphology and the peritoneal membrane function in chronic peritoneal dialysis rats. METHODS: 40 SD rats were divided randomly into 4 groups. Except control group, other groups daily received infusion of 20 mL dialysate (4.25% dialysate(HG), 1.50% dialysate(LG), Riger's solution(RG)) respectively for 8 weeks. The peritoneal membrane function was investigated by peritoneal transport test, and the rat peritoneal mesothelial cells(PMCs)morphology was analyzed by the cell imprints. RESULTS: The intraperitoneal volume and net ultrafiltration in HG and LG groups were significantly lower, with D/P_(urea) significantly higher, and C_(urea) after 4 h of dialysis significantly lower than that in RG and control groups. The density of cell population from analysis of cell imprints in HG and LG groups was significantly lower, but the mean surface area were significantly larger than that in RG and control groups. These change had no difference between HG and LG group. 5.Effect of MIF antisense oligonucleotides on expression of MIF in macrophages

AIM: To investigate the effect of antisense oligonucleotides on expression of macrophage migration inhibitory factor (MIF) in macrophages. METHODS: MIF phosphorothioate oligonucleotides was designed and synthesized. The phosphorothioate antisense, sense and missense oligonucleotides of mouse MIF was transfected into macrophages, separately. After that, macrophages were incubated with LPS. Cell culture medium was collected for MIF protein detection by EIA. Cellular RNA was extracted and the expression of MIF mRNA was examined by RT-PCR analysis. RESULTS: LPS stimulation resulted in a specific time-dependent expression of MIF derived from macrophages. MIF mRNA and MIF protein level increased at 6 h and reached a plateau at 9-12 h after LPS stimulation. Objective To explore the role of miR-98 in peripheral blood mononuclear cells (PBMC) in the pathogenesis and development of childhood asthma.Methods A total of 43 cases of asthmatic children and 30 cases of healthy controls were enrolled in the study.Peripheral blood mononuclear cells were isolated in both healthy subjects and asthmatic children in acute attack and remission stages.The expressions of miR-98 and interleukin-4(IL-4) and IL-13 mRNA were detected by real-time quantitative PCR.Results The miR-98 levels of asthmatic children in attack stage were significantly lower than those in remission stage and control group (P<0.01).The IL-4 and IL-13 mRNA levels of asthmatic children in attack stage were significantly higher than those in remission stage and control group (P<0.01).There was no significant difference of miR-98,IL-4 and IL-13 mRNA between asthmatic children in remission stage and the controls (P>0.05).Furthermore,a negative correlation was found between the expression of miR-98 and IL-5(r=-0.794,P<0.01) and between the expression of miR-98 and IL-13 mRNA (r=-0.804,P<0.01) in asthmatic children in attack stage.A positive correlation was also found between IL-4 and IL-13 mRNA in asthmatic children in attack stage (r=0.853,P<0.01).Conclusion The expression of miR-98 decreased in asthmatic children,and miR-98 might be involved in the pathogenesis and development of asthma.
7.Analysis of associated factors in secondary hyperparathyroidism of continuous ambulatory peritoneal dialysis patients

Objective To study the prevalence and associated factors of secondary hyperparathyroidism (SHPT) in continuous ambulatory peritoneal dialysis (CAPD) patients.Methods A cross-section study was performed.A total of 639 eligible participants undergoing CAPD treatment more than three months in our peritoneal dialysis center from July 2011 to January 2012 were recruited in the study.All the patients were divided into SHPT group and non-SHPT group according to the intact parathyroid hormone (iPTH) level,and the associated factors of SHPT were investigated through Logistic regression analysis.Results The prevalence of SHPT was 46.95% (300/639).Logistic regression analysis demonstrated that lower hemoglobin,hypocalcemia,hyperphosphatemia,higher alkaline phosphatase,higher Scr,higher nPCR and low calcium dialysate were independent influencing factors of SHPT.Conclusions The prevalence of SHPT is quite high in CAPD patients.Abnormal calcium-phosphorus metabolism,renal anemia,high protein diet and low calium dialysate may affect the SHPT.

9.Comparision of different fixed positions of Devine surgery in concealed penis

Objective To compare the effect of Devine surgery with different fixed positions in the treatment of concealed penis.Methods According to the different fixed positions,45 patients with concealed penis in our hospital from January 2010 to February 2015 were divided into two groups:20 patients were treated with the method that fixing the scapus penis to dermis of the penis' root (group A);the other 25 patients were treated with the method that fixing the scapus penis to pectineal fascia (group B).The surgical success rate,the penile length,and the edema time after surgery between two groups were compared.Results There was no significant difference in surgical success rate and edema time between two groups (P > 0.05).But the penile length of group B was longer than that of group A after surgery (P < 0.05).Conclusion In the treatment of concealed penis with Devine surgery,comparing to the method which fixing scapus penis to dermis of penis'root,the method which fixing scapus penis to fascia pectinea is much better in penile length and penile appearance.
10.Chromosome aberration and micronucleus formation in human peripheral blood lymphocytes induced by 12C heavy ions

Objective To investigate the biological effects of 12C heavy ions with different LETs,and to construct the dose-effect curves of chromosomal aberrations and micronucleus.Methods Human peripheral blood lymphocytes were irradiated with 0.5 - 5.0 Gy of 60Co γ-rays,29 and 148 keV/μm 12C with a dose rate of 0.5 Gy/min,respectively.The colchicine method and cytokinesis-blocking method were applied to measure chromosome aberrations and micronuclei formation,respectively. Results The chromosomal aberrations induced by 29 and 148 keV/μm of 12C ion had a linear dose response,while the dose-response of micronucleus induction followed to a linear-quadratic model.Chromosome aberration and micronucleus frequency increased with the LET of 12C ions. However, the micronucleus frequency increased slowly when the radiation doses exceeded 3.0 Gy..Chromosomal aberrations induced by 148 keV/μm 12C ions increased significantly when the time of colchicine treatment was prolonged.Conclusions Chromosome aberration and micronucleus induced by 12C ions increased with the LET,and the micronucleus frequency increased with dose less than 3 Gy.The biological effects of 12C ions were stronger than those of low LET irradiation of γ-rays.
