1.The effects of hypertonic-hyperoncotic solution on the lipopolysaccharide-stimulated ICAM-1 mRNA expression in human vascular endothelial cell

Objective To investigate the alteration in intercellular adhesion molecule (ICAM)-l mRNA expression on the surface of human vascular endothelial cells (HUVECs) induced by LPS stimulation and the effects of hypertonic-hyperoncotic solution (HHS) . Methods HUVECs were enzymatically isolated and cultured in RPMI 1640 culture medium at 37℃ for about 7 days when a monolayer of endothelial cells has grown. The primarily cultured HUVECs were mixed with LPS 100ng/ml and incubated at 37℃ for 30min, 1,4,8, 12and 24 h. 10%NaCl and 10% hydroxyethyl starch (Fresenius) were added to RPMI 1640culture medium. The HHS concentrations were 0.25% and 0.5% respectively. HUVECs were first incubated with either HHS (0.25% , 0.5% ) or isotonic solution for 10 min, 1h and 4h and then stimulated by LPS for 4h. The ICAM-1 mRNA was measured by reverse transcriptase-PCR. Results There was slight expression of ICAM-1 mRNA on the surface of HUVECs in normal condition. Objective To investigate the effect of hypertonic-hyperoncotic solution (HHS) on CD11b expression on the surface of human neutrophils which are stimulated by lipopolysaccharide (LPS) in vitro. Methods Neutrophils were isolated from fresh peripheral venous blood of healthy volunteers aged 20-4oyr find incubated with LPS 100 ng/ml al 37℃ in 5% CO2 incubator for 15, 30, 60min, 4, 12 or 24h. 3.Study on the application of early comprehensive development model in premature infants

Objective To discuss the effect of early comprehensive development mode on physical, intelligence and mental development of premature infants, and explore the feasibility of comprehensive development mode of preterm infants generally applied in child care clinic. Methods 90 cases of premature infants in intervention group from the beginning of the hospital before the intervention, continuous monitoring and comprehensive intervention including general system health, parenting guidance, family activities, growth monitoring, intelligence assessment, individualized instruction, and so on, for the timely correction of the anomalies. The 90 infants in the control group were only given conventional child health care after 1 month of corrected age. Results The nervous system abnormalities were detected 192 person-times in control group, while 129 person-times in intervention group at 3 months old, the detected rate of nervous system anomaly was higher in the control group than that in the intervention group (χ2=14.574, P=0.000). The body length of the intervention group was (74.31± 2.34) cm, the control group was (72.82 ± 2.13) cm. The body weight of the intervention group was (9.42 ± 0.93) kg, the control group was (8.11 ± 0.90) kg at 12 months old. The growth of weight and length of intervention group were higher than those of control group. The difference was statistically significant ( t=4.467, 9.602, P < 0.05). The development quotient was also assessed, the big activity sport, fine motor, adaptability, social behavior, development quotient of intervention group were (96.40±7.83) points, (95.45± 7.87) points, (98.33±10.87) points, (105.65±7.51) points, (101.45±7.36) points, and the control group were (92.53±6.57) points, (89.27±8.02) points, (94.26±9.88) points, (100.31±8.13) points, (95.65±6.93) points. The difference was statistically significant (t=2.628-5.443, P < 0.05). Conclusions Application of integrated early childhood development in premature infants can find early neurodevelopmental deviations of children and contribute to early intervention on premature infants. Using a combination of family and outpatient intervention may reduce the incidence of neurological abnormalities, improve the level of development of the intelligence assessment and promote premature infant intelligence, motor development and social adaptation ability.
4.Effects of the Different Surgery Methods on Cornea Endothelial Cell in Cataract Extractionn

Objective To compare the different influences on the cornea endothelial cell of surgery methods between human cataract extraction and phacoemulsification.Methods 25 cataract patients(36 eyes) aged 55 to 84 undergoing extra capsula extraction and phacoemulsification combined with implanation ocular lens were analyzed in rate of endothelial cell and density within 2 weeks preoperatively and postoperatively.Results The rates reduced in varying degrees.Conclusions Understanding cornea endothelial cell before operation is clinically significant for evaluating cornea edema after surgery and taking essential measures.
5.Study on the correlation between a set of single nucleotide polymorphisms of hepatitis B virus core gene and serum HBV DNA levels in patients with chronic hepatitis B

Objective To investigate the relation between a set of single nucleotide polymorphisms (SNP) in core gene of HBV in chronic hepatitis B patients and HBV DNA levels. Methods PCR restriction fragment length polymorphism(PCR-RFLP) assay and restriction enzyme Tsp509I were adopted to determine HBV SNP in HBV core gene. Nucleotide sequences of core gene were determined using the dideoxy chain termination method. HBV DNA levels were quantitated with real-time PCR. Results Five typical RFLP patterns, RFLP-C, RFLP-D, RFLP-E, RFLP-G and RFLP-C/G mixture were found and the distribution of HBV RFLP patterns was as follows: C, 61. 5% ; D, 2. 6% ; E, 9.6%; G, 16.7%; C/G mixture, 9.6%. Five SNPs, A165T, A336C, A336T, T337C and T385C, were found to be associated with RFLP patterns change and only SNP A336C or A336T caused the substitution of Glu-83 with Asp in HBcAg. The serum HBV DNA levels in RFLP-C group were higher than that in RFLP-G (P =0. 02) and RFLP-C/G group(P = 0. 006) , respectively, furthermore, the positive rate of serum ALT in RFLP-C/G group was lower than that in RFLP-C, RFLP-E and RFLP-G group, respectively. Under the condition of HBeAg-positive, the serum HBV DNA levels in RFLP-C group were higher than that in RFLP-G (P = 0. 015) and RFLP-C/G group(P =0.008) , respectively. Conclusion PCR-RFLP used in this study can be adopted to determine HBV SNPs, not genotypes in Chinese patients with chronic hepatitis B. The serum HBV DNA level in RFLP-C group higher than that in RFLP-G or RFLP-C/G group maybe associated with amino acid mutation, Glu83 Asp.
6.Initial use of trans-ulner or trans-radial approach to coronary revascularization with titanium-nitride-oxide-coated stent

Objective To evaluate the efficacy of trans-ulner or trans-radial approach to coronary revaseularization with titanium-nitride-oxide-eoated stent (TITAN2). Methods 31 patients,who were planed to receive coronary revaseularization,were selected into this study. All patients with coronary revascularization were either by trans-ulner or by trans-radial approach. The pass-through rate of stent, early thrombosis in stent and MACE at followup were recorded for TITAN2. Results 46 lesions with stenosis > 75% with coronary angiograpby in 31 patients successfully underwent coronary revaseularization with TITAN2. One stent didn't go through the lesion in RCA. The pass-through rate of stent was 97.8%. Remaining stenesis ,stent unglues,endarterium laceration ,early thrombosis in stent and coronary rupture were not discovered after stent deployment. MACE was 0 at 1-5 month follow-up. Conclusion Confirmed good efficacy in coronary revascularization with TITAN2 is observed.
7.Comparison of abnormal expression of miRNAs in peripheral blood of rheumatoid arthritis patients and osteoclasts in rat and analysis of any miRNAs

Objective:To analyze the different expressions of miRNAs in peripheral blood of rheumatoid arthritis (RA) patients and the osteoclasts in rats,and to verify the key miRNAs in RA.Methods: The miRNAs expressions in monocyte and the co-cultured osteoclasts,peripheral blood of rheumatoid arthritis (RA) patients and normal were detected by miRCURYTM LNA Array.Real time PCR was applied to verify the reliability of miRNA array.The bioinformatics software and database were applied to predict and analyze target genes.Results: miRNA array results showed that 189 miRNAs changed in RA patients compared with the normal;211 miRNAs were changed in osteoclasts group compared with monocytes group.The expressions of ten miRNAs were all abnormal in RA patients and osteoclasts in rats.The results of Real time PCR were consistent with the array.Results of bioinformatics analysis showed that the target gene of miRNA significantly enriched in signaling pathways such as VEGF,MAPK signaling pathways.Conclusion: Some miRNAs had similar abnormal expressions in different species,and these miRNAs may influence the differentiation of the osteoclasts through regulating the related signal passways.
8.Analysis of the Combination Use of CYP3A4 Inhibitors with Simvastatin or Atorvastatin in the Cardiology De-partment of A Third-grade Class A Hospital

OBJECTIVE:To provide reference for strengthening the safety of the combination use of CYP3A4 inhibitors with statins in clinical treatment. METHODS:30 cases per month(totally 300 cases)(the hospitalization time was more than 10 days) discharged from the cardiology department of a third-level class A hospital during Jan. to Oct. 2013 were selected randomly,and statistical analysis was made into general information of patients,whether to use statins and CYP3A4 inhibitor,and whether to monitor safety indicators of liver and kidney function and creatine kinase. RESULTS:Totally 291 patients were treated with statins with a total use rate of 97%,including 265 cases of atorvastatin and 11 cases of simvastatin. The CYP3A4 inhibitors were com-bined with amlodipine,diltiazem,verapammy,amiodarone,Ginkgo biloba leaf,fenofibrate and clarithromycin,etc.,and 162 (59%) patients were given the combination use of simvastatin or atorvastatin with CYP3A4 inhibitors. But the liver and kidney function,creatine kinase and other safety indicators were not monitored by laboratory examination during medication. CONCLU-SIONS:Atorvastatin and simvastatin are used commonly in patients with cardiovascular disease,and the combination use rate with CYP3A4 inhibitor is relatively higher and needs long-term medication,drug dosage should be limited according to the direction of instructions and the adverse reactions induced by drug interactions should be noticed to strengthen medication monitor and education for patients.
9.Research on the Influencing Factors of the Elderly' s Demand for the Pension Mode of "Medical-Nursing Combination":Take Urumqi as an Example

Objective:To understand the influencing factors of the elderly' s demand for the pension mode of medical-nursing combination in Urumqi. Methods: A questionnaire survey was conducted among 300 elderly people randomly selected in Urumqi. Results:Univariate analysis showed that there was significant difference between retirement monthly income, life self-care ability, type of chronic disease, the degree of understanding on the pension mode of medical-nursing combination, family support and the demand rate of the elderly for the pension mode of medical-nursing combination ( P< 0 . 05 ) . Two Logistic regression analysis showed that life selfcare ability, types of chronic diseases, the degree of understanding on the pension mode of medical-nursing combination, and family support factors were statistically significant. Conclusion:It should change the traditional concept of single pension mode, improve the medical-nursing combination service system to meet the needs of all levels of the elderly, establish a long-term care insurance system, and improve the training mechanism of the elderly.
10.A Study of Plasma Exosome Protein Spectrum in Coronary Heart Disease Patients with Phlegm Turbidity Syndrome and Healthy People

Objective To explore the specific plasma exosomal protein expression in Coronary Heart Disease(CHD)with phlegm-turbidity syndrome(PTS),so as to provide biological reference for the diagnosis of CHD with PTS.Methods A total of 20 CHD patients with PTS and 20 healthy subjects at the same period were included.Plasma exosomes were extracted from all subjects using the qEV size exclusion method.First,10 CHD cases with PTS and 10 healthy subjects were randomly selected,and differentially expressed proteins between the two groups were screened using label free non-targeted protein quantification analysis.Then,MRM targeting protein labeling technique was applied to verify the differentially expressed proteins in the remaining subjects.Results Compared with healthy controls,214 differentially expressed plasma exosomal proteins(61 up-regulated and 153 down-regulated)were found in CHD patients with PTS,mainly related to cholesterol metabolism,complement and coagulation cascade,and immune effects.ANXA6,C4BPB,F8,CFB,APOE,C9,and CLU proteins were further validated by MRM targeting protein.Conclusion CHD patients with PTS had differences in plasma exosomal protein expression from healthy controls,and the differential proteins are mainly related to cholesterol metabolism,complement,and the coagulation cascade.
