1.Effect of whole body vibration on low extremity functions and chronic diseases in the elderly

BACKGROUND: Whole body vibration (WBV) training has gained increased attention at the beginning of 21st century as a sort of exercise. WBV instruments are popularized gradually; in the meanwhile, WBV training has showed its values especially for people who are too feeble to attend conventional exercises, among whom, the elderly accounts a lot.OBJECTIVE: To summarize the effects of WBV training on low extremity functions and chronic diseases in older individuals, and to explore the underlying mechanism, thus providing reference for the study on WBV training.METHODS: Web of Science, PubMed and CNKI databases were searched for the literatures addressing WBV training published between 1998 and 2015 with the subject word of whole body vibration and the keywords of older adult, elderly,seniors, aging in English and Chinese, respectively. The articles related to the lung diseases were excluded. The subjects were more than 60 years old, the low extremity functions included neuromuscular function, balance and walking abilities, and the chronic diseases included stroke, type 2 diabetes mellitus, hypertension and Parkinson disease.RESULTS AND CONCLUSION: WBV training is likely to increase muscle strength, body balance and functional mobility.In terms of chronic diseases, WBV training contributes to muscle strength, relieving muscle stiffness, amyostasia,enancing improving some indexes of chronic diseases. WBV training is a safe and feasible exercising way for elderly people scheduled for improving the muscle strength, balance and walking abilities. Considering the existed evidences,WBV cannot replace the conventional therapy to treat chronic diseases in the elderly. Furthermore, a large number of high quality trials should be conducted in the future to clarify its effectiveness and application values.
2.The changes of bone resorption in prednisone treatment of nephrotic syndrome and the intervention of α-D3 on it

Objective To discuss the change of bone resorption of primary nephrotic syndrome (PNS) patients with prednisone treatment, andα-D3 effects on it. Methods ①30 PNS patients diagnosed and 28 healthy people in our hospital were selected;blood and urine samples before and after enough prednisone (pred) treatment were collect-ed to detect the iPTH, calcium and urinary DPD. ②30 PNS patients after 6 weeks enough prednisone treatment were randomly divided into Pred +α-D3 group and Pred group. Results ① PNS group compared with healthy group, urine DPD excretion rate was significantly increased(P<0.05),and blood iPTH was significantly increased (P<0.01);blood calcium was similar to healthy group. ② PNS patient after treatment with pred compared with that before treatment, urine DPD excretion rate was significantly increased(P<0.01),and blood iPTH was signifi-cantly increased(P<0.01). There was no obvious change in blood calcium.③Pred+α-D3 group compared with Pred group, urine DPD excretion rate was significantly decreased(P<0.01),and blood iPTH was significantly de-creased(P<0.05). There was no obvious change in blood calcium. Conclusion α-D3 can effectively reduce bone resorption in PNS patients.
3.Guide to TCM syndrome differentiation and treatment of stasis-heat syndrome

‘Stasis-heat’is a pathological factor that arises from exogenous and endogenous injuries in the development and progression of diseases.It can also be the pathogenesis at a certain stage of disease.It manifests clinically as the syndrome of‘Stasis-heat Interactivity.’Through long-term clinical practice‘Stasis-heat’has been found to spread in a variety of diseases’processes and has general clinical significance.Using‘Stasis-heat’theory in prescribing an herbal formula can significantly improve clinical efficacy,displaying the advantages of using traditional Chinese medicine to prevent and treat acute and severe diseases.According to more than 30 years of research and clinical practice,based on ongoing discussion and revision by experts,we have developed the‘Guide to TCM Syndrome Differentiation and Treatment of Stasis-heat Interactivity Syndrome,’a work that is an essential reference for both Chinese medicine and integrated Chinese and Western medicine clinicians.This article lays the groundwork in the hope that other experts will provide further input that will improve the guidelines laid out in this piece of work.
4.Mechanism of apoptosis induced by Mcl-1 inhibitor UMI-77 on gastric cancer MGC-803 cells

Objective To investigate the mechanism of apoptosis induced by UMI-77 , a novel selective inhibitor of Mcl-1, in gastric cancer MGC-803 cells. Methods MGC-803 cells were treated with UMI-77 in different concen-trations for 24 h, apoptotic rates were determined by Annexin V/PI method using flow cytometry. Mitochondrial membrane potential was determined by JC-1 staining on a flow cytometer. The activation of Caspase-9, Caspase-3 and cleavage of PARP were measured by Western blot analysis. The protein level of Bcl-2, Bcl-XL and Mcl-1 was monitored by Western blot as well. Chemically synthesized Mcl-1 siRNA was transfected into MGC-803 cells using Lipofectamine 2000 Reagent. The efficacy of gene silencing was confirmed by Western blot analysis, and opoptotic rates before and after transfection was measured by flow cytometry using Annexin V/PI staining. Results UMI-77 was effective in induction of apoptosis in gastric cancer MGC-803 cells, apoptotic rates were increased in a dose-de-pendent manner. Mitochondrial membrane potential was collapsed after UMI-77 treatment. Activation of Caspase-9, Caspase-3 and cleavage of PARP occurred at 24 h (P<0. 05). The expression level of Bcl-2 and Bcl-XL were not altered after exposure to UMI-77 , while Mcl-1 was down-regulated after 12 h ( P<0. 05 ) . Transfection with Mcl-1 siRNA successfully decreased the expression level of Mcl-1 in MGC-803 cells ( P<0. 05 ) and blocked apoptosis induced by UMI-77 ( P<0. 05 ) . Conclusion UMI-77 induces apoptosis through activation of the intrinsic path-way in gastric cancer MGC-803 cells, and knocking down Mcl-1 expression abrogates apoptosis by UMI-77.
6.Gene cloning and expression of human TSHR ectodomain in eukaryotic cells

Objective To clone and construct the plasmid containing human thyroid stimulating hormone receptor(TSHR) gene ectodomain,and then identify the immunoreactivity of the purified recombinant protein.Methods TSHR total RNA was extracted from human thyroid,and cDNA was obtained with RT-PCR technique.Human TSHR ectodomain gene(hETSHR) was cloned into pcDNA3.1(+) vector.The recombined construct was transfected into CHO cells by Lipofectin 2000.The transcript mRNA was detected by RT-PCR,and protein immunoreactivity was assayed by TSHR antibody with immunocytochemistry staining and Western blot.Results DNA sequencing results showed that the recombinant of human thyrotropin receptor ectodomain had confirmed sequence reported in GenBank.The fusion protein had the immunoreactivity.Conclusion Human thyroid stimulating hormone receptor was successfully cloned in eukaryotic expression vector and the constructor was expressed in eukaryotic cells very well.
7.Expression of TLR2 and TLR4 in the lungs of mice infected by candida glabrata

Objective To study the roles of Toll-like receptor 2(TLR2)and Toll-like receptor 4(TLR4)in the progress of pulmo-nary infection with candida glabrata in experimental mice .Methods The mice were divided into three groups ,the control group (group A) ,mice infected with candida glabrata(group B)and immunosuppressive mice infected with candida glabrata(group C) .6 mice in each group were killed at 1st and 3rd day after the success in pulmonary infection with candida glabrata .The lung tissues from each group were collected for pathological analysis and PT-PCR to detect the expression level of TLR2 and TLR4 .Protein TNF alpha(TNF-α)level was measured by ELISA method .Results The pathological analysis showed the structure was normal and there was no inflammatory reaction in lungs in group A .The infiltration of inflammatory cells ,weak injuries but no conidia or hyphae in lungs were found in group B .Most of the alveolar collapse and severe damage ,part of the expansion ,a large number of in-flammatory cells infiltration ,accompanying with conidia and hyphae accumulation were observed in group C .The expression levels of TLR2 mRNA and TNF-αprotein in mice lungs at the 1st day and the 3rd day in group C were significantly higher than in group A and B(P<0 .05) ,while group B was higher than group A(P<0 .05);The expression levels of TLR2 ,TLR4 mRNA and TNF-αprotein in group C at the 3rd day was higher than the 1st day(P<0 .01) ,meanwhile ,the expression of TLR4 was higher than in group A(P<0 .05) .Conclusion TNF-αand TLR2 expression maybe involve in the infection and process of candida glabrata ,and TLR4 may play a synergistic effect .
9.Effect of miR-340-5p targeting at BMP4 on differentiation of neural stem cells in rats

ObjectiveTo explore regulation effect of miR-340-5p on regulating bone morphogenetic protein 4(BMP4) expression and the differentiation of rat's NSCs.MethodsNSCs of rats were divided randomly into three groups: normal group (Mock),group with nonsense oligonucleotide (Anti-Con) and group with antisense oligonucleotide of miR-340-5p (Anti-miR-340-5p).The qRT-PCR was used to detect the relative expression of miR-340-5p.The expression of NF200 and MAP-2 was detected by immunocytochemical staining and immunofluorescence,respectively.Western blot was used to detect the expression of BMP4 protein.ResultsThe relative levels of miR-340-5p expression were significantly decreased in Anti-miR-340-5p group (0.14±0.01) compared with that of Mock group(1.01±0.17) and Anti-Con group(1.07±0.13) (P<0.01).Immunocytochemical staining indicated that NF200 was positive in cells of Anti-miR-340-5p group.The proportion of MAP2 positive cells was increased in Anti-miR-340-5p group compared with other groups (P<0.05).Western blot showed the increased expression of BMP4 protein in Anti-miR-340-5p group (0.84±0.09) compared with Mock group(0.53±0.04) and Anti-Con group (0.63±0.09) (P<0.05).ConclusionThe miR-340-5p may exert a potential function in regulating differentiation of NSCs into neurons through a negative regulation of BMP4.
10.Activation of caspase-4 was involved in TRAIL-induced apoptosis of gastric cancer cells

Aim To investigate the potential involve-ment of caspase-4 in TRAIL ( tumor necrosis factor-re-lated apoptosis-inducing ligand )-induced apoptosis in gastric cancer cells. Methods The effect of treatment with TRAIL /z-LEVD-fmk alone or in combination for 24 h on the apoptotic rate of gastric cancer cells was detected by FCM ( flow cytometry) using propidium io-dide DNA staining. Chemically synthesized three siR-NAs targeting caspase-4 gene were transfected into gas-tric cancer cells by Lipofectamine 2000 Reagent. The efficacy of gene silencing was confirmed by Western blot analysis . The apoptotic rates of gastric cancer cells to TRAIL before and after transfection with caspase-4 siRNA were observed by FCM. The expression level of GRP78 (78-ku glucose-regulated protein) protein was examined by Western blot, the classic endoplasmic re-ticulum stress inducer tunicamycin ( TM) was used as a control. The expression levels of caspase-4 and caspase-3 after TRAIL treatment were also measured by Western blot. Results z-LEVD-fmk decreased TRAIL-induced apoptosis of gastric cancer cells signifi-cantly(P<0. 05). As compared with negative control, the expression level of caspase-4 protein was reduced after transfection, and the apoptotic rate was also de-creased ( P <0. 05 ) . While TM induced marked up-regulation of GRP78 , treatment with TRAIL resulted in, albeit to a lesser extent, increases in GRP78, in-dicative of induction of ER stress by TRAIL. Activa-tion of caspase-4 and caspase-3 occurred early after TRAIL treatment. Conclusion Activation of caspase-4 contributes to TRAIL-induced apoptosis and is asso-ciated with induction of ER stress by TRAIL in gastric cancer cells.
