1.Cloning and expression of escherichia coli colonization factor antigen CFA/Ⅰ in escherichia coli DH5?

Objective:To clone and express colonization factor antigen Ⅰ(CFA/Ⅰ) of enterotoxigenic Escherichia coli (ETEC) in E.coli DH5?. Methods:CFA/Ⅰ gene was cloned by PCR from H10407 and recombined to a expression vector pBV220.This vector was designated pBV220- CFA/Ⅰand transformed into E.coli DH5?. The product was tested by SDS-PAGE and immunogenesis test. Results:The CFA/Ⅰ gene could be expressed in E.coli and contained strong immunogenesis by immune test. Conclusion:This study paved the way for using ETEC CFA/Ⅰ recombinant as a vaccine candidate in future.
2.The application of cyfr21-1、NSE and CEA detection in early diagnosis of lung cancer and identification of pathological types of lung cancer

Objective To explore the value of cyfr21‐1 ,NSE and CEA detection applied in early diagnosis of lung cancer and i‐dentification of pathological types of lung cancer .Methods Lung cancer patients (observation group) and benign lung lesions pa‐tients(control group) were selected from our hospital ,and then they cyfr21‐1、NSE and CEA value were compared and analyzed .Re‐sults The cyfr21‐1 ,NSE and CEA value of observation group were higher than control group ,and three indicators showed positive markers ,the difference was statistically significant (P<0 .05);Combined detection of three tumor markers specificity was highest , and compared to individual detection ,there were significant differences (P<0 .05);Cyfr21‐1 ,NSE and CEA levels had a certain gap in different pathological types of lung cancer patients .Conclusion Tumor markers cyfr21‐1 ,NSE and CEA have a certain reference value in the early diagnosis of lung cancer and lung pathology identification .
3.Influence of situational health education on self-efficacy of primipara in breast-feeding and child rearing

Objective To explore the effect of situational health education on self-efficacy of primipara in breast-feeding and child rearing. Methods One hundred and eighty-five pregnant women taking prenatal examinations in our hospital from September 2012 to August 2013 were selected into control group and they received traditional education. Another one hundred and ninety-two pregnant women taking the same prenatal examinations in our hospital from Sep 2013 to Aug 2014 were selected into experiment group and the antenatal situational health education was applied . The self-efficacy in breast-feeding and child rearing of the two groups before and after discharged from hospital (6 weeks after delivery) were compared. Result The self-efficacy in breast-feeding and child rearing of the experiment group were significantly better than those of the control group and before discharged from hospital (P<0.05). Conclusion The situational health education is important for increasing self-efficacy in breast-feeding and child rearing and is beneficial for the growth and development of the neonates.
4.Effect of inhaling penehvclidine hydyochloride in treatment of patients with acute exacerbation of chronic obstrnctive pulmonary disease

Objective To evaluate the efficiency of inhaling penehyelidine hydyoehloride(PHC)in reducing pulmonary arterial pressure and improving lung funetion in patients with acute exaeerbation of ehronie obstruetive pulmonary disease(AECOPD).Methods Sixty patients with AECOPD were randomly divided into the control group(30 patients) and the treatment group(30 patients).The patients in treatment group inhaled PHC,and patients in control group inhaled diprophylline.The pulmonary arterial pressure was measured before and after inhalation.The peak expiratory flow(PEF) and forced expiratory volume in one second(FEV1)was measured at different time points before and after inhalation. Arterial blood gas and,symptoms were also observed.Results The pulmonary arterial pressure in treatment group was reduced by 20% and reduced by 5% in the control group.FEV1 and PEF were improved after inhalation,the average increase of FEV1 was 350 ml(P<0.01),and PEF was improved by 37 L/min(P<0.05).Arterial blood gas analysis was improved,and PaO_2 increased by 18 mmHg(P<0.01).The respiratory rate and the symptoms of cough and sputum were also improved.Conclusion PHC improved the lung function of patients with AECOPD by reducing the pulmonary arterial pressure.
5.An evaluation research for new nurses six dimensional scales in pre-service training

Objective To investigate the feasibility of Six Dimensions Scale (6-D scale) of Nursing Performance used for evaluating the effect of standardized hospital-department training on new graduate nurses.Methods 6-D Scale of Nursing Performance was conducted to evaluate the nursing ability of 216 new graduate nurses (2013) of our hospital pre and post the three-month intervention,including the self-appraisal and appraisals by head nurses.Results The nursing ability was improved significantly after the training.The total score of self-appraisal 6-D scale were significantly higher than that before the training (142.34±19.92 vs 135.81±18.30),in which the subscales scores within the dimensions of leader-ship,critical care,teaching/collaboration,planning/evaluation,interpersonal relations/communication were increased significantly,while the increase of professional development was not significant.Six-D Scale and six subscales scores based on self-appraisal were higher than that based on the head nurses' appraisals.Conclusions The application of 6-D scale is scientific to evaluate the effect of standardized nurse training.We had demonstrated that standardized hospital-department training is of great benefit for new graduates to improve the nursing ability and adapt to the clinical circumstances.
6.Survey on clinical knowledge of coronary artery disease management in general practitioners in Beijing

A survey on clinical knowledge of coronary artery disease (CAD) management in general practitioners (GPs) in Beijing was conducted from March to September,2013.The questionnaire contained 4 cases (stable angina pectoris,post-percutaneous coronary intervention,acute anterior myocardial infarction and acute inferior myocardial infarction) and related clinical questions.Five hundred questionnaires were distributed and 471 valid questionnaires were returned with a recovery rate of 94.2%.The correct rate was 87.3% and 61.8% for diagnosis and treatment of emergent events in acute anterior myocardial infarction; however,the correct rate for other knowledge was ranged between 23.4% and 40.6%.The correct rate increased with the educational levels in 5/11 items(P < 0.01)and increased with the professional titles(P < 0.001)in 3/11 items,which was not associated with the attendance of training or frequency of training attendance.The results indicate that clinical knowledge of CAD management for GPs in Beijing should be improved and the training methods should be focused on the clinical capacity.
7.Coping strategies of patients with superficial bladder carcinoma and the influential factors

Objective To explore the coping strategies of patients with superficial bladder carcinoma and analyze the influential factors.Method The medical coping modes questionnaire(MCMQ)was used to do the investigation among 146 patients with superficial bladder carcinoma.Results Among the coping strategies,the scores on the factors of facing,avoidance and yielding were 2.42±0.45,2.00±0.45 and 1.52±0.50,respectively.The operative method,education level,sex and stages of chemotherapy were the main factors influencing the coping strategies.Conclusions For the postoperative patients with superficial bladder carcinoma, the coping strategies were influenced by different factors.Therefore,nursing staff should attach their highlights on patients' coping strategies and instruct the patients with active coping strategies based on the their traits.
8.Survey on general background and professional competency of general practitioners with standardization training in Beijing

One hundred and thirty general practitioners ( GP ) who received standard training ( training group ) and 189 GPs who did not receive standard training ( non-training group ) were selected with cluster random sampling method from five districts in Beijing.A questionnaire survey was conducted from April to May 2013, including basic information , working condition , professional competency and continuing education of the interviewees.The survey showed that 72.3%(94/130) in training group had physician titles;89.3%(116/130 )had work experience <10 y;97.0%(126/130)of them had bachelor degree or above;82.3%(107/130) expressed their interest in scientific research , but the 95.4% of them had no research projects;and 96% had no teaching appointment.GPs in non-training group were in a favorite position of education level and professional competency over those in training groups ; however , the latter were highly enthusiastic in further training and scientific research , and had strong potential for career development.
9.Changes in marrow fat content of non-dialysis chronic kidney disease patients assessed by T2 *-corrected water-fat MRI

Objective To detect the changes in vertebral marrow fat fraction (MFF) using T2 *-corrected water-fat MRI and to analyze the relationships between MFF with bone biomarkers of non-dialysis chronic kidney disease (CKD).Methods 78 CKD patients were divided into five groups according to the eGFR and underwent water/fat MRI to obtain MFF.The reliability of MFF measurements by two radiologists was assessed with intra-class correlation coefficient (ICC).Serum calcium,phosphorus,alkaline phosphatase,osteocalcin,intact parathyroid hormone and 25(OH)D3 were determined.Results Mean CV for MFF measurements reproducibility was 2.37%.The inter-observer agreement for MFF was excellent (ICC=0.901).The ICC for each intra-observer agreement was excellent (ICC=0.959 and 0.948,respectively).There were statistical differences in MFF among five groups of CKD.Changes of MFF were earlier than those of serum calcium,phosphorus,parathyroid hormone,alkaline phosphatase and osteocalcin.MFF was positively correlated with serum phosphorus (r =3.011,P =0.003),parathyroid hormone (r=3.852,P<0.001),and negatively associated with calcium (r=-2.767,P=0.017),25(OH)D3 (r=6.032,P<0.001),eGFR (r=-5.104,P<0.001),respectively.Multivariable regression analysis showed MFF was negatively correlated with 25(OH)D3(Sβ=-0.343)and eGFR(S(S=-0.284,P<0.001).Conclusion CKD patients had higher marrow fat.T2 *-corrected water fat MRI could serve as a useful tool to quantify marrow fat content for CKD patients.
10.Progress of PML post-translational modifications in tumor cells

The promyelocytic leukemia (PML) protein is an important part of the PML nuclear bodies (PML-NBs) structure.PML protein is crucial for the assembly of PML-NBs and recruits more than 30 different proteins, including DAXX, ATRX, and small ubiquitin-like molecules involving SUMO to the PML-NBs region.Increased evidence has emerged that a number of different proteins is involved in regulating PML activities by post-translational modifications, such as SUMO modification, ubiquitination and phosphorylation.Here, we review recent studies on the combination of PML and different proteins in the process of apoptosis, replicative senescence and DNA damage response.
