1.Effects of angiotensin Ⅱ receptor antagonist on TGF-β1 synthesis and TIMP-1 mRNA expression of hepatic stellate cells

Objective To investigate the effects of angiotensin Ⅱ (Ang Ⅱ ) and angiotensin Ⅱ type 1 receptor antagonist (AT1RA) on the transforming growth factor-β1 (TGF-β1) synthesis and tissue inhibitor of metalloproteinase-1 (TIMP-1) mRNA expression of in vitro cultured hepatic stellate cells (HSCs). Methods HSC-T6 rat hepatic stellate cell line was selected as the study model of the activated hepatic stellate cells. Cultured HSCs were randomized into control group, Ang Ⅱ group, AT1RA group and Ang Ⅱ AT1RA group. Cell culture medium was used to detect the TGF-β1 level by ELISA method. HSCs were harvested to measure the TIMP-1 mRNA expression by RT-PCR. Results TGF-β1 level of control group, Ang Ⅱ group and AngⅡ AT1RA group in cell culture medium was (7.531 ±0. 654) pg/mL, (9. 855± 1. 485)pg/mL and (7.719 ± 0.329) pg/mL respectively, Ang Ⅱ group higher than control group (P < 0.05 ), Ang Ⅱ AT1RA group lower than Ang Ⅱ group (P < 0. 05 ). TIMP-1 mRNA expression level of control group, Ang Ⅱ group and Ang Ⅱ AT1RA group in HSCs was 3. 387 ± 0. 042, 4.870 ± 0.061 and 3. 837 ± 0. 042 respectively, Ang Ⅱ group higher than control group ( P < 0. 05 ), Ang Ⅱ AT1RA group lower than Ang Ⅱ group (P < 0. 05). Conclusion AngiotensinⅡ can increase the TGF-β1 synthesis and TIMP-1 mRNA expression of hepatic stellate cells, while all these effects are inhibited by angiotensinⅡ type 1 receptor antagonist.
2.Influence of psychological care and community revisit on incidence of postpartum depression

ObjectiveTo explore the effect of psychological care and community revisit on preven-tion of postpartum depression. Methods300 cases of pregnant women were randomly divided into the intervention group and the control group with 150 cases in each group. The intervention group took psycho-logical care and community revisit, the control group received conventional care. The incidence of missing follow- up and depression in the two groups were compared using χ2 test. ResultsThe incidence of miss-ing follow-up and depression in the intervention group was significantly lower than those of the control group. ConclusionsPsychological care and community revisit can significantly reduce the occurrence of postpartum depression.
3.Effect of insulin resistance on fatty liver in high-fat diet-fed mice

[ ABSTRACT] AIM:To study the influence of insulin resistance on fatty liver in the mice fed with high-fat diet (HFD).METHODS:Male 8-week-old C57BL/6J mice were randomly divided into HFD group (with 60% calories by high saturated fatty acid) and control group (with chow diet).The mice in both groups were fed for 12 weeks.The body weight, liver weight, serum triglyceride (TG) and total cholesterol (TC), and blood glucose and insulin levels were meas-ured.Hyperinsulinemic euglycemic clamp experiment was applied to reflect insulin sensitivity .The lipid deposition in the liver was analyzed by HE staining , Sudan IV staining and measurement of liver fat content .The phosphorylation levels of IRS1 and Akt, and the protein levels of SREBP-1 and FAS were determined by Western blot to reflect the activities of insu-lin signaling and lipid synthesis .RESULTS:Compared with control group , the body weight and liver weight were signifi-cantly increased in HFD group .TG and TC contents in serum and liver tissues were remarkably increased in HFD group . High-fat diet induced insulin resistance , as evidenced by increased serum insulin levels , reduced glucose infusion rate and decreases in IRS1 and Akt phosphorylation levels .In livers of HFD group, HE staining showed that the cytoplasm of hepa-tocytes was filled with vacuoles .Sudan IV staining also displayed that many different sizes of red lipid drops existed in the hepatocytes , and the protein levels of SREBP-1 and FAS were significantly increased .In primary normal hepatocytes with exogenous oleic acid intervention for 48 h, the phosphorylation levels of IRS 1 and Akt were reduced , and the protein ex-pression of SREBP-1 and FAS was significantly increased in a dose-dependent manner .CONCLUSION: Feeding with HFD leads to insulin resistance , resulting in activation of lipid synthesis and accumulation of lipid deposition in the liver , thus inducing fatty liver .
4.The reversal effect of abnormal savda munziq total flavonoids on leukemia cell lines

Objective To investigate the reversal effect of abnormal savda munziq total flavonoids on leukemic cell lines.Methods CCK -8 method was used to detect the cytotoxicity of abnormal savda munziq total flavonoids on leukemia cells,by multivariate analysis of variance in the chemotherapy drug intervention,further calculated the reversal fold.Results CCK -8 results showed that with the increase of concentration of ASMq800(μg/mL)compared with 200(μg/mL)and 400(μg/mL),the A value of K562 /ADR cells gradually decreased,the growth curve suggested that with the increase of ASMq concentration in K562 /ADR cell,survival time was shortened,the inhibition of the cells was significantly increased.Different concentrations of ASMq on the adriamycin toxicity with increase of concen-tration of adriamycin,the inhibition of ASMq on multidrug resistance cell line was significantly higher.The reversal multiples calculated that with the increase of ASMq concentration 600 μg/mL and 900 μg/mL,the reversal index increased significantly 2.485 times and 3.651 times.Conclusion ASMq on human leukemia multidrug resistance cell line K562 /ADR could reverse the drug resistance,the reversal mechanism needs further study.
5.Effect of alpha-melanocyte in rat models of renal ischemia-reperfusion injury

BACKGROUND: Kidney ischemia-reperfusion injury often combines with acute kidney and lung injury. The expression of cutin cel growth factor receptor (KGFR) and alpha sodiumchannel protein (α-ENaC) in kidney and lung after ischemia-reperfusion injury and the protective effect of α-melanocyte require further observation and research. OBJECTIVE:To explore the therapeutic effect of α-melanocyte on the expressions of KGFR and α-ENaC in rat models of ischemia-reperfusion injury. METHODS:A total of 30 healthy male Sprague-Dawley rats were randomly divided into control group, ischemia-reperfusion group and α-MSH group. Models of renal ischemia-reperfusion injury were established by 30-minute ligation of renal artery in the ischemia-reperfusion and α-melanocyte groups. Rats in the control group were only used to expose the renal artery, no ligation. Rats in the α-melanocyte group were intraperitonealy injected with α-melanocyte (0.25 mg/kg) at 30 minutes before model establishment. Rats in the ischemia-reperfusion group were injected with 4 mL of physiological saline. RESULTS AND CONCLUSION: Compared with control group, water content of kidney and lung increased significantly in rats of ischemia-reperfusion group and a-MSH group, while the levels of KGFR and α-ENaC of kidney and lung in rats were lower (P < 0.05). Compared with the ischemia-reperfusion group, water content of kidney and lung in rats of a-MSH group decreased significantly, while the levels of KGFR and α-ENaC of kidney and lung increased gradualy (P < 0.05). Moreover, edema was significantly lessened in the rat kidney and lung. Results confirmed that after renal ischemia-reperfusion injury, KGFR and α-ENaC expression was consistent to the kidney and lung injury. α-MSH could increase the protein and mRNA expression of KGFR and α-ENaC in kidney and lung of rats, reduce the kidney and lung injury, and exert a certain protective effect.

9.Entirely different impact on the quality of nursing writing quality control way

Objective To explore the radar map in the department of, analysis of different quality control methods on the impact of nursing writing quality. Methods All medical records in August 2014 to January 2015 were analysed according to nursing group and nurse individual quality control approach. Results Temperature chart error rates were 63.43%(137/216) in the group quality control group and 29.31%(68/232) in the personal quality control group, which was statistical significance between the group quality and personal quality control (χ2= 18.581, P<0.01). Grouping radar map displayed comprehensive evaluation error rate was: group quality 2.987 1 > personal 2.985 6. Conclusions Personal quality has significant advantages in reducing error rate about the indicator temperature, especially in temperature chart, the doctor′s advice, orders and overall quality control effect is better.
10.Renin-angiotensin system blockades and contrast-induced nephropathy: a meta-analysis

Objective To evaluate the effects of renin-angiotensin system (RAS) blockades [angiotensin-converting enzyme inhibitors (ACEI) and angiotensin Ⅱ type 1 receptor blockers (ARB)]on contrast-induced nephropathy (CIN) in patients undergoing angiography.Methods Pubmed,Embase,Cochrane library,Wanfang database and CNKI were searched.The literature limited range was from their start year to July 2015.Randomized controlled trials (RCTs) and non-randomized controlled trials of renin-angiotensin system blockades in influencing CIN were assessed.Two investigators extracted data and performed quality analysis independently from all trims included.Rev man 5.3 software was used.Results 16 trials with a total of 15 897 patients were identified.There were 7490 patients who received renin-angiotensin system blockades and 8407 patients in control group.The meta analysis revealed a higher CIN incidence in ACEI/ARB group than that in control group (14.35% vs 12.13%,P=0.04,OR=1.44,95%CI 1.01-2.04).For patients with renal insufficiency,ACEI/ARB group had a higher CIN incidence than control group (12.23% vs 7.32%,P=0.02,OR=1.80,95%CI 1.10-2.94),and the serum creatinine changes in ACEI/ARB group were higher than those in control group.There was statistical difference in serum creatinine changes between groups (P=0.02,MD=0.08,95%CI 0.02-0.15).Conclusions Renin-angiotensin system blockades can increase theincidence of CIN in patients undergoing angiography.Renin-angiotensin system blockades can contribute to CIN for patients with renal insufficiency.
