1.The relationship between the remnant tumor and the epithelial-mesenchymal transition after cryoablation:an experimental study

Objective To investigate the effect of incomplete cryoablation on the biological behavior of prostatic cancer RM-1 cells and its mechanism.Methods RM-1 cells of prostatic cancer were placed in -20℃ icebox to be frozen for 5 min.After the recovery of the cell state,the RM-1 cells were frozen again for 10 min and 15 min successively.After culture for one day,the cellular morphology was microscopically examined.A total of 20 C57/BL mice were used to establish the tumor-bearing models,which were randomly and equally divided into the control group and the incomplete cryoablation group with 10 mice in each group.At scheduled time points the tumor lesion size was measured for all mice.The mice were sacrificed at 14 days,the lung tissues were collected and were stained with lE;the numbers of metastatic lesions in the lung were calculated.Transwell assay was used to test the cell migration and invasion,immuno-blotting method was adopted to determine the epithelial-mesenchymal transition-related (EMT-related) protein expression level,and the enzyme-linked immunosorbent assay (ELISA) was employed to check the secretion volume of transforming growth factor-beta (TGF-β).Results After incomplete cryoablation,RM-1 cells became disorderly arranged,their morphology was changed,and antenna structure might be formed.At 3 and 7 days after cryoablation,the tumor size in the incomplete cryoablation group was slightly smaller than that in the control group,but only the difference at 7 days after cryoablation was statistically significant between the two groups (P=0.019).At 10 and 14 days after cryoablation,the tumor volume of the two groups was almost equal.The pulmonary metastatic lesions in the incomplete cryoablation group were obviously much more than those in the control group (P<0.001).Transwell assay indicated that the cell migration and invasion ability in the incomplete cryoablation group was stronger than that in the control group (P<0.05).Immuno-blotting test revealed that,when compared with the control group,in the incomplete cryoablation group the expressions of N-cadherin,MMP-9 and Vimentin were up-regulated,while the expression of E-cadherin was downregulated.ELISA test showed that increased secretion of TGF-β was observed in the incomplete cryoablation group.Conclusion Incomplete cryoablation can enhance the migration and invasion ability of RM-1 cells,increase the number of pulmonary metastatic lesions in tumor-bearing mice,and affect the EMT-related protein expression level.
2.Clinical significance of C-reactive protein and homocysteine in type 2 diabetes with depressive disorder

Objective To investigate the changes of C- reactive protein( CRP ) and homocysteine ( Hcy)in the type 2 diabetes with depression,and its clinical significance and potential mechanism. Methods One hundred and twenty-four cases with type 2 diabetes were divided into the depression group(63 cases)and non-depression group( 61 cases ) according to the Self-Rating Depression Scale and verified by Self-Rating Anxiety Scale. The information including age,sex,education degree,body mass index,course of disease and the number of complications were recorded. The levels of CRP,Hcy,fasting plasma glucose( FPG ),glycosylated hemoglobin(HbA1c)and blood lipid were measured. The depression group was divided into mild,medium and heavy group to compared the changes of Hcy and CRP. Results The levels of Hcy,HbA1c and the number of complications in depression group were 11. 5( 8. 6,15. 6 )μmol/L,( 10. 13 ± 2. 17 )%,and 2( 1,3 ) respectively,higher than that of non-depression group(8. 6(7. 4,11. 2)μmol/L,(9. 33 ± 2. 20)%,1(0,2)), while the education degree of depression group((9. 75 ± 3. 36)years)was lower than that of non-depression group((11. 56 ± 3. 73)years),and the differences were significant( t/Z = -3. 537,0. 952,-2. 339,0. 228 respectively;P ﹤0. 05). The levels of Hcy in mild,medium and heavy depression group were(8. 75(7. 45, 10. 45)μmol/L,12. 2(8. 90,14. 40)μmol/L,19. 50(14. 33,28. 03)μmol/L respectively and the difference was significant(F =25. 963,P =0. 000). Conclusion Type 2 diabetes with depression is associated with the level of blood glucose,education degree and the course of disease. Hcy,not CRP is an independent risk factor of type 2 diabetes with depression.
3.Clinical significance of neutrophil-to-lymphocyte ratio changes in patients with castration-resistant prostate cancer after receiving cryoablation

Objective To discuss the clinical significance of peripheral neutrophil-to-lymphocyte ratio (NLR) changes in patients with castration-resistant prostate cancer (CRPC) after receiving argon-helium cryoablation.Methods A total of 33 CRPC patients,who were treated with argon-helium cryoablation at Tianjin Medical University Cancer Hospital,were included in this study.The clinical and pathological data were collected and analyzed.The following factors that might affect the postoperative overall survival (OS) of patients were analyzed with univariate and multivariate analysis:age,baseline PSA level,hemoglobin,white blood cell count,platelet count,albumin,alkaline phosphatase,NLR,platelet-to-lymphocyte ratio (PLR),hormone sensitive time,chemotherapy,bone metastasis,Gleason score,ECOG score,PSA effective rate.Results A total of 33 patients were enrolled in this study,the average age was 69 years (50-82 years) and the median survival time was 28 months (6-55 months).Univariate analysis showed that the baseline PSA level,alkaline phosphatase,NLR,hormone sensitive time,chemotherapy,bone metastases,Gleason score and PSA effective rate were significantly correlated with OS of CRPC patients after receiving cryoablation (P<0.05).Multivariate analysis showed that the baseline PSA level (P=0.003),NLR (P=0.009),Gleason score (P<0.001) were independent predictive factors for OS of CRPC patients after cryoablation therapy.Conclusion NLR can be used as a prognostic predictor for CRPC patients undergoing argon-helium cryoablation,and the increased NLR indicates a poor prognosis.(J Intervent Radiol,2017,26:237-242)
4.Argon-helium cryoablation for the treatment of hepatic metastases from nasopharyngeal carcinoma:initial results in 16 patients

Objective To evaluate the efficacy and safety of CT-guided percutaneous argon-helium cryoablation in treating hepatic metastases from nasopharyngeal carcinoma. Methods The clinical data of 16 patients with hepatic metastases from nasopharyngeal carcinoma, who had received percutaneous argonhelium cryoablation therapy, were retrospectively analyzed. The quality of life before and after therapy, and the postoperative complications were recorded. After argon-helium cryoablation therapy, the progression-free survival (PFS), the overall survival (OS), and the one-month, 3-month, 6-month, 12-month postoperative tumor local control rates were calculated. Results In all 16 patients, no severe complications occurred and the quality of life was significantly improved after argon-helium cryoablation therapy. The one-month, 3-month, 6-month, 12-month postoperative tumor local control rates were 100%, 87.5%, 80.0% and 58.3%respectively. The median PFS was 11 months (95%CI: 8.4-13.6 months), and the median survival time was19 months (95%CI: 9.2-28.8 months) . After argon-helium cryoablation therapy, the half-year, one-year and2-year survival rates were 93.8%, 75% and 43.8% respectively. Conclusion For the treatment of hepatic metastases from nasopharyngeal carcinoma, CT-guided percutaneous argon-helium cryoablation is minimally-invasive, safe and effective with reliable curative effect.
5.Study on the Spectrum-effect Relationship of Different Solvent Extracts from Trollius chinensis against Hypoxia/ reoxygenation Injury of Cardiomyocyte

OBJECTIVE:To study spectrum-effect relationship of 11 different solvent extracts from Trollius chinensis against hypoxia/reoxygenation injury of cardiomyocyte . METHODS :HPLC-MS/MS method was used to establish the fingerprints of 11 different solvent extracts from T. chinensis ,the compounds corresponding to the common peaks were identified by comparing with the substance control and literature information. MTT assay was used to detect the effects of 11 different solvent extracts from T. chinensis on the survival rate of rat myocardial H 9c2 cells injured by hypoxia/reoxygenation. The MDA content ,ROS level in cells and LDH content in the supernatant were detected by ELISA. GRA and PLS method were used to analyze the spectrum-effect relationship between the compounds corresponding to common peak and anti-hypoxia/reoxygenation injury of cardiomyocyte (drug effect). RESULTS :There were 22 common peaks in 11 different solvent extracts from T. chinensis ,and 22 compounds were identified. Compared with hypoxia/reoxygenation injury group ,survival rate of hypoxia/reoxygenation injury+S 1-S6,S9 and S 10 groups were increased significantly ,while MDA content ,ROS level and LDH content were decreased significantly (P<0.05); ROS level and LDH content of hypoxia/reoxygenation injury+S 8 group w ere decreased significantly (P<0.05). The r of GRA analysis of 22 compounds with drug effects were all higher than 0.8. CONCLUSIONS :Orientin(peak 6),vitexin(peak 11), orientin-2″-O-β-L-galacto- pyranosl (peak 4),orientin-2″-O-β-D-Pyrine xylosides (peak 5),quercetin-3-O-glucopyranoside(peak 8),vitexin-2″-O-β-L-galactoside(peak 9),hyperoside(peak 12),vitexin-2″-O-β-D-pyrine xylosides (peak 10),2″-O-(2″′- methylbutyry-loxy)-orientin(peak 15)may be the main components of anti-hypoxia/reoxygenation injury of cardiomyocytes.
6.Study on Synthesis and Identification of Rotundic Acid Esters Derivatives and in vitro Antitumor Activity

OBJECTIVE: To synthesize and identify rotundic acid esters derivatives, and to evaluate in vitro antitumor activity. METHODS: Using rotundic acid as raw material, rotundic acid esters derivatives were synthesized by 28-esterification, 3β and 23-position hydroxyl group combined with acid anhydride. The structure of rotundic acid esters derivatives were identified by HRMS, 1H-NMR, 13C-NMR and physicochemical properties. The in vitro antitumor activities were evaluated by MTT assay against paclitaxel (positive control), rotundic acid and rotundic acid esters derivatives with human cervical cancer cells HeLa, human malignant melanoma cells A375, human lung adenocarcinoma cells SPC-A1 and human liver cancer cells HepG2 (take IC50 as an indicator). RESULTS: Totally 6 kinds of rotundic acid esters derivatives were synthesized, i.e. rotundic acid methyl ester(compound 1), 3, 23-O-di(acetyl) rotundic acid methyl ester(compound 2), 3, 23-O-di(propionyl) rotundic acid methyl ester(compound 3), 3, 23-O-di(butyryl) rotundic acid methyl ester(compound 4), 3, 23-O-di(phthaloyl) rotundic acid methyl ester(compound 5) and 3, 23-O-di(succinyl) rotundic acid methyl ester(compound 6). Compared with paclitaxel, the antitumor activity of compound 5 and compound 6 to Hela, A375, HepG2 and SPC-A1 was similar to it, and there was no significant difference between them (P>0.05); the IC50 of rotuntic acid and other compounds decreased with statistical significance (P<0.05). CONCLUSIONS: In the study, 6 kinds of rotundic acid esters derivatives are synthesized and compound 5 and compound 6 have significant antitumor activity in vitro, which can provide reference for further study.
