2.Effects and related mechanisms of preconditioning with puerarin on hippocampus CA1 region neuronal injury after focal cerebral ischemia-reperfusion
Acta Universitatis Medicinalis Anhui 2015;(6):723-726
Objective To observe the effect of puerarin preconditioning on hippocampal CA1 region neuronal injury and further explore its mechanisms. Methods 30 male Sprague-Dawley rats were randomly assigned into sham, model, and puerarin preconditioning ischemia-reperfusion groups. The middle cerebral artery was occluded for 60 min by an intraluminal filament before reperfusion to induce transient focal cerebral ischemia reperfusion rats. Puer-arin preconditioning groups rats received puerarin(100 mg / kg,i. p)1 h before ischemia. The histological changes of hippocampal CA1 region neurons were measured by HE staining on the 5th day after reperfusion. The expressions of Caspase-3 and Caspase-9 proteins in the CA1 region were examined by immunohistochemistry staining. Results The results of HE staining showed that, compared with the model group, puerarin preconditioning significantly re-duced the number of cerebral ischemia induced injury of hippocampal CA1 region neurons(P < 0. 05). Immunohis-tochemistry data showed that the expressions of Caspase-3 and Caspase-9 in the hippocampal CA1 region of puerarin preconditioning group were significantly lower than that of model group(P < 0. 05). Conclusion The results indi-cate that puerarin preconditioning 1 h before ischemia exerts neuroprotective effects on focal cerebral ischemia-reperfusion induced neuronal injury. The neuroprotective mechanisms of puerarin may attribute to inhibiting the ex-pressions of Caspase-3 and Caspase-9 protein.

This study was aimed to observe effects of Albizia Julibrissin Flower Total Flavonoids (AJFTF) on the learning and memory abilities and plasma monoamine neurotransmitters 5-HT and NE content of depression model rats . A total of 90 SD rats were randomly divided into the normal group , model group , western medicine group ( Venlafaxine Hydrochloride 0 . 0125 g?kg-1 ) and high , middle and low dosage groups of AJFTF ( equivalent to the crude drug of 10 , 5 , 2 . 5 g?kg-1 ) . Depression model rats were induced by solitary cultiva-tion and chronic unpredictable stress . The learning and memory abilities of model rats were evaluated with the Morris water maze . And the plasma contents of 5-HT and NE were determined by enzyme-linked immunosor-bent assay ( ELISA ) . The results showed that AJFTF ( administered with high , middle and low dosage for 21 days ) significantly shortened the incubation period of the Morris water maze positioning and navigation ( P <0 . 05 or P < 0 . 01 ) , and increased the times of crossing the platform in space exploration ( P < 0 . 05 or P <0 . 01 ) . There was a certain dose-effect relationship . The AJFTF ( high , middle and low dosage ) increased the plasma contents of 5-HT and NE ( P < 0 . 05 ) . It was concluded that AJFTF can improve learning and memory abilities of depression model rats which indicates that AJFTF have potential antidepressant effects . Its action mechanism may be related to increase plasma monoamine neurotransmitter content of 5-HT and NE .

ObjectiveTo evaluate the methodological quality of systematic review / Meta analysis (SR/MA) of intervention randomized controlled trial (RCT) published in the Sichuan Mental Health. MethodsThe literature databases such as Wanfang Data, China National Knowledge Infrastructure (CNKI), VIP Database for Chinese Technical Periodical (VIP) and Chinese Biomedical Literature Database (CBM) were searched for the SRs/MAs of intervention RCTs published in the Sichuan Mental Health from the initial issue to the issue published on 31 June 2021. Then the methodological quality of eligible SRs/MAs were assessed using A Measurement Tool to Assess systematic Reviews 2 (AMSTAR 2). ResultsThe literature search yielded 24 full-text articles, and the mean AMSTAR 2 score of the included SRs/MAs was (5.21±3.63) with a range from 1 to 11. The total AMSTAR 2 score for SRs/MAs showed difference in terms of the publication date prior to or later than the publication of AMSTAR-2 tool (t=-5.499), number of authors ≤ 2 or ≥ 3 (t=-6.736), with or without funding support (t=3.329) and author unit nature (F=7.827), with statistical significance (P<0.01). All selected studies had deficiencies on explicit statement of a priori design and registrations, list of excluded studies and reasons for exclusion, sources of funding for the research in systematic review, potential conflicts of interest statement, and funding information of the systematic review. ConclusionThe methodological quality of SRs/MAs of intervention RCTs published in the Sichuan Mental Health varies widely, After the release of AMSTAR 2, the methodological quality has improved, but the report still needs to be further standardized to provide high-quality evidence-based evidence.

Objective:
To investigate the effects of interleukin ( IL) -10 on the proliferation of HaCaT cells and CaCl2 induced expression of differentiation markers and its possible molecular mechanisms.
Methods:
HaCaT cells were treated with various concentrations of IL-10 (0,3,10,30 ng / ml) for different time (0,24,48,72 h) ,cell proliferation was measured using MTS,and cell cycle was determined by flow cytometry.HaCaT cells were pretreated with IL-10 (final concentration 10 ng / ml) for 1 h,then incubated with or without CaCl2 (final concentration 1. 2 mmol / L) for 24,48,72 h ,Western blot was performed to detect the effect of IL-10 on the expression of HaCaT keratinocyte differentiation markers. After pretreatment of HaCaT cells with PD98059,an inhibitor of mitogen-activated kinase-ERK1 /2,and LY294002,an inhibitor of phosphatidylinositol kinase-serine / threonine kinase (PI3K-AKT) ,the total RNA and proteins were extracted separately, real time quantitative polymerase chain reaction (RT-qPCR) and Western blot were used to examine the influence of IL-10 on the expression of differentiation markers (Keratin1,Keratin5,Involucrin) .
Results :
MTS results revealed that IL-10 (30 ng / ml and lower doses) did not alter the proliferation of HaCaT cells in 72 h.Flow cytometry analysis demonstrated that IL-10 had no significant influence on cell cycle progression. The results of Western blot showed that IL-10 upregulated the expression of differentiation markers Involucrin, while there was no significant effect on Keratin1 and Keratin5 .Mechanism research analysis demonstrated that IL-10 could activate ERK1 /2 and AKT ,increase their phosphorylation levels ; RT-qPCR and Western blot results showed that PD98059 and LY294002 partially blocked IL-10 induced Involucrin expression.
Conclusion
At a particular concentration range,IL-10 has little effect on HaCaT proliferation ,but it partially upregulates the expression of differentiation marker Involucrin via the MAPKs-ERK1 /2 and PI3K-AKT pathways.
