1.Medium-term, long-term efficacy of collagenase injected to target site by CT-guided in treatment for lumbar disc herniation

Objective To explore medium-term,long-term efficacy of collagenase injected to target site by CT-guided in treatment for lumbar disc herniation. Methods 50 patients with lumbar disc protrusion were treated with CT-guided percutaneous injection of collagenase accurately reaching prominence(target site). Results All patients were followed up 36 to 60 months. The total effective rate was 82.97% (more than 36 months)and82.23%(more than 60 months). Conclusion Grasping indications well, CT-guided percutaneous injection of collagenase target site in treatment of lumbar disc herniation was accurate, safe, simple, effective, and was the preferred treatment for such patients.
3.Human bone marrow mesenchymal stem cells into vascular endothelial cells before and after differentiation induced differential expression of eNOS/activity and its metabolites

Objective To investigate the differences in eNOS gene expression,activity and its metabolites before and after human bone marrow mesenchymal stem cells (hBMSCs) are induced into vascular endothelial cells.Methods hBMSCs were induced into vascular endothelial cells.The morphological changes of the cells were observed under inverted microscope.Transwell assay was used to detect the cells' migration ability.The protein expression of eNOS was detected by immunofluorescence and Western blot.The activity of eNOS was detected by ELISA and the content of NO in cell culture supernatant was determined by nitrate reduction method.Results Compared with those in the undifferentiated group,the morphological changes of the differentiated cells were obvious.Cell migration ability increased by 238.10% (73.000±7.002 vs.21.000±4.359,P<0.05).The expression of eNOS protein increased by 114.72% (0.423±0.011 vs.0.197±0.079,P<0.05).The activity of eNOS was enhanced by 157.49% (4.967±0.073 vs.1.929±±0.103,P<0.05).The synthesis and release of NO increased by 155.67% (184.909±1.853 vs.72.323±0.426,P<0.05).Conclusion After hBMSCs are induced into endothelial cells,the expression of eNOS gene increases,their activities increase,synthesis and release of the metabolite NO increase.It may provide a basis for prevention and treatment of cardiovascular diseases with stem cells.
4.Expression of Ezrin protein in nasopharyngeal carcinoma and its effect in metastasis of nasopharyngeal carci-noma

Objective To study the Ezrin protein in nasopharyngeal carcinoma expression and its effects on metastasis of nasopharyngeal carcinoma .Methods Expression of Ezrin protein was detected by immunohistochemical method in 98 cases with nasopharyngeal carcinoma and 37 cases of chronic nasopharyngitis tissues were analyzed . Clinical staging and pathological characteristics , recurrence and metastasis following treatment in the process of 98 patients with nasopharyngeal carcinoma were observed .Results 98 cases of patients with nasopharyngeal carcino-ma research group,69 cases(70.4%) patients were positive expression of Ezrin protein .The control group nasopha ryngeal mucositis of patients ,only 4 patients showed positive expression of Ezrin protein ,the rate of positive expression was 10.8%.The differences between the two groups compared with the positive rate of Ezrin significantly (χ2 =38.42,P=0.00).By comparison on strength of patients with distant metastases of different expression of Ezrin in study group,closely related to Ezrin strong positive and tumor metastasis , Ezrin protein ( ++) and ( +++) expression group and negative (-) and weak positive (+) expression group compared with patients ,tumor migration rate was significantly higher(all P<0.05).Conclusion Ezrin protein shows significantly higher expression in tis-sues of patients with nasopharyngeal carcinoma ,the positive expression of Ezrin protein in nasopharyngeal carcinoma and with the distant metastasis .
5.Preparation and releasing evaluation in vivo and in vitro of pH-dependent Yuchangning Tablet for colon-specific delivery

Objective To prepare the pH-dependent Yuchangning Tablet for colon-specific delivery(PYTCSD) used in treating the ulcerative colitis and evaluating the releasing property in vivo and in vitro.Methods The coating prescription was screened by the in vitro delivery of matrine and oxymatrine.The in vitro releasing property of the preparation was examined by the method of in vitro delivery. The in vivo releasing property of the preparation was evaluated by the shadowgraph technique of barium sulfate.Results The preparation method of the PYTCSD was obtained.The core of the tablet was coated by the alcohol solution mixed with 3.70%(g/mL) Eudragit Ⅲ,0.37%(g/mL) DEP,and 0.93%(g/mL) talcum power.The weight of the core was increased 8%.From the in vitro delivery,matrine and oxymatrine were not detected in the simulated gastric fluid after 2 h.The quantities of matrine and oxymatrine were less 10% in the simulated intestinal fluid after 4 h.The quantities of matrine and oxymatrine were 86.5% and 86.8% in the simulated colon fluid after 1 h.On the basis of the in vivo delivery by treating eight volunteers,the PYTCSD could completely get to the ileocecum or ascending colon and disintegrate in that part.Conclusion The PYTCSD can be prepared and the preparation is significantly delivered in the specific colon.
6.Synthesis and phosphodiesterase type-5 inhibitory activity of sildenafil analogues possessing a guanidine group in the phenyl ring

In this study,a series of new sildenafil analogues 11-27 possessing a guanidine group were synthesized to investigate their PDE5 inhibitory activity and selectivity using[~3H]cGMP SPA kit in vitro and efficacy in the rat model of erection.Most of the compounds showed potent activity against PDE5,and more importantly,several compounds exhibited higher PDE5 selectivity over PDE6 than that of sildenafil.Structure-activity relationship of these sildenafil analogues was also discussed.Within this series of compounds,compound 15(IC_(50) =1.7 nmol/L)not only exhibited more potent PDE5 inhibitory activity than that of sildenafil (IC_(50) = 6.5 nmol/L),but also showed functional efficacy in the rat model of erection.
7.Effects of miR-24 on Endothelial Nitric Oxide Synthase Gene Expression and Tube Formation in Human Umbilical Vein Endothelial Cells

Objective: To investigate the effects of miR-24 on endothelial nitric oxide synthase (eNOS) gene expression with regulation and endothelial cell proliferation, migration, tube formation in human umbilical vein endothelial cells (HUVECs). Methods: Constructed high expression plasmid of miR-24 and miR-24 antisense sequence were introduced into HUVECs and the cells included in 3 groups: Control group, miR-24 group and miR-24 inhibitor group. HUVEC proliferation was detected by MTT test, migration was measured by Scratching and Transwell methods, tube formation was examined by Matrigel assay; mRNA and protein expressions of eNOS and Sp1were determined by RT-PCR and Western blot analysis respectively. Results:①Compared with Control group, miR-24 group had decreased cell proliferation by 45.45% as (0.36 ± 0.04) vs (0.66 ± 0.08),P<0.05; miR-24 group had lower speed of cell migration, decreased number of cell migration by 74.75% as (30.25±3.78) vs (119.80±10.94),P<0.01 and there was no obvious tube formation.②Compared with Control group, miR-24 group showed reduced eNOS mRNA expression by 46.2% as (0.49±0.02) vs (0.91±0.01),P<0.05, reduced protein expression by 49.07% as (0.55±0.05) vs (1.08±0.05),P<0.05; meanwhile, decreased Sp1 mRNA expression by 44.9% as (0.49±0. 01) vs (0. 89±0.02)P<0.05, decreased protein expression by 54.90% as (0.46±0.02) vs (1.02±0.04),P<0.05. In miR-24 inhibitor group, the above indexes were lower than Control group but higher than miR-24 group, the amount of tube formation and the length of tubes were similar between Control group and miR-24 inhibitor group. Conclusion: MiR-24 may inhibit HUVECs proliferation, migration, tube formation and suppress eNOS expression; Sp1 might be one of the important regulators.
8.Effect of extracurricular exercise on college students’ physical health

Objective:
To research the effect of using the campus running APP for extra-curricular exercises on the physical health of college students and the habits formation of extracurricular exercises, and to provide a reference for improving their physcal health level and cultivating exercising habit.
Methods:
Through experimental method and questionnaire investigation, 1 327 students enrolled in the year 2016 were selected from Shanghai University of Finance and Economics by using cluster sampling method. Running App was used to track their extracurvicular exercises for 7 weeks, and the results were compared before and affer running.
Results:
The 56-kilometer extracurricular running had a significant difference in the male 1 000 m running index (P<0.05), and the average time spent decreased by 3.69 s. The female test scores in 50 m, forward bends, standing long jumps and sit-ups increased by 0.12 s, 1.83 cm, 2.12 cm and 2.28 respectively, showing statistical significance compared with before running(P<0.01). There was no statistically significant difference for general assessment of male physical test between after and before running (P>0.05); The average score of general assessment of female physical test was significantly higher than that before running, which is statistically significant (P<0.01).
Conclusion
Using the campus running APP for extracurricular exercises has a positive effect on cultivating students’ sports habits and enhancing their physical health.
9. Comparison of the effect of different nutritional testing methods on preoperative nutritional risk screening for patients with head and neck cancer

Objective:
To explore the role of bioelectrical impedance analysis (BIA) and laboratory tests for nutrition related indicators in preoperative nutritional risk screening of patients with head and neck cancer.
Methods:
Totally 131 patients with head and neck cancer were selected by convenient sampling method. The nutritional risk was screened by NRS 2002 nutritional risk screening scale. The bioelectrical impedance indexes were collected by human body component analysis instrument and the nutritional related laboratory indicators were collected by electronic medical records.
Results:
The incidence of nutritional risk in patients with head and neck cancer before operation was 18.6% (22/131). Correlation analysis showed that nutrition-related laboratory indicators were not related to nutritional risk, while bioelectrical impedance indicators such as fat-free mass, skeletal muscle mass, bodi mass index (BMI), inorganic salt content, water content and protein content were significantly correlated with NRS 2002 screening results(

Exploratory experiment is the key to the experimental physiology,which is universally applicable training of innovative talents.More importantly,it is an important way to cultivate innovative talents of medicine.On the basis of the original multi-subject evaluation system,we have formed a comprehensive evaluation system through refining the scoring item for teachers and students,enriching the forms of student evaluation,and building the student self-assessment.This comprehensive evaluation system is set to promote the development of experimental physiology,especially the exploratory experiments.The practice of this evaluation system effectively improve the objectivity of the evaluation,provide a basis for reform of exploratory experiment,and then promote the training of innovative talents.
