Objective:To develop a new type of microelectrode used to measure free fluoride in minim solution. Methods:F--ion sensitive field transistor( F--ISFET) microelectrode was developed based on H+-ISFET combined with an Ag/AgCl referential electrode of microtube. The F--ISFET was used to determine the free fluoride ions in saliva and dental plaque fluid in microliter. And its property was investigated. Results:The test limit of F--ISFET was 10 -6 mol/L-10 -1mol/L, the linear test limit was 10 -6mol/L-10 -4mol/L, the sensitivity was 30-55 mV/pF, the responding time was less than 30 seconds, r2 was 0.97, the reclamation was 94%-104%. Conclusion:F--ISFET can be used to measure free fluoride ions in dental plaque.

Objective To investigate the safety and feasibility of laparoscopic hepatic left lateral lobectomy. Methods Five cases of liver cancer, 4 cases of hepatic hemangioma, and 2 cases with suspected liver tumors were included in this series. Tumors were all located at left lateral lobe, with a diameter less than 10?cm. Tumors were resected by laparoscopy under a combination of pneumoperitoneum and non-pneumoperitoneum, with hand assistance through a mini-incision. Several manoeures were used for the dissection of liver tissue, including microwave coagulation, ultrasonic dissection, tainium clips clamping, rotating suction, endogia ligation. ResultThe procedure was successful in all cases with the operation time ranging from 1.5?h to 6?h, and bleeding was 50?ml to 500?ml. The hospitalization time were from 5?d to 15?d. ConclusionLaparoscopic liver resection is safe and feasible for the left liver lateral lobe tumor when the diameter is less than 10?cm, and not involving surrounding organs.

Objective To analyse effect of Erigeron breviscapus oral liquid on peripheral blood CRP and MMP-9 in patients with acute coronary syndrome.Methods 56 patients who were diagnosed with acute coronary syndrome were collected.All patients were randomly divided into experimental group and control group, 28 cases in each group.Patients in control group received conventional treatment with western medicine, patients in experimental group were given Erigeron breviscapus oral liquid of oral treatment on the basis of control group treatment, after the treatment, the serum levels of CRP,MMP-9 and clinical curative effect were detected in all patients.Results Compared with before treatment, the levels of CPR and MMP-9 in two groups decreased after the treatment(P<0.05); Compared with control group,the serum level of CRP was lower in experimental group(P<0.05); the serum level of MMP-9 was lower in experimental group(P<0.05);the total effective rate was higher in experimental group(χ2 =4.08,P<0.05).Conclusion Erigeron breviscapus oral liquid can significantly reduce the serum levels of CRP and MMP-9 in patients with acute coronary syndrome,improve clinical therapeutic effect.

[ABSTRACT]OBJECTIVETo investigate the cytomegalovirus infection in neonates, characteristics of gap junction protein Connexin26 gene mutation and the hearing follow-up results, and to analyze their correlations. METHODS60 CMV-DNA positive and 40 CMV-DNA negative neonatal newborn from The Second Affiliated Hospital of Wenzhou Medical University and The first people's Hospital of Yongkang were screened, the blood biochemistry was analyzed, and the umbilical cord blood was reserved to detect the Connexin26 gene expression of mRNA with RT-PCR.PCR results was sequenced to track the newborn hearing, and analyze the correlations between neonatal cytomegalovirus types, the mutation of Connexin26 gene and hearing test results.RESULTS 26 cases from 60 CMV-DNA positive newborns were found with blood biochemical abnormalities. In all of the newborns, a total of 41 cases had 235delC mutation, 11 cases in the mutations for the development of hearing impairment. The results of correlation analysis showed that there were correlations between cytomegalovirus infection, gene mutation and hearing impairment.CONCLUSION Cytomegalovirus infection in neonates can lead to mutations in the Connexin26 gene, and may further lead to hearing impairment, and the probability of the mutation of Connexin26 gene and sensorineural hearing loss were higher in symptomatic cytomegalovirus infection neonates.

Objective To evaluate the procedure of diagnosis and treatment for cystitis glandularis in female. Methods Forty-seven patients were classified into 3 groups randomly after being operated by tuansurethral resection:15 cases without any other therapy(A group), 16 cases with bladder instillation of pirarubicin afterward (B group), 16 cases with bladder instillation of mitomycin afterward (C group). Symptomatic reliof and recurrence were investigated in 1-year follow-up visit. Results Forty-four cases had fulfilled the entire follow-up visit. The effective rates of the three groups were 38.5%,87.5% and 86.7% respectively (X2=13.80,P＜0.01 ), the recurrence rate was 30.8% ,0,13.3% respectively (X2=4.86,P＞0.05).Conclusions The clinical situation of cystitis glandularis has no specificity. Cystisis glandularis has diversification of the clinical symptoms, urodynamics. Transurethral resection with bladder instillation is more effective than transttrethral resection simply, the recurrence rate is lower.

Injectable lipid emulsions have been routinely used in patients since 1960s as a nutritional supplement for patients requiring parenteral nutrition. In recent years, lipid injectable emulsions have been extensively studied as a kind of novel drug carrier, also the quality problems of the lipid emulsion attract more and more attentions gradually. Large diameter tail of injectable lipid emulsions as a significant quality control indicator should pay more attention. Regarding to the defect of detecting large diameter tail of lipid injectable emulsions in our country, the purpose of this article is to summarize the techniques of detecting large diameter tail, illustrate the impacts of large lipid droplet on the quality of lipid injectable emulsions, emphasize the importance of detecting large diameter tail in lipid emulsions and provide guidance for researching and developing lipid emulsions in domestic market.

Objective:To study the psychosocial effect of fluorosis on students with dental fluorosis.Methods:115 students with different types of fluorosis were sampled randomly from fluorosis epidemic area of Xingtai city in Hebei province, including 60 of 12-year-old and 55 of 15-year-old students. Dental fluorosis was classified by Dean's Index (DI). Parents and teachers of the selected students were involved in a psychosocial questionnaire investigation which included five five-point questions, respectively.Results:Prevalence of fluorosis was 89.6% in the young students in the area. Evaluation scores by parents, which included “health of teeth”, “satisfaction of teeth” and “perceived well-being”, decreased with the decline of DI. Statistical difference was found in the answer to the five questions by parents when comparing the evaluation scores of control(without dental fluorosis) group,moderate dental fluorosis group and severe dental fluorosis group(P0.05). Difference of the evaluation scores of parents and teachers on boys and girls was not significant.Conclusion:Fluorosis has psychosocial impact on students, especially evaluated by parents.

Objective To construct the secretory expression vector of recombinant human C-reactive protein(rhCRP) for its se-cretory expression in Pichia pastoris ,rhCRP was expressed as a secretory protein and purified ,and the immunity reactivity of the purified protein was identified .Methods The DNA fragment of rhCRP which was designed and synthesized was cloned into pPICZαA vector .Recombinant plasmid pPICZαA/rhCRP was linearized by SacⅠand transformed into Pichia pastoris X-33 by elec-trotransformation .The rhCRP was secreted into the medium under the methanol induction .RhCRP was purified by Histamine affin-ity chromatography .The purified rhCRP was identified by SDS-PAGE and Western blotting ,and its immunity reactivity and stabili-ty was identified by indirect ELISA .Results The pPICZαA/rhCRP expression vector was successfully constructed .The rhCRP of 23 × 103 was inducted and successfully expressed as a secretory protein by the recombinant Pichia pastoris strains .The rhCRP was purified by one step up to 90 .42% purity ,and it was showed good immunity and stability by indirect ELISA .Conclusion The rh-CRP with higher purity and immunoreactivity was successfully obtained by using the Pichia pastoris expression system ,which pro-vided an important experimental basis for producing anti-human CRP antibodies and developing testing CRP reagent .

Objective To establish a HPLC method for the assay of psoralen and isopsoralen in different effective extracts of Fructus Psoraleae. Methods HPLC was carried out on the column of Kromasil RP-C18. The mobile phase was methanol -water(65 ∶35). The flow rate was 1.0 mL/min and the UV detection wavelength was 245 nm. Results Good linearity of psoralen was showed within the range of 10.5 ng～525 ng(r= 0.999 3)and isopsoralen within the range of 9 ng～450 ng (r= 0. 999 9). The content of psoralen and isopsoralen differed in different extractions of Fructus Psoraleae. Among them,the extract C (extracted by ethyl acetate ) contained the highest contents of psoralen and isopsoralen,while the contents of psoralen and isopsoralen were very low in the extract D (extracted by n-butyl alcohol) and E (supernatant of water extract). Conclusion The method is simple,accurate and reproducible. The anti-asthma effect and the dose-effect relationship of the different effective extracts of Fructus Psoralea need further pharmacodynamics study.
