1.Determination of Impurities in High Pure Neodymium Oxide with Inductively Coupled Plasma Mass Spectrometry

A method for the determination of impurities in high pure Nd_2O_3 employing inductively coupled plasma mass spectrometry(ICP-MS) was established, and emphasise was put on the elimination of matrix induced NdO~+ and NdOH~+ spectral interferences using cell collision technology(CCT). Using 10% O_2-10% Ar-80% He as the reaction/collision gas and with proper adjustment of the instrumental parameters, ~(159)Tb,~(165)Ho,~(163)Dy could be quantified at ~(175)TbO,~(179)DyO and ~(181)HoO respectively, while the matrix induced NdO~+ and NdOH~+ spectral interferences could be reduced by 25-70 folds. In addition, different elements were tested as internal standards for matrix effect correction under the CCT mode, and it was found that Re was best. Other impurities including La, Ce, Sm, Eu, Gd, Er, Tm, Yb and Lu were detected under conventional mode, except that Pr was detected under high instrumental resolution mode to eliminate peak tailing interference from Nd. In combination with mathematic correction, the established method can be applied for the determination of impurities in 99.999% Nd_2O_3 products with satisfactory RSDs and recoveries.
2.Determination of six heavy metals in peanuts using microwave-assisted digestion-inductively coupled plasma mass spectrometry with an octopole reaction system

Objective :
To establish a microwave-assisted digestion-inductively coupled plasma mass spectrometry (ICP-MS) with an octopole reaction system for simultaneous determination of six heavy metals in peanuts, including Cr, Ni, As, Cd, Pb, Hg.
Methods :
Peanut samples were shelled and crushed evenly, and 0.350 0 g was accurately weighed and digested with 5 mL nitric acid and 1 mL hydrogen peroxide in a digestion tank. Following microwave-assisted digestion, pure water was used to quantify the samples, and internal standards and an octopole reaction system were used to remove the interference. Then, the contents of six heavy metals were determined in peanuts by ICP-MS. The accuracy and precision of ICP-MS were evaluated using national criteria ( GBW 10013 and GBW 10044 ) and spike-and-recovery testing.
Results :
The six heavy metals showed good linearity at the selected linear range ( r≥0.999 8 ). The detection limits of ICP-MS ranged from 0.001 4 to 0.023 8 ng/mL, and the spike-and-recovery rates ranged from 94.7% to 98.8%, with the relative standard deviations ranging from 0.7% to 3.6%. In addition, the determination results of the standard reference materials were all within the normal reference range. The detection of six heavy metals was 100.0% in 60 peanut samples, and the contents of six heavy metals were all low.
Conclusion
The established ICP-MS assay is feasible for simultaneous determination of multiple heavy metals in peanuts.
3.Intervention effect of Bushen-Yixin tablet on the level of NOS, NO and Ang (1-7) in spontaneously hypertensive rat

Objective To study the variation of the level of NOS,NO and Ang (1-7) in spontaneously hypertensive rat(SHR).Methods SHR were divided randomly into 3 groups:Bushenyixin group(n=8),valsartan group (n=8),and model group (n=7).Each group was given Bushenyixin tablet,valsartan,and saline by gavage respectively.After 4 weeks,the concentration of NOS,NO,Ang (1-7) in plasma and the concentration of NOS,NO in aorta were be observed.Results The concentration of NO in plasma was significantly lower in the Bushen-Yxin group than for model group(15.500±4.375)μmol/L vs.(26.000±2.828)gmol/L,P<0.05.The concentration of NO in aorta and Ang1-7 in plasma were significantly higher in the Bushen-Yixin group than in the model group [(35.604±7.505)μmol/L vs.(17.437± 11.649) μ mol/L,(45.542 ± 12.726)ng/L vs.(25.521 ± 2.727) ng/L,P<0.05].The concentration of NO in aorta was significantly higher in Bushen-Yixin group than in the valsartan group [(21.537± 13.484) μmol/L,P<0.05].Conclusion Bushen-Yixin tablet had effects of increasing of NO level in aorta and Ang (1-7) level in plasma in spontaneously hypertensive rat.
4.Determination of Residual Solvent DMF in Imiquimod by HS-GC

Objective: To establish a new method for determining residual organic solvent DMF in imiquimod. Methods: The samples were injected into an Agilent HP-PLOT/Q capillary column(30 m × 0. 530 mm,40. 0 μm) by a headspace sampler and ana-lyzed with an FID detector, the carrier gas was nitrogen, the inlet temperature was 250℃, and the detector temperature was 270℃. The column temperature was programmed raised. Results: The resolution among the peaks of DMF and the other residual solvents could meet the requirements. There was a good linearity within the experimental concentration range. The average recovery was 94. 6%(RSD=4. 0%, n=9). The limit of quantification and the limit of detection was 4. 809μg·ml-1 and 0. 963μg·ml-1, respectively. Conclusion:The method is convenient, accurate and sensitive, which can be used in the determination of residual solvent DMF in imi-quimod.
5.The analysis of smooth pursuit test in unilateral vestibular peripheral vertigo

Objective:To analyze the result of smooth pursuit test(SPT) in unilateral vestibular peripheral vertigo and investigate its influencing factors.Method:Smooth pursuit test (SPT) and spontaneous nystagmus (SN)were examined in one hundred and eighty-five patients with unilateral peripheral vertigo(case group) and 51 normal persons(control group) by Video-Nystagmography (Synapsys, France), and the gain of SPT and SN were selected as the observation parameters in order to analyze the waveform and gain of SPT and the relativity between SN and the gain of SPT.Result:Of the 185 patients, 105(56.8%),72(38.9%) and 8(4.3%) cases producedⅠ,Ⅱ and Ⅲ waveforms respectively. Of these patients, 58(31.4%) demonstrated SN and none had Ⅳ waveform. While of 51 normal persons, 38(74.5%), 13(25.5%) persons producedⅠand Ⅱwaveforms repectively and there were no Ⅲ, Ⅳ waveforms or SN. There was statistical significance between the stong and weak gain of SPT in these two groups. Weak gain was significantly different between two groups. The stong and weak gain of SPT in case group were 0.86±0.06,0.80±0.06; 0.78±0.09, 0.65±0.1; 0.68±0.13, 0.45±0.12. The relativity between SN and the gain of SPT was positive when they had same direction(r_s=-0.63,P<0.05)and negative when opposite (r_s=0.34,P<0.05).Conclusion:Ⅰ,Ⅱ,Ⅲ three waveforms of SPT could appear in unilateral vestibular peripheral vertigo and the corresponding gains are gradually decreasing.SN is the influencing factor of SPT.

There was statistical significance between the strong and weak gain of SPT in these two groups. Weak gain was significantly different between two groups. The strong and weak gain of SPT in case group were 0.86 +/- 0.06, 0.80 +/- 0.06; 0.78 +/- 0.09, 0.65 +/- 0.1; 0.68 +/- 0.13, 0.45 +/- 0.12. CONCLUSION
I , II, III three waveforms of SPT could appear in unilateral vestibular peripheral vertigo and the corresponding gains are gradually decreasing. SN is the influencing factor of SPT.

7.SOCS3 Attenuates GM-CSF/IFN-γ-Mediated Inflammation During Spontaneous Spinal Cord Regeneration.

SOCS3, a feedback inhibitor of the JAK/STAT signal pathway, negatively regulates axonal regrowth and inflammation in the central nervous system (CNS). Here, we demonstrated a distinct role of SOCS3 in the injured spinal cord of the gecko following tail amputation. Severing the gecko spinal cord did not evoke an inflammatory cascade except for an injury-stimulated elevation of the granulocyte/macrophage colony-stimulating factor (GM-CSF) and interferon gamma (IFN-γ) cytokines. Simultaneously, the expression of SOCS3 was upregulated in microglia, and unexpectedly not in neurons. Enforced expression of SOCS3 was sufficient to suppress the GM-CSF/IFN-γ-driven inflammatory responses through its KIR domain by attenuating the activities of JAK1 and JAK2. SOCS3 was also linked to GM-CSF/IFN-γ-induced cross-tolerance. Transfection of adenovirus overexpressing SOCS3 in the injured cord resulted in a significant decrease of inflammatory cytokines. These results reveal a distinct role of SOCS3 in the regenerating spinal cord, and provide new hints for CNS repair in mammals.
8.Baitouweng Tang Inhibits Growth of Esophageal Cancer Cells Through BUB1/STAT3 Signaling Pathway

ObjectiveTo investigate the mechanism of Baitouweng Tang in inhibiting the growth of esophageal cancer (EC) cells by regulating budding uninhibited by benzimidazoles 1 (BUB1)/signal transducer and activator of transcription 3 (STAT3) signaling pathway. MethodGene chip technology was used to explore the differential gene expression between esophageal cancer tissues and normal tissues and identified differentially expressed genes. The differentially expressed genes were analyzed by bioinformatics methods. EC cells were treated with 25, 50, 100, 200, 400, 800 mg·L-1 Baitouweng Tang. EC cell viability was detected by Thiazolyl Blue (MTT) colorimetry. Cell cycle and apoptosis were measured by flow cytometry. The expression of BUB1 was measured by real time quantitative polymerase chain reaction (Real-time PCR). The protein levels of BUB1, STAT3, phosphorylated (p)-STAT3, Cyclin B1 (CCNB1), cyclin-dependent kinase 1 (CDK1), B-cell lymphoma-2 (Bcl-2), cysteinyl aspartate-specific proteinase(Caspase)-3, and Caspase-9 were measured by Western blot. The migration and invasion abilities of the cells were measured by wound-healing and Transwell invasion assays. ResultDifferentially expressed genes were primarily involved in biological processes, signaling pathways, and network construction related to cell mitosis, with BUB1 identified as a key core gene. Compared with the control group, Baitouweng Tang inhibited BUB1 expression (P<0.05,P<0.01). In vitro experiments showed that compared with the control group, Baitouweng Tang could significantly inhibit the growth (P<0.05,P<0.01), migration and invasion (P<0.05,P<0.01) of EC cells, induce apoptosis (P<0.05,P<0.01), and cause G2/M phase increase (P<0.01). After treatment with Baitouweng Tang, compared with the results in the control group, the expression of Caspase-3, and Caspase-9 in EC cells increased significantly (P<0.05,P<0.01), while the expression of Bcl-2, BUB1, CCNB1, and CDK1 decreased significantly (P<0.05,P<0.01). Moreover, the STAT3 signaling pathway was also found to play an important role in this process. ConclusionBaitouweng Tang may inhibit the growth of EC cells by downregulating BUB1 and mediating the STAT3 signaling pathway.
9.miR-495-3P targets BUB1 to regulate the effect of STAT3 signaling pathway on biological behavior of esophageal cancer cells

Objective :
To investigate the effect of miR-495-3p targeting budding uninhibited by benzimidazoles (BUB1) on signal transducer and activator of transcription 3 (STAT3) signaling pathway on biological behavior of esophageal cancer (EC) cells.
Methods :
The differentially expressed genes in EC tissues and normal tissues were screened by the cDNA microarray technique. The differentially expressed genes were analyzed by bioinformatics methods.The target genes of miRNAs were predicted by the TargetScan database and verified by a dual luciferase gene reporter assay.KYSE150 cells were divided into blank control group,NC mimics group and miR-495-3p mim- ics group.The activity of KYSE150 cells were detected by the CCK-8 method.Cell cycle and apoptosis were meas- ured by flow cytometry.The expression of BUB1 mRNA was measured by real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-qPCR) .The levels of BUB1,STAT3,phosphor (p) -STAT3,cyclin dependent kinase 1 ( CCNB1) ,cyclin dependent kinase 1 ( CDK1) ,B-cell lymphoma-2 (Bcl-2) ,cysteinyl aspar- tate-specific proteinase-3 ( Caspase-3) and cysteinyl aspartate-specific proteinase-3 ( Caspase-9) were measured by Western blot.The migration and invasion abilities of the cells were measured by wound-healing and Transwell inva- sion assays.
Results :
Differentially expressed genes were involved in biological processes,signaling pathways and network construction,which were mainly related to cell cycle.BUB1 is the key (Hub) gene,and BUB1 is the tar- get gene of miR-495-3p.In vitro experiments showed that overexpression of miR-495-3p could significantly inhibit the migration and invasion of EC cells and induce apoptosis and G2 / M phase arrest.After treatment with overex- pression of miR-495-3p,the expression of Caspase-3 and Caspase-9 in EC cells increased significantly(P<0. 01) , while the expression of Bcl-2,BUB1,CCNB1,CDK1 and p-STAT3 decreased significantly(P<0. 01) .Moreover, the STAT3 signaling pathway might play an important role in this process.
Conclusion
miR-495-3p may influence the biological behavior of esophageal cancer cells by down-regulating BUB1-mediated STAT3 signaling pathway.
