Objective To study the expression and clinical significance of ARHGAP4 in colorectal cancer Method Real?time PCR,Western blot and immunocytochemistry were used to detect the expression of ARHGAP4 in colorectal cancer tissues and cell lines ,and the correlation between its expression and clinical features of patients was analyzed Results ARHGAP4 is overexpressed in colorectal cancer tissues and cell lines and its overexpression is correlated with T stage, N stage, clinical stage, and metastasis. Conclusion ARHGAP4 may promote the progression of colorectal cancer ,and have the potential to be a novel prognosis marker.
2.The expression level of miR-125a-5p in colorectal cancer with various differentiation

Objective To identify the expression level of miR-125a-5p in colorectal cancer (CRC) with various differentiation,and to investigate its diagnostic significance.Methods Real-time polymerase chain reaction was used to analyze the expression level of miR-125a-5p in tumors and paired normal tissues of different differentiated CRC.Receiver-operating-characteristic (ROC) curve was performed to evaluate the specificity and sensitivity of using miR-125a-5p as biomarkers to discriminate CRC patients from normal persons.Results Compared with normal tissues,miR-125a-5p was down-regulated in both high and moderate differentiation,miR-125a-5p could distinguish CRC from normal persons with high sensitivity and specificity,which were 80.00 % and 90.00 %,respectively.Conclusions miR-125a-5p down-regulated in CRC indicates that miR-125a-5p contributes to tumorigenesis,and may be a potential biomarker for CRC.
3.Isolation and purification of pig islet cells

Objective To establish effective method for large-scale purification of islet cells from pig pan-cress. Methods Pig pancreas tissue was digested with collagenase P followed by purification in a HCA-Fi-coil dis continuous gradient using Cobe2991 cell separator. After isolation, the islet cell yield and purity were evaluated with light microscope with DTZ staining, and the islet function assessed by insulin release as-say in vitro. Results The number of the islets coll ected from each pancreas averaged (275 000±20 895)islet equivalents (IEQ) before purification, and (230 350±26 679) IEQ after the purification with discon-tinuous gradient centrifugation. From each gram of the pancreatic tissue, (2710±229) IEQ were obtained with an average purity of (50.2±1.95) %. The purified islets responded well to high-concentration (16.7 mmol/L) glucose stimulation with a 4. 74-fold increase of insulin secretion over the basal level (3.3 mmol/L, P <0.001). Conclusion The established method can be applicable for large-scale purifi-cation of fully functional islet cells from pig pancreas.
4.Effects of different dose of clopidogrel on serum CD62p level in patients with coronary heart disease

Objective To investigate the change of serum CD62p of coronary heart disease (CHD) patients after different doses of clopidogrel administration.Methods One hundred and ninety-one patients with CHD were selected as our subjects.Of which,95 cases were with SAP and 66 cases were with non ST segment elevation acute coronary syndrome (NST-ACS).SAP patients were randomly given clopidogrel at dose of 75 mg/d or 150 mg/d and served as A and B groups.NST-ACS patients were randomly given 300 mg clopidogrel,then randomly divided into C and D groups with sequentially taking clopidogrel at dose of 75 mg/d or 150 mg/d respectively.Thirty healthy peoples were served as E group without drug intervention.Concentrations of serum CD62p were detected by Elisa before taking clopidogrel,24 h and the fifty day of after taking clopidogrel.Results (1) Before taking clopidogrel,the serum concentrations of CD62p in CHD patients were higher (A group:(7.62 ± 2.99) ng/L,B group:(8.48 ± 3.13) ng/L,C group:(9.50 ± 3.32) ng/L,D group:(10.22 ± 5.14) ng/L than that of healthy control group ((5.49 ± 1.99) ng/L,P < 0.05).The Serum CD62p levels in SAP patients were lower than that of NST-ACS patients (P < 0.05).(2)The serum concentrations of CD62p in A and B groups at before taking clopidogrel were (7.62 ±2.99) ng/L and (8.48 ±3.13) ng/L respectively,higher than that four days after taking clopidogrel ((6.79 ± 2.51) ng/L,(6.37 ± 1.80) ng/L;t =2.390,4.520;P <0.05 or P <0.01).There was no statistical significant difference between A and B groups(P >0.05).(3) In C and D groups,the serum CD62p at before taking clopidogrel were (9.50 ±3.32) ng/L and (10.22 ±5.14) ng/L,higher than that after taking clopidogrel for four days ((8.21 ± 2.62) ng/L,(8.17 ± 2.37) ng/L; t =2.084,2.157 ; P < 0.05).No significant difference was seen between C and D groups (P > 0.05).Conclusion The serum CD62p in patients with CHD was higher than that in the healthy control.Clopidogrel administration can decrease serum CD62p in CHD patients.
5.Genomics analysis of miRNA in colon cancer tissues.

OBJECTIVETo examine the differential expression of miRNAs in colon cancer tissues and matched tumor adjacent tissues.
METHODSDifferential expression of microRNAs in twenty paired human colon cancer tissues and adjacent tissues were detected by QPCR. miRNAs with significantly differential expression (fold change >2.4 and P<0.01) were screened to analyze their accumulation by Cluster analysis. Thus correlation of miRNA and other colon cancer-associated proteins was examined.
RESULTSExpressions of 17 miRNAs were significantly reduced in colon cancer tissues. Cluster analysis showed that miR763-3, miR451 and miR99a had similar expression. Plasma CK20 level was negatively correlated with miR100 (r=-0.948), miR152a-5p (r=-0.948), miR125b (r=-0.949), miR145 (r=-0.949) and miR145*(r=-0.949) (all P<0.05).
CONCLUSIONmiR145 and other 16 miRNAs may be used as diagnostic molecular markers of colon cancer, and miR100, miR125a-5p, miR125b, miR145 and miR145* may become the molecular markers of colon cancer lymph node metastasis.

6.The teachers′ experience of nursing practice teaching collaboration between Guangdong and Macao under the background of Guangdong-Hong Kong-Macao Greater Bay Area

Objective:To explore the experience of teachers from Guangdong and Macao in nursing teaching cooperation, the existing problems in current teaching cooperation and suggestions for improvement.Methods:From January to April 2021, using descriptive qualitative method to conduct in-depth among eight clinical tutors from The Third Affiliated Hospital of Guangzhou Medical University who teach Macao nursing students, and four teachers from Kiang Wu Nursing College of Macao using the purposive sampling method. And adopted content analysis for data analysis.Results:A total of 4 themes and 2 sub-themes were analyzed: the positive impact of cross-border teaching cooperation projects including developed the nursing business of the two places and deepened the cooperative relationship between the two places; limiting the depth and breadth of knowledge transfer because of the short cross-border learning time; the imbalance between students′ abilities and teachers′ expectations; expectations for homogeneous internships for heterogeneous groups.Conclusions:Cross-border nursing teaching cooperation is an important promoter for the development of nursing education between Guangdong and Macao, but there are still deficiencies in cooperation, and it is necessary for the two places to strengthen the construction of a sharing platform for teaching resources to promote the development of nursing education in the Guangdong-Hong Kong-Macao Greater Bay Area.
7.Study on miR-490-5p and miR-363 as novel biomarkers for the diagnosis of colorectal cancer.

OBJECTIVETo identify dysregulated microRNA(miRNA) that can act as novel biomarker for colorectal cancer screening.
METHODSReal time-polymerase chain reaction was used to detect the expression profile of miRNA in tumor and paired normal tissues, and the significant dysregulated miRNA was examined by non-paired t-test. Receiver operating characteristic(ROC) curve was performed to evaluate the specificity and sensitivity of miR-363 and miR-490-5p as biomarkers to discriminate colorectal cancer patients from normal person.
RESULTSSeven-three dysregulated miRNAs were found in male samples, of which 6 miRNAs were up-regulated and other 67 miRNAs were down-regulated, while 42 dysregulated miRNAs were found in female samples, of which 5 were up-regulated and 37 were down-regulated in tumor tissues compared with normal tissues. Among above dysregulated miRNAs, 33 miRNAs had significant differences, besides, dysregulated expression level of 10 of 33 miRNAs was over 5 folds in male and female as well as mixed samples.
CONCLUSIONSThe expression pattern in female is different from that in male. miR-363 and miR-490-5p possess the potential in screening colorectal cancer patients from healthy people.

8.Role of centromere protein H in human gastric cancer cell proliferation.

OBJECTIVETo explore the role of centromere protein H (CENP-H) in the proliferation of human gastric cancer cells.
METHODSRT-PCR and Western blot analysis were employed to examine the mRNA and protein expressions of CENP-H in 7 human gastric cancer cell lines and immortalized human gastric epithelial cells (GES-1). The cells were infected with the retrovirus vectors pMSCV-CENP-H or CENP-H-RNAi to establish stable cell lines with high CENP-H expression or CENP-H expression interference. MTT assay and colony formation assay were used to examine the changes in the cell proliferation after the infection.
RESULTSCENP-H was over-expressed in gastric cancer cell lines AGS, BGC823, SGC-7901, MKN45, HGC27, MGC-803 and MKN28 at both mRNA and protein levels. The established AGS/CENP-H cell line with increased CENP-H expression showed enhanced proliferative activity, while the cell line MGC-803/CENP-H-RNAi with CENP-H expression interference showed an obviously lowered proliferation ability.
CONCLUSIONCENP-H promotes the proliferation of human gastric cancer cells, suggesting its important role in the occurrence and development of gastric cancer.

9.Effect of remimazolam on anesthetic efficacy of painless bronchoscopy in elderly patients

Objective To investigate the effect of remimazolam on anesthetic effect and postoperative cognitive function during painless bronchoscopy in elderly patients.Methods A total of 90 patients with painless bronchoscopy admitted to Wenzhou People's Hospital were selected.And randomly dividing into 45 patients in control group,45 patients in observation group.The control group was given propofol,and the observation group was given remimazolam.Mean arterial pressure(MAP),heart rate(HR),and blood oxygen saturation(SpO2)were monitored.Effective time after induction,recovery time after withdrawal and discharge time were compared.Patient's cognitive function was assessed using the mini-mental state examination(MMSE),auditory word learning test(AVLT),shape connection test(STT),and animal language fluency test(AFT).Enzyme linked immunosorbent assay(ELISA)was used to determine substance P(SP),C-reactive protein(CRP),noradrenaline(NE),tumor necrosis factor-α(TNF-α)in serum,interleukin(IL)-6 and prostaglandin E2(PGE2)concentrations.Results Compared with the control group,the MAP of the observation group patients at time points T2 and T3 showed statistically significant differences(P<0.05);HR was especially significant at T2,T3 and T6 periods(P<0.05).SpO2 at T2 and T3(P<0.05).The awakening time,discharge time of the observation were shorter than control(P<0.05).At 1 day after surgery,the MMSE score,STT score,AFT score were significantly higher(P<0.05);The serum levels of SP,PGE2 and NE and IL-6,TNF-α and CRP were decreased in the control(P<0.05).The incidence of adverse reactions was 17.78%in control group and 8.89%in observation group(χ2=7.654,P=0.031).Conclusion Remazolam is used in painless bronchoscopy in elderly patients,intraoperative hemodynamics is stable,having little impact on postoperative cognitive function,and inhibits the release of inflammatory factors and the secretion of pain mediators,which is worthy of clinical use.
