1.Inhibiting effect of ethanol extract from peanut root on rat prostate hyperplasia and its influence on Bcl-2 and Bax protein

Objective To explore the inhibitory effect of the ethanol extract from peanut root on the non-castration adult rat prostate hyperplasia induced by testosterone propionate and its mechanism .Methods 60 SD rats were randomly divided into the control ,model and high ,middle and low dose of peanut root ethanol extract treatment groups ,among them ,the control group was normally fed without medication ;the model group was subcutaneously injected by testosterone propionate (TP ,5 mL/kg/d) and simultaneously gavaged subcutaneous injection with peanut root ethanol extract (10 mL/kg/d ,5 mLg/kg/d or 1 mL/kg/d) for suc-cessive 14 d .The rats in various groups were killed on 15 d and their prostate ,spermatophore and testicle tissues were separated and weighed .The ratio of gland/body-weight(mg/g) was calculated .The expressions of Bcl-2 and Bax proteins in the prostate tissues were detected by immunohistochemistry .Results There was statistical difference in the indexes of prostate between the treatment groups(high and middle dose) and the model group(P<0 .01) ,while there was no statistical difference between the low dose group and the model group(P>0 .05) .This result showed that there was a dose-effect relationship between the inhibition effect on rat prostate hyperplasia and the ethanol extracts from peanut root .There was no statistical differences between the model group and the treatment groups in the indexes of testicle ,spermatophore or the change of weight (P>0 .05) .There was statistical difference be-tween the control group or model group and the treatment group in the expression of Bcl-2 or Bax protein(P<0 .05) ,while no sta-tistical difference between the treatment group and the control group (P>0 .05) .Conclusion Ethanol extract from peanut root has good inhibiting effect on prostate hyperplasia and its mechanism could reach the curative effect by regulating the balance relation be-tween apoptosis gene Bcl-2 and Bax protein proportion and promoting apopitosis of benign prostatic hyperplasia .
2.Expression of matrix metalloproteinase-2 in patients with renal fibrosis and its clinical significance

Objective To study the change of matrix metalloproteinase-2 (MMP-2) in the development of renal fibrosis and evaluate the diagnostic value of blood MMP-2 level in renal fibrosis.Methods Thirty three patients with chronic glomerulonephritis were divided into proliferation group(n=18) and fibrosis group(n=15) according to renal fibrosis degree in renal pathology.Another 8 healthy subjects were used as control group.The levels of pro-MMP-2 and MMP-2 in serum were measured with the method of SDS-PAGE enzymography.Then the levels of pro-MMP-2 and MMP-2 in serum were compared between various groups.Results The serum MMP-2 level in proliferation group was much higher than that in control group(P0.05).Conclusion There is a significant difference of serum MMP-2 between suffering group and normal control group.Therefore the detection of serum MMP-2 level can be conducive to understand the development of disease and estimate prognosis.
3.Role of TLR9 in pathogenesis of glomerulonephritis

Objective To investigate the role of TLR9 in the glomerulonephritis through observing the changes of Toll-like receptor 9 in the glomerulo nephritis kidney tissue with or without CpG-ODN stimulation .Methods Wistar male rats were randomly divided into normal group(N),nephritis model group(M),CpG-ODN group(CpG) and GpC-ODN group(GpC).The urine samples were collected at 2,4,6 and 8 weeks after treatment,respectively.Blood samples were collected at the end of the last urine sample was collected,and the kidney tissue was collected,then the rats were killed.24 h urine protein was measured by Coomassie light blue technique.Serum album and renal function were determined by serological method.The pathologic changes of kidney were observed by light microscope and NF-?B p65 expression was detected using immunohistochemystry,RT-PCR was performed to detect the expressions of TLR9,INF-? and IL-6 mRNA.Results The expression of TLR9 was lighter in group M,and significantly increased after CpG-ODN stimulation compared with group M.Furthermore,24 h urine protein excretion was markedly increased,serum album was markedly decreased.The histopathologic changes of kidney were more severe.The mesangial cells(MCs) proliferated diffusifully in midrange and wide range,some of the glomeruli formatted cellularity crescent,micrangium loop was limitted,mononuclear macrophile cells were seen in the mesangial region.Conclusion Inflammatory factors mediated by TLR9 can deteriorate the biochemical and histopathologic changes.The immunologic reaction mediated by TLR9 is one of the mechanisms for the glomerulonephtitis' progression.
4.Design of low-intermediate frequency electrotherapy and pain assessment system.

Aiming at the single treatment and the design separation between treatment and assessment in electrotherapy equipment, a kind of system including low-intermediate frequency treatment and efficacy evaluation was developed. With C8051F020 single-chip microcomputer as the core and the circuit design and software programming used, the system realized the random switch of therapeutic parameters, the collection, display and data storage of pressure pain threshold in the assessment. Experiment results showed that the stimulus waveform, current intensity, frequency, duty ratio of the system output were adjustable, accurate and reliable. The obtained pressure pain threshold had a higher accuracy (< 0.3 N) and better stability, guiding the parameter choice in the precise electrical stimulation. It, therefore, provides a reliable technical support for the treatment and curative effect assessment.

5.Extraction Technology Optimization for Qige Granule by Orthogonal Design

Objective:To optimize the extraction technology of Qige granule. Methods:An HPLC method was established to de-termine the contents of astragaloside IV and puerarin in the extract of Qige granules. Based on the orthogonal design of L9(34),three factors including the water amount,decocting duration and extracting times were evaluated. Results:The best extraction process was as follows:adding 12-fold amount of water and extracting 3 times with 30 min for each time. Conclusion:The extraction technology opti-mized by the components evaluation is reasonable and practicable.
6.Multiple organ dysfunction syndrome due to ingestion of fish gall bladder.

OBJECTIVETo investigate changes in renal function, urine N-acetyl-beta-D-glucosaminidase enzyme (N-AG),liver function, myocardial enzymes, the pathology of renal damage and the mechanism of acute renal failure (ARF) associated with fish gall bladder poisoning.
METHODSEleven patients with acute fish gall bladder poisoning were consecutively admitted to our hospital from September 1997 to October 1999. Renal function, urine N-AG enzyme, liver function, and myocardial enzymes were assayed before and after treatment. One patient consented to a kidney biopsy and the pathology of renal damage was observed under light and electron microscopes.
RESULTSAll patients had multiple organ dysfunction syndrome (MODS) and 11 patients suffered from ARF. Ten patients had liver dysfunction, ten patients had poisonous myocarditis, and 8 patients had gastrointestinal dysfunction. Renal function, urine N-AG enzyme, liver function, and myocardial enzymes were significantly improved after treatment compared with those of before treatment (P < 0.05). Kidney biopsy showed that the main damage site was the proximal renal tubule. All eleven patients recovered and were discharged from the hospital.
CONCLUSIONSIngestion of fish gall bladder leads to kidney damage, as well as liver, heart and gastrointestinal tract injury. The mechanism of acute renal function failure is the serious tubular damage, confirming the location of kidney damage. Necrosis of the proximal tubules plays an important role in the development of ARF. Immediate hemodialysis is the most effective treatment.
