1.The presence of u-PA receptor in human endothelial cell and its regulation induced by PMA

Urokinase-type plasminogen activator plays an important role in the dissolution of clot and the treatment of thrombosis diseases. Presently, more attention has been paid to urokinase metabolism in human body and the relationship between urokinase fibrolysis effect and its receptor in endothelial cell.Cultured endothelial cell was used in the experiment to react with labelled urokinase in the presence or absence of unlabelled urokinase. It was found that the specific bindind of labelled urokinase with endothelial cell increased with the increasing concentration of labelled urokinase and reached the saturation point from 10 to 32 ?g labelled urokinase/ ml. The maximal binding sites with urokinase per cell were 1.4?10?, and Kd value 7.5? 10~(-12)M, Meanwhile, PMA-treated endothelial cell was used to react with ~(125)I-uPA at the same conditions as discribed above. Compared with the specific binding of DMSO-treated endothelial ceil, the maximal binding sites per PMA-treated cell increased from 1.4?10~6 Per untreated cell to 2.2?10~6, and the Kd value from 7.5?10~(-12)M to 9?10~(-12)M.These results showed that urokinase receptor exists on the surface of endothelial cell, and that the number of receptor per cell could be modulated by PMA. It is possible that the urokinase binding with endothelial cell can play an important role in the very short half-life of urokinase in human body.
2.Study on Chemical Constituents of Volatile Oils in Drug Pair of Flos Lonicerae and Fructus Forsythiae by GC-MS

Objective To discuss the influence of drug compatibility on the components of the volatile oil in Flos Lonicerae and Fructus Forsythiae. Methods GC-MS system was used to analyze the volatile oil in Flos Lonicerae, Fructas Forsythiae and their combination. The effect of compatibility on the components and content of volatile oil was evaluated. Results The results of GC-MS showed that gerani]o, tetradecanoic acid methyl ester, hexadecanoic acid ethyl ester, fluoranthene, 9, 12, 15-Octadeeatrienoic acid ethyl ester and linoleic acid was detectable in Flos Lonicerae but was undetectable in the drug pair; camphene, alpha-terpinene, perillaalcohol, 2-beta-pinene, beta-hexadecanal, phellandrene was de-tectable in Fructus Forsythiae but was undetectable in the drug pair; limonene, isolongifolene, nonadecane was detectable in the drug pair but was undetectable in the two single drugs. Conclusion The compatibility of the drug pair has an effect on components and contents in the volatile oil of Flos Lonicerae and Fructus Forsythiae.
3.Protective effect of Anisodamine on ionizing radiation damage of cochlea

Objective To explore protective effect of Anisodamine against injure of cochlea hair cells induced by ionizing radiation. Methods 50 healthy guinea pigs were randomly divided into 3groups: radiation group( n = 20) , anisodamine protective group( n = 20) and control group( n = 10). 20ears were observed in each group. Tempora of guinea pigs in anisodamine protective group and radiation group were exposed to fractional doses of 6 MeV electron beam produced by linear accelerators (2 Gy/d)with a total dose of 60 Gy. Animals were injected intramuscularly with anisodamine (20 mg/kg) in protection group and equal saline solution in irradiated group, respectively, 30 min pre-irradiation.Morphological and functional changes in the guinea pig cochlea were observed. Results After radiation,the average ABR threshold value of the radiation group (52. 27 ± 2.42 dB peSPL) was significantly higher than that of the anisodamine protection group (37.65 ± 1.92 dB peSPL, t = 2.01, P< 0.05 ), and outer fair cells of cochlea reduced greatly in quantity by observing the stretched preparations of cochlea basilar membrane ( F=135. 362, P< 0.05 ). Under transmission electron microscope, outer fair cells of cochlea in the radiation group had obscure boundaries and swelled to lose their original shapes. Meanwhile, cell implement and the nucleus were obviously obnormal after radiation. In the anisodamine protection group,the boundaries of cochlea outer fair cells were very clear and the cells swelled a little. Cell implement and nuclear envelope were complete. Cochlea outer fair cells in the control group were found to line up in order with no collapse and reduction in number under scanning electron microscope, but those in the radiation group collapsed obviously, reduced in number and aligned in a state of chaos, while the outer fair cells in the anisodamine protection group regularly posed on the whole and the collapse was occasionally found.Conclusions The 60 Gy radiation at fractioned dose on the tempuses of the guinea pigs could cause damage to the cochlea hair cells. Anisodamine might have a protective effect against radiation damage.
4.“T”-shaped locking plate for posterolateral tibial plateau fractures by Carlson posterolateral approach:12-month follow-up

BACKGROUND:Single fracture or colapse of the posterolateral tibial plateau fractures is relatively rare in the clinical work. Rational choice of surgical approach and internal fixation for posterolateral plateau fracture is significant to restore the lower limb force line, maintain the joint stability and obtain good biocompatibility.
OBJECTIVE:To compare the stability and biocompatibility of Carlson posterolateral and posterior midline approaches for the treatment of posterolateral tibial plateau fractures with “T” shaped locking plate.
METHODS:From July 2011 to July 2014, 43 patients with posterolateral tibial plateau fractures, who were treated in the Affiliated Hospital of Jining Medical University, were retrospectively analyzed. Al patients were assigned to two groups according to approaches. In the Carlson posterolateral approach group, 22 cases received “T”-shaped plate insertion by Carlson posterolateral approach. In the posterior midline approach group, 21 cases received “T”-shaped plate insertion by posterior midline approach. After repair, perioperative data, fixation effects and knee function score were compared and analyzed between both groups.
RESULTS AND CONCLUSION:(1) 43 cases (43 knees) of posterolateral tibial plateau fractures were folowed up strictly. (2) No significant difference in operation time, fracture healing time, total load time, Hospital for Special Surgery score at 12 months postoperatively, tibial plateau angle and posterior slope angle immediately and 12 months postoperatively was detected between both groups (P > 0.05). (3) Significant differences in fracture exposure, blood loss, and excelent and good rate of Rasmussen at 12 months postoperatively were identified in both groups. Moreover, above indexes were better in the Carlson posterolateral approach group than in the posterior midline approach group (P< 0.05). (4) These findings confirmed that for a single fracture or colapse of the posterolateral tibial plateau fractures, two kinds of surgical approaches can achieve ful and direct exposure. Carlson posterolateral approach has good repair effect, fixation effect and biocompatibility.
5.High-performance capillary electrophoresis method for ferulic acid assay in serum of rats treated with Buyang Huanwu Decoction

Object To develop a high-performance capillary electrophoresis method to determine ferulic acid concentrations in serum of rats treated with Buyang Huanwu Decoction (BHD). Methods Capillary zone electrophoresis was applied for ferulic acid assay, quantitative determination was based on internal standard and detection was carried on by direct UV. The electrolyte buffer was composed of 25 mmol/L borax-methanol (85∶15). Capillary electrophoresis was performed using a 52 cm (30 cm to detector)?50 ?m fused-silica capillary tube. Separation voltage was 20 kV, sampling time was 3 s, detection wavelength was 320 nm, and the temperature was 30 ℃. Results Ferulic acid was successfully separated within 6 min, the recoveries were 97.2% in serum and 103.28% in BHD, respectively. RSD were 3.73% and 0.91% (n=3), respectively. Conclusion This method can supply reference for the determination of ferulic acid in serum samples and BHD.
6.Pharmacokinetics of Berberine in Puerariae and Scutellariae and Coptidis Decoction in Vivo of Dog

OBJECTIVE:To assay serum berberine in Puerariae and Scutellariae and Coptidis Decoction and coptidis rhi?zoma single decoction in dog and to analyze the effect of the compound recipe(whole recipe)on the physiological disposition of beberine.METHODS:Serum concentrations of berberine in dog were determined at different time by HPLC and the phar?macokinetic parameters were computed with WinNonlin software.RESULTS:The concentration-time curves of berberine in Puerariae and Scutellariae and Coptidis Decoction and coptidis rhizoma single decoction in vivo of dog conformed to one-compartment model,the main pharmacokinetic parameters of berberine in Puerariae and Scutellariae and Coptidis Decoction vs.in coptidis rhizoma single decoction were as follows,AUC (0-∞) (1.25?0.06)vs.(14.71?0.54)(?g?h)/ml,t max (1.92?0.31)vs.(3.03?0.07)h,C max (0.17?0.01)vs.(1.79?0.03)?g/ml.CONCLUSIONS:As compared with single-item recipe,the plasma concentration and bioavailability of berberine in the compound recipe of Puerariae and Scutellariae and Coptidis Decoc?tion were all lower.
7.Analysis of two cases of huge adrenal tumor

Objective To investigate the clinicopathological characteristics, diagnosis and treatment of huge adrenal tumor.Methods Retrospectively analysed the clinical data of the 2 patients with huge adrenal tumor more than 10 cm in diameter, who were admitted in our hospital from September 2013 to January 2015.Clinical manifestations, treatment and prognosis were evaluated.Results Two patients were both suspected of adrenocortical carcinomas.Case 1 was a 28-year-old female.Laparoscopic surgery was performed at first but was conversed to open palliative adrenalectomy due to tumor rupture and bleeding.The postoperative pathology was adrenocortical carcinoma.The patient died of gastrointestinal bleeding caused by tumor metastasis 10 months later.Case 2 was a 39-year-old female, who underwent laparoscopic adrenolectomy with tumor excision completely.The postoperative pathology showed neoplasm of malignant potential, and no tumor recurred in 15 months follow-up.ConclusionsOpen surgery should be the priority if the diameter of adrenal tumor is more than 10 cm and diagnosis as suspicious of malignancy.Huge adrenocortical carcinoma is related to poor therapeutic response,and multimodality therapy may improve the prognosis.
8.A comparison for the patency rate between arterial bypass and interventional therapy of chronic lower limb ischemia

Objective To compare the long term outcome between traditional arterial bypass and interventional therapy for chronic lower limb ischemia according to the TASC classification. Method A retrospective study was undertaken on 201 cases receiving graft bypass or interventional therapy for CLI (chronic limb ischemia) from December 2005 to December 2008. Result The patency rate at 6 month. 12 month and 24 month after operation by femoral artery stent angioplasty in therapy group (100%, 89.8%, 75%) is higher than that of angioplasty group(82.4%, 62.5%, 35.7%), P <0.05. The 2nd term patency rate(88%) of operation group (24 months after operation) is higher than that of interventional group (70.7%), P<0.05. Conclusion Arterial bypass has higher long term patency rate and is applicable for femoral artery long segment obliterate classified as TASC C, D, while for the short segment femoral artery obliterate classified as TASC A, B interventional therapy especially stenting is an alternative despite of low patency rate.

OBJECTIVE To investigate the surgical methods for the lesions involved the common carotid artery.METHODS The clinical data of 11 cases with lesions involved the common carotid artery who underwent operations were retrospectively studied.The lesions were 1 case with recurrence tumor after 3/4 partial laryngectomy,1 case with bleeding of the carotid aneurysm caused by tuberculosis,1 case with iatrogenic carotid aneurysm,3 cases with carotid body tumor,1 case with thyroid gland cancer,2 cases with neck tumor,1 case with injury of the carotid artery and 1 case with gas gangrene.RESULTS Common carotid artery was reconstructed in 2 cases after removal of the tumors.The tumors were resected using the carotid shunt in 2 cases.Common carotid artery was sutured in 1 case with neck injury.The common carotid artery was repaired in 1 case with iatrogenic carotid aneurysm after removal of the tumor.The carotid artery was dissected out from the thyroid gland cancer in 1 case.The common carotid artery was reserved in 2 cases after resection of the neck tumors. Neck drainage was performed in the case with gas gangrene.CONCLUSION The surgical methods for lesions involved the carotid artery after removal of the tumors include the reconstruction of the carotid artery, resection and suture the carotid artery,and free of the carotid artery from the tumors.
10.A multi-center study on evaluation of leukocyte differential performance by an artificial intelligence-based Digital Cell Morphology Analyzer

Objective:To evaluate the performance of an artificial intelligent (AI)-based automated digital cell morphology analyzer (hereinafter referred as AI morphology analyzer) in detecting peripheral white blood cells (WBCs).Methods:A multi-center study. 1. A total of 3010 venous blood samples were collected from 11 tertiary hospitals nationwide, and 14 types of WBCs were analyzed with the AI morphology analyzers. The pre-classification results were compared with the post-classification results reviewed by senior morphological experts in evaluate the accuracy, sensitivity, specificity, and agreement of the AI morphology analyzers on the WBC pre-classification. 2. 400 blood samples (no less than 50% of the samples with abnormal WBCs after pre-classification and manual review) were selected from 3 010 samples, and the morphologists conducted manual microscopic examinations to differentiate different types of WBCs. The correlation between the post-classification and the manual microscopic examination results was analyzed. 3. Blood samples of patients diagnosed with lymphoma, acute lymphoblastic leukemia, acute myeloid leukemia, myelodysplastic syndrome, or myeloproliferative neoplasms were selected from the 3 010 blood samples. The performance of the AI morphology analyzers in these five hematological malignancies was evaluated by comparing the pre-classification and post-classification results. Cohen′s kappa test was used to analyze the consistency of WBC pre-classification and expert audit results, and Passing-Bablock regression analysis was used for comparison test, and accuracy, sensitivity, specificity, and agreement were calculated according to the formula.Results:1. AI morphology analyzers can pre-classify 14 types of WBCs and nucleated red blood cells. Compared with the post-classification results reviewed by senior morphological experts, the pre-classification accuracy of total WBCs reached 97.97%, of which the pre-classification accuracies of normal WBCs and abnormal WBCs were more than 96% and 87%, respectively. 2. The post-classification results reviewed by senior morphological experts correlated well with the manual differential results for all types of WBCs and nucleated red blood cells (neutrophils, lymphocytes, monocytes, eosinophils, basophils, immature granulocytes, blast cells, nucleated erythrocytes and malignant cells r>0.90 respectively, reactive lymphocytes r=0.85). With reference, the positive smear of abnormal cell types defined by The International Consensus Group for Hematology, the AI morphology analyzer has the similar screening ability for abnormal WBC samples as the manual microscopic examination. 3. For the blood samples with malignant hematologic diseases, the AI morphology analyzers showed accuracies higher than 84% on blast cells pre-classification, and the sensitivities were higher than 94%. In acute myeloid leukemia, the sensitivity of abnormal promyelocytes pre-classification exceeded 95%. Conclusion:The AI morphology analyzer showed high pre-classification accuracies and sensitivities on all types of leukocytes in peripheral blood when comparing with the post-classification results reviewed by experts. The post-classification results also showed a good correlation with the manual differential results. The AI morphology analyzer provides an efficient adjunctive white blood cell detection method for screening malignant hematological diseases.
