1.Tissue culture of Swertia bimaculata

Objective In order to protect the natural resources of Swertia bimaculata which has been destroyed seriously,the method of artificial propagation by way of tissue culture have been systematically studied.Methods The stems,leaves,and stems with buds which were from the seedlings germinated from the seeds on the initial medium were taken as explants.These explants were cultured on MS culture media by adding different portions of hormones at various cultural conditions.Results The stems were the best material in speeding propagation among the three explants.The proper initial medium for the stems was MS+ BA 0.5 mg/L+saccharose 3.0%,the optimum medium for proliferation was MS+BA 0.5 mg/L+IBA 0.1 mg/L+saccharose 3.0%,and the best medium for rooting was 1/2MS+NAA 0.5 mg/L+saccharose 1.5%.Conclusion Tissue culture of S.bimaculata could make its propagation rapid,its resources preserved,and its utilization last.
2.Modified Reconstruction of Old Rupture of Finger Flexor Tendon

Objective To explore the effect of reconstruction on old rupture of finger flexor tendon. Methods There were 30 patients (54 fingers) with old rupture of finger flexor tendon. The silica gel stick was implanted at the first stage. The palmaris longus tendon wrapped in the bioabsorbable membrane replaced the silica gel stick and was implanted at the second stage. The effectiveness was assessed by total active movement (TAM). Results There were 30 fingers as excellent, 20 fingers as good among 54 fingers of 30 patients. The percent of excellent and good was 92.6%. Conclusion Modified reconstruction is effective to prevent from tendon adhesion.
3.Three-Points Approach Ablation for Treatment of Typical Atrial Flutter Guided by CARTO

Objective To evaluate the feasibility and effect of three-points ablation approach in in treatment of typical atrial flutter guided by CARTO. Methods Twenty-six patients with typical atrial flutter diagnosed by ECG and electrophysiological study (EPS) were enrolled in this study. Activation sequence mapping and linear ablation were performed in 11 patients (conventional group) . Three-points guided linear ablation with CARTO system was performed in another15 patients (three-points group) . Results There was no significant difference in the success rate between the two groups. Both the procedure and fluoroscopic time in three-points group were significantly shorter than that in conventional group [(72.66±29.82) vs (102.52±32.61) min;(4.26±2.76) vs (7.32±3.16) min] . Conclusions The three-points ablations approach is as safe and effective as conventional ablation approach in treatment of typical atrial flutter;however,the former can significantly shorten the procedure time and fluoroscopy time.
4.Catheter Ablation of Para-Hisian Atrial Tachycardia Guide by CARTO

Objective To evaluate the feasibility of catheter ablation of Para-Hisian Atrial Tachycardia guide by CARTO. Method Catheter ablation guided by CARTO was performed after activation map in three patients with Para-Hisian Atrial Tachycardia. Result Successful ablation was got at right atrial in two patients and at non-coronary in one patient. Conclusion Catheter ablation guided by CARTO is safe and efficient for Para-Hisian Atrial Tachycardia.

6.Effect of cyclooxygenase-2 inhibitors on the expression of the Acsl gene family in the ileum of rats with nonalcoholic fatty liver disease

ObjectiveTo investigate the effect and significance of cyclooxygenase-2 (COX-2) inhibitors on the expression of the Acsl gene family in the ileum of rats with nonalcoholic fatty liver disease (NAFLD). MethodsA total of 45 Sprague-Dawley rats were randomly divided into normal control group (15 rats given normal diet), NAFLD model group (15 rats given high-fat diet), and nimesulide group (15 rats given high-fat diet and nimesulide). All rats were sacrificed after 12 weeks of feeding, and then blood samples were collected from the inferior vena cava to measure total cholesterol (TC) and triglyceride (TG). HE staining and oil red O staining were performed for the liver to evaluate the degree of hepatic steatosis in each group, and quantitative real-time PCR was used to measure the mRNA expression of the Acsl family genes in the ileum. An analysis of variance was used for comparison of continuous data between multiple groups, and the least significant difference t-test was used for further comparison between two groups. ResultsCompared with the normal control group, the NAFLD model group had significant increases in serum TC and TG and marked hepatic steatosis (all P<0.05); compared with the NAFLD model group, the nimesulide group had significant reductions in serum TC and TG and degree of hepatic steatosis (all P<0.05). Compared with the normal control group, the NAFLD model group had a significant increase in the expression of COX-2 in the ileum (P<0.05), and compared with the NAFLD model group, the nimesulide group had a significant reduction in the expression of COX-2 in the ileum (P<005). Compared with the normal control group, the NAFLD model group had significant increases in the mRNA expression of Acsl3 and Acsl5 in the ileum (both P<0.05), and compared with the NAFLD model group, the nimesulide group had significant reductions in the mRNA expression of Acsl3 and Acsl5 (both P<0.05). ConclusionThe COX-2 inhibitor nimesulide can regulate the expression of the Acsl gene family in the ileum of rats with NAFLD, suggesting that COX-2 inhibitors may inhibit the progression of NAFLD through the Acsl gene.
7.Application of CRISPR/Cas9 lentiviral vector in construction of rat hepatic stellate cells with COX-2 gene knockout

ObjectiveTo obtain HSC-T6 cells with stable expression of Cas9 protein and HSC-T6-COX-2-/- cells with COX-2 gene defect by transfecting HSC-T6 cells with CRISPR/Cas9 lentiviral vector, and to provide a good method for further functional research and new strategies for the clinical treatment of liver fibrosis. MethodsThe COX-2 gene-specific sgRNAs (COX-2-sgRNA-1, COX-2-sgRNA-2, COX-2-sgRNA-3) were designed, synthesized, and connected to the GV371 vector, and the recombinant plasmid and the packaging plasmid were transfected into 293T cells to form lentivirus particles; the fluorescence method was used to measure virus titer. The most appropriate amount of the virus was calculated based on MOI. Lenti-Cas9-puro was transfected into HSC-T6 cells, and HSC-T6-Cas9 cells were screened out by puromycin; Lenti-COX-2-sgRNA-EGFP was transfected into HSC-T6-Cas9 cells to obtain HSC-T6-COX-2-/- cells. Cruiser enzyme digestion and Western blot were used to verify gene knockout at the gene and protein levels. An analysis of variance was used for comparison of continuous data between multiple groups, and the least significant difference t-test was used for further comparison between two groups. ResultsSequencing verified that the COX-2-sgRNA expression vector was constructed successfully. Recombinant expression plasmids and packaging plasmids were transfected into 293T cells to form lentivirus particles, and the fluorescence method showed a virus titer of >1×108. HSC-T6 cells with stable expression of Cas9 protein and HSC-T6-COX-2-/- cells with COX-2 gene defect were successfully constructed. The HSC-T6-Cas9 group had significantly higher relative mRNA expression of LV-Cas9-Puro than the CON group (541.93±105.76 vs 1.00±0.02, t=12.995, P<0.01). Cruiser enzyme digestion and Western blot showed that the CRISPR/Cas9 lentivirus expression vectors played a role in the target, among which COX-2-sgRNA-2 knockout had the most significant effect, and this group had a significant reduction in the protein expression level of COX-2 compared with the CON group and the NC group (both P<0.05), suggesting that COX-2-sgRNA was active. ConclusionA CRISPR/Cas9 lentivirus vector is successfully constructed for COX-2 target gene, and HSC-T6-COX-2-/- cells with stable COX-2 gene knockout are obtained.
8.Impact and changes of maternal hemoglobin on birth weight in pregnant women of Zhuang Nationality, in Guangxi

Objective To investigate the hemoglobin (Hb) levels during pregnancy and Hb changes from early pregnancy and association with birth weight on infants.Methods Mothers of Zhuang Nationality who participated in the pregnancy care program and delivered at the Pingguo County Hospital from May 2013 to May 2015 were included in this study.Retrospective analysis was applied to collect data of health care and pregnancy outcomes.Multiple regression analysis and unconditional logistic regression model were used for data analysis.Results The mean birth weight appeared as (313 5.92± 435.84) grams.The Hb levels at early pregnancy showed significantly positive association with birth weight.Results from our study demonstrated that when Hb levels increased +1 g/dl at early pregnancy,birth weight would increase 17.61(95%CI:0.60-34.67) grams,in the adjusted model.The Hb levels at late pregnancy were significantly inversely associated with birth weight.Our findings suggested that when Hb levels increased + 1 g/di at late pregnancy,birth weight would reduce 19.61(95% CI:-37.53--1.70) grams in the adjusted model.Changes in Hb from early pregnancy stages were significantly inversely associated with birth weight after adjusting for confounders and Hb levels in the early pregnancy stages.The results also indicated that when Hb levels increased a + 1 g/dl from early to late pregnancy,the birth weight would decrease 32.63 g(95% CI:-48.93--16.32).Compared to the non-anemia group,the anemia group showed significantly increase of small-for-gestational-age (SGA)(OR=1.58,95%CI:1.08-2.32) in early pregnancy.Compared to women under the most reduction status,women with the least reduction had a significantly increase of SGA (OR=1.87,95%CI:1.24-2.81) among their infants.With the magnitude of reduction on Hb concentration during pregnancy,the risk of delivering babies with SGA showed a gradual trends of increase.Conclusion Hb levels at early pregnancy were positively associated with birth weight,but the changes of Hb were inversely associated with birth weight at late pregnancy,in women of Zhuang Nationality.Anemia in early pregnancy and the low amplitude of decreased Hb concentration during pregnancy were both risk factors for newborns under less gestational ages.
9.Ultrasound and CT findings in children pancreatoblastoma

Objective To improve the imaging diagnostic performance by analyzing US and CT findings of pancreatoblastoma in children. Methods From January 2000 to December 2016, 9 patients with pancreatoblastoma proven pathologically were enrolled in the study. The clinical and imaging features in 9 cases were analyzed retrospectively, the tumor parameters compared with intraoperative and postoperative pathology were location, size, texture, capsule, calcification, adjacent organs invasion and lymph node metastasis. The parameter of tumor size was evaluated by intraclass correlation coefficient. Results The tumor diameter ranged from 30-95 mm (median diameter 40 mm). Six tumors located in the head of the pancreas, 3 tumors located in the body and tail of the pancreas. One tumor was pure cystic, 2 tumors were solid and cystic mixed, 5 tumors were solid. Calcification was seen in 5 cases. Capsule was seen in 5 cases. Blood vessels, biliary tract and bowel around tumors were infiltrated by tumors in 3 cases proven by the pathology. Lymphadenectasis was seen in 2 cases, in which 1 case was metastasis. There were no significant differences in location, nature, calcification on US and CT. The largest diameter measured by CT and US was consistent with the tumor''s actual size, ICC=0.90.Capsule was shown by US in 5 cases and CT in 3 cases. CT showed the morphology of calcification in 5 cases, lymphadenectasis in 2 cases, abdominal effusion in 2 cases, cavernous transformation of the portal vein with the extensive collaterals in 1 case, which were not shown by US. Both US and CT dit not show the surrounding tissue infiltrated by tumors in 3 cases,but showed biliary system expansion. Conclusions The typical pancreatoblastoma manifested as encapsulated solid tumor originated in the pancreas with necrosis and calcification. US examination took a slight advantage on displaying capsule of tumor than CT. CT was more clear and intuitive in showing calcification and relationship between tumor with surrounding tissues and lymph node, and no operator dependent.
