2.Utility of a multiprobe fluorescence in situ hybridization assay in the detection of bladder cancer

Objective To evaluate the clinical application of a muhiprobe FISH (multicolor fluorescence in situ hy-bridization,M-FISH) assay in voided urine specimens for detection of bladder cancer and compare the results with those afforded by urinary cytology.Methods Voided urine specimens from 100 cancer patients with or without surgery and 10 patients with prostate hyperplasia were obtained for urinary cytology and FISH.FISH was performed using a mixture of fluorescent labeler DNA probes for the centromeric regions of chromosomes 3,7,and 17 and 9p21 region.Cystoscopy with biopsy or tumor resection was performed in all patients and compared the pathological results with the cytological and FISH findings.Results The sensitivity of M-FISH were 75.6% for low grade tumors,100% for high grade tumors,with overall sensitivity of 85.5%.Urinary cytology affords an overall sensitivity of 62.9% (33.3% for low grade tumors,96.0% for high grade tumors).Significant difference in overall sensitivity was observed between M-FISH and urinary cytology (P<0.05).The specificity of M-FISH and urinary cytology was 84.6% and 87.8% respectively.No significant difference in specificity was observed between M-FISH and urinary cytology.Conclusion M-FISH improves the sensitivity obtained with urinary cytology for bladder cancer detection with similar specificity,so can detect all high grade infiltrating tumors patients.
3.Treatment of 78 Cases of Shoulder Periarthritis by Warming-needle Method

Jiansanzhen, Tianzong (SI 11), Jugu (LI 16),Jianzhen (SI 9), Binao (LI 14) and Quchi (LI 11) were given warming-needle moxibustion to treat shoulder periarthritis in 78 cases, and the result showed total effective rate was 97.4%.
4.Clinicopathological significance and expression of Numb and α-catenin in gastric carcinoma

Objective To detect the expressions of Numb and α-catenin in the gastric cancer and matched normal gastric mucosa,and to analyze their relationship with clinicopathological factors of gastric cancer and explore their roles in gastric carcinogenesis and progression.Methods Immunohistochemical method was used to detect the expressions of Numb and α-catenin in 109 cases of tumor specimens and 97 cases of matched normal gastric mucosa.The correlations between Numb,α-catenin and clinicopathological factors were analyzed.Results The positive rates of Numb and α-catenin in gastric cancer were significantly lower than those in normal gastric mucosa (60.6% ∶ 100.0%,x2 =48.361,P =0.000;76.1% ∶ 100.0%,x2 =26.480,P=0.000).The expression of Numb protein was correlated with Lauren type (x2 =17.018,P =0.000) and tubular adenocarcinoma differentiation (x2 =17.586,P =0.000).The expression of α-catenin protein was correlated with Borrmann type (x2 =6.700,P =0.035),Lauren type (x2 =11.098,P =0.001),tubular adenocarcinoma differentiation (x2 =8.203,P =0.017) and lymph node metastasis (x2 =6.402,P =0.011).The expressions of Numb and α-catenin were statistically correlated in 109 cases of gastric cancer (rk =0.184,P =0.028).Conclusion Compared with normal gastric mucosa,both Numb and α-catenin expressions are down-regulated and the two expressions are correlated in gastric cancer.Numb and α-catenin may be involved in the regulation of histological differentiation of gastric cancer by certain passages,Numb protein may be adjusted by α-catenin pathway which is involved in Wnt-β-catenin pathway,and thus plays an important role in promoting cell proliferation,invasion and metastasis in human gastric cancer.
5.Distribution and Drug-resistance of Stenotrophomonas maltophilia

OBJECTIVE To get knowledge of the distribution and drug-resistance of Stenotrophomonas maltophilia in our hospital and improve the level of diagnosis and treatment.METHODS The automatic drug sensitive test and the paper spreads medicine sensitive test were carried on to 162 strains of S.(maltophilia) with the index of the VITEK system.RESULTS In all clinical samples,S. maltophilia isolated from phlegm was the most(90%),then was from secretion and swab.The majority of 162 strains were multidrug resistant.But to the(trimethoprim)-sulfamethoxazole and ciprofloxacin,the drug resistance was low((20.4%) and 22.2%,respectively).CONCLUSIONS S.maltophilia is the prevalent pathogen of nosocomial infection in ICU.And most of it are(isolated) from phlegm.So use antibiotics correctly,(reduce) invasive examinations or treatments and strengthen drug-resistant monitoring,are helpful to prevent and control S.maltophilia nosocomial infection.
6.Application of wireless transfer technology to medical instrument development

In many medical instrumentation systems, there exists a long distance between manipulation platform and host unit. Wireless data transferring between the platform and the host could be a feasible way to set parameters. This paper introduces wireless transmitter and receiver circuit based on encoder chip PT2262 and decoder chip PT22720, and presents some typical applications in medical instrument development.
7.The curative effect of targeted siRNA of vascular endothelial growth factor combined endostatin on collagen-induced arthritis rat

Objective To investigate the effect of treatment with endostatin combined with siRNA targeting VEGF on rats with collagen-induced arthritis (CIA).Methods Two mg/ml bovinetype Ⅱ collagen was injected into the rat footpad to build up the animal model of CIA.The experimental animal models were treated with endostatin combined with siRNA targeting VEGF 18 days later after immunization and the treatment ended 32 day later.The efficacy was evaluated by the weight,foot and ankle volume of rats.The levels of VEGF in plasma were detected by enzyme-linked immunosorbent assay (ELISA).The VEGF distribution within synovial tissue was detected and examined by immunohistochemical technique.The pathological changes of CIA were evaluated by the pathological changes of the biopsied ankle joints.Student's t test was used to evaluate the experimental data.Results The ELISA test showed that comparing with the model group (17.5±0.3),the endostatin group (15.7±0.3) ng/L and the endostatin combined siRNA targeting VEGF group (14.7±0.5) ng/L showed a significant efficacy in the treatment of CIA in rats (P<0.05).The endostatin group (135±27) and the endostatin combined siRNA targeting VEGF group (126±71) were different in the number of VEGF in plasma and the VEGF distribution within synovial tissue (P<0.05),the symptoms of arthritis in these rats were reduced than the model group.Conclusion Endostatin combined siRNA targeting VEGF has good therapeutic effect on rats with CIA.
8.Analysis of Organophosphorus and Pyrethroid Pesticides Multi-residues in Vegetables

Objective To investigate the pesticide residues in vegetables in Changchun of China. Methods 10 kinds of organophsphorus and pyrethroid pesticide residues in 8 kinds of vegetables were determined by capillary gas chromatography in 2003-2004. Results 4 kinds of organophosphorus (methyl parathion, DDVP, parathion, phorate) and 2 kinds of pyrethroid pesticides (Cypermethrin, Fenvalerate) were detected in the vegetables in 2003. 4 kinds of pesticides were detected in spinach,tomato, leek, capsicum, celery and cucumber, but not in cabbage and cole. The parathion residues and the rate of exceeded limit in spinach, capsicum was 0.007 mg/kg, 0.036 mg/kg and 9.09% respectively. The phorate residues in tomato was 0.0150 mg/kg, the rate of exceed limit was 12.5%. The most popular pesticide residued in these vegetables was methyl parathion which was detected in 5 kinds of vegetables, DDVP and cypermethrin was detected in 3 kinds of vegetables. 4 kinds of organophsphorus (DDVP, Methamidophos, Omethoate, Chlorpyrifos) and 3 kinds of pyrethroids pesticides (Cypermethrin, Deltamethrin, Fenvalerate) residues were detected in 8 kinds of vegetables in 2004. 6 kinds of pesticide were detected in leek, pesticides were also detected in the other vegetables. Methamidophos residue in leek was 0.705 0-22.560 0 mg/kg, the rate of exceed limit was 100%, omethoate residue and the rate of exceed limit in leek and capsicum was 0.219 5-0.414 5 mg/kg, 0.0170 mg/kg and 100%,14.29% respectively. Fenvalerate was detected in 8 kinds of vegetables, Cypermethrin in 5 kinds of vegetables. Conclusion Generally speaking, the problem on the organophosphorus and pyrethroids pesticide residues existed in vegetables in Changchun, China is more serious.
9.Cationic liposome-mediated bcl-xl gene transfection into human keratocytes.

The efficiency and safe range of Lipofectamine2000 (LF2000)/bcl-xl applied in human keratocytes, the optimal ratio of LF2000/bcl-xl and the bcl-xl gene expression in human keratocytes were investiaged. By using trypan-blue staining, the effects of LF2000 and bcl-xl on the survival rate of the cultured human keratocytes were measured respectively. By using semi-quantitative RT-PCR, the efficiency and the expression of LF2000-mediated bcl-xl transfection into keratocytes were examined. The results showed that the survival rate of human keratocytes had no signficant change in the presence of LF2000 (20 microg/ml) or bcl-xl (10 microg/ml) for 24 h. LF2000 could effectively mediate the transfection of exogenous gene bcl-xl into human keratocytes. The best transfection efficiency could be obtained when the ratio of bcl-xl/LF2000 was 1:8. One day after transfection, the positive cells for bcl-xl could be detectable, and the positive rate reached the peak-on the posttransfection day 3 (48.3%), then gradually decreased. Fifteen days after transfection, there were few positive cells. It was suggested that LF2000 could effectively transfer the exogenous gene bcl-xl into human keratocytes without obvious toxicity during a concentration range. LF2000/bcl-xl may be likely to play an important role in gene therapy of human keratocytes.

10.Expression, purification and biological activities assay of human C1q and tumor necrosis factor related protein-6

Aim To prepare soluble human C1 q and tumor necrosis factor related protein-6 in Escherichia coli and analyze the bioactivity. Methods Recombi-nant plasmid was transformed into E. coli expression strain, and the recombinant protein Trx-hCTRP6 was expressed induced by IPTG and then purified. Results Trx-hCTRP6 was expressed efficiently and purified using Ni-NTA affinity chromatography and Superdex G-75 column. The purified Trx-hCTRP6 was shown to be active under in vivo and in vitro assay conditions. Con-clusion Active Trx-hCTRP6 is efficiently prepared from E. coli protein expression system.
