1.Synthesis of lactosaminated carboxymethylchitosan and its action of plasmid DNA delivery

Objective To study the synthesis of lactosaminated carboxymethylchitosan (Lac-CMCS) and its action of delivering plasmid DNA into cells. Methods The lactosaminated carboxymethylchitosan was synthesized by reductive amination and was conjugated with plasmid contained antisense x gene. This complex was delivered into cells and these cells were cultured in 37℃,5%CO_(2). Testing the inhibition on HBV by determining HBsAg and HBeAg produced by HepG2.2.15 cells with ELISA technique. Results The molar ratio between lactose and CMCS was 15∶1 in this covalence conjugate. The best mass ratio of DNA to Lac-CMCS was 1∶10 in DNA/Lac-CMCS which was able to deliver gene into cells, and the ma-ximal inhibition rate was 83.6%. Conclusion In vitro, the lactosaminated carboxymethylchitosan can effectively deliver DNA into cells.
2.Changes of serum C-reactive protein and neuron-specific enolase levels in patients with acute cerebral infarction and relative study

Objective To investigate the changes of serum C-reactive protein(CRP)and neu-ron-specific enolase(NSE)levels in patients with acute cerebral infarction(ACI)and their relations with in farct size,severity of neurological impairment.Methods The levels of serum CRP,NSE were measured in 60 patients(ACI group)within 72 hours after infarction and 40 normal controls(control group).The severity of neurological impairment of patients with ACl was assessed by National Institutes of Health Stroke Scale (NIHSS)at admission.Results The mean levels of serum CRP and NSE in ACI group were higher than those in control group, (6.95±2.34)mg/L vs(2.21±1.05)mg/L,P<0.01, (22.39±4.72)μg/L vs(7.16±2.63)μg/L,P<0.01,respectively.Higher levels of CRP and NSE were obviously associated with larger infarct size and more severe neurological impairment.The changes of CRP and NSE levels correlated with scores of NIHSS(r=0.51,r=0.44,all P<0.01)significantly.CRP levels were positively correlated with those of NSE(r=0.57,P<0.01).Conclusion The levels of serum CRP and NSE can be used as an important biological marker to estimate the size of infarct and the severity of neurological impairment in ACI.
3.Reconstruction following total gastrectomy

Total gastrectomy is a very important mode of therapy for gastric cancer.Many different types of reconstruction have been proposed,but there is no definitive conclusions as to which procedure is the best because of the postoperative complications,such as dumping syndrome,regurgitant esophagitis,malnutrition and so on.In this review,the author summarized recent clinical studies addressing gastrointestinal reconstruction following total gastrectomy.There is still some debate on preserving duodenal passage and the pouch reconstruction can provide improvement of the quality of life in patients receiving total gastrectomy.
4.Effects of propofol against LPS-induced inflammation injury of Ana-1 cells

Objective To study the protective effect of propofol on lipopolysaccharide (LPS)-induced Ana-1 cell damage.Methods The LPS-induced inflammation damage of Ana-1 cells was es-tablished as a model.Ana-1 cells were divided into control group(group C),LPS group(group L), LPS+propofol 10μmol/L group(group LP1),LPS+propofol 20μmol/L group(group LP2),LPS+propofol 40 μmol/L group(group LP3),and LPS+propofol 80 μmol/L group(group LP4),respec-tively.Cell survival rate was assessed using MTT,interleukin 1βand 6,and tumor necrosis factors (TNF-α)were determined.Results Survival rate of Ana-1 cells in group L was lower than that in group C,while it was significantly improved in group LP1,LP2,LP3,LP4 in a dose-dependent man-ner (P <0.01);IL-1β,IL-6,TNF-αrelease was significantly higher in group L than in group C;IL-1β,IL-6,TNF-αrelease significantly reduced in group LP1,LP2,LP3,LP4 (P <0.01)in a dose-dependent manner (P <0.01).Within the selected dose range in this experiment,group LP4 had the best protection effect.Conclusion Propofol can reduce inflammation damage of Ana-1 cells caused by LPS.
5.Clinical applicaton of carotid endarterectomy

Objective To study the indications of carotid endarterectomy(CEA) and perioperative managements . Methods The clinical data of 11 patients with transient ischemia attack (TIA) and carotid artery stenosis who underwent CEA were reviewed retrospectively. Results After CEA, the TIA symptoms disappeared in all patients, and the chronic symptoms of cerebral ischemia had obvious improvement in 4 cases . There were no serious postoperative complications, such as cerebral hemorrhage, hemiplegia, etc. Conclusions CEA may be considered when one or both sides of carotid stenosis more than 50% is discovered by Duplex scanning, digital subtraction angiography or MRA in patients with TIA. Stage operation for bilateral carotid occlusive lesion is safer. Paying attention to perioperative managements is important for reducing operative complications.

7.Accuracy and utility of CT guided biopsy of bone lesion

[Objective]To evaluate the accuracy and value of CT guided biopsy of bone lesion.[Method]A total of 137 cases of CT guided biopsy were analyzed,including 1 case of cervical vertebrae,22 cases of thoracic vertebrae,47 cases of lumbar vertebra,8 cases of sacrum,9 cases of pelvis,2 cases of scapula and 48 cases of extremities.The biopsies were completed in local anesthesia under CT guidance,using trephine needle or Tru-cut needle.[Result]The puncture accuracy was 100%,the general diagnostic accuracy was 89.0%(122/137),in which the diagnostic accuracies of metastasis lesion,primary bone tumor and benign lesion were 95.8%(46/48),85.7%(36/42)and 85.1%(40/47)respectively.[Conclusion]CT guided biopsy was a safe and accurate method with little trauma and complication,having great value in deciding treatment of bone lesions.
8.TGF-?1 induced actin cytoskeletal reorganization in human dental pulp cells

Objective:To determine the effects of TGF-?1 on the cytoskeleton in cultured human dental pulp cells(HPCs).Methods:Human dental pulp cells were cultured from dental pulp tissue explants digested with collagenase I. Semi-confluent cultures of the cells maintained under serum deprivation were treated with 20 ng/ml of TGF-?1 for 30 min, 1, 6 and 24 h respectively.Then cells were processed for BODYPY-phalloidin direct fluorescence examination of the actin filaments,DAPI direct fluorescence of the nucleus and Rhodamine Red(TM) indirect immunofluorescence of tubulin-?. Confocal laser scanning microscopy was used to investigate the changes of actin filaments and microtubules.Results:Disintegration and reorganization of actin filaments were observed in human dental pulp cells treated by TGF-?1 at 20 ng/ml. Actin filaments assembly was found near the cell membrane,specially after 30 min exposure.Disintegration of actin filaments was most obvious after 6 h treatment.Actin filaments were reorganized after 24 h exposure.Microtubules mainly remained intact in the cells during TGF-?1 treatment.Conclusion:TGF-?1 at 20 ng/ml may induce actin cytoskeletal reorganization in human dental pulp cells.

10.Epidemiological analysis and countermeasures discussion on imported ma-laria in Zhangjiagang City

Objective To analyze the epidemiological characteristics of imported malaria in Zhangjiagang City. Methods The epidemiological data were collected and retrospectively analyzed for the distribution,cost,and exit-entry mode and port of imported malaria cases in Zhangjiagang City from 2005 to 2015. Results There were 25 imported malaria cases in Zhangjia-gang City from 2005 to 2015,and among them,there were 16 cases of falciparum malaria(64%),6 cases of vivax malaria (24%),and 3 cases of ovale malaria(12%);there was 1 cases of critically ill(4%),there were 8 cases of serious ill(32%) and 16 cases of mild ill(64%). The time of onset was in accordance with the circular distribution. The peak of the incidence of the imported malaria was one month earlier than that of the domestic infection. The seasonal peak was gentle ,and there was also the occurrence in the non-epidemic season in the city. The imported malaria patients were mainly from Africa ,followed by South-east Asia and Oceania. Conclusion The information technology should be applied to improve the key population coverage on the basis of improving the ability of malaria diagnosis and treatment of medical staff and the multi-sector 's cooperation for the im-ported malaria prevention and control in Zhangjiagang City.
