2.Expression of microRNA on peripheral blood cells in patients with systemic lupus erythematosus

Objective To study the expression of 2 bone morphogenetic protein (BMP) related microRNA (miR-21,miR-155) in SLE patients,and to analyze their correlation with clinical features.Methods Peripheral blood was obtained from 59 SLE patients and 25 healthy controls,and total RNA was extracted and reverse transcribed into complementary DNA (cDNA).Real-time PCR technique was used to detect gene expression at transcrptional level.Disease activity was determined by SLEDAI score.Patients were divided into different groups based on their manifestations or antibody profiles,then comparison of miRNA expression was carried out using Mann-Whitney test.Results The expression levels of miR-21 and miR-155 were increased in active SLE patients (SLEDAI≥8) as compared to those with mild disease (SLEDAI≤7) or healthy controls (both P<0.05).Patients with positive anti-dsDNA or lupus nephritis had higher expression levels of miR-21 and miR-155 than those without (P<0.05).Conclusion The miR-21 and miR-155 expressions are elevated in active SLE patients and are associated with anti-dsDNA and renal involvement.The results suggest that these two miRNAs might play a role in the pathogenesis of SLE disease.
3.The study of the structure and function in the posterior cingulated cortex to heroin abuser by magnetic resonance imaging

Objective To explore the abnormalities in the posterior cingulated cortex structure by voxel based morphometry(VBM)and changes of functional connectivity by resting-state functional magnetic resonance imaging(fMRI)in long-term heroin addictions.Methods High-resolution volumetric T1-weighted images and resting state fMRI examination were performed on 13 long-term heroin addicts and 14 matched healthy volunteers.The T1-weighted images were processed using optimized voxel-based morphometry to investigate abnormalities in the gray matter,at the sarne time,the posterior cingulated cortex was setted as the region-of-interest(ROI)seed,with which a whole-brain voxel temporal correlation in low frequency fMRI fluctuations was calculated,to find changes of the posterior cingulated cortex functional connectivity.Results Compared with the control group,gray matter concentration significantly decreased in the posterior cingnlated cortex,at the same time,the functional connectivity between the left posterior cingulated cortex and the right temporal lobe was increased,but the functional connectivity between the left posterior eingulated cortex and the left thalamus,the left parietal lobe,the left cerebrum were all decreased in heroin addicts.At the same time,the functional connectivity between the fight posterior cingulated cortex and the left thalamus,the left parietal lobe,the left frontal lobe were decreased in heroin addicts(t testing,P<0.01).Conclusion The structure and the functional connectivity in posterior cingulated cortex in long-term heroin addicts are changed,which may affect the dysfunction of the addictive brain and the function of reward mechanism.
4.Factors affecting microvascular complications among patients with type 2 diabetes mellitus

Objective :
To identify the factors affecting microvascular complications among patients with type 2 diabetes (T2DM), so as to provide insights into the management of microvascular complications of T2DM.
Methods:
T2DM patients hospitalized in the Department of Endocrinology of a tertiary hospital in Weifang City, Shandong Province from January 2021 to January 2022 were enrolled, and subjects' basic information, lifestyle and medical history were collected using questionnaire surveys. Fasting insulin, fasting blood glucose and glycated hemoglobin were measured, and factors affecting microvascular complications were identified among T2DM patients using a multivariable logistic regression model and a decision tree model.
Results:
Totally 1 003 T2DM inpatients were enrolled, including 515 men (51.35%) and 488 women (48.65%), and the prevalence of microvascular complications was 40.18%. Multivariable logistic regression analysis showed that age of 60 years and older (OR=2.510, 95%CI: 1.441-4.374), T2DM duration of 10 years and longer (OR=3.205, 95%CI: 2.242-4.581), fasting insulin of lower than 3.21 μIU/mL (OR=1.749, 95%CI: 1.239-2.469), using of agents or insulin to control blood glucose (OR=1.880, 95%CI: 1.143-3.092), glycated hemoglobin level of 7% and higher (OR=1.751, 95%CI: 1.172-2.615) as factors affecting microvascular complications among T2DM patients. Decision tree analysis identified course of T2DM as a major factor affecting the risk of microvascular complications among T2DM patients, and the prevalence of microvascular complications was 70.22% among T2DM patients with disease course of 10 years and longer and fasting insulin of lower than 3.21 μIU/mL or 16.32 μIU/mL and higher, 44.23% among T2DM patients with disease course of 5 to 10 years and at ages of 60 years and older, and 43.10% among T2DM patients with disease course of less than 5 years and fasting insulin of lower than 3.21 μIU/mL.
Conclusion
Advanced age, long course of T2DM, low fasting insulin and high glycated hemoglobin may increase the risk of microvascular complications among T2DM patients.
5.Study on regulatory effect of Kaixin San on endogenous melatonin biosynthesis in rat depression model.

OBJECTIVETo study the effect of Kaixin San on the rate-limiting enzyme in biosynthesis of melatonin (MT) and pineal body in rat depression model.
METHODThe unpredictable chronic mild stress was used to establish the rat depression model for 21 days. The rats were divided into the normal control group, the model group, Kaixin San low, medium and high dose groups (KXS 65, 130, 260 mg x kg x d(-1)) and the trazodone group. All groups were administered at 30 min after modeling each day. Rats were sacrificed and the pineal glands were isolated immediately after acquisition tail venous blood at 2:00a. m on the 22nd day. The plasma was analyzed for melatonin content by using a rat metabolic panel Milliplex kit. The pineal glands were analyzed for AANAT and HIOMT mRNA levels by Real-time quantitative PCR and for AANAT and HIOMT activity by a radiometric assay simultaneously.
RESULTThe plasma MT concentration, expression of AANT and HIOMT mRNA, activity of AANAT in rat pineal glands of the model group were significantly lower than the control group (P < 0.05), but the activity of HIOMT showed not change. Compared with the model group, all of Kaixin San groups showed increase in MT concentration in plasma (P <0. 05) , with the medium dose group revealing the highest level. Besides, the medium dose group displayed significant increase in AANAT, HIOMT mRNA level and AANAT activity (P < 0.05), but no increase in HIOMT activity.
CONCLUSIONKaixin San can regulate AANAT activity of pineal bodyand regulate MT biosynthesis in rat depression model.

6.Study on the role of multiple mediating effects on scientific research creativity of nursing postgraduates

Objective:To explore the role of multiple mediating effects on scientific research creativity of nursing postgraduates.Methods:In the study, 127 nursing postgraduates from three affliated hospitals of Chongqing Medical University were selected by convenience sampling. The investigation scale included general information questionnaire, tutor support scale, nursing scientific research self-efficacy scale, autonomy motivation scale and scientific research creativity scale. The questionnaire of nursing postgraduates was collected through the Sojump platform, and the data of 127 postgraduates were effectively collected in strict accordance with the inclusion and exclusion criteria. The difference analysis and correlation analysis were carried out by SPSS.Results:The correlation analysis showed that nursing postgraduates' scientific research self-efficacy and autonomy motivation were significantly positively correlated with tutor support and scientific research creativity ( P<0.05). Multiple linear hierarchical regression analysis showed that nursing research self-efficacy and autonomy motivation played a partial mediating role between tutor support and scientific research creativity ( P<0.05), in which the mediating role explained 13.1% and 4.2% of the variation respectively. The structural equation model further verified the role of multiple mediating effects on the scientific research creativity of nursing postgraduates. Conclusion:The scientific research creativity of nursing postgraduates is closely related to tutor support, scientific research self-efficacy and autonomy motivation. Tutors can enhance nursing postgraduates' scientific research self-efficacy and autonomy motivation by improving the level of tutor support, so as to improve the scientific research creativity of postgraduates.
7.Characteristic Analysis of "Imprinting Template" for Pungent Herbs Based on Molecular Connectivity Index and Matching Frequency Total Statistical Moment

ObjectiveTo analyze the fingerprint of six pungent herbs based on the molecular connectivity index(MCI)and the matching frequency total statistical moment method, and to study the division and integration of the "imprinting template" of their volatile components, so as to find the common "imprinting template" characteristics of the pungent herbs. MethodThe volatile components of six pungent herbs were extracted by steam distillation, and their fingerprints were established by gas chromatography-mass spectrometry(GC-MS) with a programmed temperature increase(80 ℃ for 5 min, 5 ℃·min-1 to 200 ℃ for 5 min, 2 ℃·min-1 to 230 ℃ for 10 min), a splitting ratio of 20∶1, an electron bombardment ion source(EI) and the detection range of m/z 35-650, and the average MCI and total statistical moment parameters of the fingerprints were calculated. Then the matching frequency method was used to classify, integrate and confirm the chromatographic peaks of the fingerprints of six pungent herbs. ResultThe average zero order, first-order and second-order MCI values of the volatile components of Pogostemonis Herba, Artemisiae Argyi Folium, Atractylodis Rhizoma, Asari Radix et Rhizoma, Magnoliae Flos and Schizonepetae Herba were 9.02, 5.28 and 5.05, respectively. The average values of peak number, total zero-order moment, total first-order moment and total second-order moment were 60, 169×107, 22.49 min and 36.82 min2, respectively. The 20 integrated imprinting templates were obtained by the matching frequency method for the six pungent herbs, among which three were common imprinting templates with the retention times of (25.97±0.21),(26.90±0.20),(31.64±1.24) min, respectively, and the representative components were valencene,β-elemene, caryophyllin, etc. ConclusionMCI combined the matching frequency total statistical moment can divide and integrate the characteristics of imprinting templates of six pungent herbs, and find their common chromatographic imprinting characteristics, which can provide a reference for the determination of effective substances of pungent herbs.
