1.The effects of lactoferrin on the proliferation,migration and osteogenic differentiation of human periodontal ligament cells

Objective:To observe the effects of lactoferrin(LF)on the proliferation,migration and osteogenic differentiation of human periodontal ligament cells(HPDLCs)in vitro.Methods:HPDLCs were cultured and identified.The proliferation and migration of HP-DLCs cultured with 0,10 and 20 μg/ml lactoferrin respectively were tested by MTT assay,Transwell assay and scratch test.The os-teogenic differentiation of the cells was evaluated using alizarin red staining and real-time PCR.Results:Lactoferrin at 10 and 20μg/ml increased the proliferation(P <0.05),increased the quantities of mineralized nodules and the expression of alkaline phospha-tase(ALP),osteocalcin(OCN)and osteopontin(OPN)(P <0.05).Conclusion:Lactoferrin promotes the proliferation,migration and osteogenic differentiation of HPDLCs.
2.Effect of Shuanglong pill on expression of vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) on aortic atherosclerotic plaque

ObjectiveTo study the effect of Shuanglong pill on expression of vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) on aortic atherosclerotic plaque.MethodsAs atherosclerotic(AS) models of rabbits were made by immune injury plus hyperlipid diet, all rabbits were divided into blank control group(group Ⅰ,6), experimental control group(group Ⅱ, 6), captopril control group(group Ⅲ, 6; 10 mg/kg·d,12 weeks), and Shuanglong pill group(group Ⅳ, 6; 2.52 g/kg·d,12 weeks). Quantitive and qualitative analysis of expression of VEGF and BFGF on aortic atherosclerotic plaque were performed.ResultsExpressive area, density and density index of VEGF and bFGF on aortic atherosclerotic plaques were significantly decreased in group Ⅲ and Ⅳ than in group Ⅱ(P<0.01). There were significant decrease for VEGF expression in group Ⅳ than in group Ⅲ(P<0.01).Pathologic staining showed that there were no foam cells formation and no stained yellow in group Ⅰ, but mass foam cells formation and widespread stained yellow in group Ⅱ. There were a small quantity of foam cells formation and little stained yellow in group Ⅲ and group Ⅳ.ConclusionShuanglong pill can delay/reverse AS progression by inhibiting VEGF and bFGF expression in atherosclerotic plaque as well as captopril.
3.Effect of Shuanglong pill on morphology and haemodynamics in experimental myocardial infarction rats

ObjectiveTo explore the effect of Shuanglong pill which can vivid blood and relax thread pulse on cardial morphology and haemodynamics in experimental myocardial infarction (MI) rats.Methods66 rats ligated left coronary artery were randomly divided into large dose group of Shuanglong pill (LDG), small dose group of Shuanglong pill (SDG), the normal control (NC) and the control group of MI. Half in each group received 2 week treatment and the other received 4 week treatment, and were spiled in femoral and common carotid artery respectively. Parameters of cardiac haemodynamics were monitored by eight lead physiological recorder. ResultsLeft ventricular systolic maximum velocity in LDG, SDG and DC was more significantly elevated than in MI, after 2 weeks and 4 weeks of treatment, but it was lower than NC. There were no significant differences among LDG, SDG and DC. Ventricular systolic maximum velocity in LDG was more significantly elevated than in MI after 2 week treatment.ConclusionTreating MI with Shuanglong pill can improve left ventricular systolic maximum velocity, and diastolic phase function of left ventricular also elevate during the acute stage of MI by using large dose.
4.A case of hydroa vacciniforme-like primary cutaneous CD8-positive T-cell lymphoma

39℃)developed at the progressive stage of this disease.Physical examination showed variously sized,round or oval,atrophic and variola-like scars along with scattered erythematous patches,papules, necrosis and crusts on the face and extremities.The face was edematous,and there were some edematous and erythematous plaques with a necrotic center on the legs and arms.Histological examination revealed a massive infiltration with atypical CD8~+lymphocytes around the vessels and appendages in dermis.A diagnosis of CD8~+cutaneous T-cell lymphoma(CTCL)was made.Glucocorticoid and immunosuppressants were effective in controlling the condition.Up to the time of the writing,there has not been any definite evidence of systemic involvement.
5.Wnt/β-catenin pathway might underlie the MET in trans- differentiation from MSC to MSC-derived neuron.

OBJECTIVETo observe MET-associated alteration during the trans-differentiation from MSCs to neuron-like cells, and to explore the possible molecular mechanism.
METHODSBone marrow MSCs were isolated from rat femur and purified in continuous cell culture. After induced differentiation to neuron-like cells by the combination of butylated hydroxyanisole (BHA) and dimethyl sulfoxide (DMSO), cells were tested by comparative polymerase chain reaction (PCR) for the relative expression of MET biomarkers and transcription factors, and for cell cycle by flow cytometry. Meanwhile, target genes of Wnt/β-catenin pathway were also analyzed by comparative PCR to determine the possible involvement.
RESULTSIn MSC-induced neuron-like cells, MET-associated transcription factors such as Snail, Slug, ZEB1, ZEB2, and Twist were significantly attenuated in expression level. The Mesenchymal marker Vimentin expression level was increased. Membrane protein E-cad was slightly down-regulated, while N-cad level was marginally elevated. Percentage of proliferating cells (S phase in cell cycle) markedly shrank from 40.42% for MSCs to 6.76% for MSC-derived neuron. Additionally, Wnt/β-catenin target genes β-catenin and c-myc were decreasingly expressed.
CONCLUSIONChemically induced trans-differentiation from MSC to neuron caused similar MET-featured alteration in gene expression and proliferation to known MET, which might be underlied by deactivation of Wnt/β-catenin pathway.

6.Risk Factors of Contrast-induced Acute Kidney Injury in Patients Undergoing Emergency Percutaneous Coronary Intervention

Background:Previous studies of contrast-induced acute kidney injury (CI-AKI) were mostly based on selective percutaneous coronary intervention (PCI) cases,and risk factors of CI-AKI after emergency PCI are unclear.The aim of this study was to explore the risk factors of CI-AKI in a Chinese population undergoing emergency PCI.Methods:A total of 1061 consecutive patients undergoing emergency PCI during January 2013 and June 2015 were enrolled and divided into CI-AKI and non-CI-AKI group.Univariable and multivariable analyses were used to identify the risk factors of CI-AKI in emergency PCI patients.CI-AKI was defined as an increase in serum creatinine ≥25% or ≥0.5 mg/dl (44.2 tmol/L) above baseline within 3 days after exposure to contrast medium.Results:The incidence of CI-AKI in patients undergoing emergency PCI was 22.7% (241/1061).Logistic multivariable analysis showed that body surface area (BSA) (odds ratio [OR] 0.213,95% confidence interval [CI]:0.075-0.607,P =0.004),history of myocardial infarction (MI) (OR 1.642,95% CI:1.079-2.499,P =0.021),left ventricular ejection fraction (LVEF) (OR 0.969,95% CI:0.944-0.994,P =0.015),hemoglobin (Hb) (OR 0.988,95% CI:0.976-1.000,P =0.045),estimated glomerular filtration rate (OR 1.027,95% CI:1.018-1.037,P < 0.001),left anterior descending (LAD) stented (OR 1.464,95% CI:1.000-2.145,P =0.050),aspirin (OR 0.097,95%CI:0.009-0.987,P =0.049),and diuretics use (OR 1.850,95% CI:1.233-2.777,P =0.003) were independent predictors of CI-AKI in patients undergoing emergency PCI.Conclusion:History of MI,low BSA,LVEF and Hb level,LAD stented,and diuretics use are associated with increased risk of CI-AKI in patients undergoing emergency PCI.
7.Intervention on Apoptosis of Renal Tubular Epithelial Cell in Chronic Lead Poisoning Rats by Quqian Granules

Objective:To investigate the protective effect of Yiqi Huazhi recipe Quqian granules on rat renal tubular cell apoptosis induced by lead poisoning. Methods:Totally 60 Wistar rats were divided into 2 groups, 12 in the control group and the others in the model group. Chronic lead poisoning model was made by drinking 0. 02% lead acetate water for 60 days. Then the lead poisoning rats were randomly divided into four groups, high-dose Quqian granules group (3. 0 g·kg-1·d-1), low-dose Quqian granules group (0. 6 g·kg-1 ·d-1 ) , positive control group ( calcium disodium edentate plus procaine, im, 50 mg·kg-1 ·d-1 ) and model group. Seven treatment courses were carried out in the first three groups with every 4-d as one course and 4-d withdrawal period between every two courses. After 60 days, the change of lead in blood and kidney was observed by atomic absorption spectrometry,the apoptosis of kidney tissues was studied by TUNEL, the expression of Bcl-2 protein was detected by immunohistochemical methods and the expression of p53 was studied by Western Blotting. Results:Compared with the control group, the body weight, hemoglobin and the expression of Bcl-2 in the model group were decreased significantly(P<0.01)those in, and Pb in blood(0.990 ±0.443)μg·ml-1, Pb in kidney(51.33 ± 5. 16)μg·ml-1 , the apoptosis of tubular epithelial cell(4. 148 ± 0. 414) and the expression of p53 protein (1. 868 ± 0. 139) were significantly higher (P<0. 05). Compared with the those in model group, the body weight, hemoglobin and Bcl-2 in high-dose group were increased significantly(P<0.01), and the blood lead level (0.082 ±0.015)μg·ml-1, the kidney lead level (6.38 ±0.97)μg ·ml-1 , the apoptosis of tubular epithelial cell(1. 412 ± 0. 109) and p53 protein expression(1. 164 ± 0. 172) were significantly lower (P<0. 05). Conclusion:Lead may induce high expression of p53,low expression of Bcl-2 and promote the apoptosis of renal tubular epithelial cells. It is proven that Yiqi Huazhi recipe Quqian granules can inhibit the expression increase of p53 and the expression de-crease of Bcl-2 resulting in the reduction of the renal tubular apoptosis to allivate the renal injury caused by lead.
8.MEDCHART application software in management of the smoking cessation

Objective To investigate the effectiveness of Varenicline combined with MEDCHART application software for health education and follow-up in patients at smoking cessation clinic.Methods A total of 104 cases who had smoking cessation counseling and intervention at smoking cessation clinic from February 2014 to January 2015 were divided randomly into MEDCHART APP group (n=51) and conventional group (n=53).Both groups were intervened by brief admonishment and given Varenicline.The 9-12 weeks and 26th week protracted continuous abstinence rates were observed.Results There were no significant difference in 9-12 week protracted withdrawal rate between 26th week APP and conventional groups (56.9% vs.45.3%),the 26th week abstinence rates in group MEDCHART APP (41.2%) were significantly higher than the conventional group (20.8%)(x2=5.088,P<0.05).There was significant difference in full course completion rates between the two groups;15 cases (29.4%) completed the course in group MEDCHART APP,whereas 5 cases (9.4%) did so in conventional group (x2=4.405,P<0.05).Conclusion MEDCHART APP can strengthen health education and follow-up,which helps extend the time for Varenicline,increase 26th week smoking cessation withdrawal rate.It is a useful tool for smoking cessation education and follow-up.
9.Optimization on alcohol precipitation techniques of Liuwei Dihuang decoction by response surface methodology.

The theoretical basis of the alcohol precipitation process control was provided, the alcohol precipitation was optimized and the relationship equation was got. The monod glycoside, loganin and paeoniflorin were used as the evaluation indexes to determine the impact factors of alcohol precipitation techniques of Liuwei Dihuang decoction by the Plackett-Burman experimental design and the levels of non-significant factors were identified. Then, Box-Behnken response surface methodology was used to research and discuss the critical process parameters influence the effect of alcohol precipitation and draw interaction between key process parameters and the correlation equation with index components. Through the establishment and solving the quadratic regression model of composite score, the optimum preparation conditions of alcohol precipitation techniques of liuwei were as follows: stirring speed was 580 r x min(-1), standing time was 17 hours, alcohol concentration was 34%, the density of Liuwei Dihuang decoction was 1.13. The response surface methodology for optimized alcohol precipitation techniques of Liuwei Dihuang decoction is. reasonable and feasible.
