Objective:To study the satisfaction of the physical self of the junior students in Guangzhou. Methords:582 middle school students were measured by the physical self scale. Results: The junior students in Guangzhou felt a medium satisfaction at their physical self, and the highest satisfaction was appearance(3.9?0.8),then the lowest was sexual characters(3.0?0.7). 2.Orthotopic transplantation of human amniotic mesenchymal stem cells for treatment of cerebral infarction in rats

BACKGROUND: Preliminary experimental study found that the human amniotic mesenchymal stem cells (hAMSCs)transplantation can improve nerve injury symptoms of rats with cerebral infarction.OBJECTIVE: To observe the survival, colonization and differentiation of hAMSCs in the infarct area of cerebralinfarction rats.METHODS: Sixty Sprague-Dawley rats were randomly assigned into hAMSCs transplantation, model or shamoperation groups (n=20/group). Animal models of middle cerebral artery occlusion were produced in the model andtransplantation groups by Zea-Longa method. One day after modeling, rats in the hAMSCs transplantation groupwere given in situ transplantation of 10 μL of hAMSCs (2×106) into the damaged striatum and cortex, while those inthe model and sham operation group were given the same volume of PBS. Within 1 week after transplantation, ratneurological defects were assessed and changes in their body mass were continuously monitored. Two weeks aftertransplantation, TTC staining was used to observe cerebral infarct size, hematoxylin-eosin staining was used forpathological observation of brain tissues, and immunofluorescent staining was used to detect expression ofneuron-specific nuclear protein.RESULTS AND CONCLUSION: With time, weight loss was increased while neurologic deficit scores were graduallyreduced in the hAMSCs and model groups. Compared with the model group, the weight loss and neurologic deficitscores were lower in the hAMSCs group,; however, there was a significant difference in the neurologic deficit scoresbut not in the weight loss between the two groups. Additionally, the hAMSCs significantly reduced infarct size,attenuated pathologic injury, and decreased the number of inflammatory cells. Immunofluorescence stainingshowed that the hAMSCs were observed at 1 week after transplantation under inverted luorescence microscope,and gradually differentiated into nerve cells at 2 weeks after transplantation. In conclusion, transplanted hAMSCsmay migrate to and survive in the cerebral infarct region, and differentiate into nerve cells in situ in rats with cerebralinfarction.
3.Antidepressant-like effect of schisandrin in mice

OBJECTIVE To explore the antidepressant effect and the underlying mechanisms of schisandrin (SCH), a component of the fruits of Schizandra chinesis. METHODS The forced swimming test (FST) and tail suspension test (TST) in mice were used to evaluate the antidepressant activity of SCH (5, 10, and 30 mg · kg-1) following single administration intragastrically, and the locomotor activity was investigated to exclude its neural excitatory effects. Effects of SCH on neural monoamine systems were studied in two pharmacological models, including reserpine induced monoamine depletion test and yohimbine toxicity potentiation test. RESULTS In behavioral despair models, SCH (30 mg·kg-1) signif?icantly decreased the immobility time in the TST and FST (P<0.05) compared with normal control group. Results of the locomotor activity experiment showed that SCH had no excitatory or inhibitory actions on the central nervous system. In the reserpine reversal experiment, SCH (30 mg · kg-1) antagonized thepalpebral ptosis and akinesia symptoms caused by reserpine(2.5 mg · kg-1) treatment (P<0.05) compared with model group, but had little effect on the drop of the anal temperature. Moreover, SCH did not increase the lethality caused by subcutaneous injection of yohimbine (30 mg · kg-1)at the threshold lethal dosage. CONCLUSION SCH exerts potential antidepressant-like effect in mice.
4.Adaptive responses on chromosome aberration and DNA breakage of peripheral lymphocytes from workers exposed to thorium and rare earth mixed dust in Baotou Steel Plant

Objective To explore if the occupational exposure to low dose thorium could induce adaptive response in peripheral lymphocytes.Methods 40 individuals.who exposed to thorium and rare earth mixed dust(exposure group) or control in Baotou Steel Plant, were selected, and chromosome aberrations were analyzed.Then the peripheral blood samples were irradiated in vitro with 2 Gy60Co γ-rays,and unstable chromosome aberration or DNA stand breakage analysis using single cell gel electrophoresis was performed. Results The dicentrics before 2 Gy exposure in exposure group was higher than that in control(P>0.05). 5.Autophagy is involved in pulmonary artery endothelial cell apoptosis induced by cigarette smoke extract

AIM: To investigate the role of autophagy in the apoptosis of human pulmonary artery endothelial cells (HPAECs) induced by cigarette smoke extract (CSE).METHODS: HPAECs were cultured routinely.HPAECs were treated with CSE at different concentrations, and the cell viability was detected by MTT assay.HPAECs were divided into control group, CSE group, 3-methyladenine (3-MA) group and 3-MA+CSE group.The autophagy was observed under fluorescence microscope with monodansylcadaverine (MDC) staining.Annexin V/propidium iodide staining and Hoechst 33342 staining were employed to detect apoptosis.In addition, the protein levels of LC3, beclin-1 and cleaved caspase-3 were determined by Western blot.RESULTS: MDC staining showed the increased production of autophagic vacuoles was observed in CSE group.The results of Western blot showed that the expression levels of autophagy-related proteins LC3 and beclin-1 were increased, while 3-MA pretreatment inhibited the expression of these proteins and the production of autophagic vacuoles.Observation with Annexin V/propidium iodide staining and Hoechst 33342 staining showed that the apoptotic rate in CSE group was significantly higher than that in control group, and pretreatment with 3-MA induced further increase in the cell apoptosis.The protein level of cleaved caspase-3 in CSE group was significantly higher than that in control group (P<0.05), and 3-MA+CSE treatment induced the further increase in the protein level of cleaved caspase-3.CONCLUSION: CSE induces autophagy and apoptosis in the HPAECs.Inhibition of autophagy promotes the apoptosis induced by CSE in HPAECs, which can be achieved through activation of caspase-3.
6.Changes of lymphocytes and regulatory T cell subset of mice influenced by 60 Co γ-ray irradiation

Objective To analyze the influence of ionizing radiation on the lymphocytes and its regulatory T cells in mice.Methods Mice were administered with whole body irradiation of γ-rays at different doses,and lymphocytes were separated from thymus and spleen,then the number of total cells were counted and the percentages of CD4 + T and CD4 + FOXP3 + CD25 + Treg lymphocytes were analyzed by using FACS.Results The lymphocyte numbers in thymus and spleen decreased in dosedependent manner and reached to the minimum at 4 d after irradiation (F =118.08,144.01,P < 0.05).Exposure to higher dose(more than 1 Gy) decreased Treg number time-dependently in thymus,however increased it in spleen.On the contrary,exposure to lower dose (less than 0.75 Gy) increased Treg number in thymus.Besides,the percentage of Treg cells increased dose-dependently(in thymus,F =5.16,89.44,3.01,P < 0.05 ; in spleen,F =52.02,32.13,27.45,P < 0.05).Conclusions The radiation responses of lymphocytes and their Treg subpopulation vary with the different doses.Treg cells are resistant to high dose irradiation,however,their differentiation could be induced by low dose irradiation.In addition,the different time-dependent responses of lymphocytes and their subpopulation to ionizing radiation indicate the difference of lymphocyte maturation,differentiation and emigration.
7.Micronucleated reticulocytes in the peripheral blood of mice exposed to 60Co γ-rays

Objective To detect the changes of the percentage of micronucleated reticulocytes (MN-RET) in the peripheral blood of mice exposed to 60Co γ-rays,in order to provide evidence for a new biomarker of radiation biodosimetry.Methods ICR mice were irradiated in whole body with 0,0.5,1,2,4 and 8 Gy at a dose rate of 0.24 Gy/min.Peripheral blood was collected for MN-RET assay using a flow cytometry.Results The percentage of peripheral MN-RET increased steadily with irradiation doses up to 2 Gy and then had a downtrend beyond 2 Gy.The changes of MN-RET observed with a microscope were consistent with the results from flow cytometry.The dose response of the MN-RET fitted to a lineal model (R2 =0.9063),and the MN-RET at 2 Gy was significantly higher than that of nonirradiated control (t =-2.856,P < 0.05).Conclusion Percentage of peripheral M N-RET could be an early,rapid and high-throughput radiation bio-dosimeter in certain range of doses.
8.Three-dimensional element analysis of swan-like memory compressive connector for treating fractures and nonunion of humerus

Objective:To study the biomechanical basis of swan like memory compressive connector (SMC) for treating fractures and nonunions, and the facilitating effects of dynamic memorial stress of SMC for bone healing. Methods:By computer emulation and three dimensional (3D) finite element analysis, the biomechanical behavior of humeral type connector for fixing humerus was emulated and analyzed. The finite element model of humeral type connector was divided into 3 487 units. The number of nodes was 5 397. The element was a unit with 20 nodes, 4 faces. The finite element model of humerus was divided into 5 783 units, the number of nodes was 9 863. And the finite element was a 3D unit with 10 nodes, 4 faces.Results:When the connector fixated humerus, it came in for tension stress in its inner surface, and compressing stress in its outer surface. The maximum first structural major stress of metamorphosed compressive part was 224.5 MPa and -34 MPa, far less than its utmost stress and fatigue limit. The initiative memorial bone holding force for maintaining axial stability was 125.05 N, and the longitudinal initiative memorial compression force was 196 N. The stress distribution in fixed humerus was even, the stress in most nodes was positive stress. Conclusion:The SMC has good anti fatigue and reuse characters. The dynamic memorial compressive stress field is good for the stability of fixation and enhancement of bone healing.
9.Clinical observation on the efficacy enhancing and toxicity attenuating effect of nuzhen yangyin granule to the anti-parkinsonism therapy mainly with Medopa.

OBJECTIVETo observe and assess the efficacy enhancing and toxicity attenuating effect of Nuzhen Yangyin Granule (NYG) to the anti-parkinsonism (paralysis agitans) therapy with Medopa and Artane.
METHODSAdopting the randomized double-blinded method, the effect of adding NYG to 30 patients with Parkinsonism in the treated group, who already received anti-Parkinsonism treatment but showing decreased response to Medopa and Artane and with obvious adverse reaction, was observed and controlled by 30 patients treated by adding placebo.
RESULTSThe total effective rate in the treated group and the control group was 86.7% and 56.7% respectively, the total syndrome improving rate was 90% and 56.7% respectively and the toxicity attenuating rate 90% and 43.3% respectively, comparison between the two groups showed significant difference (P < 0.05 or P < 0.01). NYG also showed markedly effective in reducing the adverse reactions of Medopa and Artane on digestive, neuro-psychiatric and cardiovascular system.
CONCLUSIONNYG has obvious efficacy enhancing and toxicity attenuating effects caused by the anti-Parkinsonism treatment with Medopa and Artane.

10.Diagnosis of hematolymphoid malignancy by using effusion fluid cytology specimens: a study of 33 cases.

OBJECTIVETo study the diagnostic accuracy of hematolymphoid malignancy by using effusion fluid cytology specimens and to evaluate the values of immunocytochemistry for this assay.
METHODSThe cytospin preparations/smears and cell block sections of effusion cytology specimens from 33 cases of hematolymphoid malignancy were retrospectively reviewed. Immunocytochemical study was performed. In selected cases, in-situ hybridization for Epstein-Barr virus-encoded RNA and immunoglobulin and T-cell receptor gene rearrangement study were carried out as indicated.
RESULTSThere were 33 cases of hematolymphoid malignancy, including 12 cases of T-lymphoblastic leukemia/lymphoma, 16 cases of mature B cell neoplasm (including 9 cases of diffuse large B-cell lymphoma, 2 cases of Burkitt lymphoma, 2 cases of plasmacytoma/multiple myeloma, 2 cases of B-small lymphocytic leukemia/lymphoma and 1 case of mantle cell lymphoma), 3 cases of mature T or NK-cell neoplasm (including 1 case of extranodal nasal NK/T-cell lymphoma, 1 case of angioimmunoblastic T-cell lymphoma and 1 case of T-cell prolymphocytic leukemia), 1 case of myeloid sarcoma and 1 case of mast cell sarcoma. Amongst the 33 cases studied, 16 represented disease relapses, including 8 cases of diffuse large B-cell lymphoma, 2 cases of plasmacytoma/multiple myeloma, 2 cases of B-small lymphocytic leukemia/lymphoma, 1 case of T-lymphoblastic leukemia/lymphoma, 1 case of angioimmunoblastic T-cell lymphoma, 1 case of mantle cell lymphoma and 1 case of mast cell sarcoma. The remaining 17 cases showed serous effusion as the primary manifestation, with the diagnosis primarily made upon cytologic examination. The cytologic findings seen in all the 33 cases studied were in agreement with the corresponding histologic diagnosis.
CONCLUSIONSDiagnosis of hematolymphoid malignancy by effusion fluid cytology specimens is possible, especially when coupled with the clinical history, immunophenotype, in-situ hybridization and gene rearrangement study findings. This is especially so for cases with disease relapses.
