2.Experimental Study of the Interference of Hongjingtian in Cytokines in Hippocampus of Post Stroke Depression Rats

Objective To study the interference of Hongjingtian in cytokines in hippocampus of post stroke depression(PSD) rats.Methods The healthy SD rats were randomly divided into five groups:simple focal cerebral ischemia group,PSD group,Hongjingtian group of three doses [2.5,5,7.5 g/(kg?d)].PSD rats model was established.Hongjingtian group was given Hongjingtian and other groups were given normal saline.The ethological score of depression was evaluated.Pathological changes of hippocampus were observed by HE staining and the content of TNF-?,IL-1? and IL-6 in hippocampus were measured.Results Compared with normal group,the expression of the content of TNF-? and IL-1? of other groups increased,while the content of IL-6 decreased,the ethological score of depression decreased.HE staining of hippocampus showed neurocyte damaged obviously.The difference between them was significant(P0.05).Conclusion The steady state of network of cytokines in PSD experienced disequilibrium and neurocyte of hippocampus were damaged worse than that in simple focal cerebral ischemia.Hongjingtian has neuroprotective effects on PSD rats,its mechanism may through improving the damaged extent of neurocyte of hippocampus by way of reducing the content of TNF-?,IL-1? and increasing the content of IL-6 in hippocampus.
3.Combination of urine sediment quantitative analyzer and urine dry chemistry analyzer in detection of urine cell

Objective To evaluate the efficacy of IQ200 urine sediment quantitative analyzer(IQ200) and Mejer700 urine dry chemistry analyzer(Mejer700) in detecting urine erythrocyte(RBC)and leukocyte(WBC) .Methods The WBC and RBC in 1 000 u‐rine specimens were detected by using IQ200 ,Mejer700 and sediment microscopy respectively .Taking sediment microscopy as the golden standard ,the sensitivity ,specificity and accuracy of IQ200 and Mejer700 in detecting urine RBC and WBC were analysed .Re‐sults No statistically significant differences of positive rates of RBC and WBC detected by using the three methods were found (P>0 .05) .When IQ200 and Mejer700 were combined ,the sensitivities of detection of RBC and WBC test were improved .Conclu‐sion The sensitivities of RBC and WBC detection by using IQ200 and Mejer700 are high .Combing the two methods the sensitivity and accuracy is higher ,which could improve work efficiency .Combination of the three methods should be recommended ,in order to improve the quality of urine analysis .
4.Study on the Distribution of Human Leukocyte Antigen B27 Positive Expression in Uygur and Han Nationalities in Xinjiang

Objective To study the relationship between the positive expression of human leukocyte antigen B27 (HLA-B27) in different age groups and genders in Uygur and Han nationality in Xinjiang,and explore the clinical value of HLA-B27 in different ethnic groups.Methods From January 2013 to November 2016,1 416 cases of Uygur and Han patients with positive expression of HLA-B27 were detected by flow cytometry.There were 369 cases of Uygur and 1 047 cases of Han.Results Independent samples t-test showed that the positive expression of HLA-B27 in Uygur and Han population was statistically significant (165.22±8.262 vs 163.99±8.113,t=2.479,P=0.013).In male,there was a significant difference in the positive expression of HLA-B27 between Uygur and Han population (165.40 ± 8.237 vs 163.99 ± 8.164,t =2.187,P =0.029).In the age of 41~60 years old and >60 years old,the positive expression of HLA-B27 in Uygur was higher than that in Han nationality (166.18 ± 7.650 vs 164.53 ± 8.018,t =2.215,P =0.027;171.63 ± 8.134 vs 167.40 ± 9.469,t =2.126,P=0.035).There was no significant difference in the positive expression of HLA-B27 between Uygur and Han nationality in women,as well as in the age of 20 years and 21~40 years (t=-0.029~1.257,all P>0.05).Conclusion The investigation showed that the positive expression of HLA-B27 in Uygur was higher than that in Han nationality.The content of HLA-B27 positive expression has racial difference.
5.Study on Determination Method of Total Flavonoids in Gansu Astragali Radix and ;Hedysarum Polybotrys

Objective To study the determination method of total flavonoids in Gansu Astragali Radix and Hedysarum Polybotrys. Methods Calycosin glycosides etc. was selected as reference substances, comparison on the difference of absorption curves was done by ultraviolet spectroscopy and colorimetric method (NaNO2-Al(NO3)3-NaOH, AlCl3, Mg(Ac)2, NaOH, phosphomolybdic acid, HCl-Mg power). Results With colorimetric method, the maximum absorption wavelength of referrence and the test was inconsistent. The absorption peak shape was also different. With UV method, Calycosin glycosides in band Ⅱ (260 nm) showed a shoulder absorption. Astragali Radix and Hedysarum Polybotrys also showed characteristic shoulder absorptions in band Ⅱ with absorption wavelength at 263 nm and 265 nm. So the sample absorption wavelength is basically the same as that of the control sample. Conclusion Colorimetries usually used for determination of total flavonoids are not suitable for the comparison determination of Gansu Astragali Radix and Hedysarum Polybotrys. It is suitable for determining the contents of total flavonoids in samples by UV spectrophotometry at the band Ⅱ, which is the characteristic absorption band of isoflavone compound.

Objective:To know the work status of clinical pharmacy in medical institutions of Guizhou province. Methods:Ques-tionnaires were used to analyze the situation of clinical pharmacy in 108 medical institutions of Guizhou province. Results: A total of 246 questionnaires were taken back, and among the 231 valid questionnaires were received including gradeⅡor above hospitals. The main contents of clinical pharmacy work carried out in medical institutions included 7 aspects: pharmacists ’ participation in ward rounds, which accounted for 47. 11%; pharmacists’ participation in case consultation, which accounted for 16. 65%; pharmacists’ participation in teaching practice, which accounted for 38. 84%; pharmacists’ participation in prescription evaluation and analysis, which accounted for 72. 73%;pharmacists’ participation in antimicrobial drug monitoring and drug use evaluation, which accounted for 62. 37%;pharmacists’ participation in drug counsultation and education, which accounted for 58. 68%;pharmacists’ participation in adverse drug reaction monitoring and supervision, which accounted for 77. 32%. Conclusion:The development of clinical pharmacy in Guizhou province still lags behind, and the number of clinical pharmacists is insufficient, which can’ t meet the growing demand for personalized medicine. In particular, the development of clinical pharmacy is restricted by the limited pharmaceutical service. The cog-nition degree of pharmacist group in Guizhou province has been improved. However, the number and the service quality of clinical pharmacists need to be improved further.
8.Effect of RNA interference on EGF receptor expression of non-small-cell lung cancer A549 cell line.

OBJECTIVETo investigate changes in biologic properties of non-small-cell lung cancer (NSCLC) A549 cells whose EGF receptor (EGFR) expression was suppressed by short interference RNA (siRNA).
METHODSA549 cells were transfected with synthetic EGFR sequence-specific siRNA by Lipofectamine. EGFR expression was examined by Western blot and flow cytometry. The biological features of the transfected A549 cells were assessed by cell cycle analysis, colony formation and chemosensitivity assay.
RESULTSSequence-specific siRNAs targeting EGFR significantly down-regulated its expression in A549 cells. Cell growth and colony formation were inhibited by 85.0% and 63.3%, respectively, as compared to the non-sequence-specific siRNA treated cells. Decreased EGFR expression was accompanied by 12.7% increase in A549 cells in G(0)-G(1) phase and 6.6% decrease in S-phase. The EGFR sequence-specific siRNA transfected A549 cells were much more sensitive to the cytotoxic effect of cisplatin with a 77.2% decrease in IC(50) compared to the non-sequence-specific iRNA transfected A540 cells.
CONCLUSIONDown regulation of EGFR expression of NSCLC by sequence-specific siRNA may be considered as an additional option in the treatment of EGFR over-expressing cancers, including NSCLC.

9.Monitoring of Oxidation Process of Human Milk Lipid Substitutes by Molecular Fluorescence Spectroscopy

The fatty acid contents in soybean oil, corn oil, coconut oil, sunflower seed oil and fish oil were detected by gas chromatography. According to the characteristics of fatty acids of raw oil and the relevant regulations of GB 10765-2010,the formula of 6 kinds of human milk lipid substitutes was designed by Matlab software and Excel linear programming. At the same time, the Schaal oven accelerated oxidation method was used to study the oxidation process of 6 formulations using synchronous fluorescence technique combined with traditional chemical reagents (oxidation index:peroxide value,value of fennel and total oxidation value). By statistical analysis of data, the statistical relationship between fluorescence intensity and oxidation index was explored. The results showed that the main fatty acids of coconut oil were lauric acid and myristic acid,and its saturated fatty acid content was the highest 93.75% ±0.06%;while all the other oils were mainly composed of palmitic acid,linoleic acid and other components,wherein the fish oil had the highest content of unsaturated fatty acid. With the increase of oxidation time,the fluorescence intensity decreased gradually and the oxidation index value increased continuously. The correlation analysis and regression analysis results showed that the change of oxidation index and the fluorescence intensity of 6 samples had negative correlation, and the fluorescence intensity and the peroxide value and total oxidation value was quadratic linear correlation, and anisidine value as a linear correlation had strong correlation.
10.Breeding of Yeast Fusant for Efficient Ethanol Fermentation from Xylose

Yeast strains with improved ethanol yield are important for efficient bioconversion of lignocellulosic biomass for fuel ethanol.Candida shehatae CICC1766 was adapted to 4%(v/v)ethanol,and then subjected to UV mutagenesis.One respiration deficient mutant Rd-5 with improved xylose fermentation capability was selected.Protoplasts of Rd-5 were inactivated by UV treatment,followed by the PEG-mediated protoplast fusion with a Saccharomyces cerevisiae strain with good ethanol-fermenting capability.The xylose fermenting capability of the fusants was investigated,and the fusant F6 demonstrated good ethanol fermentation performance,producing 18.75g/L ethanol from 50g/L xylose with an ethanol yield of 0.375 or 73.4% of its theoretical value of 0.511.Comparing with its parent Candida shehatae strain,the ethanol yield of F6 was increased by 28%.
