1.The detection of expression and significance of TGase3 mRNA in esophageal carcinoma by tissue microarray and in situ hybridization methods

Objective To study the expression of TGase3 mRNA in esophageal carcinoma and to investigate it's role and the significance of in the carcinogenesis and progression of esophageal carcinoma.Methods The expression of TGase3 mRNA in 70 ca8es of esophageal carcinoma tissue and 10 cases of normal esophageal tissue were detected by tissue miemarray(TMA)and in situ hybridization(ISH)methods.Results The loss expression rate of TGase3mRNA in esophageal carcinoma tissue was 68.57%(48/70),which was significantly higher than in normal control(P<0.05).The expression of TGase3 mRNA was no correlation with patients'sex,age,tumor site,lymph node metastasis.But it's significant correlation with clinical stage,histology grade,Pathology grade(P<0.05).Conclusion There was a high frequency of TGase3 mRNA expression loss in esophageal carcinoma,TGase3 mRNA gene may be involved in carcinogenesis and progression of esophageal carcinoma.

3.Clinical observation of locally advanced pancreatic cancer treated by compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy

Objective To study the recent clinical efficacy of compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy in the treatment of locally advanced pancreatic cancer.Methods 48 cases of untreated locally advanced pancreatic cancer were randomly divided into treatment group (compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy) and control group (three-dimensional conformal intensity modulated radiationtherapy),there were 24 cases in each group.Three-dimensional conformal intensity modulated radiation therapy used 95 % isodose coverage PTV.Using 6 MV X-ray irradiation,2 Gy/day,5 times/week,the total radiation dose was 50 Gy/25 times/5 weeks to complete.Treatment group:one week before treatment infusion of compound matrine injections 20 ml/time,l/d,21-day was one cycle.Control group:simple intensity modulated radiotherapy.Results After treatment,the clinical benefit response rate of the treatment group was 87.5 % (21/24),the control group was 70.83 % (17/24),the difference was statistically significant (P =0.045).Term efficacy rate of the treatment group was 79.17 % (19/24),the control group was 50.00 % (12/24),the difference was statistically significant (P =0.035).Adverse reactions were hematologic toxicity,leukopenia occurred in the treatment group was 16.67 % (4/24),but the control group was 45.83 % (11/24),the difference was statistically significant (P =0.029).Conclusion The effects of compound matrine injections combined with three-dimensional conformal intensity modulated radiotherapy for locally advanced pancreatic cancer are good,they can relieve pain,improve short-term efficacy,reduc side effects,and improve the quality of life.

Objective To identify the factors influencing the response to clopidogrel.Methods Seventy-two patients with acute coronary syndrome who were treated at Beijing Tongren Hospital from September,2005 to April,2006 were eligible for this study.10 ?mol/L ADP induced platelet aggregation was measured before and 7 days after clopidogrel treatment by the traditional optical aggregometry.According to the inhibition of platelet aggregation(?A),the population was divided into low response group and normal response group.Then some factors possibly influencing the response to clopidogrel were analyzed by specific statistical methods.Results Baseline characteristic showed that compard with that of normal response group,the body mass index and the percentage of obesity and STEMI were lower in low response group,while its percentage of hypertension and diabetes were higher(P
5.Taxonomy and Application of Micromonospora

Micromonospora distributes extensively in environment. But most of them have not been known because the limitation of isolation and taxonomic approach. The phenotypic information such as morphological, cultural and physiological characteristics have been applied widely in the taxonomy of Micromonospora. The functions of molecular methods become more and more important in the taxonomy of Micromonospra accompanying with the development of the polyphasic taxonomy. Micromonospora is one of the most important sources in finding new bioactive compounds, some of them can yield antibiotics, such as gentamicins, rafomicins and newmicins, some of them can degrade natural rubber and fibrin. It indicated in recent years’ study that some bioactive compounds with particular chemical construction could be found from Micromonospora. These compounds can identify cancer cells and kill them effectively.

Objective:To study the effect of different doses of oxycodone on the serum thromboxane A2 (TXA2),plasma endothelin (ET) levels and immune function of patients underwent laparoscopic cholecystectomy.Methods:90 patients of elective laparoscopic cholecystectomy who were treated from August 2013 to August 2016 in our hospital were selected and divided into 3 groups by random number table,with 30 cases in each group.At the beginning of operation,they were given intravenous oxycodone,group A was given 0.1 mg/kg oxycodone,group B was given 0.2 mg/kg oxycodone,group C was given 0.3 mg/kg oxycodone.The changes of hemodynamics,serum TXA2 and ET levels were compared between the three groups at T0 (after admission),T1 (after anesthesia induction),T2 (after intubation),T3 (gallbladder separation),T4 (end of surgery) and the changes of immune function was compared at T0,T5 (postoperative 2h),T6(postoperative 1d),T7 (postoperative 3d);and the extubation time,recovery time,hypotensor,additional analgesics situationin and adverse reactions were recorded.Results:The diastolic pressure (DBP),systolic blood pressure (SBP),heart rate (HR) in three groups at T2,T3 point was significantly higher than T0 point(P<0.05),the DBP,SBP and HR in the B,C groups were significantly lower than the A group at T2 and T3 point(P<0.05);the TXA2 in three groups at T2,T3,T4 point was significantly higher than T0 point(P<0.05),and A group>B group>C group,there was significant difference between the two groups (P<0.05);the ET in three groups at T2,T3 point was significantly higher than T0 point(P<0.05),the ET in the B,C group was significantly higher than the A group at T2,T3 point(P< 0.05);the CD3+,CD4+,CD8+ and CD4+/CD8+ of the three groups at T5 point were significantly lower than that ofT0 point (P<0.05),the CD3+,CD4+,CD8+ and CD4+/CD8+ in A group were significantly lower than that of B,C group at T5 point (P<0.05);the extubation time in C group was significantly longer than that of A,B group(P<0.05);the total incidence of adverse reactions in C group was significantly higher than that the A,B group (P<0.05).Conclusion:In the laparoscopic cholecystectomy,application of 0.2 mg/kg oxycodone had little effect on hemodynamics,serum TXA2,ET levels and immune function with higher safety.

Objective To detect the pathological state of metabolic syndrome and the expression of microRNA-205 in women with endometrial cancer.Methods Thirty-eight cases of patients with endometrial cancer were collected including waist circumference,weight,blood pressure.Collected preoperative fasting blood,postoperative endometrial lesions and endometrium tissue adjacent to carcinoma paraffin section.Analyzed the metabolic syndrome in patients with pathological state indicators,extracted the total RNA of endometrium tissue and the level of microRNA-205 was detected.Results The prevalence of metabolic syndrome was 44.74% (17/38).The abnormal rate of high-density lipoprotein cholesterol,blood pressure,triglycerides,waist circumference,fasting blood sugar successively were 52.6% (20/38),36.8% (14/38),34.2% (13/38),34.2% (13/38) and 26.3% (10/38).The expression level of microRNA-205 in endometrial carcinoma was higher than that in normal endometrium tissues,there was significant difference (P < 0.01).Conclusions Metabolic syndrome is closely associated with the metabolic syndrome and its components.The expressions of microRNA-205 may participate in the occurrence and development of endometrial carcinoma,and it may be as an early screening of molecular markers.
9.Development of the mouse spinal cord and neuroapoptosis

Objective To investigate the neural proliferation , differentiation and apoptosis of the developing spinal cord of the mouse and to discuss the mechanism of spinal cord ’ s development .Methods 5-Bromodeoxyuridine ( BrdU) assay was used to mark the proliferative neural stem cells , and the immunofluorescent stainings ( DCX, NeuN and Caspase8) were carried out to visualize the newborn neurons , mature cells and apoptotic cells in the spinal cord with 173 mice arrange from E18 to P90.Results BrdU positive neural stem cells appeared evenly in the spinal cord at early days . With age increasing , the neural stem cells differentiated into neuroglial cells and neurons .The newborn neurons in the subventricular zone migrated toward the intermediate zone ( putative gray matter ) and differentiated into mature neurons gradually .With neurons ’ concentrating towards the center , the gray matter formed an “H” shape .In the meantime , with neural differentiation , some apoptotic neurons appeared among the newborn neurons and mature neurons . Double immunostaining showed that most apoptotic neurons were newborn neurons , suggesting the neuroapoptosis more likely occurred in newborn neurons .The statistical data showed that the number of DCX , NeuN and Caspase-8 positive cells reduced with age increasing , suggesting neural differentiation and neuroapoptosis decreased during spinal cord ’ s development .Conclusion Neural proliferation , neural differentiation and neuroapoptosis occur in developing spinal cord . They work together to regulate the formation and development of the spinal cord .
