Chinese Traditional and Herbal Drugs 2013;44(7):863-868
Objective: To evaluate the hypoglycemic effect of aqueous extract from Taraxaci Herba (AETH) on postprandial blood glucose (PBG) in diabetic rats induced by Streptozotocin (STZ) and to explore the mechanism. Methods: The PBG within 120 min was measured after 7 d ig administration of AETH (400, 200, and 100 mg/kg) in normal rats and diabetic model rats induced by STZ, and ELISA was performed to detect the serum insulin of rats in each group. The euglycemic clamp assay was performed to investigate the effect of AETH (400 mg/kg) on the insulin sensitivity in normal rats, and the glucose infusion rate (GIR) was detected. The 50% inhibiting concentration (IC50) against α-glucosidase was measured in vitro, using pNPG as substrate. Caco-2 cells were preincubated for 72 h with AETH (200 and 100 mg/L), and then the capacities of Caco-2 monolayer on maltose hydrolysis and glucose absorption were measured. Results: The 7 d ig administration of AETH decreased the PBG significantly in diabetic rats induced by STZ, but had no effect on serum insulin level and GIR. AETH inhibited α-glucosidase in vitro, and the IC50 was higher than that of acarbose. AETH (200 mg/L) inhibited the capacities of Caco-2 monolayer on both maltose hydrolysis and glucose absorption. Conclusion: AETH has the hypoglycemic effect on PBG in diabetic rats, and the mechanism is related to the inhibition of maltose hydrolysis and glucose absorption in small bowel.

Neonatal mortality has been significantly decreased because of the development of neo-natal respiratory support techniques. Nevertheless, constant and high frequency ventilators have some limita-tions to solve all problems in neonatal respiratory failure. Special mechanical ventilation, mainly including he-liox and partial liquid ventilation, provides new respiratory support for newborn infants. The clinical signifi-cance of heliox and partial liquid ventilation need further investigation.

Objective: By observing the differences in Western Ontario and McMaster Universities osteoarthritis index (WOMAC) and visual analog scale (VAS) scores in moxibustion treatment for moderate-to-severe primary knee osteoarthritis (KOA) with moxa of different storage years (3-year moxa and 1-year moxa from Qichun, Huanggang City, Hubei Province, China) through a randomized clinical trial, to objectively evaluate the differences in therapeutic efficacy of moxibustion with moxa of different storage years. Methods: A total of 63 patients with moderate-to-severe KOA who met the inclusion criteria were randomized into moxibustion group 1 and moxibustion group 2 by central randomization method, with 32 cases in moxibustion group 1 and 31 cases in moxibustion group 2. Moxibustion group 1 was treated with moxa stored for 3 years, and moxibustion group 2 was treated with moxa stored for 1 year. Dubi (ST 35), Neixiyan (EX-LE 4) and Heding (EX-LE 2) were selected in both groups, and the treatment lasted 20 min per time, 3 times a week. The immediate efficacy was compared after 6 times of treatment, and long-term efficacy was compared at follow-up 4 weeks after the end of treatment. Results: During the treatment, there were 2 dropouts in moxibustion group 1, and 1 dropout in moxibustion group 2. The total effective rate in the two groups was 83.3% and 60.0%, respectively. Followed up at 4 weeks after the end of treatment, the total effective rate in the two groups was 80.0% and 66.7%, respectively. There were no statistical differences between the two groups (both P>0.05). After treatment and 4 weeks after the end of treatment, the WOMAC and VAS scores in both groups decreased significantly compared with those before treatment (all P<0.01); the scores of stiffness item of WOMAC in moxibustion group 1 were lower than those in moxibustion group 2 (both P<0.05); there were no statistical differences in the scores of pain item and dysfunction item of WOMAC, and VAS scores between the two groups (all P>0.05). Conclusion: Moxibustion with moxa of different storage years (stored for 3 years and 1 year) both can improve the pain, stiffness and motor function in patients with moderate-to-severe KOA. While moxa stored for 3 years has a better therapeutic efficacy in improving stiffness of the knee joint than that stored for 1 year.

Objective To investigate the expression of occludin in the intestinal epithelial cells of neonatal rats with necrotizing enterocolitis ( NEC) . The influence of vitamin D on NEC and the expression of occludin was also explored. Methods Sixty neonatal Wistar rats from different litters were divided into four groups 48 hours after birth:10 in the breast-fed+ control group,10 in the breast-fed +vitamin D group,20 in the NEC+ control group and 20 in the NEC+ vitamin D group. In groups with NEC,neonatal rats were sep-arated from their mothers and treated with rat milk substitute+hypoxia+hypothermia. In groups with vitamin D treatment, paricalcitol was given through intraperitoneal injection 30 minutes before, one day and two days after the induction of NEC. The tissue was harvested from the ileum of the rats 72 hours after induction of NEC. Microscopically, the tissue was assessed according to the Nadler scoring system. The expression and location of occludin was observed after immunofluorescence staining. Western blotting was performed and quantified to compare the expression of occludin among groups. Results In NEC rats, the intestinal structure was destructed. Separation of the submucosal or muscular layer, discharge or disappearance of the villa was accompanied by necrosis of the intestinal cells. Vitamin D treatment could significantly alleviate those chan-ges,with the Nadler score 1. 70 ± 0. 21 compared to 2. 90 ± 0. 23 without vitamin D treatment ( P<0. 01 ) . The expression of occludin was decreased in NEC rats and vitamin D could partly rescue this decrease (P<0. 01). The distribution of occludin was sparse and discontinuous in NEC rats while after vitamin D treatment, the distribution was maintained even and continuous. Conclusion The expression of occludin in intestinal epithelial cells is significantly reduced in NEC rat. Vitamin D treatment may inhibit the development of NEC by maintaining the expression of occludin.
