1. Pituitary adenylate cyclase-activating polypeptide modulates immune function of LPS-stimulatd dendritic cells

Objective: To investigate whether pituitary adenylate cyclase-activating polypeptide (PACAP) can modulate the immune function of mouse LPS-stimulated dendritic cells. Methods: DCs were derived from bone marrow of C57BL/6 using rmGM-CSF and rmIL-4, then the DCs were exposed to different stimuli for indicated time periods. The stimulated DCs and their culture supernatant were subjected to ELISA and FACS analysis. The total RNA was extracted from the stimulated DC for RT-PCR and RNase Protection Assay. Results: PACAP significantly inhibited the production of IL-2, IL-12,TNF-α and MIP-2 in LPS-stimulated DCs (P<0. 05,P<0. 01,P<0. 01 ,P<0. 01.respectirely), but the inhibition of LPS-induced IL-6, MIP-1α and MIP-1β production was not apparent. Conclusion: PACAP can negatively modulate the immunity of LPS-stimulated DCs.
2. Chemical constituents from Tibetan medicine Swertia chirayita

Objective: To investigate the chemical constituents of Swertia chirayita. Methods: Column chromatography, such as silica gel, MCI, Sephadex LH-20 were used to isolate and purify the compounds. Physicochemical properties and spectroscopic methods were used to elucidate their structures. Results: Twelve compounds, including 2 xanthones, 4 triterpenoids, 3 secoiridoids, and 3 other compounds, the chemical constituents were isolated from the ethyl acetate fraction from 85% ethanol extract of S. chirayita, and identified as bellidifolin (1), norbellidifolin (2), oleanolic acid (3), 4-epi-hederagenin (4), 2-epi-corosolic acid (5), ursolic acid (6), amarogentin (7), swerimilegenin I (8), erythrocentaurin (9), pyrocatechol (10), syringic acid (11), and 4-hydroxy-3-methoxybenzoic acid (12). Conclusion: Compounds 4, 5, and 11 are isolated from genus Swertia for the first time, compounds 8 and 9 are found from S. chirayita for the first time.
3.Effects of perindopril on bone metabolism in a rat model of retinoic acid-induced osteoporosis

BACKGROUND: Renin-angiotensin-aldosterone system existed in bone tissue. Recent studies on antihypertensive drugs found that angiotensin converting enzyme inhibitor type antihypertensive drug was possibly effective for osteoporosis. Perindopril is one of the commonly used antihypertensive drugs. Whether perindopril affected bone metabolism or could be used in anti-osteoporosis has not been reported.
OBJECTIVE: To observe effects of perindopril on bone metabolism in a rat model of osteoporosis induced by retinoic acid.
METHODS: Fifty Sprague-Dawley rats were randomly divided into five groups, with ten in each group. In the model group and each perindopril groups, rats were intragastricaly administered retinoic acid solution 80 mg/kg per day. After successful model establishment, rats in different perindopril groups were intragastrical y administered perindopril 2, 4 and 8 mg/kg per day, once a day, for 42 consecutive days. In the normal control and model groups, rats were given an equal volume of distil ed water. Serum calcium, phosphorus, alkaline phosphatase, bone mass and bone mineral density were detected in each group. Expression of bone specific alkaline phosphatase and tartrate-resistant acid phosphatase mRNA in bone tissue was determined.
RESULTS AND CONCLUSION: Compared with the model group, after treatment with perindopril, serum calcium and phosphorus levels were increased, alkaline phosphatase activities were significantly decreased, bone mass and bone mineral density were obviously increased in rats with retinoic acid-induced osteoporosis. Expression of bone specific alkaline phosphatase mRNA was higher in the perindopril 8 mg/kg group than in the perindopril 2 and 4 mg/kg groups and model group. Tartrate-resistant acid phosphatase mRNA expression was higher in the perindopril 8 mg/kg group than in the model group. These results indicated that perindopril could improve partial bone metabolic biochemical markers in osteoporosis rats, promoted bone formation by up-regulating bone specific alkaline phosphatase mRNA expression, and had a certain preventive effect on retinoic acid-induced osteoporosis.
4.The analysis of pulmonary infect flora distribution and risk factors of stroke patients with tracheotomy in intensive care unit

Objective To investigate pulmonary infect flora distribution characteristics of stroke patients with tracheotomy in Intensive care unit (ICU) and analyze the risk factors.Methods A total of 792 cases of ICU stroke patients was selected,including 426 cases of tracheotomy patients,and 366 cases of non-tracheotomy patients.The incidence of lung infections was compared.Bacteriological examination was used for tracheotomy bacteriological studies for lung infected stroke patients with tracheotomy.Bacteria infection's characteristics was observed.The risk factors were analyzed.Results (1) The tracheotomy patient 's lung infection rate was 23.00%,higher than 9.56% of the non-tracheotomy patients,the difference was statistically significant (X2 =19.125,P <0.05);(2) For lung infection-occurred patients with tracheotomy of ICU,gram-negative bacteria infection rate was 63.97%,significantly higher than 19.85% of gram-positive bacteria and 16.18% of fungi,the difference was statistically significant (x2 =18.255,17.042,P < 0.01);(3) Lung infection rate of ICU stroke tracheotomy patients with unconscious,dysphagia,hospitalization time > 14 d,blood glucose levels ≥≥7.8 mmol/L,and based diseases was significantly higher than that of patients with conscious,non-dysphagia,hospitalization time ≤<14 d,blood sugar level <7.8 mmol/L,and no based diseases (P < 0.05);(4) Consciousness,dysphagia,hospitalization,blood sugar levels,and the underlying disease were the independent risk factors of lung infection in ICU stroke tracheotomy patients (OR1 =11.528,OR2 =8.046,OR3 =15.174,OR4 =7.795,ORs =10.784,P <0.05).Conclusions Gram-negative bacteria is the main reason for pulmonary infections in ICU stroke tracheotomypatients patients with stroke.State of consciousness,invasive treatment,hospitalization,blood sugar levels,and the underlying disease are the independent risk factors.

Vascular endothelial growth factor (VEGF) is an endothelial cell-specific mitogen.Studies have shown that VEGF is closely associated with ischemic stroke,and this makes it possible to intervene in ischemic stroke from the level of VEGF and its receptor.This article reviews the biological effect of VEGF and its receptor,mechanism of action involving in various stages of ischemic stroke,and the therapeutic prospect in ischemic stroke.
6.Value of MRI in qualitative and quantitative diagnosis of bone marrow infiltration in lymphoma

Objective To evaluate the value of MRI in qualitative and quantitative diagnosis of bone marrow involvement in patients with lymphoma. Methods 28 diagnosed lymphoma with bone marrow infiltration and 31 healthy spines as controls were included.MRI performance and the signal intensity ratio on T1WI (SIR1) of the spine bone marrow and cerebrospinal fluid were analyzed and statistical analyses were performed.Results Qualitative diagnosis among these patients,was shown than MRI results indicated that 27 cases had abnormal signal. The sensitivity of MRI was 96.4 %. There were four main patterns of marrow infiltration in MRI,containing nodular pattern 21.4 % (6/28),scattered pattern 53.6 % (15/28),mottled pattern 14.3 % (4/28) and uniform pattern 7.1% (2/28) respectively.By quantitative diagnosis the SIR1 of study group (1.251±0.253) was apparently lower than that in the control group (2.625±0.434) with statistical significance (t =15.022,P < 0.001).The results of multiple comparisons showed that the SIR1 of mild degree (1.390±0.172),moderate degree (0.982±0.790) and severe degree (0.908±0.122) patients with lymphoma compared with the normal controls had significant differences (all P =0.000),and difference between mild and moderate degree had statistical significance (LSD,P =0.012),so did the difference between mild and severe degree (LSD,P =0.025).However,no significant difference could be seen between moderate and severe degree (LSD, P =0.757). Positive linear correlation was existed between the persentages of SIR1 and tumor cells in bone marrow (r =-0.765, P < 0.001). Conclusion As a noninvasive and direct-viewing technique, MRI presents a global view of bone marrow with high sensitivity in detecting bone marrow involvement in lymphoma. To a certain extent, it could be possible to estimate the infiltration degree and evaluation of the tumor burden in bone marrow by quantitative measurement of MRI.
7.Spectral CT characterizing the pathological type of gastric cancer

Objective To evaluate spectral CT imaging in characterizing the pathological type and the differentiation of gastric cancer.Methods Ninety-one patients diagnosed of gastric cancer were retrospectively analyzed.Patients were all underwent triple-phase enhanced scan using single source dualenergy CT on gemstone spectral imaging(GSI) mode.Three types of images were reconstructed for analysis:the water concentration (WC),iodine concentration (IC),and normalized iodine concentration (NIC).The patients were divided into groups of mucinous carcinoma (MUC),including mucinous adenocarcinoma and signet ring cell carcinoma,and non-mucinous gastric carcinoma (non-MUC).Independent-samples t test was used for statistical analysis.Results There were 24 patients of MUC and 67 patients of non-MUC.The IC and NIC of the non-MUC were significantly higher than the MUC in vein phase (VP) and in parenchymal phase (PP) [(21.33 ±6.31) × 100 vs (16.94 ±6.13) × 100 μg/ml,0.55 ±0.17 vs 0.42 ±0.14 in VP,and(20.65 ±5.49) × 100 vs (18.07 ±4.51) × 100 μg/ml,0.72 ±0.20 vs 0.57 ±0.12 in PP,respectively t =-2.948,-3.362,-2.261,-4.326,P <0.05].The IC and NIC of the signet ring cell carcinoma were statistically higher than those of the mucinous adenocarcinoma [(19.36 ± 4.75) × 100 vs (12.10 ±5.92) ×100μg/ml,0.49 ± 0.09 vs0.28±0.11 in VP,and (19.88±3.28) ×100 vs (14.45±4.62) ×100 μg/ml,0.63 ±0.08 vs 0.47 ±0.11 in PP,respectively t =3.253,5.180,3.339,3.850,P <0.01].For the poorly differentiated and well /moderately differentiated adenocarcinoma,the IC and NIC of the former were higher than those of the latter in VP and PP [(23.48 ±6.49) × 100 vs (18.98 ±5.26) × 100 μg/ml,0.61±0.16vs0.48±0.15 in VP,and (22.95±5.51) ×100 vs (18.28±4.47) ×100 μg/ml,0.81 ± 0.21 vs 0.63 ± 0.15 in PP,respectively t =3.098,3.249,3.766,4.117,P < 0.01].The between-group differences of WC,IC and NIC in artery phase were no significant (P > 0.05).Conclusions Different pathological types could determine different IC.The spectral CT imaging may be helpful for characterizing the pathological type of gastric cancer.

OBJECTIVE:To study the stability of omeprazole sodium solution for injection under the influence of metal ion calcium and magnesium.METHODS:HPLC was adopted to determine the variation of the content of omeprazole sodium solution acted by metal ion calcium and magnesium of different concentrations in different time.The change in colour and luster was also observed.RESULTS:Omeprazole sodium solution tended to disintegrate rapidly along with the prolonging of time,and increase with the increasing of concentration;when Mg2+ was added,change in color was seen,with no degradation substance produced;when Ca2+was added,change in color was also seen,with white deposit produced.CONCLUSION:Ca2+ and Mg2+ show significant influence on the stability of omeprazole sodium solution.
