Objective: To study the antioxidative and hepatoprotective activities of low molecular fucoidan oligosaccharides(LMFO) from Laminaria japonica in mice. Methods: Mice were pretreated with LMFO(50?100?150 mg/kg ig respectively, 10 days),and then 0.2 % CCl 4 10 ml/kg ig and D-GalN(600 mg/kg)+LPS(lipopolysaccharide,1 ?g/kg) ig respectively in two model groups to induce liver injury. Liver injury was assessed by quantifying activities of plasma GPT, SOD, GSH-Px and MDA content. Results: The increase of plasma GPT activity was significantly inhibited by LMFO in two liver injury models, suggesting that LMFO had good protective effect on the hepatocytes. LMFO had good antioxidative effect in mice with liver injury induced by CCl 4 and D-GalN+LPS as indicated by decreased MDA content and increased activities of plasma SOD and GSH-Px. Conclusion: LMFO is protective against CCl 4-induced and D-GalN+ LPS induced liver injury in mice and its effect may be due to its antioxidative activities in vivo.

Research factors are a very important element in any research design. Research factors include experimental and non-experimental factors. The former is the general term used to describe the similar experimental conditions that researchers are interested in, while the latter are other factors that researchers have little interest in but may influence the result. This article mainly focuses on the following issues: the definition of research factors, the selection and arrangement of experimental factors and non-experimental factors, the interaction between research factors, the standardization of research factors and the common mistakes frequently made by researchers.

Observed index is a very important element in a research design, because it is a specific reflection of the effects of research factors on the research subjects and is indispensable in any research. Generally, there are two types of observed indexes: the indexes that reflect natural attributes, habits or states of the research subjects and the indexes that reflect the effects of different drugs or treatments on research subjects. This article mainly introduces the definition, characteristics, selection and observation of research indexes and the major and minor indexes.

Chinese Journal of Neurology 2008;41(3):164-167
Objective To established a diagnostic methods to idenfity Niemann-Pick disease type C (NPC) in China. Methods Two patients aged 5 and 20 years respectively who presented progressive neurologic regression and splenomegaly were subjected to filipin staining of cultured skin fibroblasts and genetic analyses of NPC1 gene. Results Although there were differences in onset ages and clinical presentations, filipin staining of the cultured skin fibroblasts confirmed the clinical diagnosis of NPC, showing an intense punctate pattern of fluorescence concentrating around the nuclei, consistent with the accumulation of unesterified cholesterol in NPC cells. Genetic sequence analysis further verified the results of filipin staining. The case 1 was compound heterozygous for M1142T(3425T>C),R1186H(3557T>C)and case 2 for Q88H(264G>T),469_470insGT.The latter 2 mutations were novel, and the possibility of polymorphisms was not supposed by analyzing 134 DNA samples obtained form normal controls. Conclusions Filipin stainning of the cultured skin fibroblasts is a reliable method to clinically diagnose NPC with a sensitivity. Genetic diagnose should be performed where genetic analysis is allowed.
