Background and Purpose:TIP30 is a newly found tumor suppressing protein that has low level expression in many kinds of tumor cell lines,and it inhibits the growth of tumor cells of different origin.In this article,we want to probe into the value of a newly found tumor suppressor gene TIP30 as a molecular marker for the prediction of relapse or metastasis of lung cancer patients.Methods:In our present study,immunohistochemical analyses of a retrospective database of pathologic specimens were used to demonstrate the expression of TIP30 protein in NSCLC.Results:The low expression of TIP30 protein in NSCLC tumor tissue was statistically significant in the clinical stage,relapse or metastasis or 5 year disease-free survival rate,whereas low levels of TIP30 expression did not relate to histologic type and histologic differentiation.Conclusions:TIP30 protein may be used as a molecular marker to identify and predict the relapse or metastasis of NSCLC,and may provide a new clue for the clinical doctor to evaluate the prognosis of patients with NSCLC.

OBJECTIVETo study the dynamic changes of capillaries and inflammatory cells in different regions of brain in rat with middle cerebral artery obstruction (MCAO), and the effects of acupuncture in different frequencies on them.

METHODSIn reference to Zea-Longa's method, rat model of MCAO was established by thread-ligation. Shuigou point (DU26), the main acupoint for "awakening brain and opening apertures", was stimulated by high (180 times/s) or low (60 times/s) frequency puncturing 5 s every 12 h for 6 times totally. The amount of capillaries (AC) and inflammatory cells (AIC) in brain cortex (BC), hippocampus (Hp) and corpus striatum (CS) was counted.

RESULTSChanges in AC and AIC of all brain regions (except for CS) in rats immediately after modeling were statistically insignificant (P > 0.05). But 72 h later, AC in CS decreased, AC in Hp, AIC in BC and AIC in Hp increased significantly in the modeled rats, showing significant difference to the normal level, but AIC reduced to approach the normal. As compared with the rats un-intervened, AIC in BC and Hp was decreased in rats intervened with high frequency puncturing, AC and AIC in CS were increased in rats intervened by slow frequency puncturing (both P < 0.05).

CONCLUSIONAmount of capillaries and inflammation cells are changed dynamically in MCAO rats after brain ischemia, showing evident brain regional specificity; the ischemic improving effects of acupuncture in different frequencies are various in their action rings, also showing brain regional specificity.

The study of small drug molecules interacting with nucleic acids is an area of intense research that has particular relevance in our understanding of relative mechanism in chemotherapeutic applications and the association between genetics (including sequence variation) and drug response. In this contribution, we demonstrate how the sequence-specific binding of an anticancer drug Dacarbazine (DTIC) to single base (A-G) mismatch could be sensitively detected by combining electrochemical detection with biosensing surface based on gold nanoparticles.

Objectives To evaluate the influences of age and sex on dose-response and recovery time course of atracurium. Methods Seventy-two patients (38 men and 34 women), ASA grade I, aged 15 ～ 59 years,scheduled for elective surgery were included in the study. All patients were divided into three groups according to their ages: Group 1, 32 patients aged 15 ～ 29 years; Group 2, 21 patients aged 30 ～ 40 years, and Group 3, 19 patients aged 41 ～ 59 years. General anesthesia was maintained with 60% nitrous oxide in oxygen, thiopentone and fentanyl. Neuromuscular function was assessed by train-of-four(TOF) stimulation of accelerometry and the percentage depression of the first response (T1) of the TOF stimulus was used as the study parameter. The dose-response relations of atracurium were determined by the cumulative dose-response technique. Results As compared to older and female patients, dose-response curves of atracurium in young and male patients were shifted in a parallel fashion to the left. The dose-response relation of atracurium correlated closely with patients′ ages and sexes by multiple linear regression analysis.There were significant differences of ED50、ED90 and ED95, and clinical duration and total duration of action following administration of atracurium of same doses between the three age groups as well as men and women. Conclusions Age and gender could significantly affect the pharmacodynamics of atracurium.

AIMTo investigate the absorption and distribution of recombinant hirudin-2 (rHV2) in the GI tract in rats after oral administration.

METHODSUsing HPLC, fluorescence spectrophotometry and confocal laser scanning microscopy to measure the amount of intact rHV2 absorbed into gastrointestinal mucosa and blood.

RESULTSHPLC spectrum showed that there were intact rHV2 molecules in plasma after 1 h of oral administration. After oral administration for 3 h, 1.2%-26.8% of fluorescence was found in the GI tract in rats. Chromatographic analysis showed that 2.27%-38.75% of fluorescence recovered from the GI tract luminal contents and mucosa was eluted at the void volume of a Sephadex G-25 column. Microscopic examination showed that FITC-rHV2 was taken up throughout the whole small intestine but the ileum appeared to be a preferred site for FITC-rHV2 transport in rats.

CONCLUSIONrHV2 may partially survive in the GI lumen and subsequently absorbed in active form by gastrointestinal tract.

OBJECTIVETo investigate the influence of Rehmannia glutinosa oligosaccharides (ROS) on the proliferation of 3T3-L1 adipocytes and insulin resistance.

METHOD3T3-L1 preadipocytes were cultured, the proliferation of 3T3-L1 preadipocytes was detected by MTT method. Insulin resistant 3T3-L1 adipocytes cell model was induced by dexamethasone and the change of glucose concentration in cell culture was determined after ROS treatment.

RESULTIn the high glucose DMEM culture media, MTT method showed that the absorbance at 570nm of 3T3-L1 preadipocytes was increased and that of 3T3-L1 adipocytes was decreased. ROS significantly increased glucose consumption in 3T3-L1 preadipocytes and adipocytes culture in a concentration-dependent manner. ROS improved the sensitivity of 3T3-L1 adipocytes to insulin.

CONCLUSIONROS can promote the proliferation of 3T3-L1 preadipocytes, inhibite the proliferation of 3T3-L1 adipocytes, and also, significantly improve insulin resistance induced by dexamethasone.

OBJECTIVETo observe the clinical efficacy of acupuncture at Hegu (LI 4) on central facial nerve paralysis after ischemic stroke, and explore dose-effect relationship among different stimulation intensities of acupuncture at Hegu (LI 4) as well as its optimal treatment plan.

METHODSAccording to different acupuncture stimulation intensities which were based on treatment time and needle insertion direction, fifty patients were randomly divided into a Hegu 1 group, a Hegu 2 group, a Hegu 3 group, a Hegu 4 group and a control group, ten cases in each one. Different stimulation intensities of acupuncture at Hegu (LI 4) combined with facial paralysis acupoints, including Yingxiang (LI 20), Dicang (ST 4), Jiache (ST 6) and Quanliao (SI 18), were applied in Hegu 1 to 4 groups; meanwhile acupuncture at stroke acupoints, including Neiguan (PC 6), Shuigou (GV 26) and Sanyinjiao (SP 6), and medication treatment were adopted. Except acupuncture at Hegu (LI 4), the treatment of the control group was identical as Hegu groups. The treatment duration lasted for 14 days. The House-Brackmann facial never grading systems (H-B), Toronto facial grading system (TFGS), degrees of facial never paralysis (DFNP), facial disability index (FDI) and clinical efficacy were compared among groups.

RESULTS(1) Compared before the treatment, H-B, TFGS, DFNP and physical function score in FDI were all improved significantly in the Hegu 1 to 4 groups (all P < 0.05), but social function score in FDI was not obviously improved (all P > 0.05); all the scores in the control group were not evidently changed (all P > 0.05). (2) Compared with the control group, differences of H-B before and after treatment in the Hegu 1 to 4 groups, differences of TFGS in the Hegu 2 group and differences of DFNP in the Hegu 1 and Hegu 2 group were significantly improved (all P < 0.05). The differences of any scale among Hegu 1 to 4 groups were not significant (all P > 0.05), in which the most evident change was found in Hegu 2 group. (3) The total effective rate was 90.0% (9/10), 100.0% (10/10), 90.0% (9/10) and 80.0% (8/10) in Hegu 1 to 4 groups, which were significantly higher than 60.0% (6/10) in the control group (all P < 0.05).

CONCLUSIONAcupuncture at Hegu (LI 4) has affirmative clinical efficacy on central facial nerve paralysis after ischemic stroke, in which oblique insertion along the opposite direction of meridian for 5 s of twirling manipulation has the best clinical effect.

AIMTo investigate the permeation mechanism of paclitaxel by enhancers and preparation factors.

METHODSThe fluidity of mucous membrane and membrane protein conformation changes were determined by using electron spin resonance (ESR) when mucous membrane was treated by several enhancers. At the same time, the factors of penetration of lower dissolution drug across the intestinal mucous membrane were studied in three formulas inclusion complex, microemulsion and injection.

RESULTSPolyethylene glycol (PEG) 1500, hydroxypropyl-beta-cyclodextrin (HP-beta-CD) and phospholipid as enhancers could reinforce the permeation of paclitaxle because of loosening of protein conformation in intestinal mucous membrane. Paclitaxel-HP-beta-CD inclusion complex and paclitaxel microemulsion as vehicle could significantly increased permeation kinetic rate of paclitaxel with fluid diffuse method.

CONCLUSIONCharacteristics of enhancing intestinal absorption of poor dissolution drug had been provided with enhancer the change of membrane fluid.

AIMTo perpare and identify irisquinone hydroxypropyl-beta-cyclodextrin inclusion complex (irisquinone-HP-beta-CD), as well as to study the inclusion mechanism and molecule stoichiometry between irisquinone and hydroxypropyl-beta-cyclodextrin.

METHODSIrisquinone-HP-beta-CD was prepared by freeze-drying technique. The ratio of host and guest was also studied in inclusion process by mol gradient and continuing variational methods. At the same time, the inclusion complex was identified by X-ray diffraction (XRD) and differential scanning calorimetry (DSC).

RESULTSIt was demonstrated that the solubility of irisquinone was enhanced markedly by inclusion with HP-beta-CD when stoichiometry was 2:1 of host and guest at 25 degrees C, 35 degrees C and 45 degrees C.

CONCLUSIONThe solubility and stability of irisquinone could be increased by preparing the inclusion complex with hydroxypropyl-beta-cyclodextrin.

AIMTo prepare the target drug delivery systems(TDDS), albendazole polybutycyanocrylate nanoparticles (ABZ-PBCA-NP), its pharmaceutical characters and tissue distributions were simultaneously investigated.

METHODSAlbendazole nanoparticles were prepared with the emulsification-polymerization method and the drug-load mechanism of polybutycyanocrylate nanoparticles was studied with the equal-tempaerature adsorption principle. The dialyse dynamic of albendazole from ABZ-PBCA-NP was investigated in four formulations in vitro. The tissue distribution of albendazole in different drug vehicles was studied with isotope labelling experiment.

RESULTSABZ-PBCA-NP and ABZ-PVP-PBCA-NP fit to the Higuchi and bi-exponent function in vitro respectively. The drug loaded in nanoparticles was abide by the Langmuir adsorption equation. Targeting index of albendazole in liver and spleen in mice are 11.4 and 3.9 after ig 3H-ABZ-PBCA-NP. The bioavailability of albendazole nanoparticle and suspension are 76.0% and 36.9% respectively.

CONCLUSIONThe absorptive capability of drug was enhance when 4% PVP was added into the nanoparticle, and its release time was lengthen. At the same time, the nanoparticles vehicles increase the albendazole bioavailability.
