Background Leber hereditary optic neuropathy (LHON) is a disease characterized by maternal inheritance.A number of mitochondrial DNA (mtDNA) mutation has been thought to be associated with this disease.Objective This study was to investigate the clinical and molecular genetic properties of LHON in two Chinese families.Methods Forty subjects from two Chinese families with LHON were enrolled in Affiliated First Hospital of Zhengzhou University,including 28 maternal members (10 of these members are LHON and 12 controls from two families.All the participants had a complete ophthalmic examination including visual acuity,direct ophthalmoscopy,color sensation and visual evoked potentials.MtDNA was extract from the whole blood sample of all participants.PCR-DNA sequencing was performed to detect the point mutation of the G11778A,T14484C,and G3462A for each subject.Written informed consent was obtained from each subject prior to this study.Results Only G11778A point mutation was identified in all 28 maternal members from the two families.No point mutation of G11778A was identified in non-maternal members,and no point mutation of the T14484C and G3462A were found in the two families.Conclusions The inherited pattern of these two families shows typical clinical and genetic features of LHON.LHON patients with G1 1778A mutation have a poor prognosis of visual acuity.

Objective To reproduce an experimental model of microsurgical reversible obstruction jaundice by micro-surgical technique. Methods Eight Banna miniature pigs were equally and randomly assigned into model group and control group. An improved Foley catheter was put into gall bladder of the pigs in the control group. A biliary calculus removal balloon tube was inserted into the common bile duct through the cystic duct in the experimental group to establish either partial or total biliary obstruction. The obstruction could be relieved by withdrawing the balloon tube from the common bile duct. Biliary tract visualization was conducted to evaluate the biliary obstruction, and venous blood samples were collected to determine the liver function (such as serum levels of TBIL and ALT etc.) in all pigs on the 2nd, 4th and 6th day after operation. Results Cholangiography showed a progressive dilation of both common hepatic duct and intrahepatic bile duct in the experimental group when the partial biliary obstruction was induced. During the periods when the total biliary obstruction was created the common bile duct, the common hepatic duct and intrahepatic bile duct of the animals in the experimental group were dilated progressively too. Both intra- and extra- hepatic bile ducts changed significantly compared to the control group. The dilatation subsided following the relief of obstruction. There was no dilatation of the bile duct in the control group. The serum levels of TBIL and ALT were significantly higher in the experimental group than that in the control group, and they were lowered after the relief of obstruction. Conclusion A microsurgical reversible obstruction jaundice model has been successfully reproduced with a biliary calculus removal balloon tube. It may provide a reliable experimental method for researches on the pothogenesis and treatment of obstruction jaundice.

Objective To understand the job burnout conditions of nurses with different education levels in Ningyang county,analyze the influence of education level on job burnout,in order to find strategies to relieve the job burnout,reduce burnout,increase working enthusiasm,then further lower nurse turnover rate and stabilize of the nurse team.Methods A total of 150 nurses in the first hospital of Ningyang county participated in the investigation using Maslach Burnout Inventory (MBI) in December 2012.The survey data underwent statistical analysis.Results The results show that in the dimension of emotional exhaustion,education level showed main effect.While in the dimension of depersonalization and personal accomplishment,the education level showed no main effect.Conclusions Education levels have a significant impact on job burnout of nurses,mainly in the emotional exhaustion dimension.Hospital managers should pay attention to job burnout of nurses with college degree,reduce their occupational burnout through positive changes in personal and organizational factors.

Objective:To prepare celecoxib nanosuspension ( CXB-NSs) and study the pharmacokinetics of CXB-NSs in rats. Methods:CXB-NSs were prepared by an anti-solvent precipitation and high pressure homogenization method. The particle size, polydispersion index ( PdI) and zeta potential of the nanosuspension were studied. Totally 12 Wistar rats were randomly divided into CXB-NSs group and CXB suspension group, and gastric drug dose was 100 mg·kg-1 . CXB concentration in plasma was determined by HPLC and the pharmacokinetic parameters were calculated by 3P97 software. Results: The particle size, polydispersion index, zeta potential of CXB-NSs was (442. 5 ± 61. 9) nm, 0. 312 ± 0. 057 and ( -31. 6 ± 3. 9) mV, respectively. Objective To observe the effect of application of Lamaze childbirth method in childbirth. Methods The observation group(110 cases)was trained by Lamaze childbirth method in late pregnancy period. The control group (110 cases) was not trained by Lamase childbirth method in antepartum period, and were given regular observation and nursing. Mastering of natural childbirth knowledge, pain intensity, childbirth time, mode of delivery and amount of bleeding post- childbirth 2h were observed in the two groups. Results The observation group mastered more natural birth knowledge than the control group, childbirth pain was obviously reduced, natural birth rate was higher than the control group, the first and second labor time was shorter than the control group, the amount of bleeding was obviously less than the control group 2 hours post childbirth. Conclusions Pregnant women need Lamaze childbirth method training before childbirth and instruction of maternity nurses. These may relieve pain, shorten childbirth time and reduce amount of bleeding post childbirth by application of Lamaze childbirth method.

Objective To investigate the relationship between the quantified electroencephalograph (EEG) and end-tidal desflurane concentration and characteristics of desflurane absorption during low-flow desflurane anesthesia. Methods Fifteen ASA I - Ⅱ patients with a mean age of (51.6?11.9)yr, weighing (59.6?7.9)kg scheduled for operation performed under combined general and epidural anesthesia were studied. The patients were premedicated with phenobarbital 0.1g and atropine 0.5mg.Epidural catheter was placed and a test dose of 1.0% lidocaine and 0.2% tetracaine was given before induction of general anesthesia. General anesthesia was induced with fentanyl 2-3?g?kg-1, thiopental 4-7mg?kg-1 and succinyl choline 1.5-2.0 mg?kg-1 and maintained with desflurane, intermittent iv boluses of vecuronium and epidural anesthesia. Desflurane anesthesia was started with high-flow for 15min followed by low flow. During high-flow wash-in, FGF was 4L/min, VT was set at 8-10ml?kg-1, PETCO2 was maintained at 4.2-4.6kPa(32-35mm Hg) and desflurane vaporizer was set at 5% .During low-flow FGF 0.5L/min. ObjectiveTo analyze the internet-based cognitive behavior therapy (ICBT) intervent medical university students' social anxiety,and provide experiences and theoretical basis for future social anxiety interventions.Methods45 medical students of social anxiety were voluntarily divided into the one-to-one intervention group,one-to-five intervention group and control group.The control group did not intervene,one-to-one intervention group and one-to-five intervention group were interfered single person or team by cognitive behavioral therapy,respectively.The level and symptom of social anxiety of the 3 groups were assessed before and after the intervention by social phobia inventory(SPIN) and Social anxiety symptoms interview inventory.ResultsAfter the intervention,the scores of SPIN and the 3 subscales within it (fear,avoidance and physiological) of one-to-one ICBT group and one-to-five ICBT group were lower than those of the control group (SPIN scores:one-to-one ICBT group 15.60 ±7.87,one-to-five ICBT group 14.87 ± 8.83,control group 20.20 ± 11.46).Variance analysis showed that,in addition to physiological subscale(F =0.641,P ＞ 0.05 ),the scores of SPIN,fear and avoidance subscales had significant differences among the3 groups (F=3.764,P=0.018; F=3.606,P=0.021 ; F=5.444,P=0.005).In the analysis of participants'social anxiety symptoms,after treatment,in one-to-one ICBT group,there was 1 participant changed into subthreshold social anxiety and 1 participant into normal in the 2 threshold social anxiety participants.All the 3 subthreshold social anxiety participants changed into normal.In one-to-five ICBT group,there was 1 threshold social anxiety participant lapsed to subthreshold social anxiety,1 subthreshold social anxiety participants still remained in subthreshold social anxiety,2 symptomatic social anxiety participants changed into normal.But there were no significant changes in control group.ConclusionsAll the two methods of intervention have a certain effect.The effect of one-to-one intervention therapy is most significant.
