1.The research progress on stereotactic body radiation therapy for stageⅠnon-small cell lung cancer

Image guided stereotactic body radiation therapy ( SBRT) has emerged as a promising technolo-gy for early-stage non-small cell lung cancer ( NSCLC) increasingly ,particularly for patients that are unable to tolerate or deny operation ,with a trend to replace surgical resection .Abundant clinical research results have dem-onstrated that SBRT is safe to yield local control rate of more than 90%without excessive toxicity in early -stage NSCLC.In this review ,we discuss the research progress on treating stage I NSCLC with SBRT .
2.Universal Newborn Hearing Screening in Beijing in 2010 and Its Related Factors

Objective To describe the results of the Universal Newborn Hearing Screening (UNHS) in Beijing in 2010; and investigatethe factors related with the results. Methods The result of the UNHS investigated by 106 maternity hospitals were reported. These hospitalswere investigated with the self-made questionnaire. Results In 2010, 159888 infants (91.99%) were screened in 106 maternity hospitals inBeijing as the 1st stage UNHS. 10.32% of them failed OAE tests. 72.30% received the 2nd stage UNHS and 13.27% of them failed testsagain. The referral rate was 0.99% in the end. The screening rate of both the 1st and 2nd stage was more in the newborn with Beijing householdregistration than in those with other household registration, as well as the hospitals in urban than in rural areas. For the rate fail in the1st stage UNHS, it was more in hospitals in urban than in rural areas, with distortion product otoacoustic emission (DPOAE) than with transientevoked otoacoustic emissions (TEOAE), by the bed than in the special room, and screening only once than screening again. ConclusionThe quality of UNHS is as high as in the developed countries in the newborn with Beijing household registration. To further improvethe screening rate, it is important to focus on the newborn with other household registers and the hospitals in rural areas. The instrument, personnelqualifications, environment and operation need to be further standardized.

4.An in vitro analytical method based on bio-thermal activity for the determination of dissolution rate of Chinese medicine solid preparation.

To explore the new pattern of Chinese medicine solid preparations (CMSP) in vitro dissolution, a method testing the bio-thermal activity in combination with UPLC was used. Microcalorimetry was used to obtain the characteristic metabolic growth power-time curves and a series of biothermodynamic parameters of the inhibition of Staphylococcus aureus by Yinhuang tablet dissolving solutions at the pH 6.8 (phosphate buffer) dissolution medium at different times. From these results, the cumulative dissolution of Yinhuang tablet based on bio-thermal activity was obtained. The dissolution rates of two components of chlorogenic acid and baicalin were determined by UPLC method. Then f2 similar factor method was used to evaluate the relevance of these two methods. The result showed that f2 values all were more than 50, indicating that there is a good correlation between the two methods of measuring the dissolution rate. It is feasible to determine CMSP in vitro dissolution by using bio-thermal activity, and to provide new evaluation methods for controlling the quality of CMSP.

7.Mutation analysis of the STAT3 gene in a patient with hyper-IgE syndrome

Objective To analyze mutations of the STAT3 gene in a patient with hyper-IgE syndrome (HIES).Methods Clinical data were collected and blood samples were obtained from a 14-year-old patient with hyper-IgE syndrome (HIES) and her parents.Genomic DNA was extracted and subjected to PCR for the amplification of the entire encoding and splice sites of the STAT3 gene followed by bidirectional sequencing.Meanwhile,amplified ribosomal DNA restriction analysis was carried out.Results A heterozygous missense mutation A1843G,which caused a K615E substitution,was found in exon 19 encoding the SH2 domain of the STAT3 gene in the patient,but not in either of her parents.The result of amplified ribosomal DNA restriction analysis was consistent with the findings mentioned above.Conclusion The novel K615E missense mutation in the STAT3 gene may contribute to the development of HIES.
8.Long-term effect of intensive glucose control on diabetic retinopathy in type 2 diabetes patients

Objective To observe the effect of long-term intensive glucose control therapy on diabetic retinopathy in outpatients with type 2 diabetes mellitus.Methods Forty-nine patients with type 2 diabetes mellitus were randomly assigned to participate in the trial from 2002 to 2007,receiving either intensive (24 cases) or standard glucose control (25 cases).The patients were examined by the same ophthalmologists to identify any new diabetic retinopathy (DR).After 5 years of intensive glucose control,all of the patients were asked to attend our clinic every 6 months,but no attempts were made to maintain their previously assigned therapies.Hemoglobin A1c (HbA1 c) was measured regularly.In November of 2009,a retinal examination was carried out by the same ophthalmologist who worked in the trial.The visual acuity,lens,vitreous body and fundus were examined after pupil dilation to identify diabetic retinopathy (DR).Fundus fluorescein angiography and retinal laser photocoagulation were carried out when necessary.Results In the second year after enrollment in the trail,the median HbA1c level of the intensive-therapy group was significantly lower than that of the standard-therapy group [(6.3 ± 0.6) % vs.(7.2 ± 1.2) %,t =2.09,P < 0.05],and was maintained in a controlled level throughout the following 4 years.During the post-trial monitoring,no new case of of macula edema or diabetic associated blindness occurred in either intensive or standard-therapy group,the whole occurrence of micro-aneurysms,fundus hemorrhage,as well as those who needed retinal laser photocoagnlation and lowering in visual acuity in intensive-therapy group was lower than that in the standard-therapy group (3 vs.14,1 vs.7,2 vs.4,3 vs.11,respectively ;9 vs.36,totally,x2 =4.719,P < 0.05).During the first post-trial monitoring,there was no difference in median HbAlc level between intensive-therapy group and standard-therapy group [(7.2 ± 1.1) % vs.(7.3 ± 1.3) %,t =0.25,P =0.806],which was sustained in the following years.In the trail,no new case of fundus hemorrhage or diabetic associated blindness occurred in intensive-therapy group during the five-year period of therapy.Number of new episodes of micro-aneurysm,macula edema were less in intensive-therapy group than that in standard-therapy group,number of new episodes of lowering in visual acuity,and those who needed retinal laser photocoagnlation,were significantly less in intensive-therapy group than that in standardtherapy group(15 vs.25,4 vs.23,Z =-4.459,P < 0.05) during five-year follow-up.Conclusions The benefit of reduced incidence of diabetic retinopathy in intensive glucose can be maintained because of the legacy effect.

10.In vitro stability of Endostar

Aim: To compare the stability of Endostar~(TM) and endostatin under different temperatures and pH using polyacrylamide gel electrophoresis( PAGE) and Western blot and to compare the activity of Endostar kept at 4 ℃ and 37 ℃ by inhibition of endothelial cell proliferation. Methods: Endostar and endostatin expressed by Phicha pastoris were kept at 4 ℃, and 37 ℃ for 96 hours, respectively. The electrophoresis of the samples was detected by reduced and non-reduced PAGE. The results were further confirmed by Western blot with rabbit anti-Endostar polyclonal antibody. The inhibitory effect of Endostar stored at different temperatures on HUVEC was also exam-ined by cell-based assay. Results: Single band at 20 kD was detected in all lanes of SDS-PAGE gel loaded with endostatin and Endostar samples under reducing condition. In acidic native PAGE with three different pH values, endostatin showed a smear characteristic, whereas Endostar showed a unique band in acidic non-continuous native PAGE. Although the smear phenomenon was also observed under two conditions of constant native elec-trophoresis, the major band of Endostar could be detected. Similar electrophoretic behavior was found for endosta-tin and Endostar stored at both 37 ℃ and 4 ℃ . Western blot showed similar results to those by PAGE. Furthermore, Endostar stored at these two temperatures also had identical inhibitory effect on proliferation of HUVEC. Conclusion: Endostar and endostatin exhibit similar thermostability at regular conditions, but Endostar was more stable than endostatin expressed in P. pastoris under acidic condition.
