1. Spent culture supernatant of Lactobacillus acidophilus influences contraction of isolated intestinal smooth muscle of rabbits

Objective: To observe the influence of spent culture supernatant (SCS) of Lactobacillus acidophilus strain LA14 on the contraction of isolated intestinal smooth muscle and to discuss the related mechanism. Methods: The ileum samples of rabbits were prepared and the contraction frequency and amplitude of intestinal smooth muscle were observed as the normal control. Then the SCS, bacterium suspension, and SCS with bacterium suspension were added by an accumulative dose to the culture media (0.3 ml per times, at an interval of 6 min), respectively. Four minutes after each administration, the contractive curves were recorded for 2 min. The influences of various groups of Lactobacillus acidophilus on the contraction of isolated intestinal smooth muscle were observed. The effect of SCS on M cholinoceptor was observed by adding in order pilocarpine, atropine or SCS, and pilocarpine. Results: After continuous administration of SCS or SCS with bacterium suspension (0.6-1.5 ml), the contraction frequency of the intestinal smooth muscle was significantly lowered compared with before administration (P<0.05 or P<0.01), and there were no significant differences between before administration and other volume groups (P>0.05). Within the range of 0.3-1.5 ml, the SCS, bacterium suspension, and SCS with bacterium suspension resulted in no significant difference in reducing the contraction amplitude, except for SCS with bacterium suspension at 1.5 ml(P<0.05). SCS or atropine significantly inhibited pilocarpine-induced increase of contraction amplitude(P<0.05 or P<0.01). SCS also reduced the contraction frequency of the intestinal smooth muscle(P<0.01). Conclusion: SCS of Lactobacillus acidophilus may inhibit the peristalsis of the intestinal smooth muscle of rabbits by blocking M cholinoceptor.
3.Effect of Guilingji Capsule on the fertility, liver functions, and serum LDH of male SD rats exposed by 900 mhz cell phone.

OBJECTIVETo observe the effect of Guilingji Capsule (GC) on the fertility, liver functions, and serum lactate dehydrogenase (LDH) of adult male SD rats exposed by 900 MHz cell phone.
METHODSTotally 18 adult male SD rats and 36 adult female rats in child-bearing period were selected and randomly divided into three groups according to weight equilibrium principle, i.e., the normal group, the radiated group, and the GC group, 6 males and 12 females in each group. Male rats in the normal group and all female rats were not radiated. Male rats in the radiated group and the GC group received radiation for 4 h per day, lasting for 18 successive days. Rats in the GC group received GC suspension at the daily dose of 0. 15 g/kg by gastrogavage at the same time. Equal volume of normal saline was administrated to other male rats. Then male rats were mated with corresponding female rats from the 14th radiation night to the 18th radiation night in the ratio of 1:2. Male rats were killed following on the next morning of ending the radiation. Female rats were normally fed and then killed before delivery. The pregnant outcomes of female rats in responding groups (the rates of pregnancy and the number of death fetus, birth weight, body length, and tail length) were observed and compared. Serum alanine aminotransferase (ALT), aspartate transferase (AST), AST/ALT, and LDH levels of the male rats were detected by colorimetry. Histological and morphological changes of liver were observed by HE staining.
RESULTSCompared with the normal group, the pregnancy rates of female rats decreased and the number of death fetus increased, the serum LDH level obviously increased in the radiated group (P < 0.05). Serum levels of ALT, AST, and AST/ALT were no significantly changed in the radiated group. The hepatocyte nuclear atrophy and cytoplasm vacuolar degeneration appeared. Compared with the radiated group, the pregnancy rates increased, the number of death fetus dropped, and the serum level of LDH decreased in the GC group (P < 0.05). There was no obvious change in serum levels of ALT, AST, or AST/ALT. The hepatocyte nuclear atrophy and cytoplasm vacuolar degeneration were significantly attenuated. The histomorphological structures recovered to normal basically in the GC group.
CONCLUSIONSThe pregnancy rates could be decreased, the number of death fetus increased, histomorphological structures abnormal, and serum LDH level increased by exposure toy GSM 900 MHz cell phone. GC could prevent and treat the aforesaid lesion. But there was no statistical difference in serum ALT or AST levels.

5.Study on davallia mariesil flavones improving condition of osteoporosis via Notch signaling pathway

Objective:To evaluate the effect and mechanism of davallia mariesil flavones (DMF) improving osteoporosis via Notch signaling pathway.Methods:(1) 120 cases patients with osteoporosis in our hospital from January 2016 to January 2017 were analyzed,and divided into experiment group (60 cases) and control group (60 cases) using digital random grouping method.Experiment group was treated with DMF combined with D-calfor.Control group was treated with D-calfor only.After treatment, the levels of serum calcium,serum phosphor,TNF-α,MCP-1 and IL-6 in serum were detected to evaluate the clinical efficacy.(2) Bone marrow stromal cells were separated and cultivate.NC group:DMEM(contain 10% FBS).RA group:RA(0.4 mmol/L).DMF+RA group:DMEM(contain DMF)+RA(0.4 mmol/L).Jaggedl+RA group:Jaggedl(1 000 μg/L)+RA(0.4 mmol/L).Jaggedl+DMF+RA group:Jaggedl(1 000 μg/L)+DMEM(contain DMF)+RA(0.4 mmol/L).DAPT+RA group:DAPT(16 μmol/L)+RA(0.4 mmol/L). DAPT+DMF+RA group:DAPT(16 μmol/L)+DMEM(contain DMF)+RA(0.4 mmol/L).Western blotting assays and PCR were performed to assess mRNA and protein levels of Notch-1,Hes-1.Results: (1) In clinical study,the effective rate in treatment group was obviously higher than control group (91.67%>76.67%,P<0.05).The levels of serum calcium and serum phosphor in the experiment group was higher than in the control group (P<0.05).The levels of TNF-α,MCP-1 and IL-6 in the experiment group was lower than in the control group (P<0.05).(2) In experimental study,compared with the RA group,the expressions of Notch-1,Hes-1 mRNA and protein were upregulated in Jaggedl+RA group,but were downregulated in DAPT+RA group,DMF+RA group (P<0.05). Compared with the Jaggedl+RA group,the expressions of Notch-1,Hes-1 mRNA and protein were downregulated in Jaggedl+DMF+RA group (P<0.05).Compared with the DAPT+RA group,the expressions of Notch-1,Hes-1 mRNA and protein were downregulated in DAPT+DMF+RA group (P<0.05).Conclusion:DMF could improve the condition of osteoporosis.The mechanism may be associated with inhibiting the Notch signaling pathway by DMF.
6.A Meta-analysis of the Risk of Secondary Infection of Tocilizumab in the Treatment of COVID-19

Objective Meta-analysis was conducted to assess the risk of secondary infection caused by tocilizumab(TCZ)in the treatment of Corona Virus Disease 2019(COVID-19),in order to provide an evidence-based basis for the safety of tocilizumab in patients with COVID-19.Methods Cochrane Library,PubMed,Web of Science,CNKI,SinoMed and Wanfang databases were searched in computer to collect randomized controlled trial and cohort study of treating COVID-19 with tocilizumab from December 19,2019 to December 30,2022.A meta-analysis of the results of each study was performed using RevMan 5.4.1 software.Results A total of 1691 references were screened and eighteen studies involving 3933 patients were included.The incidence of secondary infection in the tocilizumab with the standard treatment group and standard treatment group was 19.14%(331/1729)and 12.11%(267/2204),respectively.Meta-analysis showed that the tocilizumab + standard treatment group had a higher incidence of secondary infection than the standard treatment group[RR = 1.35,95%CI(1.05,1.74),P = 0.02].The results of the subgroup analysis showed that the risk of secondary infection with different doses of tocilizumab was different.The incidence of secondary infection was significantly higher in the subgroup with doses of 400~800 mg/d tocilizumab than in the standard care group[RR = 1.48,95%CI(1.19,1.84),P = 0.0004].The incidence of secondary infection in subgroups with doses of≤400 mg/d tocilizumab was also significantly higher than that in the standard treatment group[RR = 1.87,95%CI(1.28,2.72),P = 0.001].However,there was no statistical significance between the subgroup 6~8 mg/kg tocilizumab and the standard treatment group.Conclusions Tocilizumab may increase the risk of secondary infection in patients with COVID-19 compared with standard treatment,and the benefits and risks of tocilizumab should be carefully evaluated before clinical administration.Moreover,large and high-quality studies are needed for further evaluation.
7.Expression of phosphoprotein P2 of Cysticercus cellulosae in Pichia pastoris and its application.

Cysticercosis is caused by the metacestode form of Taenia solium-Cysticercus cellulosae and it causes great economic losses and threatens the people's health. There are some problems on how to control cysticercosis, in order to resolve the key problem that the native antigen to diagnose and prevent cysticercosis is very limited and is not satisfied, Pichia pastoris Expression System was used to express recombinant P2 protein. The interested P2 gene was got by digesting the pGEM - P2 vector using restriction endonuclease, then it was inserted into the secretory pPIC9K Pichia pastoris expression vector and transformed into E. coli. Positive recombinant plasmids were selected sequenced and named pPIC9K-P2 and it was linearized by Sal I and Bgl II, then the linear DNA transfored into Pichia pastoris GS115 by electroporation. The recombinant expression vector pPIC9K - P2 integrated into GS115 via homologous recombination between the transforming DNA and regions of homology within the genome. The transformants were screened for multicopy recombinants using G418 and were distinguished for Mut phenotypes by MD and MM. Two different phenotypes were generated-HIS+ MUT+ (Methanol utilization plus) and HIS+ MUT(S) (Methanol utilization slow). PCR analysis of the multicopy recombinants indicated that the P2 gene was integrated within the genome of pichia Pastoris. The multicopy recombinants were named GS115/pPIC9K - P2HIS+ MUT+ and GS115/pPIC9K-P2HIS+ MUT(S), both HIS+ MUT+ and HIS+ MUT(S) were induced with methanol. The results of SDS-PAGE and Western blot demonstrated that the culture supernatant of the induced Pichia pastoris contained P2 protein which was accumulated up to 33 % of total proteins in the culture supernant and its molecular weight is 12.6kD. The results of the clinical study indicated that the expression P2 protein could be used to diagnose human cysticercosis and swine cysticercosis as diagnosis antigen.

8.An etiological study on acute respiratory infection among inpatient children in Suzhou.

OBJECTIVETo investigate the pathogen of children with acute respiratory infection (ARI) in Suzhou and to provide some evidences for clinical diagnosis and treatment.
METHODSThe nasopharyngeal secretion samples from 2492 inpatient children with ARI, during the period of November 2005 to May 2007, were investigated for respiratory syncycial virus (RSV), influenza virus A and B, parainfluenza virus type 1, 2, 3 and adenovirus by both the indirect immunofluorescence assay and virus isolation. Human metapneumovirus (hMPV) were examined by reverse-transcription polymerase chain reaction (RT-PCR) at the same time.
RESULTSOf 2492 samples tested, 961 (38.6%) were positive. The total positive rate of virus pathogens in children with ARI was found related to age, season and respiratory disease. The detection rates by age were: 50.0% (412/824), 43.4% (190/438), 30.5% (207/679)and 27.6% (152/551), chi(2) = 96.5002, P < 0.01; the detection rates by season were : 46.7% (366/784), 13.8% (66/478), 13.8% (59/428) and 58.6% (470/802), chi(2) = 392.3279, P < 0.01; the detection rates by disease were (acute upper respiratory infection, acute laryngitis, throat-trachea-bronchitis, bronchial pneumonia, pneumonia genuine, bronchiolitis, bronchial asthma): 21.4% (30/140), 73.7% (14/19), 32.0% (8/25), 36.9% (598/1620), 13.1% (8/61), 66.1% (216/327) and 29.0% (87/300), chi(2) = 162.1276, P < 0.01. There was no association between the total positive rate and sex. The detection rates by sex were: 39.0% (588/1508) for male and 37.9% (373/984) for female, chi(2) = 0.2962, P > 0.05. The peak of RSV appeared from December to March. There was the highest RSV detection rate 50.2% (164/327) with bronchiolitis. The hMPV can be detected all year around. The peak of hMPV appeared in winter and the detection rate was 13.2% (106/802).
CONCLUSIONRSV and hMPV are the main respiratory viral pathogens in Suzhou. Detection of viral pathogens in children with respiratory infection could give fast, accurate diagnostic evidence, and help avoid antibiotics abuse.

9.The inhibitory action of the antisense oligodeoxynucleotide to the expression of vascular endothelial growth factor by radiotherapy in a prostate cancer cell line.

OBJECTIVESTo investigate mechanism for the increasing level of serum vascular endothelial growth factor(VEGF) in tumour patients during radiotherapy and the inhibitory action of the antisense oligodeoxynucleotide (AS-ODN) to the expression of VEGF protein by radiotherapy in the prostate cancer cell line (PC3M).
METHODSTo observe the changes of serum VEGF in the prostate cancer patients during radiotherapy dynamically and the inhibitory action of the antisense oligodeoxynucleotide to the expression of VEGF by radiotherapy in PC3M.
RESULTSThe changes of serum VEGF in three patients receiving radiotherapy had been observed continuously. The levels of serum VEGF began to increase when the patients received radiotherapy and rised up to peak value after fifteen days, then declined to the range of pre-radiotherapy. Irradiating the PC3M cells with X-rays significantly increased the VEGF expression and secretion. The expression of VEGF protein in the group treated by VEGF AS-ODNs and X-ray irradiation decreased significantly than the group treated only by X-ray irradiation.
CONCLUSIONSThe induction of VEGF protein expression by X-ray irradiation in tumor cells may result in the increasing of the VEGF in the prostate cancer patients during radiotherapy and the induction can be blocked by VEGF AS-ODNs.

Objective:To study the international situation of technology innovation and transfer, to improve the performance in the health field of China.Methods:Collect data and status quo of innovation and transformation of scientific and technological achievements in 10 World-class research universities and hospitals through literature review, and conduct summary analysis.Results:According to the study, the health field is the main arena of technology innovation and transfer in which the status of the main body of innovation of hospitals cannot be ignored, and the transformation ability of some hospitals is higher than universities.Technology innovation and transfer in the health field is characterized by multiple disciplines, long-term and high cost, and it is necessary to build a full ecological chain from research and development (R&D) to production and commercialization.Conclusions:The dominant position of the health field and hospitals in technology innovation and transfer needs to be emphasized.Interdisciplinary collaboration and innovative economy building should be strengthened.
