Chinese Traditional and Herbal Drugs 2018;49(20):4743-4747
Objective To study the effect of different cutting time and drying methods on the index components of pharmacopeia in different fresh-cutting Aurantii Fructus, and investigate the difference of component and efficacy between the preferred fresh-cutting Aurantii Fructus and the traditional slices. Methods The HPLC system consisted of a Diamonsil C18 (250 mm × 4.6 mm, 5 μm) with acetonitrile/phosphoric acid water (20∶80, pH 3.0) as the mobile phase, the flow rate was 1.0 mL/min, column temperature was 30 ℃, detection wavelength was 283 nm, and the injection volume was 20 μL. The content of naringin and neohesperidin in fresh-cutting Aurantii Fructus with different processing methods was determined to optimize the cutting time and drying method. At the same time, the effects of sclices from fresh-cutting and traditional process on the small intestine propulsion rate, gastric emptying rate, gastrin, and hemorheology were determined. Results The content of naringin and neohesperidin of the fresh-cutting Aurantii Fructus that dried in sun was higher than that dried at 60 ℃ in oven at the corresponding days from the same origin, and the highest content was from fresh-cutting slices dried in sun for 3 d. There was no significant difference in the indexes of small intestine propulsion rate, gastric emptying rate, and gastrin between the preferred fresh-cuting Aurantii Fructus and the traditional ones, but the indexes of hemorheology showed significant difference (P < 0.05). Conclusion The preferred fresh-cutting Aurantii Fructus (processed by half cutting horizontally, drying for 3 d, and then cutting into thin slices and drying in the sun) showes no significant difference in gastrointestinal motility indexes when compared with the traditional ones.

Abstract: The loop-mediated isothermal amplification (LAMP) technique is a technique for the specific and efficient amplification of target fragments at a constant temperature using two pairs of specially designed primers and a strand displacement activity DNA polymerase. LAMP technique is a simple, rapid, specific, sensitive and cost-effective nucleic acid amplification method, and therefore has a promising future in the field rapid detection of Mycobacterium tuberculosis and grassroots applications. In this review, the basic principles and characteristics of the LAMP technique, the main molecular markers for the diagnosis of tuberculosis, and the use of different molecular markers and various types of novel techniques in the diagnosis of pulmonary tuberculosis, extrapulmonary tuberculosis, and drug-resistant tuberculosis were described. The LAMP technique has been widely used in the diagnosis of tuberculosis with high sensitivity and specificity, but the technique still has some shortcomings. This paper reviews the progress of its application in tuberculosis in recent years and provides an outlook on its development, with a view to providing a rational research direction for rapid diagnosis of tuberculosis in a resource-limited environment.

To identify whether a highly repeated GT sequence from DCA1 promoter from Dunaliella salina, which have been proved to be a salt-inducible promoter in our previous study, would be a salt-inducible regulation element, different primers were designed to amplify 6 different-length fragments of DCA1 promoter from D.salina by PCR.After these fragments were respectively inserted into the HindⅢ-BamH I sites of the vector pU?GUS,serial expression vectors containing the gus gene were generated.D.salina cells transformed with these recombinant plasmids by electroporation were grown in liquid media containing different concentrations of sodium chloride respectively.GUS enzyme activity was measured histochemically and fluorometrically. The results revealed that 3 fragments containing GT repeated sequence drove the external gus gene expression and the expression pattern of the gus gene was regulated by the concentrations of sodium chloride.Additionally, the 2 fragments without tandem GT sequence drove the gus gene expression, but the expression pattern of the gus gene wasn't regulated by the concentration of sodium chloride; Also, the upstream fragment of the tandem GT sequence wasn't able to drive the gus gene expression. In conclusion, the highly repeated GT sequence from the DCA1 promoter plays an important role in the salt-inducible regulation of DCA1 promoter from D.salina and might be a novel salt-inducible element.

Cysticercosis is caused by the metacestode form of Taenia solium-Cysticercus cellulosae and it causes great economic losses and threatens the people's health. There are some problems on how to control cysticercosis, in order to resolve the key problem that the native antigen to diagnose and prevent cysticercosis is very limited and is not satisfied, Pichia pastoris Expression System was used to express recombinant P2 protein. The interested P2 gene was got by digesting the pGEM - P2 vector using restriction endonuclease, then it was inserted into the secretory pPIC9K Pichia pastoris expression vector and transformed into E. coli. Positive recombinant plasmids were selected sequenced and named pPIC9K-P2 and it was linearized by Sal I and Bgl II, then the linear DNA transfored into Pichia pastoris GS115 by electroporation. The recombinant expression vector pPIC9K - P2 integrated into GS115 via homologous recombination between the transforming DNA and regions of homology within the genome. The transformants were screened for multicopy recombinants using G418 and were distinguished for Mut phenotypes by MD and MM. Two different phenotypes were generated-HIS+ MUT+ (Methanol utilization plus) and HIS+ MUT(S) (Methanol utilization slow). PCR analysis of the multicopy recombinants indicated that the P2 gene was integrated within the genome of pichia Pastoris. The multicopy recombinants were named GS115/pPIC9K - P2HIS+ MUT+ and GS115/pPIC9K-P2HIS+ MUT(S), both HIS+ MUT+ and HIS+ MUT(S) were induced with methanol. The results of SDS-PAGE and Western blot demonstrated that the culture supernatant of the induced Pichia pastoris contained P2 protein which was accumulated up to 33 % of total proteins in the culture supernant and its molecular weight is 12.6kD. The results of the clinical study indicated that the expression P2 protein could be used to diagnose human cysticercosis and swine cysticercosis as diagnosis antigen.

Chinese Journal of Cardiology 2007;35(1):47-50
OBJECTIVETo discuss the clinical epidemiology features and change trend of aortic dissection (AD).
METHODSRetrospective analysis of consecutive 339 patients with AD over 10 years in Guangdong Cardiovascular Institution. Hospital records and prognosis were compared between two five-year periods.
RESULTS339 cases with AD were hospitalized during the past 10 years. The mean age was 55.7 +/- 11.2. The male/female ratio was 4.75 to 1. Hypertension was present in 71.7% of all patients. Heavy smoking history was elicited in 52.2% of all the patients. Type I dissection were identified in 32.3% of all the cases, type II in 5.1%, and type III in 62.6%. In-hospital mortality of acute type A dissection was 35.3%, acute type B dissection 8.0%. In two five-year periods, the total number of cases increased by 165%, among which type I was the fastest, increased by 270%. Changes of mean age and male/female ratio were not significant. For acute AD, changes of prognosis in one year improved, but did not reach statistical significance.
CONCLUSIONSThis study provides insight into current regional profiles of AD. The number of hospitalized patients with AD is increasing dramatically. The mean age of the first-attack is much younger and the male ration is much higher than that reported by other regional researchers. Limited by sample size, one year prognosis of acute AD dissection improved, but did not reach statistical significance. These data support the urgent need for further improvement in prevention and treatment of AD.
