1.Percutaneous CT-guided fixation of sacroiliac joint with cannulated lag screw for treating pathologic pain of sacroiliac joint

BACKGROUND: Most patients with posterior pelvic and sacroiliac metastasis are terminally ill. Their treatment is usually palliative and directed toward relieving pain and improving the quality of life with the least possible intervention.OBJECTIVE: To investigate the possibility of percutaneous CT-guided fixation of sacroiliac joint with cannulated lag screw treatment for pathologic pain of sacroiliac joint.DESIGN: Preoperative and postoperative auto-control clinical trial was conducted.SETTING and PARTICIPANTS: The study was completed in Ningbo Sixth Hospital. Eight patients(4 males and 4 females, aged 12 years to 83 years)with metastatic tumor of sacroiliac joint were selected for our study.METHODS: After treatment with percutaneous CT-guided fixation of sacroiliac joint with cannulated lag screw, the sacroiliac joint of the 8 patients became pathologically unstable. Enneking pain scale was obtained preoperatively and postoperatively.MAIN OUTCOME MEASURES: Pain recovery was assessed preoperatively and postoperatively.RESULTS: After 2. 5 years' follow-up, pain of the patients was relieved with a range of 0 to 5(mean 3.2).CONCLUSION: Percutaneous cannulated lag screws with CT guidance help alleviate the patients' pain due to pathologically unstable sacroiliac joint.

Methods The cell viability was detected by MTT assay and morphological observation. The effect of(-)clausenamide on the PC12 cell apoptosis was analyzed by flow cytometry. Then western blotting and confocal microscope was used for the further study of effect of(-)clausenamide on the protein expression of GSK-3?,Bax and Bcl-2. 3.Investigation on the reference interval of platelet-related parameters in different detection systems

Objective To investigate the reference interval of platelet‐related parameters including mean platelet volume (MPV) ,platelet distribution width (PDW) ,platelet hematocrit (PCT) .Methods Used Sysmex XN‐9000 assembly line and PEN‐TRA 120 hematology analyzer to analysis the venous blood cells for 3 415 cases of 16 to 90 years old physical examination personnel in Mianyang Central Hospital ,the platelet‐related parameters results of 2 718 standard healthy population by screening were statis‐tically analyzed .Results Platelet‐related parameters 95% reference interval (2 .5% -97 .5% ) of XN assembly line :M PV 9 .9-14 . 7 fL ,PDW 11 .2% -24 .3% ,PCT 0 .15% -0 .30% ;Platelet‐related parameters of 95% reference interval of PENTRA 120:MPV 8 .4-12 .7 fL ,PDW 13 .8% -24 .8% ,PCT 0 .12% -0 .26% ;comparison between the instruments :MPV ,PDW ,PCT results showed significant differences (P<0 .05);comparison between men and women :PCT test results of XN assembly line were differ‐ent (P<0 .05) ,MPV ,PDW ,PCT results of PENTRA 120 were different (P<0 .05);comparison between age groups :PCT result of all ages between men and women were different in XN assembly line (P<0 .05) ,in PENTRA 120 ,MPV ,PDW ,PCT results be‐tween ages were different in men (P<0 .05) ,PCT results between ages were different in women (P<0 .05) .Conclusion Each la‐boratory should establish the laboratory supporting biological reference interval of platelet‐related parameters based on different in‐struments ;in practical applications ,should set the reference interval for different gender or age .
4.Discussing displacemental trend on nontractional side of canine mandible with finite element method when distraction osteogenesis with unilateral incomplete osteotomy.

OBJECTIVETo discuss the trend of displacement on the side of nondistraction when distraction osteogenesis with unilateral incomplete osteotomy of canine mandible.
METHODSThe method of finite element analysis was used to simulate distraction osteogenesis with unilateral incomplete osteotomy and the displacement of six marked point such as the temporomandibular joint and the angle of mandible and coracoid process and teeth were observed on nondistractional side when the transport disc was distracted with a distance and without a distance.
RESULTSWhen the transport disc was distracted without a distance, the Von Mises stress was 0 in the six marked point of nondisractional side and their displacement in 3 dimensions (X, Y, Z axis) was 0. When the transport disc was distracted with 1 mm width, all marked point had some displacement.
CONCLUSIONWhen distracted and observed from the side of distraction, the mandible at the side of nondistraction has the trend of contrarotation round the center of the transverse ridge mid-point of condyle process on the plane of sagittal, whereas it has the trend of parallel movement approximately in the direction of opposite side on the plane of coronary.

5.Protective effect of puerarin against calcium overload after focal cerebral ischemia injury in rats.

OBJECTIVETo observe the temporal and spatial changes in the distribution of Ca2+ in the rat brain following focal cerebral ischemia injury and explore the protective effect of puerarin against calcium overload.
METHODSFocal cerebral ischemia was induced by middle cerebral artery occlusion in rats. After cerebral ischemia, puerarin was administered in the rats at different time points. The volume of ischemic cerebral tissue was assessed by TTC staining, and the fluorescence intensity of Ca2+ in the cortex and corpora striata was determined under laser scanning confocal microscope.
RESULTSThe fluorescence intensity of Ca2+ in the infracted cortex and corpora striata begun to increase 2 h after the ischemia and was further enhanced with the prolongation of the ischemic time. No significance was found in the fluorescence intensity of Ca2+ between the cortex and corpora striata. The fluorescence intensity of Ca2+ in the infarcted corpora striata was obviously higher than that in the cortex after ischemia. Compared with that in the ischemic model group, the fluorescence intensity of Ca2+ in the infarcted cortex and corpora striata decreased significantly at 2 and 12 h following puerarin intervention (P<0.05).
CONCLUSIONPuerarin treatment can relieve calcium overload, reduce cerebral ischemic volume and play a neuroprotective role against focal cerebral ischemia. Twelve hours following cerebral ischemic injury may be the time window for administering puerarin intervention.

Objective TolearnthesituationofsmokingtriesanditsinfluencingfactorsamongundergraduatesinWenzhou City.Methods Withdifferentprofessions,510undergraduatesfromgrade1to3intwouniversitieswereselectedby stratified random sampling and investigated using a self-developed questionnaire.The influencing factors for smoking initiationofstudentswereanalyzedusingchi-squaretestandlogisticregression.Results Atotalof510undergraduates were investigated and 505 finished the questionnaires.The prevalence of smoking tries was 33.47%,and the prevalence among male students was higher than female students (50.80%vs.1 6.47%,P<0.01 ).The prevalence of smoking tries among non medical students was higher than that of medical students (40.08% vs.27.1 3%,P<0.01 ).The current smoking rate was 9.31%,and the main reason for this behaviors was social need (53.1 9%).Regression analysis showed that female(OR=0.38,95%CI:0.24-0.62)was a protective factor of smoking tries while the peer smoking behavior was theriskfactorsmokingtries(OR=3.46,95%CI:2.44-4.90).Conclusion Theprevalenceofsmokingtries, willingness to smoke and secondhand smoke exposure were high among the universities students.Education of tobacco control should be strengthened,and the tobacco control of peer-education model can be promoted.
7.Pancreatic Neuroendocrine Neoplasms: Correlation between MR Features and Pathological Tumor Grades

This study investigated the accuracy of MRI features in differentiating the pathological grades of pancreatic neuroendocrine neoplasms (PNENs).A total of 31 PNENs patients were retrospectively evaluated,including 19 cases in grade 1,5 in grade 2,and 7 in grade 3.Plain and contrastenhanced MRI was performed on all patients.MRI features including tumor size,margin,signal intensity,enhancement patterns,degenerative changes,duct dilatation and metastasis were analyzed.Chi square tests,Fisher's exact tests,one-way ANOVA and ROC analysis were conducted to assess the associations between MRI features and different tumor grades.It was found that patients with older age,tumors with higher TNM stage and without hormonal syndrome had higher grade of PNETs (all P<0.05).Tumor size,shape,margin and growth pattern,tumor pattern,pancreatic and bile duct dilatation and presence of lymphatic and distant metastasis as well as MR enhancement pattern and tumor-topancreas contrast during arterial phase were the key features differentiating tumors of all grades (all P<0.05).ROC analysis revealed that the tumor size with threshold of 2.8 cm,irregular shape,pancreatic duct dilatation and lymphadenopathy showed satisfactory sensitivity and specificity in distinguishing grade 3 from grade 1 and grade 2 tumors.Features ofperipancreatic tissue or vascular invasion,and distant metastasis showed high specificity but relatively low sensitivity.In conclusion,larger size,poorlydefined margin,heterogeneous enhanced pattern during arterial phase,duct dilatation and the presence of metastases are common features of higher grade PNENs.Plain and contrast-enhanced MRI provides the ability to differentiate tumors with different pathological grades.
8.The vaginal microecology in gynecological outpatients: a report from China

Objective To study the vaginal microecology of the patients in the outpatient department of Obstetrics and gynecology in China.Methods A multicenter cross-sectional study was conducted in gynecologic clinic of 9 collaborative hospitals in China.200 consecutive patients were collected in each hospital and their vaginal microecology combined with related factors were analyzed.Results A total of 2 093 specimens were collected in this study.The detection rate of Trichomonas was 5.5%(115/2 093). The detection rate of Candida mycelia was 15.9%(333/2 093), with germinal spores was 4.1%(86/2093).The detection rate of bacterial vaginosis was 18.8%(394/2 093).The distribution results of vaginal flora in patients showed that the normal flora accounted for only 27.3%(571/2 093).The normal flora with the insufficiency of H2O2 accounted for 23%(480/2 093).The bacteria inhibiting flora accounted for 3.8%(79/2 093).The abnormal microflora(non BV type)accounted for 14.9%(312/2 093).The abnormal microflora(BV intermediate type)accounted for 13.4%(280/2 093).The abnormal microflora(BV type)accounted for 17.6%(369/2 093).The average pH of vaginal discharge was 4.58 ±0.495.There was no significant difference of the incidence of trichomonas and bacterial vaginosis between north and south of the Yangtze river, while the detection rate of fungal hyphae and the fungal spores is significantly higher in the south than that in the north.The analysis results of factors affecting the microecology showed that age and contraception methods were two important factors.The patients′age from bacteria inhibition group was 49.64 +16.68 which was significantly higher than that of the other microecology groups.The proportion of abnormal microflora of patients from the oral contraceptive group was 40%(20/50).The proportion of abnormal microflora of patients from IUD group was 36.6%(63/172).Compared with these two contraception methods, the proportion of abnormal microflora of patients from condoms usage group was 27.8%(91/327)which was significantly lower.The incidence of abnormal leucorrhea in the normal group was 37.7%, which was significantly lower than that of other abnormal groups.Conclusion This study showed the vaginal microecology status of the Chinese outpatient ′s clinic and found that the vagina microecology was related to age, region and contraceptive methods.The typical manifestation of microecological abnormality is the increase of leucorrhea.(Chin J Lab Med,2018, 41:287-291)
9.Differentiation of mesenchymal stem cells into dopaminergic neuron-like cells in vitro.

OBJECTIVETo explore the way to induce mesenchymal stem cells (MSCs) to differentiate into dopaminergic neurons in vitro.
METHODSMSCs were obtained from rat bone marrow, cultured and passaged. MSCs used in this experiment had multipotency, which was indirectly proved by being induced to differentiate into chondrocytes and adipocytes. MSCs were cultured in medium containing 0.5 mmol/L IBMX for 2 days. Then the medium was replaced with induction medium, which contained GDNF, IL-1beta, mesencephalic glial-cell-conditioned medium and flash-frozen mesencephalic membrane fragments. The surface markers of the differentiated neurons, such as NSE, nestin, MAP-2a, b and TH were detected by immunocytochemistry and Western blot after MSCs were cultured in induction medium for 7 days and 15 days.
RESULTSMSCs differentiated into neural progenitors and expressed nestin after MSCs were incubated with medium containing IBMX for 2 d. After the medium was replaced with induction medium containing many inducing agents, MSCs differentiated into neuron-like cells and dopaminergic neuron-like cells and expressed NSE, MAP-2a, b and TH. The percentage of NSE-positive cells, MAP-2a, b-positive cells and TH-positive cells was 30.032 +/- 2.489%, 41.580 +/- 5.101% and 34.958 +/- 5.534%, respectively after MSCs were induced in medium containing GDNF, IL-1beta, mesencephalic glial-cell-conditioned medium and flash-frozen mesencephalic membrane fragments for 15 days.
CONCLUSIONMSCs can differentiate into dopaminergic neuron-like cells and are a new cell source for the treatment of neurodegeneration diseases and have a great potential for wide application.

10.Effects of estrogen-related receptor alpha (ERRα) on proliferation and metastasis of human lung cancer A549 cells.

Estrogen-related receptor alpha (ERRα) plays an important role in the development of hormone-dependent cancers, but its roles in lung cancer remain elusive. The present study was aimed to investigate the effects of ERRα on the proliferation and metastasis of lung cancer A549 cells. The mRNA and protein levels of ERRα were detected in lung cancer A549 and MCF-7 cells and bronchial epithelial BEAS-2B cells by qRT-PCR and Western blotting, respectively. ERRα plasmid transfection and XCT-790 (an inverse agonist of ERRα) were used to up-regulate or down-regulate ERRα expression in A549 cells, respectively. The viability of A549 cells was measured by cell counting kit-8 (CCK-8) and the motility of A549 cells by wound healing assay and Transwell migration/invasion assay. The epithelial markers E-cadherin (E-Cad) and zona occludin-1 (ZO-1), the mesenchymal markers fibronectin (FN) and vimentin (Vim) and the transcription factors (Snail, Zeb1 Twist and Slug) were further detected at mRNA and protein levels by qRT-PCR and Western blotting, respectively. The results showed that ERRα promoted the growth of lung cancer A549 cells in vitro. XCT-790 significantly inhibited the migration and invasion of A549 cells. Over-expression of ERRα promoted the epithelial-to-mesenchymal transition (EMT) of A549 cells, down-regulated the epithelial makers E-Cad and ZO-1, and up-regulated the mesenchymal makers FN and Vim. Silencing of Slug, but not other transcription factors, significantly abolished the ERRα-induced EMT of A549 cells. It was suggested that ERRα promoted the migration and invasion of A549 cells by inducing EMT, and Slug was involved in the process. Targeting ERRα might be an efficient approach for lung cancer treatment.
